    Education, Culture and Learning in Response to Conflict and Protracted Crisis

    Panel Session on ‘Education, Culture and Learning in Response to Conflict and Protracted Crisis’ at the Annual CIES Conference 2021. Chaired by Moskal, five papers addressed both the lived experiences and actions - in policy and practice - in conflict, post-conflict and UK contexts. A discussant (Odena) offered reflections on common threads and issues to reconsider, drawing on his experience leading The Arts of Inclusion network. Presenters examined youth policies in Colombia and Mexico (Arizpe & Gormally), experiences of doctoral students from Middle-Eastern conflict zones in the UK (Pacheco), skills development of refugees in India (Dagar), Arabic teaching with refugees in Italy (Aldegheri), and workshops with Syrian and Iraqi refugees at a UK university (Moskal). These studies engage in discussions of issues confronting youth in particular, their wellbeing, aspirations, learning and skills development. The papers contribute understanding to education in crises, one of the major global challenges in achieving the Sustainable Development Goals (SDG4). Education is crucial in establishing peace-building processes, and for the wellbeing and economic integration of all individuals affected by conflict. Formal and non-formal learning has a strategic role to play in peace building, and promotion of equity and diversity

    Testing the cognitive-behavioural maintenance models across DSM-5 bulimic-type eating disorder diagnostic groups: A multi-centre study

    The original cognitive-behavioural (CB) model of bulimia nervosa, which provided the basis for the widely used CB therapy, proposed that specific dysfunctional cognitions and behaviours maintain the disorder. However, amongst treatment completers, only 40–50 % have a full and lasting response. The enhanced CB model (CB-E), upon which the enhanced version of the CB treatment was based, extended the original approach by including four additional maintenance factors. This study evaluated and compared both CB models in a large clinical treatment seeking sample (N = 679), applying both DSM-IV and DSM-5 criteria for bulimic-type eating disorders. Application of the DSM-5 criteria reduced the number of cases of DSM-IV bulimic-type eating disorders not otherwise specified to 29.6 %. Structural equation modelling analysis indicated that (a) although both models provided a good fit to the data, the CB-E model accounted for a greater proportion of variance in eating-disordered behaviours than the original one, (b) interpersonal problems, clinical perfectionism and low self-esteem were indirectly associated with dietary restraint through over-evaluation of shape and weight, (c) interpersonal problems and mood intolerance were directly linked to binge eating, whereas restraint only indirectly affected binge eating through mood intolerance, suggesting that factors other than restraint may play a more critical role in the maintenance of binge eating. In terms of strength of the associations, differences across DSM-5 bulimic-type eating disorder diagnostic groups were not observed. The results are discussed with reference to theory and research, including neurobiological findings and recent hypotheses

    Circulating Cell-Free DNA in Dogs with Mammary Tumors: Short and Long Fragments and Integrity Index

    Circulating cell-free DNA (cfDNA) has been considered an interesting diagnostic/prognostic plasma biomarker in tumor-bearing subjects. In cancer patients, cfDNA can hypothetically derive from tumor necrosis/apoptosis, lysed circulating cells, and some yet unrevealed mechanisms of active release. This study aimed to preliminarily analyze cfDNA in dogs with canine mammary tumors (CMTs). Forty-four neoplastic, 17 non-neoplastic disease-bearing, and 15 healthy dogs were recruited. Necrosis and apoptosis were also assessed as potential source of cfDNA on 78 CMTs diagnosed from the 44 dogs. The cfDNA fragments and integrity index significantly differentiated neoplastic versus non-neoplastic dogs (P<0.05), and allowed the distinction between benign and malignant lesions (P<0.05). Even if without statistical significance, the amount of cfDNA was also affected by tumor necrosis and correlated with tumor size and apoptotic markers expression. A significant (P<0.01) increase of Bcl-2 in malignant tumors was observed, and in metastatic CMTs the evasion of apoptosis was also suggested. This study, therefore, provides evidence that cfDNA could be a diagnostic marker in dogs carrying mammary nodules suggesting that its potential application in early diagnostic procedures should be further investigated

Context: The discovery of EU-protected Geomalacus maculosus in commercial plantations requires an understanding of the implications of forestry practices for the species within the context of sustainable forest management. Aims: 1.Compare Geomalacus maculosus captures across mature exotic Sitka spruce-dominated stands, previously clear-felled stands and adjacent peatland habitats.2.Assess the suitability, for forest managers, of population estimate models for G. maculosus.3.Assess the implications of felling by comparing relative abundances of G. maculosus directly before and after clear-felling at a mature exotic Sitka spruce-dominated stand. Methods: Geomalacus maculosus catches were compared at four sites across two to three mature (43–45 years old) conifer stands per site, one clear-felled stand per site, and one adjacent peatland per site using refuge traps and hand searching. Capture-mark-recapture studies were undertaken to estimate population sizes. A BACIP (Before-After-Control-Impact-Paired) analysis was undertaken in one forest stand at one forest site to determine impacts of a clear-felling event. Results: Mean catches of Geomalacus maculosus adults in the mature forest stands were over 10 and 11 times greater than mean catches on peatland and clear-fell stands, respectively. The Schnabel model for estimating population size was most suited for mature forest stands but could not be utilised for the other habitats. BACIP analysis showed a significant impact of clear-felling with a 95% reduction in mean G. maculosus catches after a clear-felling event where none of the individuals marked prior to felling were recaptured compared to 21% recapture rates at the control site. Greater tree circumference in mature conifer stands correlated with greater catches. Conclusion: Guidelines are needed to ensure the protection of Geomalacus maculosus in commercial forestry. Interventions could include patch/tall stump retention at final felling and/or translocation of the protected species

    Simultaneous quantitative assessment of circulating cell-free mitochondrial and nuclear DNA by multiplex real-time PCR

    Quantification of circulating nucleic acids in plasma and serum could be used as a non-invasive diagnostic tool for monitoring a wide variety of diseases and conditions. We describe here a rapid, simple and accurate multiplex real-time PCR method for direct synchronized analysis of circulating cell-free (ccf) mitochondrial (mtDNA) and nuclear (nDNA) DNA in plasma and serum samples. The method is based on one-step multiplex real-time PCR using a FAM-labeled MGB probe and primers to amplify the mtDNA sequence of the ATP 8 gene, and a VIC-labeled MGB probe and primers to amplify the nDNA sequence of the glycerinaldehyde-3-phosphate-dehydrogenase (GAPDH) gene, in plasma and serum samples simultaneously. The efficiencies of the multiplex assays were measured in serial dilutions. Based on the simulation of the PCR reaction kinetics, the relative quantities of ccf mtDNA were calculated using a very simple equation. Using our optimised real-time PCR conditions, close to 100% efficiency was obtained from the two assays. The two assays performed in the dilution series showed very good and reproducible correlation to each other. This optimised multiplex real-time PCR protocol can be widely used for synchronized quantification of mtDNA and nDNA in different samples, with a very high rate of efficiency

    Aflatoxin-Induced TP53 R249S Mutation in HepatoCellular Carcinoma in Thailand: Association with Tumors Developing in the Absence of Liver Cirrhosis

    Primary Liver Cancer (PLC) is the leading cause of death by cancer among males in Thailand and the 3rd among females. Most cases are hepatocellular carcinoma (HCC) but cholangiocarcinomas represent between 4 and 80% of liver cancers depending upon geographic area. Most HCC are associated with chronic infection by Hepatitis B Virus while a G→T mutation at codon 249 of the TP53 gene, R249S, specific for exposure to aflatoxin, is detected in tumors for up to 30% of cases. We have used Short Oligonucleotide Mass Analysis (SOMA) to quantify free circulating R249S-mutated DNA in plasma using blood specimens collected in a hospital case:control study. Plasma R249S-mutated DNA was detectable at low concentrations (≥67 copies/mL) in 53 to 64% of patients with primary liver cancer or chronic liver disease and in 19% of controls. 44% of patients with HCC and no evidence of cirrhosis had plasma concentrations of R249S-mutated DNA ≥150 copies/mL, compared to 21% in patients with both HCC and cirrhosis, 22% in patients with cholangiocarcinoma, 12% in patients with non-cancer chronic liver disease and 3% of subjects in the reference group. Thus, plasma concentrations of R249S-mutated DNA ≥150 copies/mL tended to be more common in patients with HCC developing without pre-existing cirrhosis (p = 0.027). Overall, these results support the preferential occurrence of R249S-mutated DNA in HCC developing in the absence of cirrhosis in a context of HBV chronic infection
