Herein, we tested cell therapy using early outgrowth cells (EOCs) or their conditioned media (CM) to protect the retina of diabetic animal models (type 1 and type 2) and assessed the mechanisms by in vitro study. Control and diabetic (db/db) mice (8 weeks of age) were randomized to receive a unique intravenous injection of 5x10(5)EOCs or 0.25 ml thrice weekly tail-vein injections of 10x concentrated CM and Wystar Kyoto rats rendered diabetic were randomized to receive 0.50 ml thrice weekly tail-vein injections of 10x concentrated CM. Four weeks later, the animals were euthanized and the eyes were enucleated. Rat retinal Muller cells (rMCs) were exposed for 24 h to high glucose (HG), combined or not with EOC-conditioned medium (EOC-CM) from db/m EOC cultures. Diabetic animals showed increase in diabetic retinopathy (DR) and oxidative damage markers; the treatment with EOCs or CM infusions significantly reduced this damage and re-established the retinal function. In rMCs exposed to diabetic milieu conditions (HG), the presence of EOC-CM reduced reactive oxygen species production by modulating the NADPH-oxidase 4 system, thus upregulating SIRT1 activity and deacetylating Lys-310-p65-NF.B, decreasing GFAP and VEGF expressions. The antioxidant capacity of EOC-CM led to the prevention of carbonylation and nitrosylation posttranslational modifications on the SIRT1 molecule, preserving its activity. The pivotal role of SIRT1 on the mode of action of EOCs or their CM was also demonstrated on diabetic retina.     A motivação é um dos elementos essenciais para uma aprendizagem eficiente e de qualidade. Um aluno motivado aprende melhor, uma vez que todo aluno é motivado pelo êxito ou inibido pelo fracasso. No ensino a distância a motivação dos alunos é o centro das atenções no processo educacional, uma vez que se tem o reconhecimento de que a aprendizagem é um processo pessoal, sistemático e reflexivo e que depende do despertar do aluno para suas potencialidades, que pode ser sozinho ou com ajuda do tutor. O presente estudo relata uma pesquisa realizada com os três cursos de capacitação a distância, do quarto módulo, oferecidos aos funcionários do Ministério da Saúde em parceria com a Universidade Federal de Santa Catarina – UFSC, sendo que esta parceria existe desde 2007 para os cursos de capacitação. Os cursos são oferecidos em módulos de quarenta e cinco e de sessenta horas. Este estudo tem com o objetivo investigar a postura motivadora do tutor na visão dos alunos que estão finalizando os cursos do quarto módulo de capacitação. Trata-se de um estudo de caso, com uma pesquisa bibliográfica e descritiva, tendo uma abordagem quantitativa. Este estuda destaca que os alunos sentem-se motivados a continuar perseverantes nos cursos de capacitação em estudo-aprendizagem através de uma comunicação clara e contínua, uma vez que a dificuldade com o uso das tecnologias de informação e comunicação (internet, AVEA) são os maiores fatores de dificuldade do aluno do Mistério da Saúde

    Antioxidant vitamin supplementation on muscle adaptations to resistance training: A double-blind, randomized controlled trial.

    Objectives The aim of this study was to examine whether antioxidant vitamin supplementation with vitamin C (VitC) and vitamin E (VitE) affects the hypertrophic and functional adaptations to resistance training in trained men. Methods This was a double-blind, randomized controlled trial in which participants were supplemented daily with VitC and VitE ( n = 12) or placebo ( n = 11) while completing a 10-wk resistance training program accompanied by a dietary intervention (300 kcal surplus and adequate protein intake) designed to optimize hypertrophy. Body composition (dual-energy x-ray absorptiometry), handgrip strength, and one-repetition maximum (1-RM), maximal force (F0), velocity (V0), and power (Pmax) were measured in bench press (BP) and squat (SQ) tests conducted before and after the intervention. To detect between-group differences, multiple-mixed analysis of variance, standardized differences, and qualitative differences were estimated. Relative changes within each group were assessed using a paired Student's t test. Results In both groups, similar improvements were produced in BP 1-RM , SQ 1-RM SQ, and BP F0 (P < 0.05) after the resistance training program. A small effect size was observed for BP 1-RM (d = 0.53), BP F0 (d = 0.48), and SQ 1-RM (d = –0.39), but not for SQ F0 (d = 0.03). Dominant handgrip strength was significantly increased only in the placebo group (P < 0.05). According to body composition data, a significant increase was produced in upper body fat-free mass soft tissue (FFMST; P < 0.05) in the placebo group, whereas neither total nor segmental FFMST was increased in the vitamin group. Small intervention effect sizes were observed for upper body FFSMT (d = 0.32), non-dominant and dominant leg FFMST (d = –0.39; d = –0.42). Although a significant increase in total body fat was observed in both groups (P < 0.05) only the placebo group showed an increase in visceral adipose tissue (P < 0.05), showing a substantial intervention effect (d = 0.85). Conclusions The data indicated that, although VitC/VitE supplementation seemed to blunt upper body strength and hypertrophy adaptations to resistance training, it could also mitigate gains in visceral adipose tissue elicited by an energy surplus.pre-print1,83 M

    Aproximación del comportamiento de ladrillos cerámicos huecos mediante métodos numéricos

    En la provincia de Misiones, Argentina, se destaca el uso frecuente de mampuestos conocidos como Ladrillos Cerámicos Huecos (LCH), los cuales son de fabricación industrial, moldeados con máquinas extrusoras, cocidos en hornos continuos. En términos de comportamiento mecánico y estructural es un elemento de rotura frágil, el cual suele estar solicitado en forma global a compresión dentro de las mamposterías que conforma, con valores de resistencia que dependen de varios factores propios de su proceso de fabricación. En cuanto a su geometría es destacable la aparente interacción entre las paredes internas y externas que conforman una estructura de marcos. El objetivo del presente trabajo es analizar mediante una herramienta de simulación numérica, el comportamiento mecánico de los mampuestos de la región, en forma aislada, considerando las diferentes variables que intervienen en su forma y composición material. Para ello, los ladrillos cerámicos individuales son modelados a través del método de los elementos finitos en el software SAP 2000 versión 14.1.0. El material es considerado según propiedades mecánicas y geométricas que son obtenidas a través de ensayos experimentales normalizados. Con los resultados experimentales de deformaciones y resistencia a compresión se calibra el modelo numérico, con el que se obtiene información sobre el comportamiento de interacción entre las paredes horizontales y verticales del ladrillo a efectos de poder avanzar en la formulación de macro modelos conformados por elementos finitos de área.Publicado en: Mecánica Computacional vol.     Association between autism spectrum disorder and diabetes: systematic review and meta-analysis

    There is mixed evidence on the link between autism spectrum disorder (ASD) and diabetes. We conducted the first systematic review/meta-analysis on their association. Based on a pre-registered protocol (PROSPERO: CRD42021261114), we searched Pubmed, Ovid, and Web of Science databases up to 6 December 2021, with no language/type of document restrictions. We assessed study quality using the Newcastle-Ottawa Scale (NOS). We included 24 studies (total: 3427,773 individuals; 237,529 with ASD and 92,832 with diabetes) in the systematic review and 20 in the meta-analysis (mean stars number on the NOS: 5.89/10). There was a significant association, albeit characterized by significant heterogeneity, when pooling unadjusted OR (1.535, 95% CI = 1.109-2.126), which remained significant when restricting the analysis to children and type 2 diabetes, but became non-significant when considering adjusted ORs (OR: 1.528, 95% CI = 0.954-2.448). No significant prospective association was found (n = 2) on diabetes predicting ASD (HR: 1.232, 0.826-11.837). Therefore, the association between ASD and diabetes is likely confounded by demographic and clinical factors that should be systematically investigated in future studies
