    Secular stagnation and the demographic transition: a factor model approach

    Secular stagnation is a tale of ageing economies permanently constrained in a below full-employment steady-state due to aggregate demand shortfalls. Using a factor model combined with a VAR for seven key macro-series augmented with demographics, we investigate the link between the demographic transition and macroeconomic trends. We find that, while a single variable offers an insufficient representation of the recent population dynamics, two factors can account for almost the entire variance of the five variables that best describe the demographic transition. Our results indicate that demographics have exerted significant deflationary and contractionary pressures in the Euro Area and Japan since 1970

    XII Expedição Científica do Departamento de Biologia - Pico 2005.Uma breve caracterização da flora vascular da ilha do Pico é feita a partir da análise do seu catálogo de plantas vasculares (Pereira et al., 2006). A flora vascular Picoense fora de cultivo compreende no momento 642 unidades taxonómicas diferentes (taxa) representadas por 634 espécies, 375 géneros e 118 famílias. O número de taxa introduzidos representa 70,5% do total dos taxa presentes, enquanto a percentagem de taxa nativos se cifra apenas pelos 26,0%. No entanto 43,1% dos taxa nativos que ocorrem no Pico são endémicos dos Açores ou da Macaronésia. A distribuição das espécies nos grandes grupos taxonómicos (Pteridophyta, Gymnospermae, Dicotyledoneae e Monocotyledoneae) difere com significado estatístico entre as espécies nativas e introduzidas. A contribuição das espécies introduzidas é maior a nível das dicotiledóneas e menor a nível dos pteridófitos quando comparada com a distribuição das espécies nativas.ABSTRACT: A short characterization of the vascular flora of Pico Island is made from the analysis of it’s checklist of vascular plants (Pereira et al., 2006). The flora of Pico Island is at the moment represented by 642 different taxa, 634 species, 375 genera and 118 families. The number of introduced species represents 70,5% of the total species while the native species correspond only to 26,2 %. About 43,1% of the native taxa are endemic to Azores or Macaronesia. The species distribution on the main taxonomic groups (Pteridophyta, Gymnospermae, Dicotyledoneae e Monocotyledoneae) is significantly different between native and introduced species.     X-linked Adrenal Hypoplasia Congenita: report of two families and a new NR0B1 mutation

    Background: X-linked Adrenal Hypoplasia Congenita (X-linked AHC) tipically manifests as adrenal insufficiency in a bimodal clinical presentation (5–60 days and 2–13 years) and, at pubertal age, hypogonadotropic hypogonadism (HH) in males. It is caused by mutations in NR0B1 (present in Xp21.2), a gene with a critical role in the development of adrenals and hypothalamic-pituitary-gonadal (HPG) axis. It represents ~1% of all causes of primary adrenal insufficiency under age 18. We present two kindreds with NR0B1 mutations, one with a previously unreported mutation. Clinical case - family A: The proband presented with adrenal crisis at day 18 of life. Family history was relevant for an uncle death as a newborn. Hormonal assays revealed elevated ACTH (34,5 pmol/L, reference: T, leading to a premature stop codon, p.Lys362*, in exon 1. His mother and sister were both asymptomatic carriers for this mutation. This mutation, not described previously, leads probably to a non-functional truncated protein. Clinical case - family B: Kindred B had two males who presented with adrenal crisis at the newborn age (proband: 14 days; brother: 16 days). At 6 months of age, the proband had elevated ACTH (193.2 pmol/L, reference: G; p.Tyr81*, in exon 1, present in the two affected males. His mother and one sister were also asymptomatic carriers. Conclusion: Our kindreds highlights the clinical aspects of the rare X-linked AHC and its challenging differential diagnosis. The diagnosis was based on …. Completar … and an apparently X-linked mode of inheritance. Molecular analysis of the NR0B1 gene allowed the identification of the molecular defect in both families. Two different nonsense mutation in exon 1 were identified, giving rise most probably to truncated non-functional proteins, confirming the disease severity or the clinical phenotype of the affected boys. Mutation identification was relevant for genetic counseling of the parents, as well as for the patient’s sisters

    A review on lipase immobilization on hydrophobic supports incorporating systematic mapping principles

    A review of the literature covering research on the immobilization of lipases on hydrophobic supports was performed using systematic mapping (SM) concepts. This approach consists of a rigorous review methodology used to catalog evidence, identify gaps at the frontier of knowledge, unknown trends, and research groups. Our results showed a wide variety of available lipases, including commercial, wild-type and recombinant strains. However, the most commonly used lipases are lipases from Thermomyces lanuginosus (TLL), Candida rugosa (CRL) or Rhizomucor miehei (RML) and lipase B from Candida antarctica (CALB). A wide variety of supports with different degrees of hydrophobicity were identified and the supports activated with a layer of octyl or octadecyl groups were the most used. The advantages of lipase immobilization on these supports were discussed. Among them, the immobilization, purification, stabilization and hyperactivation of lipases in a single step stand up. Moreover, problems related to lipase immobilization by interfacial activation were highlighted (mainly enzyme release). Strategies to overcome these problems include immobilization on heterofunctional supports or intermolecular crosslinking of enzymes immobilized by physical and/or chemical agents. The possibility of increasing the capacity of supports by lipase multilayer immobilization was also discussed.     Developing tools for the e-learning platform mathE

    MathE (mathe.pixel-online.org) is an e-learning platform for higher education developed and implemented by a consortium of seven institutional partners from five European countries. The aim of the project is to enhance the quality of teaching and improve pedagogies and assessment methods by facilitating the identification of students’ gaps in Math, providing appropriate digital tools and promoting self-evaluation with immediate feedback. The Polytechnic Institute of Bragança (IPB), in Portugal, is one of the consortium members: sixteen of its teachers collaborate in the development of this platform, being responsible for thirteen of the topics/subtopics in which the platform is structured. Such topics cover a wide range of contents, from linear transformations to integration, from graph theory to probabilities. The articulation of the topics of the MathE collection corresponds to the canonic mathematics content of engineering, business and education degrees. The MathE platform is organized into three main sections: Student´s Assessment, MathE Library and Community of Practice. So far, IPB has already developed a collection of around 800 questions for the student´s assessment section and is currently developing the MathE Library. More than 350 students from IPB are using the MathE platform; some offered as volunteers, whose role is testing the behavior of the platform as well as looking for bugs and other details that require improvement, while others are already using the platform in their study.     Reuma.pt/vasculitis - the Portuguese vasculitis registry

    BACKGROUND: The vasculitides are a group of rare diseases with different manifestations and outcomes. New therapeutic options have led to the need for long-term registries. The Rheumatic Diseases Portuguese Register, Reuma.pt, is a web-based electronic clinical record, created in 2008, which currently includes specific modules for 12 diseases and > 20,000 patients registered from 79 rheumatology centres. On October 2014, a dedicated module for vasculitis was created as part of the European Vasculitis Society collaborative network, enabling prospective collection and central storage of encrypted data from patients with this condition. All Portuguese rheumatology centres were invited to participate. Data regarding demographics, diagnosis, classification criteria, assessment tools, and treatment were collected. We aim to describe the structure of Reuma.pt/vasculitis and characterize the patients registered since its development. RESULTS: A total of 687 patients, with 1945 visits, from 13 centres were registered; mean age was 53.4 ± 19.3 years at last visit and 68.7% were females. The most common diagnoses were Behçet's disease (BD) (42.5%) and giant cell arteritis (GCA) (17.8%). Patients with BD met the International Study Group criteria and the International Criteria for BD in 85.3 and 97.2% of cases, respectively. Within the most common small- and medium-vessel vasculitides registered, median [interquartile range] Birmingham Vasculitis Activity Score (BVAS) at first visit was highest in patients with ANCA-associated vasculitis (AAV) (17.0 [12.0]); there were no differences in the proportion of patients with AAV or polyarteritis nodosa who relapsed (BVAS≥1) or had a major relapse (≥1 major BVAS item) during prospective assessment (p = 1.00, p = 0.479). Biologic treatment was prescribed in 0.8% of patients with GCA, 26.7% of patients with AAV, and 7.6% of patients with BD. There were 34 (4.9%) deaths reported. CONCLUSIONS: Reuma.pt/vasculitis is a bespoke web-based registry adapted for routine care of patients with this form of rare and complex diseases, allowing an efficient data-repository at a national level with the potential to link with other international databases.     SARS-CoV-2 introductions and early dynamics of the epidemic in Portugal

    Genomic surveillance of SARS-CoV-2 in Portugal was rapidly implemented by the National Institute of Health in the early stages of the COVID-19 epidemic, in collaboration with more than 50 laboratories distributed nationwide. Methods By applying recent phylodynamic models that allow integration of individual-based travel history, we reconstructed and characterized the spatio-temporal dynamics of SARSCoV-2 introductions and early dissemination in Portugal. Results We detected at least 277 independent SARS-CoV-2 introductions, mostly from European countries (namely the United Kingdom, Spain, France, Italy, and Switzerland), which were consistent with the countries with the highest connectivity with Portugal. Although most introductions were estimated to have occurred during early March 2020, it is likely that SARS-CoV-2 was silently circulating in Portugal throughout February, before the first cases were confirmed. Conclusions Here we conclude that the earlier implementation of measures could have minimized the number of introductions and subsequent virus expansion in Portugal.     Health-related quality of life in patients with type 1 diabetes mellitus in the different geographical regions of Brazil : data from the Brazilian Type 1 Diabetes Study Group

    Background: In type 1 diabetes mellitus (T1DM) management, enhancing health-related quality of life (HRQoL) is as important as good metabolic control and prevention of secondary complications. This study aims to evaluate possible regional differences in HRQoL, demographic features and clinical characteristics of patients with T1DM in Brazil, a country of continental proportions, as well as investigate which variables could influence the HRQoL of these individuals and contribute to these regional disparities. Methods: This was a retrospective, cross-sectional, multicenter study performed by the Brazilian Type 1 Diabetes Study Group (BrazDiab1SG), by analyzing EuroQol scores from 3005 participants with T1DM, in 28 public clinics, among all geographical regions of Brazil. Data on demography, economic status, chronic complications, glycemic control and lipid profile were also collected. Results: We have found that the North-Northeast region presents a higher index in the assessment of the overall health status (EQ-VAS) compared to the Southeast (74.6 ± 30 and 70.4 ± 19, respectively; p < 0.05). In addition, North- Northeast presented a lower frequency of self-reported anxiety-depression compared to all regions of the country (North-Northeast: 1.53 ± 0.6; Southeast: 1.65 ± 0.7; South: 1.72 ± 0.7; Midwest: 1.67 ± 0.7; p < 0.05). These findings could not be entirely explained by the HbA1c levels or the other variables examined. Conclusions: Our study points to the existence of additional factors not yet evaluated that could be determinant in the HRQoL of people with T1DM and contribute to these regional disparities

    Genome of Herbaspirillum seropedicae Strain SmR1, a Specialized Diazotrophic Endophyte of Tropical Grasses

    The molecular mechanisms of plant recognition, colonization, and nutrient exchange between diazotrophic endophytes and plants are scarcely known. Herbaspirillum seropedicae is an endophytic bacterium capable of colonizing intercellular spaces of grasses such as rice and sugar cane. The genome of H. seropedicae strain SmR1 was sequenced and annotated by The Paraná State Genome Programme—GENOPAR. The genome is composed of a circular chromosome of 5,513,887 bp and contains a total of 4,804 genes. The genome sequence revealed that H. seropedicae is a highly versatile microorganism with capacity to metabolize a wide range of carbon and nitrogen sources and with possession of four distinct terminal oxidases. The genome contains a multitude of protein secretion systems, including type I, type II, type III, type V, and type VI secretion systems, and type IV pili, suggesting a high potential to interact with host plants. H. seropedicae is able to synthesize indole acetic acid as reflected by the four IAA biosynthetic pathways present. A gene coding for ACC deaminase, which may be involved in modulating the associated plant ethylene-signaling pathway, is also present. Genes for hemagglutinins/hemolysins/adhesins were found and may play a role in plant cell surface adhesion. These features may endow H. seropedicae with the ability to establish an endophytic life-style in a large number of plant species
