    Diarylureas: New Promising Small Molecules against Streptococcus mutans for the Treatment of Dental Caries

    Dental caries is a biofilm-mediated disease that represents a worldwide oral health issue. Streptococcus mutans has been ascertained as the main cariogenic pathogen responsible for human dental caries, with a high ability to form biofilms, regulated by the quorum sensing. Diarylureas represent a class of organic compounds that show numerous biological activities, including the antimicrobial one. Two small molecules belonging to this class, specifically to diphenylureas, BPU (1,3-bis[3,5-bis(trifluoromethyl)phenyl]urea) and DMTU (1,3-di-m-tolyl-urea), showed interesting results in studies regarding the antimicrobial activity against the cariogenic bacterium S. mutans. Since there are not many antimicrobials used for the prevention and treatment of caries, further studies on these two interesting compounds and other diarylureas against S. mutans may be useful to design new effective agents for the treatment of caries with generally low cytotoxicit

    Hepatitis E Virus Occurrence in Pigs Slaughtered in Italy

    In Europe, foodborne transmission has been clearly associated to sporadic cases and small clusters of hepatitis E in humans linked to the consumption of contaminated pig liver sausages, raw venison, or undercooked wild boar meat. In Europe, zoonotic HEV-genotype 3 strains are widespread in pig farms but little information is available on the prevalence of HEV positive pigs at slaughterhouse. In the present study, the prevalence of HEV-RNA positive pigs was assessed on 585 animals from 4 abattoirs located across Italy. Twenty-one pigs (3.6%) tested positive for HEV in either feces or liver by real-time RT-PCR. In these 21 pigs, eight diaphragm muscles resulted positive for HEV-RNA. Among animals collected in one abattoir, 4 out of 91 plasma tested positive for HEV-RNA. ELISA tests for the detection of total antibodies against HEV showed a high seroprevalence (76.8%), confirming the frequent exposure of pigs to the virus. The phylogenetic analyses conducted on sequences of both ORF1 and ORF2 fragments, shows the circulation of HEV-3c and of a novel unclassified subtype. This study provides information on HEV occurrence in pigs at the slaughterhouse, confirming that muscles are rarely contaminated by HEV-RNA compared to liver, which is the most frequently positive for HEV

    Recently, motion tracking techniques to acquire human body movements have broadened their range of applications from sports training, to rehabilitation, to ergonomics issues. For industrial applications, motion capture systems are largely used to acquire workers movements to improve working conditions, comfort, safety, etc. Several tracking solutions have been provided to analyze trunk and upper limbs movement based on different sensing technologies such as optical systems, audio systems, radar systems, magnetic systems, inertial systems and mechanical motion systems. Motion tracking with inertial sensors has been an active research area due to its several advantages. In this paper the authors introduce a motion tracking method of arbitrary human upper body motion. Low cost wearable inertial sensors and CPU unit are used in the proposed approach to track the upper body movement in 3D space and in real time

    Complexes of Ruthenium(II) as Promising Dual-Active Agents against Cancer and Viral Infections

    : Poor responses to medical care and the failure of pharmacological treatment for many high-frequency diseases, such as cancer and viral infections, have been widely documented. In this context, numerous metal-based substances, including cisplatin, auranofin, various gold metallodrugs, and ruthenium complexes, are under study as possible anticancer and antiviral agents. The two Ru(III) and Ru(II) complexes, namely, BOLD-100 and RAPTA-C, are presently being studied in a clinical trial and preclinical studies evaluation, respectively, as anticancer agents. Interestingly, BOLD-100 has also recently demonstrated antiviral activity against SARS-CoV-2, which is the virus responsible for the COVID-19 pandemic. Over the last years, much effort has been dedicated to discovering new dual anticancer-antiviral agents. Ru-based complexes could be very suitable in this respect. Thus, this review focuses on the most recent studies regarding newly synthesized Ru(II) complexes for use as anticancer and/or antiviral agents

    Implantable device-related endocarditis detected by point-of-care echocardiography in a patient with dyspnea and chest pain

    We describe a case of a 70-year-old man who presented to the emergency department with dyspnea and chest pain. He had history of atrial fibrillation, chronic obstructive pulmonary disease, prosthetic mitral valve and pacemaker implantation. He had a sepsis related to multiple bilateral septic emboli pointed out by chest computed tomography scan. Point of care echocardiography showed a pacemaker-related endocarditis with large vegetation on the right ventricular lead. The patient started empiric antibiotic therapy and he was addressed to complete hardware removal

    Nine-Year Nationwide Environmental Surveillance of Hepatitis E Virus in Urban Wastewaters in Italy (2011–2019)

    Hepatitis E virus (HEV) is an emerging causative agent of acute hepatitis worldwide. To provide insights into the epidemiology of HEV in Italy, a large-scale investigation was conducted into urban sewage over nine years (2011&ndash;2019), collecting 1374 sewage samples from 48 wastewater treatment plants located in all the 20 regions of Italy. Broadly reactive primers targeting the ORF1 and ORF2 regions were used for the detection and typing of HEV, followed by Sanger and next generation sequencing (NGS). Real-time RT-qPCR was also used to attempt quantification of positive samples. HEV RNA detection occurred in 74 urban sewage samples (5.4%), with a statistically significant higher frequency (7.1%) in central Italy. Fifty-six samples were characterized as G3 strains and 18 as G1. While the detection of G3 strains occurred in all the surveillance period, G1 strains were mainly detected in 2011&ndash;2012, and never in 2017&ndash;2019. Typing was achieved in 2 samples (3f subtype). Viral concentrations in quantifiable samples ranged from 1.2 &times; 103 g.c./L to 2.8 &times; 104 g.c./L. Our results suggest the considerable circulation of the virus in the Italian population, despite a relatively small number of notified cases, a higher occurrence in central Italy, and a noteworthy predominance of G3 strains

    Ozone affects pollen viability and NAD(P)H oxidase release from Ambrosia artemisiifolia pollen

    Air pollution is frequently proposed as a cause of the increased incidence of allergy in industrialised countries. We investigated the impact of ozone (O3) on reactive oxygen species (ROS) and allergen content of ragweed pollen (Ambrosia artemisiifolia). Pollen was exposed to acute O3 fumigation, with analysis of pollen viability, ROS and nitric oxide (NO) content, activity of nicotinamide adenine dinucleotide phosphate (NAD[P]H) oxidase, and expression of major allergens. There was decreased pollen viability after O3 fumigation, which indicates damage to the pollen membrane system, although the ROS and NO contents were not changed or were only slightly induced, respectively. Ozone exposure induced a significant enhancement of the ROS-generating enzyme NAD(P)H oxidase. The expression of the allergen Amb a 1 was not affected by O3, determined from the mRNA levels of the major allergens. We conclude that O3 can increase ragweed pollen allergenicity through stimulation of ROS-generating NAD(P)H oxidase

    <i>Lavandula x intermedia</i> and <i>Lavandula angustifolia</i> essential oils: phytochemical composition and antimicrobial activity against foodborne pathogens

    <p>Four cultivars (cv) of <i>Lavandula x intermedia</i> (‘Abrialis’, ‘Alba’, ‘Rinaldi Ceroni’ (R.C.) and ‘Sumiens’) were cultivated in Italy and their essential oils (EOs) were distilled from Alfalfa Mosaic Virus-free plants. These EOs and one from <i>L. angustifolia</i> Miller were chemically characterised by GC-MS and GC-FID. Antimicrobial activity was evaluated against <i>Listeria monocytogenes</i> (24 strains) and <i>Salmonella enterica</i> (10 food strains). Minimal inhibitory concentrations (MIC) ≥ 10.0 μL/mL inhibited <i>Salmonella</i> (cv ‘R.C.’ was the most active); MIC of 0.3 μL/mL for cv ‘Abrialis’ and ‘R.C.’ inhibited <i>L. monocytogenes</i>, revealing noticeable activity, especially on clinical strains. This activity appears related to EOs composition. Particularly cv ‘Abrialis’ and ‘R.C.’ showing the highest antimicrobial activity, were rich in the specific constituents: linalool (38.17 and 61.98%), camphor (8.97 and 10.30%), 1,8-cineole (6.89 and 8.11%, respectively). These EOs could find potential applications in food biopreservation and in surface decontamination, even in hospitals, and deserve deeper investigations.</p
