    Measuring automatic retrieval: A comparison of implicit memory, process dissociation, and speeded response procedures

    Abstract Using the stem completion task, we compared estimates of automatic retrieval from an implicit memory task, the process dissociation procedure, and the speeded response procedure. Two standard manipulations were employed. In Experiment 1, a depth of processing effect was found on automatic retrieval using the speeded response procedure although this effect was substantially reduced in Experiment 2 when lexical processing was required of all words. In Experiment 3, the speeded response procedure showed an advantage of full versus divided attention at study on automatic retrieval. An implicit condition showed parallel effects in each study, suggesting that implicit stem completion may normally provide a good estimate of automatic retrieval. Also, we replicated earlier findings from the process dissociation procedure, but estimates of automatic retrieval from this procedure were consistently lower than those from the speeded response procedure, except when conscious retrieval was relatively low. We discuss several factors that may contribute to the conflicting outcomes, including the evidence for theoretical assumptions and criterial task differences between implicit and explicit tests

    Characterization of highly stable liposomal and immunoliposomal formulations of vincristine and vinblastine

    Liposome and immunoliposome formulations of two vinca alkaloids, vincristine and vinblastine, were prepared using intraliposomal triethylammonium sucroseoctasulfate and examined for their ability to stabilize the drug for targeted drug delivery in vivo. The pharmacokinetics of both the encapsulated drug (vincristine or vinblastine) and liposomal carrier were examined in Sprague Dawley rats, and the in vivo drug release rates determined. Anti-HER2 immunoliposomal vincristine was prepared from a human anti-HER2/neu scFv and studied for targeted cytotoxic activity in cell culture, and antitumor efficacy in vivo. Nanoliposome formulations of vincristine and vinblastine demonstrated similar pharmacokinetic profiles for the liposomal carrier, but increased clearance for liposome encapsulated vinblastine (t 1/2 = 9.7 h) relative to vincristine (t 1/2 = 18.5 h). Immunoliposome formulations of vincristine targeted to HER2 using an anti-HER2 scFv antibody fragment displayed a marked enhancement in cytotoxicity when compared to non-targeted liposomal vincristine control; 63- or 253-fold for BT474 and SKBR3 breast cancer cells, respectively. Target-specific activity was also demonstrated in HER2-overexpressing human tumor xenografts, where the HER2-targeted formulation was significantly more efficacious than either free vincristine or non-targeted liposomal vincristine. These results demonstrate that active targeting of solid tumors with liposomal formulations of vincristine is possible when the resulting immunoliposomes are sufficiently stabilized

    The Magnitude of Global Marine Species Diversity

    Background: The question of how many marine species exist is important because it provides a metric for how much we do and do not know about life in the oceans. We have compiled the first register of the marine species of the world and used this baseline to estimate how many more species, partitioned among all major eukaryotic groups, may be discovered. Results: There are ∼226,000 eukaryotic marine species described. More species were described in the past decade (∼20,000) than in any previous one. The number of authors describing new species has been increasing at a faster rate than the number of new species described in the past six decades. We report that there are ∼170,000 synonyms, that 58,000–72,000 species are collected but not yet described, and that 482,000–741,000 more species have yet to be sampled. Molecular methods may add tens of thousands of cryptic species. Thus, there may be 0.7–1.0 million marine species. Past rates of description of new species indicate there may be 0.5 ± 0.2 million marine species. On average 37% (median 31%) of species in over 100 recent field studies around the world might be new to science. Conclusions: Currently, between one-third and two-thirds of marine species may be undescribed, and previous estimates of there being well over one million marine species appear highly unlikely. More species than ever before are being described annually by an increasing number of authors. If the current trend continues, most species will be discovered this century

    Do temperature and water depth influence microcrustacean hatching responses from floodplain wetland sediments?

    Microcrustacea in ephemeral wetlands produce dormant eggs to escape prolonged dry conditions. These eggs can hatch on inundation, although in most cases not all eggs hatch during a single wetting event. Incomplete hatching can reflect bet-hedging strategies, but also the presence or absence of environmental cues that stimulate hatching. This study examines the effects of environmental cues likely to change for wetlands in the future, namely, temperature and water depth. Surface sediments collected from dry anabranches of the Macintyre River floodplain (eastern Australia) were inundated under two temperature regimes (warm and cool) in microcosms of two depths (shallow and deep). Hatched microcrustacea were sampled for 6 weeks. The abundance and assemblage composition of microcrustacea varied by temperature but not by depth. Although the total abundance was greater under warm conditions, the effect of temperature diminished over time. Temperature also had a greater effect on non-ostracods, with 144% more non-ostracods being hatched under warm than under cool conditions. Thus, changes to temperature during inundation periods arising from global climate change or river regulation are likely to influence the abundance and composition of microcrustacean assemblages, especially among non-ostracods, which will influence food availability for larval and juvenile native fish and, hence, recruitment

    A gateway event is a change to a system that leads to the possibility of huge increases in kinds and levels of complexity. It opens up a whole new kind of phase space to the systemÕs dynamics. Gateway events during evolution of life on earth include the appearance of eukaryotes (organisms with a cell nucleus), an oxygen atmosphere, multi-cellular organisms and grass. Gateway events during the development of mathematics include each invention of a new class of numbers (negative, irrational, imaginary, ...), and dropping Euclid's parallel postulate. A gateway event produces a profound and fundamental change to the system: Once through the gateway, life is never the same again. We are currently poised on the threshold of a significant gateway event in computation: That of breaking free from many of our current Òclassical computationalÓ assumptions. Our Grand Challenge for computer science is to journey through the gateway event obtained by breaking our current classical computational assumptions, and thereby develop a mature science of Non-Classical Computatio

    Evaluating wetland vegetation responses to flow regimes is challenging because of the inherently complex, variable and dynamic nature of wetland vegetation in space and time. We propose four principles to guide the development of management objectives and evaluation approaches to support adaptive management of wetland vegetation in flow-managed systems. First, we assert a need for more explicit, direct and defensible alignment of management objectives, targets and indicators to reflect broader ecological, sociocultural and economic values, and the underlying ecosystem functions that support them. Second, we propose a framework for indicator selection across multiple spatiotemporal scales and levels of ecological organisation, from individuals to landscape mosaics (vegscapes). Third, we emphasise the need to evaluate vegetation condition and responses to environmental flows in relation to a more nuanced understanding of temporal flow dynamics. Finally, we discuss the importance of considering the effects of non-flow variables that can modify vegetation responses to environmental flows. We highlight key knowledge needs required to support the implementation of these principles, particularly the urgency of improving our understanding of ecological, sociocultural and economic values of wetland vegetation and the attributes and functions that support these values
