    Observations Save the Day at the Wachovia Cultural Center Excavation

    In the observational method, gaps in the available information are filled by observations. Such observations were invaluable during the construction of a 54-story building with an 8-story underground parking deck in Charlotte, North Carolina, which required an excavation nearly 100-feet deep into rock. Cracks appeared in an adjacent brick office building by the time the north side of the excavation had reached a depth of about 40 feet. Blast-induced settlement was initially suspected. As excavation continued, some soilfilled weathered joints, seams, and fractures in the rock sidewalls were observed, and displacement of the cracks increased. Inclinometer casings were installed along the street bordering the north sidewall. After increasing deformation, the casings began to shear at a depth of about 47 feet. A clay-filled seam dipping into the excavation at an angle of 10 to 15 degrees was discovered in the north sidewall. A row of grouted No. 18 bars was installed in drill holes angled across the clay seam. After analysis of a sliding block model revealed the need for additional capacity, a row of shear pins was installed in the street behind the north sidewall. No further movement was observed

    Walking the Talk: A Multistakeholder Exploration of Organizational Authenticity, Employee Productivity, and Post-Merger Performance

    Does consistency between how a firm treats employees (what it does) and its espoused employee-oriented values (what it says) affect employee productivity? Furthermore, given that the stakeholder theory perspective holds that what happens to one stakeholder influences other stakeholders, does this sort of consistency vis-à-vis a firm’s customers also influence employee productivity? We empirically investigate the influence of organizational authenticity—defined as consistency between a firm’s espoused values and realized practices—in the context of a merger, and specifically during post-merger integration. Our findings show that a lack of organizational authenticity in terms of both under-promising and over-promising to both employees and customers is associated with lower productivity, which in turn is related to long-term merger performance, thus affecting outcomes for shareholders. These findings support the importance of authenticity and should therefore be of interest to executives responsible for ensuring the consistency between what a firm says and what it does, as well as those who participate in and study the merger integration process. In particular, we propose stakeholder theory as a helpful lens for examining the merger integration process as well as other joint actions such as strategic alliances

    Evaluation of the Family Start programme: Report on findings of the impact evaluation

    The purpose of this impact evaluation was to assess the impact that Family Start participation is having on the wellbeing of New Zealand children and their whānau. The evaluation examined a range of health, educational, and social outcomes for children, including separate analyses for Māori and Pasifika children. The evaluation was informed by a Bridging Cultural Perspectives approach which comprises the He Awa Whiria and Negotiated Spaces models. Reviewers representing the three knowledge streams (Māori, Pasifika, Pākehā) worked together to assist with the interpretation of the findings at an aggregate level, and through the lens of each worldview

    Computational approaches for understanding the diagnosis and treatment of Parkinson's disease

    This study describes how the application of evolutionary algorithms (EAs) can be used to study motor function in humans with Parkinson’s disease (PD) and in animal models of PD. Human data is obtained using commercially available sensors via a range of non-invasive procedures that follow conventional clinical practice. EAs can then be used to classify human data for a range of uses, including diagnosis and disease monitoring. New results are presented that demonstrate how EAs can also be used to classify fruit flies with and without genetic mutations that cause Parkinson’s by using measurements of the proboscis extension reflex. The case is made for a computational approach that can be applied across human and animal studies of PD and lays the way for evaluation of existing and new drug therapies in a truly objective way

    Developmental language disorders and risk of recidivism among young offenders

    Background Although factors such as adverse family background have been widely examined, little is known about the prevalence or potential impact of developmental language disorder (DLD) on risk of recidivism in young people with history of criminal justice system contact. Methods A total of 145 young offenders participated. An adversity score was constructed based on information found in youth justice service records. Data collected included standardised measures of expressive and receptive language, nonverbal IQ and the inventory of callous–unemotional traits. Survival analysis was performed to examine differences in reoffending risk between young offenders with and without DLD. Results The cumulative incidence of reoffending within a year of the young person's court order was markedly raised in the DLD group (62%; 95% CI 52, 72) versus the non‐DLD group (25%; 95% CI 16, 39). Furthermore, in the final multivariable survival analysis the independent elevation in risk linked with DLD was not greatly attenuated with adjustment for nonverbal IQ, adversity score, age at first offence, number of previous offences and deprivation score. DLD was the most significant predictor with an adjusted hazard ratio of 2.61 (95% CI 1.80, 3.78). Conclusions Young offenders with DLD are more than twice as likely to reoffend than their unaffected offending peers. DLD is a powerful predictor of recidivism above and beyond other known risk factors

    Influence of corners in excavations on damage assessment

    This paper provides guidance on quantifying the extent of corner effects in excavations and their impact on damage assessment. The corner effects’ extent is of great importance in making early decisions during project planning and preliminary design, particularly in relation to stakeholder engagement and placement of instruments. By using empirical relations, one is able to provide an equation, validated against the literature and additional numerical models, for estimating the extent of corner effects for a particular excavation geometry. Furthermore, two more equations for quantifying the damage of excavations to adjacent structures are presented and validated against two case studies in the literature. The proposed equations are also useful in the context of early stages of project development. Finally, a simple study shows the different effects of corners in sections parallel and perpendicular to a retaining wall. This highlights that corner effects may actually induce additional damage due to the introduction of a movement gradient, as opposed to the common previous perception that assumed that they were always conservative as they reduced absolute movements

    Methylation matters: binding of Ets-1 to the demethylated Foxp3 gene contributes to the stabilization of Foxp3 expression in regulatory T cells

    The forkhead-box protein P3 (Foxp3) is a key transcription factor for the development and suppressive activity of regulatory T cells (Tregs), a T cell subset critically involved in the maintenance of self-tolerance and prevention of over-shooting immune responses. However, the transcriptional regulation of Foxp3 expression remains incompletely understood. We have previously shown that epigenetic modifications in the CpG-rich Treg-specific demethylated region (TSDR) in the Foxp3 locus are associated with stable Foxp3 expression. We now demonstrate that the methylation state of the CpG motifs within the TSDR controls its transcriptional activity rather than a Treg-specific transcription factor network. By systematically mutating every CpG motif within the TSDR, we could identify four CpG motifs, which are critically determining the transcriptional activity of the TSDR and which serve as binding sites for essential transcription factors, such as CREB/ATF and NF-κB, which have previously been shown to bind to this element. The transcription factor Ets-1 was here identified as an additional molecular player that specifically binds to the TSDR in a demethylation-dependent manner in vitro. Disruption of the Ets-1 binding sites within the TSDR drastically reduced its transcriptional enhancer activity. In addition, we found Ets-1 bound to the demethylated TSDR in ex vivo isolated Tregs, but not to the methylated TSDR in conventional CD4+ T cells. We therefore propose that Ets-1 is part of a larger protein complex, which binds to the TSDR only in its demethylated state, thereby restricting stable Foxp3 expression to the Treg lineage

    The Role of Purported Mucoprotectants in Dealing with Irritable Bowel Syndrome, Functional Diarrhea, and Other Chronic Diarrheal Disorders in Adults

    Chronic diarrhea is a frequent presenting symptom, both in primary care medicine and in specialized gastroenterology units. It is estimated that more than 5% of the global population suffers from chronic diarrhea. and that about 40% of these subjects are older than 60 years. The clinician is frequently faced with the need to decide which is the best therapeutic approach for these patients. While the origin of chronic diarrhea is diverse, impairment of intestinal barrier function, dysbiosis. and mucosal micro-inflammation are being increasingly recognized as underlying phenomena characterizing a variety of chronic diarrheal diseases. In addition to current pharmacological therapies, there is growing interest in alternative products such as mucoprotectants, which form a mucoadhesive film over the epithelium to reduce and protect against the development of altered intestinal permeability, dysbiosis, and mucosal micro-inflammation. This manuscript focuses on chronic diarrhea in adults, and we will review recent evidence on the ability of these natural compounds to improve symptoms associated with chronic diarrhea and to exert protective effects for the intestinal barrier
