    A fruit diet rather than invertebrate diet maintains a robust innate immunity in an omnivorous tropical songbird

Diet alteration may lead to nutrient limitations even in the absence of food limitation, and this may affect physiological functions, including immunity. Nutrient limitations may also affect the maintenance of body mass and key life history events that may affect immune function. Yet, variation in immune function is largely attributed to energetic trade-offs rather than specific nutrient constraints. 2. To test the effect of diet on life history traits, we tested how diet composition affects innate immune function, body mass and moult separately and in combination with each other, and then used path analyses to generate hypotheses about the mechanistic connections between immunity and body mass under different diet composition. 3. We performed a balanced parallel and crossover design experiment with omnivorous common bulbuls Pycnonotus barbatus in out-door aviaries in Nigeria. We fed 40 wild-caught bulbuls ad libitum on fruits or invertebrates for 24 weeks, switching half of each group between treatments after 12 weeks. We assessed innate immune indices (haptoglobin, nitric oxide and ovotransferrin concentrations, and haemagglutination and haemolysis titres), body mass and primary moult, fortnightly. We simplified immune indices into three principal components (PCs), but we explored mechanistic connections between diet, body mass and each immune index separately. 4. Fruit fed bulbuls had higher body mass, earlier moult and showed higher values for two of the three immune PCs compared to invertebrate fed bulbuls. These effects were reversed when we switched bulbuls between treatments after 12 weeks. Exploring the correlations between immune function, body mass and moult, showed that an increase in immune function was associated with a decrease in body mass and delayed moult in invertebrate fed bulbuls, while fruit fed bulbuls maintained body mass despite variation in immune function. Path analyses indicated that diet composition was most likely to affect body mass and immune indices directly and independently from each other. Only haptoglobin concentration was indirectly linked to diet composition via body mass. 5. We demonstrated a causal effect of diet composition on innate immune function, body mass and moult: bulbuls were in better condition when fed on fruits than invertebrates, confirming that innate immunity is nutrient specific. Our results are unique because they show a reversible effect of diet composition on wild adult birds whose immune systems are presumably fully developed and adapted to wild conditions – demonstrating a short-term consequence of diet alteration on life history traits.Publisher PDFPeer reviewe

    Can agricultural cultivation methods influence the healthfulness of crops for foods

    The aim of the current study was to investigate if there are any health eïŹ€ects of long-term consumption of organically grown crops using a rat model. Crops were retrieved over two years from along-term ïŹeld trial at three diïŹ€erent locations in Denmark, using three diïŹ€erent cultivation systems(OA, organic based on livestock manure; OB, organic based on green manure; and C, conventional with mineral fertilizers and pesticides)with two ïŹeld replicates. The cultivation system had an impact on the nutritional quality, aïŹ€ecting Îł-tocopherol, some amino acids, and fatty acid composition. Additionally, the nutritional quality was aïŹ€ected by harvest year and location. However, harvest year and location rather than cultivation system aïŹ€ected the measured health biomarkers. In conclusion, the diïŹ€erences in dietary treatments composed of ingredients from diïŹ€erent cultivation systems did not lead to signiïŹcant diïŹ€erences in the measured health biomarkers, except for a signiïŹcant diïŹ€erence in plasma IgGl evels

    Study protocol: can a school gardening intervention improve children's diets?

    BACKGROUND: The current academic literature suggests there is a potential for using gardening as a tool to improve children's fruit and vegetable intake. This study is two parallel randomised controlled trials (RCT) devised to evaluate the school gardening programme of the Royal Horticultural Society (RHS) Campaign for School Gardening, to determine if it has an effect on children's fruit and vegetable intake. METHOD/DESIGN: Trial One will consist of 26 schools; these schools will be randomised into two groups, one to receive the intensive intervention as "Partner Schools" and the other to receive the less intensive intervention as "Associate Schools". Trial Two will consist of 32 schools; these schools will be randomised into either the less intensive intervention "Associate Schools" or a comparison group with delayed intervention. Baseline data collection will be collected using a 24-hour food diary (CADET) to collect data on dietary intake and a questionnaire exploring children's knowledge and attitudes towards fruit and vegetables. A process measures questionnaire will be used to assess each school's gardening activities. DISCUSSION: The results from these trials will provide information on the impact of the RHS Campaign for School Gardening on children's fruit and vegetable intake. The evaluation will provide valuable information for designing future research in primary school children's diets and school based interventions. TRIAL REGISTRATION: ISRCTN11396528

    Stress induced polarization of immune-neuroendocrine phenotypes in Gallus gallus

    Immune-neuroendocrine phenotypes (INPs) stand for population subgroups differing in immune-neuroendocrine interactions. While mammalian INPs have been characterized thoroughly in rats and humans, avian INPs were only recently described in Coturnix coturnix (quail). To assess the scope of this biological phenomenon, herein we characterized INPs in Gallus gallus (a domestic hen strain submitted to a very long history of strong selective breeding pressure) and evaluated whether a social chronic stress challenge modulates the individuals’ interplay affecting the INP subsets and distribution. Evaluating plasmatic basal corticosterone, interferon-Îł and interleukin-4 concentrations, innate/acquired leukocyte ratio, PHA-P skin-swelling and induced antibody responses, two opposite INP profiles were found: LEWIS-like (15% of the population) and FISCHER-like (16%) hens. After chronic stress, an increment of about 12% in each polarized INP frequency was found at expenses of a reduction in the number of birds with intermediate responses. Results show that polarized INPs are also a phenomenon occurring in hens. The observed inter-individual variation suggest that, even after a considerable selection process, the population is still well prepared to deal with a variety of immune-neuroendocrine challenges. Stress promoted disruptive effects, leading to a more balanced INPs distribution, which represents a new substrate for challenging situations.Fil: Nazar, Franco Nicolas.     Seasonal Patterns of Invasive Pneumococcal Disease

    Pneumococcal infections increase each winter, a phenomenon that has not been well explained. We conducted population-based active surveillance for all cases of invasive pneumococcal disease in seven states; plotted annualized weekly rates by geographic location, age, and latitude; and assessed correlations by time-series analysis. In all geographic areas, invasive pneumococcal disease exhibited a distinct winter seasonality, including an increase among children in the fall preceding that for adults and a sharp spike in incidence among adults each year between December 24 and January 7. Pneumococcal disease correlated inversely with temperature (r –0.82 with a 1-week lag; p<0.0001), but paradoxically the coldest states had the lowest rates, and no threshold temperature could be identified. The pattern of disease correlated directly with the sinusoidal variations in photoperiod (r +0.85 with a 5-week lag; p<0.0001). Seemingly unrelated seasonal phenomena were also somewhat correlated. The reproducible seasonal patterns in varied geographic locations are consistent with the hypothesis that nationwide seasonal changes such as photoperiod-dependent variation in host susceptibility may underlie pneumococcal seasonality, but caution is indicated in assigning causality as a result of such correlations

    A Knowledge-based approach to wildlife management

    We present a method for developing a knowledge base to give wildlife managers timely access to pertinent information. This knowledge-based approach to species management in Namibia focuses on anthologies and management strategies for high-value mammal species.To manage wildlife resources sustainably access to an appropriate knowledge base is essential. The strategic species management approach promotes planning on local, national, and international levels. A hypermedia Information System for Rare Species Management (IRAS) and a decision support system (Wildlife Introduction Advisor) are described. Unlike print media, which become dated, hypermedia is easily updated. The non-sequential nature of hypermedia allows information extraction according to the readers needs. Due to this flexibility hypermedia supports adaptive management and information sharing. Usability evaluation of the IRAS system disproved our assumption that established wildlife conservation categories such as habitat, distribution, and numbers are appropriate and sufficient to structure species information. This result led to a restructuring of the information according to management objectives. Evaluation of the Wildlife Introduction Advisor indicates that the model performs reliably and is robust towards input error. Human experts seem to be susceptible to bias. The economic benefits associated with wildlife translocations demand accountability through a well-defined, transparent distribution process.     Unambiguous detection of nitrated explosive vapours by fluorescence quenching of dendrimer films

    Unambiguous and selective standoff (non-contact) infield detection of nitro-containingexplosives and taggants is an important goal but difficult to achieve with standard analyticaltechniques. Oxidative fluorescence quenching is emerging as a high sensitivity method fordetecting such materials but is prone to false positives—everyday items such as perfumeselicit similar responses. Here we report thin films of light-emitting dendrimers that detectvapours of explosives and taggants selectively—fluorescence quenching is not observed for arange of common interferents. Using a combination of neutron reflectometry, quartz crystalmicrobalance and photophysical measurements we show that the origin of the selectivity isprimarily electronic and not the diffusion kinetics of the analyte or its distribution in the film.The results are a major advance in the development of sensing materials for the standoffdetection of nitro-based explosive vapours, and deliver significant insights into the physicalprocesses that govern the sensing efficacy

    Fluorescence studies on new potential antitumoral benzothienopyran-1-ones in solution and in liposomes

    Fluorescence properties of four new potential antitumoral compounds, 3-arylbenzothieno[2,3-c]pyran-1-ones, were studied in solution and in lipid membranes of dipalmitoyl phosphatidylcholine (DPPC), egg yolk phosphatidylcholine (Egg-PC) and dioctadecyldimethylammonium bromide (DODAB). The 3-(4-methoxyphenyl)benzothieno[2,3-c]pyran-1-one (1c) exhibits the higher fluorescence quantum yields in all solvents studied. All compounds present a solvent sensitive emission, with significant red shifts in polar solvents for the methoxylated compounds. The results point to an ICT character of the excited state, more pronounced for compound 1c. Fluorescence (steady-state) anisotropy measurements of the compounds incorporated in liposomes of DPPC, DODAB and Egg-PC indicate that all compounds have two different locations, one due to a deep penetration in the lipid membrane and another corresponding to a more hydrated environment. In general, the methoxylated compounds prefer hydrated environments inside the liposomes. The 3-(4- fluorophenyl)benzothieno[2,3-c]pyran-1-one (1a) clearly prefers a hydrated environment, with some molecules located at the outer part of the liposome interface. On the contrary, the preferential location of 3-(2-fluorophenyl)benzothieno[2,3-c]pyran-1-one (1b) is in the region of lipid hydrophobic tails.     Real-Time Fluorescence Loop Mediated Isothermal Amplification for the Diagnosis of Malaria

    BACKGROUND: Molecular diagnostic methods can complement existing tools to improve the diagnosis of malaria. However, they require good laboratory infrastructure thereby restricting their use to reference laboratories and research studies. Therefore, adopting molecular tools for routine use in malaria endemic countries will require simpler molecular platforms. The recently developed loop-mediated isothermal amplification (LAMP) method is relatively simple and can be improved for better use in endemic countries. In this study, we attempted to improve this method for malaria diagnosis by using a simple and portable device capable of performing both the amplification and detection (by fluorescence) of LAMP in one platform. We refer to this as the RealAmp method. METHODOLOGY AND SIGNIFICANT FINDINGS: Published genus-specific primers were used to test the utility of this method. DNA derived from different species of malaria parasites was used for the initial characterization. Clinical samples of P. falciparum were used to determine the sensitivity and specificity of this system compared to microscopy and a nested PCR method. Additionally, directly boiled parasite preparations were compared with a conventional DNA isolation method. The RealAmp method was found to be simple and allowed real-time detection of DNA amplification. The time to amplification varied but was generally less than 60 minutes. All human-infecting Plasmodium species were detected. The sensitivity and specificity of RealAmp in detecting P. falciparum was 96.7% and 91.7% respectively, compared to microscopy and 98.9% and 100% respectively, compared to a standard nested PCR method. In addition, this method consistently detected P. falciparum from directly boiled blood samples. CONCLUSION: This RealAmp method has great potential as a field usable molecular tool for diagnosis of malaria. This tool can provide an alternative to conventional PCR based diagnostic methods for field use in clinical and operational programs
