Identification of a Novel Self-Sufficient Styrene Monooxygenase from Rhodococcus opacus 1CP.
Sequence analysis of a 9-kb genomic fragment of the actinobacterium Rhodococcus opacus 1CP led to identification of an open reading frame encoding a novel fusion protein, StyA2B, with a putative function in styrene metabolism via styrene oxide and phenylacetic acid. Gene cluster analysis indicated that the highly related fusion proteins of Nocardia farcinica IFM10152 and Arthrobacter aurescens TC1 are involved in a similar physiological process. Whereas 413 amino acids of the N terminus of StyA2B are highly similar to those of the oxygenases of two-component styrene monooxygenases (SMOs) from pseudomonads, the residual 160 amino acids of the C terminus show significant homology to the flavin reductases of these systems. Cloning and functional expression of His10-StyA2B revealed for the first time that the fusion protein does in fact catalyze two separate reactions. Strictly NADH-dependent reduction of flavins and highly enantioselective oxygenation of styrene to (S)-styrene oxide were shown. Inhibition studies and photometric analysis of recombinant StyA2B indicated the absence of tightly bound heme and flavin cofactors in this self-sufficient monooxygenase. StyA2B oxygenates a spectrum of aromatic compounds similar to those of two-component SMOs. However, the specific activities of the flavin-reducing and styrene-oxidizing functions of StyA2B are one to two orders of magnitude lower than those of StyA/StyB from Pseudomonas sp. strain VLB120
Integrated systems for rapid point of care (PoC) blood cell analysis
Counting the different subpopulations of cells in a fingerprick of human blood is important for a number of clinical point-of-care (PoC) applications. It is a challenge to demonstrate the integration of sample preparation and detection techniques in a single platform. In this paper we demonstrate a generic microfluidic platform that combines sample processing and characterisation and enumeration in a single, integrated system. Results of microfluidic 3-part differential leukocyte (granulocyte, lymphocyte and monocyte) counts, together with erythrocyte and thrombocyte (platelet) counts, in human blood are shown and corroborated with results from hospital clinical laboratory analysis
Automating Agential Reasoning: Proof-Calculi and Syntactic Decidability for STIT Logics
This work provides proof-search algorithms and automated counter-model extraction for a class of STIT logics. With this, we answer an open problem concerning syntactic decision procedures and cut-free calculi for STIT logics. A new class of cut-free complete labelled sequent calculi G3LdmL^m_n, for multi-agent STIT with at most n-many choices, is introduced. We refine the calculi G3LdmL^m_n through the use of propagation rules and demonstrate the admissibility of their structural rules, resulting in auxiliary calculi Ldm^m_nL. In the single-agent case, we show that the refined calculi Ldm^m_nL derive theorems within a restricted class of (forestlike) sequents, allowing us to provide proof-search algorithms that decide single-agent STIT logics. Rodent herpesvirus Peru encodes a secreted chemokine decoy receptor
Viruses have long been studied not only for their pathology and associated disease but also as model systems for understanding cellular and immunological processes. Rodent herpesvirus Peru (RHVP) is a recently characterized rhadinovirus related to murine gammaherpesvirus 68 (MHV68) and Kaposi's sarcoma-associated herpesvirus (KSHV) that establishes acute and latent infection in laboratory mice. RHVP encodes numerous unique proteins that we hypothesize might facilitate host immune evasion during infection. We report here that open reading frame (ORF) R17 encodes a high-affinity chemokine binding protein that broadly recognizes human and murine CC and C chemokines. The interaction of R17 with chemokines is generally characterized by rapid association kinetics, and in the case of CCL3, CCL4, CCL5, CCL24, and XCL1, extremely stable complexes are formed. Functionally, R17 potently inhibited CCL2-driven chemotaxis of the human monocytic cell line THP-1, CCL3-driven chemotaxis of peripheral blood mononuclear cells, and CCL2-mediated calcium flux. Our studies also reveal that R17 binds to glycosaminoglycans (GAGs) in a process dependent upon two BBXB motifs and that chemokine and GAG binding can occur simultaneously at distinct sites. Collectively, these studies suggest that R17 may play a role in RHVP immune evasion through the targeted sabotage of chemokine-mediated immune surveillance
A randomised feasibility study to investigate the impact of education and the addition of prompts on the sedentary behaviour of office workers
Abstract Background Office workers have been identified as being at risk of accumulating high amounts of sedentary time in prolonged events during work hours, which has been associated with increased risk of a number of long-term health conditions. There is some evidence that providing advice to stand at regular intervals during the working day, and using computer-based prompts, can reduce sedentary behaviour in office workers. However, evidence of effectiveness, feasibility and acceptability for these types of intervention is currently limited. Methods A 2-arm, parallel group, cluster-randomised feasibility trial to assess the acceptability of prompts to break up sedentary behaviour was conducted with office workers in a commercial bank (n = 21). Participants were assigned to an education only group (EG) or prompt and education group (PG). Both groups received education on reducing and breaking up sitting at work, and the PG also received hourly prompts, delivered by Microsoft Outlook over 10 weeks, reminding them to stand. Objective measurements of sedentary behaviour were made using activPAL monitors worn at three time points: baseline, in the last 2 weeks of the intervention period and 12 weeks after the intervention. Focus groups were conducted to explore the acceptability of the intervention and the motivations and barriers to changing sedentary behaviour. Results Randomly generated, customised prompts, delivered by Microsoft Outlook, with messages about breaking up sitting, proved to be a feasible and acceptable way of delivering prompts to office workers. Participants in both groups reduced their sitting, but changes were not maintained at follow-up. The education session seemed to increase outcome expectations of the benefits of changing sedentary behaviour and promote self-regulation of behaviour in some participants. However, low self-efficacy and a desire to conform to cultural norms were barriers to changing behaviour. Conclusions Prompts delivered by Microsoft Outlook were a feasible, low-cost way of prompting office workers to break up their sedentary behaviour, although further research is needed to determine whether this has an additional impact on sedentary behaviour, to education alone. The role of cultural norms, and promoting self-efficacy, should be considered in the design of future interventions. Review of Flywheel based Internal Combustion Engine Hybrid Vehicles
Hybrid vehicles of different configurations and utilizing different energy storage systems have existed in development for many decades and more recently in limited production. They can be grouped as parallel, series or complex hybrids. Another classification is micro, mild and full hybrids which makes the distinction on the basis of functionality. The common energy storage systems in hybrid vehicles are batteries, supercapacitors and high speed flywheels. This paper aims to review a specific type of hybrid vehicle which involves the internal combustion engine (ICE) as the prime mover and the high speed flywheel as an energy storage device. Such hybrids are now attracting considerable interest given their potential for low cost. It is hence timely to produce a review of research and development in this subject. The flywheel is coupled to the drive line with a continuous variable transmission (CVT). The CVT can be of various types such as electrical, hydraulic or mechanical but usually in this case it is a non-electrical one. Different configurations are possible and the paper provides a timeline of the development of such powertrains with various examples. These types of hybrid vehicles have existed as prototypes for many decades and the authors believe that their development has reached levels where they can be considered serious contenders for production vehicles
Stabilization of mesoporous nanocrystalline zirconia with Laponite
The mesoporous nanocrystalline zircoina was synthesized via solid state reaction-structure directing method in the presence of Laponite. The introduction of Laponite renders the higher thermal stability and lamellar track to the zirconia. Laponite acts as inhibitor for crystal growth and also hard template for the mesostructure. The role of Laponite is attributed to the interaction between the zirconia precursors and the nano-platelets of Laponite via the bridge of hydrophilic segments of surfactant. It results in the formation of Zr-O-Mg-O-Si frameworks in the direction of Laponite layer with the condensation of frameworks during the calcination process, which contributes the higher stability and lamellar structure to the nano-sized zirconia samples
The combination of sibling victimization and parental child maltreatment on mental health problems and delinquency
This study examined how the combination of sibling victimization and parental child maltreatment is related to mental health problems and delinquency in childhood and adolescence. Co-occurrence, additive associations, and interactive associations of sibling victimization and parental child maltreatment were investigated using a sample of 2,053 children aged 5–17 years from the National Survey of Children’s Exposure to Violence. The results provide primarily evidence for additive associations and only suggest some co-occurrence and interactive associations of sibling victimization and child maltreatment. Evidence for co-occurrence was weak and, when controlling for the other type of maltreatment, only found for neglect. Sibling victimization was related to more mental health problems and delinquency over and above the effect of child abuse and neglect. Moderation by sibling victimization depended on child age and was only found for the relation between both types of child maltreatment by parents and delinquency. For mental health, no interactive associations were found. Hyperbaric oxygen therapy for long COVID:a prospective registry
A potential beneficial effect of hyperbaric oxygen therapy (HBOT) on complaints of long COVID was found, leading to increased demand for this treatment despite many remaining clinical questions and lack of formal guideline recommendations and reimbursement. A registry was set up in order to gain more insight into patient characteristics and (long-term) outcomes of long COVID patients undergoing HBOT. Patient-reported outcome measures were collected at baseline, after treatment, at 3-month and 1-year follow up. This publication includes the 3-month results. The primary outcome measures were the mental and physical component score (MCS/PCS) of the SF-36 questionnaire. A clinically relevant positive or negative response was defined as a ≥ 10 point increase or decrease of in MCS and/or PCS after 3 months. Secondary outcomes included the EQ-5D, severity of complaints and ability to work. In this prospective registry, 56–63% of long term-ill patients had a clinically relevant improvement in MCS/PCS 3 months after HBOT. However, 13–19% of the patients had a clinically relevant deterioration in MCS/PCS. Symptoms that showed most improvement were predominantly in the cognitive domain. This indicates that HBOT may have a positive effect on complaints of long COVID, but alertness for worsening of the condition should be exercised.</p
