    On the coherent rotation of diffuse matter in numerical simulations of galaxy clusters

    We present a study on the coherent rotation of the intracluster medium and dark matter components of simulated galaxy clusters extracted from a volume-limited sample of the MUSIC project. The set is re-simulated with three different recipes for the gas physics: (i)(i) non-radiative, (ii)(ii) radiative without AGN feedback, and (iii)(iii) radiative with AGN feedback. Our analysis is based on the 146 most massive clusters identified as relaxed, 57 per cent of the total sample. We classify these objects as rotating and non-rotating according to the gas spin parameter, a quantity that can be related to cluster observations. We find that 4 per cent of the relaxed sample is rotating according to our criterion. By looking at the radial profiles of their specific angular momentum vector, we find that the solid body model is not a suitable description of rotational motions. The radial profiles of the velocity of the dark matter show a prevalence of the random velocity dispersion. Instead, the intracluster medium profiles are characterized by a comparable contribution from the tangential velocity and the dispersion. In general, the dark matter component dominates the dynamics of the clusters, as suggested by the correlation between its angular momentum and the gas one, and by the lack of relevant differences among the three sets of simulations.Comment: 12 pages, updated to match the MNRAS versio

    Epigenetic control of EMT/MET dynamics: HNF4α impacts DNMT3s through miRs-29

    Background and aims: Epithelial-to-mesenchymal transition (EMT) and the reverse mesenchymal-to-epithelial transition (MET) are manifestations of cellular plasticity that imply a dynamic and profound gene expression reprogramming. While a major epigenetic code controlling the coordinated regulation of a whole transcriptional profile is guaranteed by DNA methylation, DNA methyltransferase (DNMT) activities in EMT/MET dynamics are still largely unexplored. Here, we investigated the molecular mechanisms directly linking HNF4α, the master effector of MET, to the regulation of both de novo of DNMT 3A and 3B. Methods: Correlation among EMT/MET markers, microRNA29 and DNMT3s expression was evaluated by RT-qPCR, Western blotting and immunocytochemical analysis. Functional roles of microRNAs and DNMT3s were tested by anti-miRs, microRNA precursors and chemical inhibitors. ChIP was utilized for investigating HNF4α DNA binding activity. Results: HNF4α silencing was sufficient to induce positive modulation of DNMT3B, in in vitro differentiated hepatocytes as well as in vivo hepatocyte-specific Hnf4α knockout mice, and DNMT3A, in vitro, but not DNMT1. In exploring the molecular mechanisms underlying these observations, evidence have been gathered for (i) the inverse correlation between DNMT3 levels and the expression of their regulators miR-29a and miR- 29b and (ii) the role of HNF4α as a direct regulator of miR-29a-b transcription. Notably, during TGFβ-induced EMT, DNMT3s' pivotal function has been proved, thus suggesting the need for the repression of these DNMTs in the maintenance of a differentiated phenotype. Conclusions: HNF4α maintains hepatocyte identity by regulating miR-29a and -29b expression, which in turn control epigenetic modifications by limiting DNMT3A and DNMT3B levels

    HDAC4 regulates satellite cell proliferation and differentiation by targeting P21 and Sharp1 genes

    Skeletal muscle exhibits a high regenerative capacity, mainly due to the ability of satellite cells to replicate and differentiate in response to appropriate stimuli. Epigenetic control is effective at different stages of this process. It has been shown that the chromatin-remodeling factor HDAC4 is able to regulate satellite cell proliferation and commitment. However, its molecular targets are still uncovered. To explain the signaling pathways regulated by HDAC4 in satellite cells, we generated tamoxifen-inducible mice with conditional inactivation of HDAC4 in Pax7(+) cells (HDAC4 KO mice). We found that the proliferation and differentiation of HDAC4 KO satellite cells were compromised, although similar amounts of satellite cells were found in mice. Moreover, we found that the inhibition of HDAC4 in satellite cells was sufficient to block the differentiation process. By RNA-sequencing analysis we identified P21 and Sharp1 as HDAC4 target genes. Reducing the expression of these target genes in HDAC4 KO satellite cells, we also defined the molecular pathways regulated by HDAC4 in the epigenetic control of satellite cell expansion and fusion

    Solid State NMR Study of the Mixing Degree Between Ginkgo Biloba Extract and a Soy-Lecithin-Phosphatidylserine in a Composite Prepared by the Phytosome® Method

    Leaves extract of Ginkgo biloba, known in China since the most ancient times, has been widely used in the area of senile dementia thanks to its improving effects on cognitive function. A promising formulation of this botanical ingredient consists in a Ginkgo biloba-soy-lecithin-phosphatidylserine association obtained by the Phytosome® process. The precise assess- ment of the mixing degree between Ginkgo biloba and soy-lecithin-phosphatidylserine in this formulation is an important piece of information for understanding the reasons of its final performances. To this aim in the present study we carried out for the first time a Solid State Nuclear Magnetic Resonance investigation on Ginkgo biloba-soy-lecithin-phosphatidylserine association, on its constituents and on a mechanical mixture. The analysis of different observables highlighted a very intimate mixing (domains of single components not larger than 60 nm) of Ginkgo biloba and soy-lecithin-phosphatidylserine in their association obtained by Phytosome® process, together with a slight modification of their molecular dynamics, not observed in the case of the mechanical mixture

    The influence of planning and response inhibition on cognitive functioning of Non-Psychotic unipolar depressed suicide attempters

    Depression is one of the main risk factors for suicide. However, little is known about the intricate relationships among depressive symptomatology in unipolar depression, suicide risk, and the characteristics of executive dysfunction in depressed patients. We compared 20 non-psychotic unipolar depressed suicide attempters to 20 matching depressed non-attempters and to 20 healthy controls to further investigate the possible differences in neuropsychological performance. Depressed subjects were controlled for current suicidal ideation, and their neuropsychological profile was assessed using a range of measures of executive functioning, attention, verbal memory, processing speed, and psychomotor speed. Depressed groups were outperformed by healthy controls. Depressed attempters presented more cognitive impairment than depressed non-attempters on a simple Go/No-go response inhibition task and performed better than non-attempters on the Tower of London planning task. Depressed attempters were clearly distinguished by a deficit in response inhibition (Go/No-go commission errors). The normative planning performance (Tower of London extra moves) of the suicide attempters was unexpected, and this unanticipated finding calls for further research. Normative planning may indicate an increased risk of suicidal behavior.info:eu-repo/semantics/publishedVersio

    Theatre is a valid add-on therapeutic intervention for emotional rehabilitation of parkinson's disease patients

    Conventional medical treatments of Parkinson's disease (PD) are effective on motor disturbances but may have little impact on nonmotor symptoms, especially psychiatric ones. Thus, even when motor symptomatology improves, patients might experience deterioration in their quality of life. We have shown that 3 years of active theatre is a valid complementary intervention for PD as it significantly improves the well-being of patients in comparison to patients undergoing conventional physiotherapy. Our aim was to replicate these findings while improving the efficacy of the treatment. We ran a single-blinded pilot study lasting 15 months on 24 subjects with moderate idiopathic PD. 12 were assigned to a theatre program in which patients underwent "emotional" training. The other 12 underwent group physiotherapy. Patients were evaluated at the beginning and at the end of their treatments, using a battery of eight clinical and five neuropsychological scales. We found that the emotional theatre training improved the emotional well-being of patients, whereas physiotherapy did not. Interestingly, neither of the groups showed improvements in either motor symptoms or cognitive abilities tested by the neuropsychological battery. We confirmed that theatre therapy might be helpful in improving emotional well-being in PD

    Role of the JP45-Calsequestrin Complex on Calcium Entry in Slow Twitch Skeletal Muscles

    We exploited a variety of mouse models to assess the roles of JP45-CASQ1 (CASQ, calsequestrin) and JP45-CASQ2 on calcium entry in slow twitch muscles. In flexor digitorum brevis (FDB) fibers isolated from JP45-CASQ1-CASQ2 triple KO mice, calcium transients induced by tetanic stimulation rely on calcium entry via La3+- and nifedipine-sensitive calcium channels. The comparison of excitation-coupled calcium entry (ECCE) between FDB fibers from WT, JP45KO, CASQ1KO, CASQ2KO, JP45-CASQ1 double KO, JP45-CASQ2 double KO, and JP45-CASQ1-CASQ2 triple KO shows that ECCE enhancement requires ablation of both CASQs and JP45. Calcium entry activated by ablation of both JP45-CASQ1 and JP45-CASQ2 complexes supports tetanic force development in slow twitch soleus muscles. In addition, we show that CASQs interact with JP45 at Ca2+ concentrations similar to those present in the lumen of the sarcoplasmic reticulum at rest, whereas Ca2+ concentrations similar to those present in the SR lumen after depolarization-induced calcium release cause the dissociation of JP45 from CASQs. Our results show that the complex JP45-CASQs is a negative regulator of ECCE and that tetanic force development in slow twitch muscles is supported by the dynamic interaction between JP45 and CASQs
