    Polyvalent horse F(Ab`)2 snake antivenom: Development of process to produce polyvalent horse F(Ab`)2 antibodies anti-african snake venom

    A method to obtain polyvalent anti-Bitis and polyvalent-anti-Naja antibodies was developed by immunizing horses with B. arietans, B. nasicornis, B. rhinoceros, N. melanoleuca and N. mossambicacrude venoms. Antibody production was followed by the ELISA method during the immunization procedure. Once the desired anti-venom antibody titers were attained, horses were bled and the immunoglobulins were separated from the sera by (NH4)2SO4 precipitation, cleaved with pepsin and filtered through a 30 kDa ultrafiltration membrane. F(ab´)2 fragments were further purified by Q-Fast Flow chromatography, concentrated by molecular ultrafiltration and sterilized by filtration through 0.22 m membranes. The resulting F(ab´)2 preparations were rich in intact L and in pieces of H IgG(T) chains, as demonstrated by electrophoresis and Western blot and exhibited high antibody titers, as assayed by the ELISA method. In addition, the preparations possess a significant capacity to neutralize the lethality of venoms, as estimated by ED50 determination in mouse assay and are free of toxic substances, pyrogen and bacterial or fungal contaminations

    Retention of copper(II) metal ions in a silicon-glass microfluidic device

    This work describes the construction of a silicon microchip for retention of copper(II) metal ions. Conventional photolithographic process was applied to transfer the generated pattern to silicon wafers. Using Reactive Ion Etching (RIE), SF6 based, channels 50 mu m wide and 10 mu m deep were produced. The channels were sealed with borosilicate glass using anodic bonding process. The surface of the channels were modified with N-(beta-aminoethyl)-gamma-aminopropyl-trimetoxysilane through a silanization reaction to promote the adsorption of copper(II) ions. An amperometric detector was placed at the microchip outlet and copper(II) ions were detected by a gold electrode at 0 V (against Ag/AgCl(KCl sat.) reference electrode). Copper(II) ions were retained and eluted with HCl 50 mu mol L-1 in a micro-flow system at a flow rate about 100 mu L min(-1). Reproducibility in the peak area and height were about 4.6 % and 10 %, respectively, for three consecutive injections of 600 mu L of 10 mu mol L-1 copper(II) sample.1881531153

    Regulatory T cells (Treg) play a critical role in immune tolerance. The scarcity of Treg therapy clinical trials in humans has been largely due to the difficulty in obtaining sufficient Treg numbers. We performed a preclinical investigation on the potential of mesenchymal stromal cells (MSCs) to expand Treg in vitro to support future clinical trials. Human peripheral blood mononuclear cells from healthy donors were cocultured with allogeneic bone marrow-derived MSCs expanded under xenogeneic-free conditions. Our data show an increase in the counts and frequency of CD4+ CD25high Foxp3+ CD127low Treg cells (4- and 6-fold, respectively) after a 14-day coculture. However, natural Treg do not proliferate in coculture with MSCs. When purified conventional CD4 T cells (Tcon) are cocultured with MSCs, only cells that acquire a Treg-like phenotype proliferate. These MSC-induced Treg-like cells also resemble Treg functionally, since they suppress autologous Tcon proliferation. Importantly, the DNA methylation profile of MSC-induced Treg-like cells more closely resembles that of natural Treg than of Tcon, indicating that this population is stable. The expression of PD-1 is higher in Treg-like cells than in Tcon, whereas the frequency of PDL-1 increases in MSCs after coculture. TGF-β levels are also significantly increased MSC cocultures. Overall, our data suggest that Treg enrichment by MSCs results from Tcon conversion into Treg-like cells, rather than to expansion of natural Treg, possibly through mechanisms involving TGF-β and/or PD-1/PDL-1 expression.     Covid-19 Dynamic Monitoring and Real-Time Spatio-Temporal Forecasting

    Background: Periodically, humanity is often faced with new and emerging viruses that can be a significant global threat. It has already been over a century post—the Spanish Flu pandemic, and we are witnessing a new type of coronavirus, the SARS-CoV-2, which is responsible for Covid-19. It emerged from the city of Wuhan (China) in December 2019, and within a few months, the virus propagated itself globally now resulting more than 50 million cases with over 1 million deaths. The high infection rates coupled with dynamic population movement demands for tools, especially within a Brazilian context, that will support health managers to develop policies for controlling and combating the new virus. / Methods: In this work, we propose a tool for real-time spatio-temporal analysis using a machine learning approach. The COVID-SGIS system brings together routinely collected health data on Covid-19 distributed across public health systems in Brazil, as well as taking to under consideration the geographic and time-dependent features of Covid-19 so as to make spatio-temporal predictions. The data are sub-divided by federative unit and municipality. In our case study, we made spatio-temporal predictions of the distribution of cases and deaths in Brazil and in each federative unit. Four regression methods were investigated: linear regression, support vector machines (polynomial kernels and RBF), multilayer perceptrons, and random forests. We use the percentage RMSE and the correlation coefficient as quality metrics. / Results: For qualitative evaluation, we made spatio-temporal predictions for the period from 25 to 27 May 2020. Considering qualitatively and quantitatively the case of the State of Pernambuco and Brazil as a whole, linear regression presented the best prediction results (thematic maps with good data distribution, correlation coefficient >0.99 and RMSE (%) <4% for Pernambuco and around 5% for Brazil) with low training time: [0.00; 0.04 ms], CI 95%. / Conclusion: Spatio-temporal analysis provided a broader assessment of those in the regions where the accumulated confirmed cases of Covid-19 were concentrated. It was possible to differentiate in the thematic maps the regions with the highest concentration of cases from the regions with low concentration and regions in the transition range. This approach is fundamental to support health managers and epidemiologists to elaborate policies and plans to control the Covid-19 pandemics

    O presente trabalho tem como objetivo analisar, ? luz da Sociolingu?stica Variacionista, a varia??o dos pronomes de segunda pessoa do singular, tu e voc?, no falar ressaquinhense, identificando os fatores lingu?sticos e extralingu?stico que influenciam o uso de uma ou outra forma pronominal. O corpus constitui-se de 24 entrevistas e foi estratificado em fun??o da ?rea geogr?fica, faixa et?ria e sexo. Os dados gerais indicaram a predomin?ncia de voc? sobre o tu em Ressaquinha (MG). Em seguida, o programa GoldVarb X selecionou 4 grupos de fatores que favorecem o uso de tu na seguinte ordem de relev?ncia: tipo de refer?ncia (espec?fica), contexto sint?tico (sujeito), tipo de discurso (direto), faixa et?ria (19 a 35 anos). Outras vari?veis, tais como: contexto frasal, sexo e ?rea geogr?fica n?o foram selecionadas por tal programa computacional, mas demonstraram que o pronome tu ? mais recorrente nas frases interrogativas, nas falas masculinas e na zona urbana, respectivamente.The paper aims to describe and analyze, in the light of Variationist Sociolinguistics, the variation of second person singular pronouns (Tu and Voc?) in the town of Ressaquinha (MG), identifying the linguistic and extralinguistic factors that influence the use of one or another pronominal form. The corpus consisted of 24 interviews and was stratified according to geographical area, age and gender. The data provided indicates a predominance of Voc? about Tu in Ressaquinha (MG). The GoldVarb X program selected four factors that favor the use of Tu, in the following order of relevance: reference type (specific); syntactic context (subject); type of speech (direct); age group (19 to 35 years). Other variables, such as: such a program did not select phrasal context, gender and geographical area, but they showed that the pronoun Tu is recurrent in the interrogative phrases, in the masculine speeches and in the urban zone

    COVID-SGIS: A Smart Tool for Dynamic Monitoring and Temporal Forecasting of Covid-19

    Background: The global burden of the new coronavirus SARS-CoV-2 is increasing at an unprecedented rate. The current spread of Covid-19 in Brazil is problematic causing a huge public health burden to its population and national health-care service. To evaluate strategies for alleviating such problems, it is necessary to forecast the number of cases and deaths in order to aid the stakeholders in the process of making decisions against the disease. We propose a novel system for real-time forecast of the cumulative cases of Covid-19 in Brazil. / Methods: We developed the novel COVID-SGIS application for the real-time surveillance, forecast and spatial visualization of Covid-19 for Brazil. This system captures routinely reported Covid-19 information from 27 federative units from the Brazil.io database. It utilizes all Covid-19 confirmed case data that have been notified through the National Notification System, from March to May 2020. Time series ARIMA models were integrated for the forecast of cumulative number of Covid-19 cases and deaths. These include 6-days forecasts as graphical outputs for each federative unit in Brazil, separately, with its corresponding 95% CI for statistical significance. In addition, a worst and best scenarios are presented. / Results: The following federative units (out of 27) were flagged by our ARIMA models showing statistically significant increasing temporal patterns of Covid-19 cases during the specified day-to-day period: Bahia, Maranhão, Piauí, Rio Grande do Norte, Amapá, Rondônia, where their day-to-day forecasts were within the 95% CI limits. Equally, the same findings were observed for Espírito Santo, Minas Gerais, Paraná, and Santa Catarina. The overall percentage error between the forecasted values and the actual values varied between 2.56 and 6.50%. For the days when the forecasts fell outside the forecast interval, the percentage errors in relation to the worst case scenario were below 5%. / Conclusion: The proposed method for dynamic forecasting may be used to guide social policies and plan direct interventions in a cost-effective, concise, and robust manner. This novel tools can play an important role for guiding the course of action against the Covid-19 pandemic for Brazil and country neighbors in South America

    The biogas chain and local sustainability: an environmental socioeconomic analysis of energy obtained from urban solid-waste from the Caximba land-fill in Curitiba, Parana, Brazil

    Urban solid waste-derived renewable energy sources are currently being discussed as strategic choices; however, they have not yet been significantly included in world and/or Brazilian energetic matrixes. Feasibility studies must be carded out on the use of renewable sources and on related socioeconomic and environmental impacts. The research method was exploratory and was aimed at assessing the feasibility of generating energy from the Caximba (Parana, Brazil) sanitary landfill biogas chain, taking its socioeconomic and environmental sustainability into consideration; this landfill contains around 8 million tons of solid waste. The results showed that generating renewable energy was not economically feasible in the technological conditions defined by this case-study due to the plant's low-scale production. However, biogas production could become economically, environmentally and socially feasible if combined with carbon credit trading.1934839

    We show that in the context of two-dimensional sigma models minimal coupling of an ordinary rigid symmetry Lie algebra g\mathfrak{g} leads naturally to the appearance of the "generalized tangent bundle" TMTMTM\mathbb{T}M \equiv TM \oplus T^*M by means of composite fields. Gauge transformations of the composite fields follow the Courant bracket, closing upon the choice of a Dirac structure DTMD \subset \mathbb{T}M (or, more generally, the choide of a "small Dirac-Rinehart sheaf" D\cal{D}), in which the fields as well as the symmetry parameters are to take values. In these new variables, the gauge theory takes the form of a (non-topological) Dirac sigma model, which is applicable in a more general context and proves to be universal in two space-time dimensions: A gauging of g\mathfrak{g} of a standard sigma model with Wess-Zumino term exists, \emph{iff} there is a prolongation of the rigid symmetry to a Lie algebroid morphism from the action Lie algebroid M×gMM \times \mathfrak{g}\to M into DMD\to M (or the algebraic analogue of the morphism in the case of D\cal{D}).     Lipidic characterization of Santa Ines lamb shoulder

    The edible portion of the shoulder of 12 castrated and 12 non-castrated Santa Ines lambs slaughtered at different ages (84, 168, 210, 252 days) were used. The shoulders were chemically analyzed to determine the quantity of total lipids, cholesterol, and fatty acids composition. Castrated and non-castrated lambs gained body weight (p = 0.0393, p = 0.0017) and half carcass weight (p = 0.0240, p = 0.0017), respectively. The shoulder weight was increased in the carcasses of non-castrated lambs (p = 0.0110). The edible portion of the shoulder of castrated lambs presented higher total lipids (16.09 g.100 g(-1)). The cholesterol content was influenced by castration (p = 0.0001) reducing with age. Castrated animals presented higher content of C18:1 T11, CLA, and C18:0. The shoulder weight is only increased with increasing age in the carcasses of non-castrated lambs. Castration influences the cholesterol content of the shoulder; however, both castrated and non-castrated lambs had their cholesterol contents reduced with increasing age. Castration and age interfered in the estearic acid concentration of the edible portion of lamb shoulder.31250851

    OBJECTIVE:To develop and validate conceptual and operational definitions for the defining characteristics of the respiratory nursing diagnoses, ineffective breathing pattern, impaired gas exchange and impaired spontaneous ventilation, in newborns.METHODS:This was a methodological study of conceptual validation of the defining characteristics of three respiratory nursing diagnoses, by consensus analysis of a committee of five specialist nurses, and then a group of five non-nursing professionals, using the Delphi technique.RESULTS:After two rounds of evaluation, consensus was obtained that was equal to or greater than 80% on all of the definitions, which were then considered validated.CONCLUSION:The definitions developed for the defining characteristics of three nursing diagnoses were validated with a high level of consensus.OBJETIVO:Elaborar e validar definições conceituais e operacionais para as características definidoras dos diagnósticos de enfermagem respiratórios, Padrão Respiratório Ineficaz, Troca de Gases Prejudicada e Ventilação Espontânea Prejudicada em recém-nascidos.MÉTODOS:Estudo metodológico, de validação conceitual das características definidoras dos três diagnósticos de enfermagem respiratórios por meio da análise de consenso de um comitê de cinco enfermeiras especialistas e de cinco profissionais não enfermeiros, utilizando a técnica Delphi.RESULTADOS:Após duas rodadas de avaliação, obteve-se consenso igual ou superior a 80% na totalidade das definições, sendo consideradas validadas.CONCLUSÃO:As definições elaboradas para as características definidoras dos três diagnósticos de enfermagem foram validadas com elevado grau de consenso.Universidade Federal de São Paulo (UNIFESP) Escola Paulista de EnfermagemUNIFESP, EPESciEL
