    FUSE observations of the HI interstellar gas of IZw18

    We present the analysis of FUSE observations of the metal-deficient dwarf galaxy IZw18. We measured column densities of HI, NI, OI, ArI, SiII, and FeII. The OI/HI ratio (log(OI/HI)=-4.7^{+0.8}_{-0.6}) is consistent with the O/H ratio observed in the HII regions (all uncertainties are 2-sigma). If the oxygen is depleted in the HI region compared to the HII regions, the depletion is at most 0.5dex. This is also consistent with the log(O/H) ratios ~-5 measured with FUSE in the HI regions of other blue compact dwarf galaxies. With log(NI/OI)=-2.4^{+0.6}_{-0.8}, the measured NI/OI ratio is lower than expected for primary nitrogen. The determination of the NII column density is needed to discriminate between a large ionization of NI or a possible nitrogen deficiency. The neutral argon is also apparently underabundant, indicating that ionization into ArII is likely important. The column densities of the other alpha-chain elements SiII and ArI favor the lower edge of the permitted range of OI column density, log(N(OI))~16.3.Comment: Accepted for publication in A&

    Exosomes as Hedgehog carriers in cytoneme-mediated transport and secretion

    The Hedgehog signalling pathway is crucial for development, adult stem cell maintenance, cell migration and axon guidance in a wide range of organisms. During development, the Hh morphogen directs tissue patterning according to a concentration gradient. Lipid modifications on Hh are needed to achieve graded distribution, leading to debate about how Hh is transported to target cells despite being membrane-tethered. Cytonemes in the region of Hh signalling have been shown to be essential for gradient formation, but the carrier of the morphogen is yet to be defined. Here we show that Hh and its co-receptor Ihog are in exovesicles transported via cytonemes. These exovesicles present protein markers and other features of exosomes. Moreover, the cell machinery for exosome formation is necessary for normal Hh secretion and graded signalling. We propose Hh transport via exosomes along cytonemes as a significant mechanism for the restricted distribution of a lipid-modified morphogen.PostprintPeer reviewe

    Tubulin Binds to the Cytoplasmic Loop of TRESK Background K+ Channel In Vitro.

    The cytoplasmic loop between the second and third transmembrane segments is pivotal in the regulation of TRESK (TWIK-related spinal cord K+ channel, K2P18.1, KCNK18). Calcineurin binds to this region and activates the channel by dephosphorylation in response to the calcium signal. Phosphorylation-dependent anchorage of 14-3-3 adaptor protein also modulates TRESK at this location. In the present study, we identified molecular interacting partners of the intracellular loop. By an affinity chromatography approach using the cytoplasmic loop as bait, we have verified the specific association of calcineurin and 14-3-3 to the channel. In addition to these known interacting proteins, we observed substantial binding of tubulin to the intracellular loop. Successive truncation of the polypeptide and pull-down experiments from mouse brain cytosol narrowed down the region sufficient for the binding of tubulin to a 16 amino acid sequence: LVLGRLSYSIISNLDE. The first six residues of this sequence are similar to the previously reported tubulin-binding region of P2X2 purinergic receptor. The tubulin-binding site of TRESK is located close to the protein kinase A (PKA)-dependent 14-3-3-docking motif of the channel. We provide experimental evidence suggesting that 14-3-3 competes with tubulin for the binding to the cytoplasmic loop of TRESK. It is intriguing that the 16 amino acid tubulin-binding sequence includes the serines, which were previously shown to be phosphorylated by microtubule-affinity regulating kinases (MARK kinases) and contribute to channel inhibition. Although tubulin binds to TRESK in vitro, it remains to be established whether the two proteins also interact in the living cell

    Inequality, social capital, and varieties of capitalism in Latin America

    This paper aims to research whether the varieties of capitalism impact the relationship between social capital and inequality in developed economies (coordinated and liberal economies) and Latin America (hierarchical economies) in the period from 2000 to 2016. To do that, the study uses a panel analysis of developed economies and Latin America, that is, a fixed effect panel static analysis. The model includes one dependent variable (Gini index) and ten independent variables (generalised trust, informal economy, years of schooling, union density, market capitalization, annual gross domestic product, gross domestic product per capita, health expenditure, feminine labour force, and gross capital). The findings show that the relationship between inequality and social capital is negative and significant and that hierarchical economies (Latin America) amplified such a relationship

    TIA: A forward model and analyzer for Talbot interferometry experiments of dense plasmas

    Producción CientíficaInterferometry is one of the most sensitive and successful diagnostic methods for plasmas. However, owing to the design of most common interferometric systems, the wavelengths of operation and, therefore, the range of densities and temperatures that can be probed are severely limited. Talbot–Lau interferometry offers the possibility of extending interferometry measurements to x-ray wavelengths by means of the Talbot effect. While there have been several proof-of-concept experiments showing the efficacy of this method, it is only recently that experiments to probe High Energy Density (HED) plasmas using Talbot–Lau interferometry are starting to take place. To improve these experimental designs, we present here the Talbot-Interferometry Analyzer (TIA) tool, a forward model for generating and postprocessing synthetic x-ray interferometry images from a Talbot–Lau interferometer. Although TIA can work with any two-dimensional hydrodynamic code to study plasma conditions as close to reality as possible, this software has been designed to work by default with output files from the hydrodynamic code FLASH, making the tool user-friendly and accessible to the general plasma physics community. The model has been built into a standalone app, which can be installed by anyone with access to the MATLAB runtime installer and is available upon request to the authors

    Tranilast increases vasodilator response to acetylcholine in rat mesenteric resistance arteries through increased EDHF participation

    Background and Purpose: Tranilast, in addition to its capacity to inhibit mast cell degranulation, has other biological effects, including inhibition of reactive oxygen species, cytokines, leukotrienes and prostaglandin release. In the current study, we analyzed whether tranilast could alter endothelial function in rat mesenteric resistance arteries (MRA). Experimental Approach: Acetylcholine-induced relaxation was analyzed in MRA (untreated and 1-hour tranilast treatment) from 6 month-old Wistar rats. To assess the possible participation of endothelial nitric oxide or prostanoids, acetylcholineinduced relaxation was analyzed in the presence of L-NAME or indomethacin. The participation of endothelium-derived hyperpolarizing factor (EDHF) in acetylcholine-induced response was analyzed by preincubation with TRAM-34 plus apamin or by precontraction with a high K+ solution. Nitric oxide (NO) and superoxide anion levels were measured, as well as vasomotor responses to NO donor DEA-NO and to large conductance calcium-activated potassium channel opener NS1619. Key Results: Acetylcholine-induced relaxation was greater in tranilast-incubated MRA. Acetylcholine-induced vasodilation was decreased by L-NAME in a similar manner in both experimental groups. Indomethacin did not modify vasodilation. Preincubation with a high K+ solution or TRAM-34 plus apamin reduced the vasodilation to ACh more markedly in tranilastincubated segments. NO and superoxide anion production, and vasodilator responses to DEA-NO or NS1619 remained unmodified in the presence of tranilast. Conclusions and Implications: Tranilast increased the endothelium-dependent relaxation to acetylcholine in rat MRA.     Comparative study of continuous hourly energy consumption forecasting strategies with small data sets to support demand management decisions in buildings

    Producción CientíficaBuildings are one of the largest consumers of electrical energy, making it important to develop different strategies to help to reduce electricity consumption. Building energy consumption forecasting strategies are widely used to support demand management decisions, but these strategies require large data sets to achieve an accurate electric consumption forecast, so they are not commonly used for buildings with a short history of record keeping. Based on this, the objective of this study is to determine, through continuous hourly electricity consumption forecasting strategies, the amount of data needed to achieve an accurate forecast. The proposed forecasting strategies were evaluated with Random Forest, eXtreme Gradient Boost, Convolutional Neural Network, and Temporal Convolutional Network algorithms using 4 years of electricity consumption data from two buildings located on the campus of the University of Valladolid. For performance evaluation, two scenarios were proposed for each of the proposed forecasting strategies. The results showed that for forecasting horizons of 1 week, it was possible to obtain a mean absolute percentage error (MAPE) below 7% for Building 1 and a MAPE below 10% for Building 2 with 6 months of data, while for a forecast horizon of 1 month, it was possible to obtain a MAPE below 10% for Building 1 and below 11% for Building 2 with 10 months of data. However, if the distribution of the data captured in the buildings does not undergo sudden changes, the decision tree algorithms obtain better results.     TRESK channel contribution to nociceptive sensory neurons excitability: modulation by nerve injury

    <p>Abstract</p> <p>Background</p> <p>Neuronal hyperexcitability is a crucial phenomenon underlying spontaneous and evoked pain. In invertebrate nociceptors, the S-type leak K<sup>+ </sup>channel (analogous to TREK-1 in mammals) plays a critical role of in determining neuronal excitability following nerve injury. Few data are available on the role of leak K<sub>2P </sub>channels after peripheral axotomy in mammals.</p> <p>Results</p> <p>Here we describe that rat sciatic nerve axotomy induces hyperexcitability of L4-L5 DRG sensory neurons and decreases TRESK (K2P18.1) expression, a channel with a major contribution to total leak current in DRGs. While the expression of other channels from the same family did not significantly change, injury markers ATF3 and Cacna2d1 were highly upregulated. Similarly, acute sensory neuron dissociation (<it>in vitro </it>axotomy) produced marked hyperexcitability and similar total background currents compared with neurons injured <it>in vivo</it>. In addition, the sanshool derivative IBA, which blocked TRESK currents in transfected HEK293 cells and DRGs, increased intracellular calcium in 49% of DRG neurons in culture. Most IBA-responding neurons (71%) also responded to the TRPV1 agonist capsaicin, indicating that they were nociceptors. Additional evidence of a biological role of TRESK channels was provided by behavioral evidence of pain (flinching and licking), in vivo electrophysiological evidence of C-nociceptor activation following IBA injection in the rat hindpaw, and increased sensitivity to painful pressure after TRESK knockdown in vivo.</p> <p>Conclusions</p> <p>In summary, our results clearly support an important role of TRESK channels in determining neuronal excitability in specific DRG neurons subpopulations, and show that axonal injury down-regulates TRESK channels, therefore contributing to neuronal hyperexcitability.</p

    Ionization disequilibrium in K- and L-shell ions

    Producción CientíficaTime-gated Sc K-shell and Ge L-shell spectra are presented from a range of characterized thermodynamic states spanning ion densities of 1e19-1e20cm-3 and plasma temperatures around 2000eV. For the higher densities studied and temperatures from 1000 to 3000 eV, the Sc and Ge x-ray emission spectra are consistent with steady-state calculations from the modern atomic kinetics model SCRAM. At the lower ion densities achieved through plasma expansion, however, the model calculations require a higher plasma temperature to reproduce the observed Ge spectrum. We attribute this to ionization disequilibrium of the Sc because the ionization time scales exceed the hydrodynamic timescale when the inferred temperatures diverge.This work has been supported by the Research Grant No. PID2019-108764RB-I00 from the Spanish Ministry of Science and Innovation
