    Investigation of fiber/matrix adhesion: test speed and specimen shape effects in the cylinder test

    The cylinder test, developed from the microdroplet test, was adapted to assess the interfacial adhesion strength between fiber and matrix. The sensitivity of cylinder test to pull-out speed and specimen geometry was measured. It was established that the effect of test speed can be described as a superposition of two opposite, simultaneous effects which have been modeled mathematically by fitting two parameter Weibull curves on the measured datas. Effects of the cylinder size and its geometrical relation on the measured strength values have been analyzed by finite element method. It was concluded that the geometry has a direct influence on the stress formation. Based on the results achieved, recommendations were given on how to perform the novel single fiber cylinder test

    CARE MODEL IN PRIMARY HEALTH CARE: ACCESS AND COMPREHENSIVE CARE DURING THE COVID-19 PANDEMIC

    Objective: To analyze the care model in Primary Health Care from the perspective of access and comprehensive care.Method: Qualitative research, with data collected from May to December 2021 in each regional health center in the state of Paraná through recorded interviews using a semi-structured script with 26 managers. IRAMUTEQ  software was used to process and group the data into five classes. The results were analyzed using dialectical hermeneutics.Results: Two classes showed actions related to the attributes of access and comprehensiveness present in the care model. Actions to increase access included the participation of a multi-professional team to promote comprehensive care and the use of technological resources while maintaining the biomedical model. Final considerations: During the pandemic, there was adaptation and resilience on the part of managers who organized the care model and the inclusion of technological tools to facilitate access and continuity of care

    ExacTrac Dynamic workflow evaluation: Combined surface optical/thermal imaging and X‐ray positioning

    In modern radiotherapy (RT), especially for stereotactic radiotherapy or stereotactic radiosurgery treatments, image guidance is essential. Recently, the ExacTrac Dynamic (EXTD) system, a new combined surface-guided RT and image-guided RT (IGRT) system for patient positioning, monitoring, and tumor targeting, was introduced in clinical practice. The purpose of this study was to provide more information about the geometric accuracy of EXTD and its workflow in a clinical environment. The surface optical/thermal- and the stereoscopic X-ray imaging positioning systems of EXTD was evaluated and compared to cone-beam computed tomography (CBCT). Additionally, the congruence with the radiation isocenter was tested. A Winston Lutz test was executed several times over 1 year, and repeated end-to-end positioning tests were performed. The magnitude of the displacements between all systems, CBCT, stereoscopic X-ray, optical-surface imaging, and MV portal imaging was within the submillimeter range, suggesting that the image guidance provided by EXTD is accurate at any couch angle. Additionally, results from the evaluation of 14 patients with intracranial tumors treated with open-face masks are reported, and limited differences with a maximum of 0.02 mm between optical/thermal- and stereoscopic X-ray imaging were found. As the optical/thermal positioning system showed a comparable accuracy to other IGRT systems, and due to its constant monitoring capability, it can be an efficient tool for detecting intra-fractional motion and for real-time tracking of the surface position during RT

    The MOF-containing NSL complex associates globally with housekeeping genes, but activates only a defined subset

    The MOF (males absent on the first)-containing NSL (non-specific lethal) complex binds to a subset of active promoters in Drosophila melanogaster and is thought to contribute to proper gene expression. The determinants that target NSL to specific promoters and the circumstances in which the complex engages in regulating transcription are currently unknown. Here, we show that the NSL complex primarily targets active promoters and in particular housekeeping genes, at which it colocalizes with the chromatin remodeler NURF (nucleosome remodeling factor) and the histone methyltransferase Trithorax. However, only a subset of housekeeping genes associated with NSL are actually activated by it. Our analyses reveal that these NSL-activated promoters are depleted of certain insulator binding proteins and are enriched for the core promoter motif ‘Ohler 5’. Based on these results, it is possible to predict whether the NSL complex is likely to regulate a particular promoter. We conclude that the regulatory capacity of the NSL complex is highly context-dependent. Activation by the NSL complex requires a particular promoter architecture defined by combinations of chromatin regulators and core promoter motifs

    ANÁLISE DA COBERTURA VACINAL E HESITAÇÃO VACINAL DE CRIANÇAS DO LITORAL DO PARANÁ DE 2015 A 2021

    O objetivo do estudo foi analisar a cobertura vacinal (CV) e a ocorrência da hesitação vacinal, sobre a perspectiva dos cuidadores, na vacinação de crianças de 0 a 10 anos residentes no litoral do Paraná nos municípios que compreendem a primeira regional de saúde (1ª RS - Paranaguá: Antonina, Guaraqueçaba, Guaratuba, Matinhos, Morretes, Paranaguá e Pontal do Paraná), no período de 2015 a 2021. Trata-se de um estudo quantitativo transversal realizado em duas etapas: a primeira foi a coleta e análise de dados secundários referente a cobertura vacinal referentes às vacinas recomendadas para crianças de 0 a 10 anos, a segunda foi a aplicação de questionário online para verificar a hesitação vacinal de pais/responsáveis. Os resultados referentes a CV possibilitam observar que todos os imunobiológicos que foram analisados sofreram queda na cobertura no período de análise, mas principalmente nos anos de 2020 e 2021 (anos pandêmicos) não chegando a 80% de cobertura em grande parte dos municípios de 1ªRS Paranaguá. A hesitação vacinal foi confirmada pela baixa CV, mas em relação aos determinantes sociais analisados, encontrou-se associação estatisticamente significativa com a baixa escolaridade. Conclui-se que ocorreu queda na CV no litoral do Paraná desde 2016 e que houve piora durante a pandemia de COVID-19, a hesitação vacinal foi observada de maneira sutil pois foi analisada em um número baixo de pessoas em relação a população total da região.Palavras-chave: Saúde da criança; Vacinação; Vacinas; Recusa de Vacinação

    Critical analysis of information security culture definitions

    This article aims to advance the understanding of information security culture through a critical reflection on the wide-ranging definitions of information security culture in the literature. It uses the hermeneutic approach for conducting literature reviews. The review identifies 16 definitions of information security culture in the literature. Based on the analysis of these definitions, four different views of culture are distinguished. The shared values view highlights the set of cultural value patterns that are shared across the organization. An action-based view highlights the behaviors of individuals in the organization. A mental model view relates to the abstract view of the individual’s thinking on how information security culture must work. Finally, a problem-solving view emphasizes a combination of understanding from shared value-based and action-based views. The paper analyzes and presents the limitations of these four views of information security culture definitions

    GAD1 mRNA Expression and DNA Methylation in Prefrontal Cortex of Subjects with Schizophrenia

    Dysfunction of prefrontal cortex in schizophrenia includes changes in GABAergic mRNAs, including decreased expression of GAD1, encoding the 67 kDa glutamate decarboxylase (GAD67) GABA synthesis enzyme. The underlying molecular mechanisms remain unclear. Alterations in DNA methylation as an epigenetic regulator of gene expression are thought to play a role but this hypothesis is difficult to test because no techniques are available to extract DNA from GAD1 expressing neurons efficiently from human postmortem brain. Here, we present an alternative approach that is based on immunoprecipitation of mononucleosomes with anti-methyl-histone antibodies differentiating between sites of potential gene expression as opposed to repressive or silenced chromatin. Methylation patterns of CpG dinucleotides at the GAD1 proximal promoter and intron 2 were determined for each of the two chromatin fractions separately, using a case-control design for 14 schizophrenia subjects affected by a decrease in prefrontal GAD1 mRNA levels. In controls, the methylation frequencies at CpG dinucleotides, while overall higher in repressive as compared to open chromatin, did not exceed 5% at the proximal GAD1 promoter and 30% within intron 2. Subjects with schizophrenia showed a significant, on average 8-fold deficit in repressive chromatin-associated DNA methylation at the promoter. These results suggest that chromatin remodeling mechanisms are involved in dysregulated GABAergic gene expression in schizophrenia

    MODELO ASISTENCIAL EN LA ATENCIÓN PRIMARIA A LA SALUD: ACCESO Y ATENCIÓN INTEGRAL DURANTE LA PANDEMIA DE COVID-19

    Objetivo: analizar el modelo asistencial en Atención Primaria a la Salud desde la perspectiva del acceso y la atención integral.Método: investigación cualitativa, con datos recogidos de mayo a diciembre de 2021, en cada centro regional de salud del estado de Paraná, a través de entrevistas grabadas, utilizando un guion semiestructurado con 26 gestores. Se utilizó el software IRAMUTEQ para procesar y agrupar los datos en cinco clases. Los resultados se analizaron utilizando la hermenéutica dialéctica.Resultados: dos clases mostraron acciones relacionadas con los atributos de acceso e integralidad, presentes en el modelo de atención. Las acciones para aumentar el acceso incluyeron la participación de un equipo multiprofesional para promover la atención integral y la inclusión de recursos tecnológicos, manteniendo el modelo biomédico. Consideraciones finales: durante la pandemia, hubo adaptación y resiliencia por parte de los gestores que organizaron el modelo de atención, incluyendo herramientas tecnológicas para facilitar el acceso y la continuidad de la atención
