STAGE-SPECIFIC EFFECTS OF PITX2 INACTIVATION DURING SKELETAL MYOGENESIS

Durante el desarrollo embrionario, estructuras transitorias llamadas somitos dan lugar al dermomiotomo epitelial, origen de los precursores de la musculatura esquelética del tronco. Las células progenitoras musculares multipo- tentes (MPC) que expresan Pax3 surgen del dermomiotomo y adquieren su identidad definitiva a través de los factores reguladores miogénicos (MRF) Myf5, Mrf4 y MyoD. Además, las células madre musculares (células satélite) del cuerpo y las extremidades también surgen de somitos, en común con el músculo con el que están asociadas. Evidencias previas han revelado que el factor de transcripción Pitx2 podría ser un factor clave dentro de las vías moleculares que controlan el destino de los progenitores musculares derivados de somitos. Sin embargo, la posición jerárquica ocupada por Pitx2 dentro de la cascada genética que controla la miogénesis derivada de los somitos sigue sin resolverse. Para obtener información sobre este problema, hemos generado dos ratones mutantes condicionales para inactivar específicamente Pitx2 en progenitores musculares multipotentes Pax3+ (Pax3Cre+/Pitx2loxp/loxp) y en los progenitores comprometidos miogénicos (Myf5Cre+/Pitx2loxp / loxp). Nuestros análisis revelaron que la inacti- vación de Pitx2 en los precursores de Pax3+ conduce a una miogénesis deteriorada, mientras que la pérdida de Pitx2 en las células miogénicas Myf5+ tiene un impacto en el número de células madre satélite musculares que alcanzan su nicho en el músculo adulto con graves consecuencias en la regeneración muscular.During embryonic development, transitory structures called somites give rise to an epithelial dermomyotome, the source of skeletal muscles precursors of the trunk. Multipotent muscle progenitor cells (MPCs) that express Pax3 arise from the dermomyotome and acquire their definitive identity via the myogenic regulatory factors (MRFs) Myf5, Mrf4, and MyoD. Moreover, the muscle stem cells (satellite cells) of the body and limbs also arise from somites, in common with the muscle that they are associated with. Previous evidences have revealed that the transcription factor Pitx2 might be a player within the molecular pathways controlling somite-derived muscle progenitors’ fate. However, the hierarchical position occupied by Pitx2 within the genetic cascade that control somite-derived myogenesis remain unsolved. To get insight into this issue, we have differentially generated two conditional Pitx2 mutant mice to specifically inactivate Pitx2 in multipotent Pax3+ muscle progenitors (Pax3Cre+/Pitx2loxp/loxp mice) and in myogenic committed progenitors (Myf5Cre+/Pitx2loxp/loxp mice). Our analyses revealed that Pitx2 inactivation in Pax3+ precursors lead to impaired myogenesis while the loss of Pitx2 in Myf5+ myogenic cells have an impact in the number of muscle satellite stem cells that reach their niche in the adult muscle with severe consequences in muscle regeneration.Tesis Univ. A novel programmable lysozyme-based lysis system in Pseudomonas putida for biopolymer production

Indexación: Scopus; Web of Science.Cell lysis is crucial for the microbial production of industrial fatty acids, proteins, biofuels, and biopolymers. In this work, we developed a novel programmable lysis system based on the heterologous expression of lysozyme. The inducible lytic system was tested in two Gram-negative bacterial strains, namely Escherichia coli and Pseudomonas putida KT2440. Before induction, the lytic system did not significantly arrest essential physiological parameters in the recombinant E. coli (ECPi) and P. putida (JBOi) strain such as specific growth rate and biomass yield under standard growth conditions. A different scenario was observed in the recombinant JBOi strain when subjected to PHA-producing conditions, where biomass production was reduced by 25% but the mcl-PHA content was maintained at about 30% of the cell dry weight. Importantly, the genetic construct worked well under PHA-producing conditions (nitrogen-limiting phase), where more than 95% of the cell population presented membrane disruption 16 h post induction, with 75% of the total synthesized biopolymer recovered at the end of the fermentation period. Geomorfología y paisaje en el ordenamiento territorial: valorizando el corredor inferior del río Mapocho

Considering the roles how biological corridor of the lower hydrological system of the Mapocho River, pointing to their integration in the urban planning of the respective communes from an ecologic, environmental and social perspective, is carried on a research about their geomorphological and landscape characteristics and the processes that are leading the evolution of the landscape. With this knowledge and from an ecosystemic perspective of the river territory, the limits of different homogeneous units were determined. Any geomorphological unit recognized, meaning also landscape systems, was characterized by its geology, sedimentology, soils and drainage, hydro-morphodynamic processes and human intervention, which allows through the determination of the quality and fragility of the geomorphic-landscape units, to establish land use proposals with their corresponding scopes on qualifications and restrictions. The resulting mapping contains implicit a proposal for land-use planning from the ecological planning point of view.    Considerando las funciones de corredor biológico del sistema hidrológico inferior del Río Mapocho, y apuntando a su integración a la planificación urbana de la intercomuna respectiva desde un punto de vista ecológico-ambiental y social, se reconocen sus características geomorfológicas y paisajísticas, y se establecen los procesos que lideran la evolución del modelado de las formas naturales. Con esta base se delimita el territorio fluvial desde una perspectiva ecosistémica y se determinan los límites de las diferentes unidades homogéneas resultantes. Cada unidad geomorfológica, en alta correspondencia con el sistema de paisajes, es caracterizada en función de su geología, sedimentología, suelos y drenaje, hidro-morfodinámica e intervención humana, lo cual permite mediante la determinación de la calidad y fragilidad de ellas, establecer propuestas de uso del suelo con sus correspondientes alcances sobre aptitudes y restricciones. La cartografía resultante contiene implícita una propuesta de ordenamiento del territorio desde el punto de vista de la planificación ecológica.  

Consideraciones en torno al Liber Contemplationis in Deum

El objetivo de este breve trabajo ha sido presentar algunas características que nos han llamado la atención del amplísimo Liber Contemplationis in Deum. Estos aspectos son, desde un punto de vista formal, el uso de esquemas, generalmente como paralelismos muy cuidados en todo el Liber; la pasión desbordante que se refleja en la ampulosidad y variedad de adjetivos con los que invoca a Dios; y, sobre todo y subyacente en todo el texto, la continua manifestación de su hondo pesar y de su arrepentimiento como pecador, la necesidad de hacer partícipes de su fe a los infieles yel canto a la esperanza en la misericordia y piedad de Dios, expresado en el punto culminante de los capítulos.The aim of this brief study is to offer some characteristics that have caught our attention in the very broad Liber contemplationis in Deum. These aspects are, from a formal point of view, the use of schemes, generally in the form of very carefully designed parallel patterns throughout the Liber; the overwhelming passion that is reflected in the wealth and variety of adjectives that invoke God; and above all, and underlying all the text, the continuous manifestation of Llull's deep regret and repentance as a sinner, the need to share his faith with infidels and the song of hope andtrust in the mercy and pity of God, which marks the culminating points of each chapter
