Metal ion-N7 coordination in a ribozyme branch domain by NMR
The N7 of purine nucleotides presents one of the most dominant metal ion binding sites in nucleic acids. However, the interactions between kinetically labile metal ions like Mg2+ and these nitrogen atoms are inherently difficult to observe in large RNAs. Rather than using the insensitive direct N-15 detection, here we have used (2)J-H-1,N-15]-HSQC (Heteronuclear Single Quantum Coherence) NMR experiments as a fast and efficient method to specifically observe and characterize such interactions within larger RNA constructs. Using the 27 nucleotides long branch domain of the yeast-mitochondrial group II intron ribozyme Sc.ai5 gamma as an example, we show that direct N7 coordination of a Mg2+ ion takes place in a tetraloop nucleotide. A second Mg2+ ion, located in the major groove at the catalytic branch site, coordinates mainly in an outer-sphere fashion to the highly conserved flanking GU wobble pairs but not to N7 of the sandwiched branch adenosine. Entanglement of charge transfer, hole doping, exchange interaction and octahedron tilting angle and their influence on the conductivity of La1-xSrxFe0.75Ni0.25O3-{\delta}: A combination of x-ray spectroscopy and diffraction
Substitution of La by Sr in the 25% Ni doped charge transfer insulator LaFeO3
creates structural changes that inflect the electrical conductivity caused by
small polaron hopping via exchange interactions and charge transfer. The
substitution forms electron holes and a structural crossover from orthorhombic
to rhombohedral symmetry, and then to cubic symmetry. The structural crossover
is accompanied by a crossover from Fe3+-O2--Fe3+ superexchange interaction to
Fe3+-O2--Fe4+ double exchange interaction, as evidenced by a considerable
increase of conductivity. These interactions and charge transfer mechanism
depend on superexchange angle, which approaches 180{\deg} upon increasing Sr
concentration, leading an increased overlap between the O (2p) and Fe/Ni (3d)
orbitals
Correlation of high temperature X-ray photoemission spectral features and conductivity of epitaxially strained (La0.8Sr0.2)0.95Ni0.2Fe0.8O3/SrTiO3(110)
Reversible and irreversible discontinuities at around 573 K and 823 K in the
electric conductivity of a strained 175 nm thin film of
(La0.8Sr0.2)0.95Ni0.2Fe0.8O3-{\delta} grown by pulsed laser deposition on
SrTiO3 (110) are reflected by valence band changes as monitored in
photoemission and oxygen K-edge x-ray absorption spectra. The irreversible jump
at 823 K is attributed to depletion of doped electron holes and reduction of
Fe4+ to Fe3+, as evidenced by oxygen and iron core level soft x-ray
spectroscopy, and possibly of a chemical origin, whereas the reversible jump at
573 K possibly originates from structural changes
Adenosine generation catalyzed by CD39 and CD73 expressed on regulatory T cells mediates immune suppression
The study of T regulatory cells (T reg cells) has been limited by the lack of specific surface markers and an inability to define mechanisms of suppression. We show that the expression of CD39/ENTPD1 in concert with CD73/ecto-5′-nucleotidase distinguishes CD4+/CD25+/Foxp3+ T reg cells from other T cells. These ectoenzymes generate pericellular adenosine from extracellular nucleotides. The coordinated expression of CD39/CD73 on T reg cells and the adenosine A2A receptor on activated T effector cells generates immunosuppressive loops, indicating roles in the inhibitory function of T reg cells. Consequently, T reg cells from Cd39-null mice show impaired suppressive properties in vitro and fail to block allograft rejection in vivo. We conclude that CD39 and CD73 are surface markers of T reg cells that impart a specific biochemical signature characterized by adenosine generation that has functional relevance for cellular immunoregulation
Testing in the incremental design and development of complex products
Testing is an important aspect of design and development which consumes significant time and resource in many companies. However, it has received less research attention than many other activities in product development, and especially, very few publications report empirical studies of engineering testing. Such studies are needed to establish the importance of testing and inform the development of pragmatic support methods. This paper combines insights from literature study with findings from three empirical studies of testing. The case studies concern incrementally developed complex products in the automotive domain. A description of testing practice as observed in these studies is provided, confirming that testing activities are used for multiple purposes depending on the context, and are intertwined with design from start to finish of the development process, not done after it as many models depict. Descriptive process models are developed to indicate some of the key insights, and opportunities for further research are suggested
Innovation and Crowdsourcing Contests
In an innovation contest, an organizer seeks solutions to an innovation-related problem from a group of independent agents. Agents, who can be heterogeneous in their ability levels, exert efforts to improve their solutions, and their solution qualities are uncertain due to the innovation and evaluation processes. In this chapter, we present a general model framework that captures main features of a contest, and encompasses several existing models in the literature. Using this framework, we analyze two important decisions of the organizer: a set of awards that will be distributed to agents and whether to restrict entry to a contest or to run an open contest. We provide a taxonomy of contest literature, and discuss past and current research on innovation contests as well as a set of exciting future research directions
Experience and gender effects in acquisition experiment with value messages
In the bargaining experiment, the privately informed seller of a company sends a value message to the uninformed potential buyer who proposes a price for acquiring the company. Participants are constantly either seller or buyer and interact over 30 rounds with randomly changing partners. How are overstating the value of the company, underpricing the received value message and acceptance of price offers affected by experience and gender (constellation)? We control via treatments for awareness of gender (constellation) and show that gender (constellation) matters and that the main experience effects apply across gender (constellations)
Sex differences in cerebral venous sinus thrombosis after adenoviral vaccination against COVID-19
Introduction: Cerebral venous sinus thrombosis associated with vaccine-induced immune thrombotic thrombocytopenia (CVST-VITT) is a severe disease with high mortality. There are few data on sex differences in CVST-VITT. The aim of our study was to investigate the differences in presentation, treatment, clinical course, complications, and outcome of CVST-VITT between women and men. Patients and methods: We used data from an ongoing international registry on CVST-VITT. VITT was diagnosed according to the Pavord criteria. We compared the characteristics of CVST-VITT in women and men. Results: Of 133 patients with possible, probable, or definite CVST-VITT, 102 (77%) were women. Women were slightly younger [median age 42 (IQR 28–54) vs 45 (28–56)], presented more often with coma (26% vs 10%) and had a lower platelet count at presentation [median (IQR) 50x109/L (28–79) vs 68 (30–125)] than men. The nadir platelet count was lower in women [median (IQR) 34 (19–62) vs 53 (20–92)]. More women received endovascular treatment than men (15% vs 6%). Rates of treatment with intravenous immunoglobulins were similar (63% vs 66%), as were new venous thromboembolic events (14% vs 14%) and major bleeding complications (30% vs 20%). Rates of good functional outcome (modified Rankin Scale 0-2, 42% vs 45%) and in-hospital death (39% vs 41%) did not differ. Discussion and conclusions: Three quarters of CVST-VITT patients in this study were women. Women were more severely affected at presentation, but clinical course and outcome did not differ between women and men. VITT-specific treatments were overall similar, but more women received endovascular treatment.</p
