    Earpiece with sensors to measure/monitor multiple physiological variables

    An apparatus and method for positioning sensors relative to one another and anatomic features in a non-invasive device for measuring and monitoring multiple physiological variables from a single site uses an earpiece incorporating a shielded pulse oximetry sensor (POS) having a miniaturized set of LEDs and photosensors configured for pulse oximetry measurements in the reflectance mode and located in the earpiece so as to position the POS against a rear wall of an ear canal. The earpiece also includes a thermopile of no larger than 7 mm. in diameter located on the earpiece to so as to position the thermopile past a second turn of an external auditory meatus so as to view the tympanic membrane. The thermopile includes a reference temperature sensor attached to its base for ambient temperature compensation

    Experience recruits MSK1 to expand the dynamic range of synapses and enhance cognition

    Experience powerfully influences neuronal function and cognitive performance, but the cellular and molecular events underlying the experience-dependent enhancement of mental ability haveremained elusive. In particular, the mechanisms that couple the external environment to the genomic changes underpinning this improvement are unknown. To address this we have used male mice harbouring an inactivating mutation of mitogen- and stress-activated protein kinase 1 (MSK1), a BDNF-activated enzyme downstream of the MAPK pathway. We show that MSK1 is required for the full extent of experience-induced improvement of spatial memory, for the expansion of the dynamic range of synapses, exemplified by the enhancement of hippocampal LTP and LTD, and for the regulation of the majority of genes influenced by enrichment. In addition, and unexpectedly, we show that experience is associated with an MSK1-dependent downregulation of key MAPK and plasticity related genes, notably of EGR1/Zif268 and Arc/Arg3.1, suggesting the establishment of a novel genomic landscape adapted to experience. By coupling experience to homeostatic changes in gene expression MSK1, represents a prime mechanism through which the external environment has an enduring influence on gene expression, synaptic function and cognition

D.)--Columbia Univeristy.Includes bibliographical references (p. 245-246)."The development of a mass etching program for the recovery of silicified fossils from Permian limestone of west Texas has resulted in the accumulation of a large number of fossil gastropods and has made possible the present systematic study of the pleurotomarian gastropods. Most of the collections used herein were made by, or under the direction of, G. Arthur Cooper, Norman D. Newell, and J. Brookes Knight. These collections were obtained from the Wolfcamp, Leonard, and Word formations of the Glass Mountains of Texas and from the Hueco, Bone Spring, and Cherry Canyon formations of the Sierra Diablo and Guadalupe Mountain region. Comments on stratigraphy are made for the orientation of the reader. Little original stratigraphic work has been done, except to verify or corroborate correlations based on other fossil groups. A few of the paleoecological relationships are discussed, with particular emphasis on molluscan assemblages in the Glass Mountains. An instance is cited of a relationship between a Heliospongia and Glyptotomaria (Glyptotomaria) marginata. Statistical methods used include those based on univariate analysis and on allometric growth line comparisons. This latter type of analysis is based on the relative growth line, as estimated by the reduced major axis. Discrimination was based, in part, on a comparison of slope and position differences of the relative growth line between samples. Studies of the logarithmic spiral show that it can be useful in the study of pleurotomarians and may be treated statistically. The value of the logarithmic spiral formula as a discriminatory character must be weighed against the time and labor required in its derivation. Gross measurements of the spiral, such as the use of suture ratios, tend to reduce expenditure of time, but they also reduce accuracy. Some of the directional and non-directional variations within species and genera are noted. This study attempts to show some aspects of pleurotomarian speciation during the Permian. These are particularly well illustrated in the genus Tapinotomaria. Studies of population structure made it possible to understand some of the changes thought to be responsible for most speciation in the families studied. Emphasis is placed on ontogenetic growth characters. The bulk of this paper is devoted to an analysis and description of the rich and heretofore unknown Permian pleurotomarian faunules. The general increase in knowledge about Permian representatives of the superfamily Pleurotomariacea is briefly discussed. Two families, Portlockiellidae and Phymatopleuridae, are diagnosed and discussed. The family Eotomariidae Wenz, 1938, is redefined and expanded. Thirteen genera are discussed herein: Lamellospira and Lacunospira are new; Eirlysia, Tapinotomaria, Discotomaria, Callitomaria, and Shwedagonia are discussed for the first time (see Batten, 1956). Tapinotomaria and Shwedagonia have been discussed at some length because of their interesting evolutionary developments. Five previously described genera are also included, Euconospira, Spiroscala, and Glyptotomaria are redefined; Phymatopleura and Paragoniozona are discussed.     The Kinase Function of MSK1 Regulates BDNF Signaling to CREB and Basal Synaptic Transmission, But Is Not Required for Hippocampal Long-Term Potentiation or Spatial Memory

    The later stages of long-term potentiation (LTP) in vitro and spatial memory in vivo are believed to depend upon gene transcription. Accordingly, considerable attempts have been made to identify both the mechanisms by which transcription is regulated and indeed the gene products themselves. Previous studies have shown that deletion of one regulator of transcription, the mitogen- and stress-activated kinase 1 (MSK1), causes an impairment of spatial memory. Given the ability of MSK1 to regulate gene expression via the phosphorylation of cAMP response element binding protein (CREB) at serine 133 (S133), MSK1 is a plausible candidate as a prime regulator of transcription underpinning synaptic plasticity and learning and memory. Indeed, prior work has revealed the necessity for MSK1 in homeostatic and experience-dependent synaptic plasticity. However, using a knock-in kinase-dead mouse mutant of MSK1, the current study demonstrates that, while the kinase function of MSK1 is important in regulating the phosphorylation of CREB at S133 and basal synaptic transmission in hippocampal area CA1, it is not required for metabotropic glutamate receptor-dependent long-term depression (mGluR-LTD), two forms of LTP or several forms of spatial learning in the watermaze. These data indicate that other functions of MSK1, such as a structural role for the whole enzyme, may explain previous observations of a role for MSK1 in learning and memory

    Numerical simulations of shocks encountering clumpy regions

    We present numerical simulations of the adiabatic interaction of a shock with a clumpy region containing many individual clouds. Our work incorporates a sub-grid turbulence model which for the first time makes this investigation feasible. We vary the Mach number of the shock, the density contrast of the clouds, and the ratio of total cloud mass to inter-cloud mass within the clumpy region. Cloud material becomes incorporated into the flow. This "mass-loading" reduces the Mach number of the shock, and leads to the formation of a dense shell. In cases in which the mass-loading is sufficient the flow slows enough that the shock degenerates into a wave. The interaction evolves through up to four stages: initially the shock decelerates; then its speed is nearly constant; next the shock accelerates as it leaves the clumpy region; finally it moves at a constant speed close to its initial speed. Turbulence is generated in the post-shock flow as the shock sweeps through the clumpy region. Clouds exposed to turbulence can be destroyed more rapidly than a similar cloud in an "isolated" environment. The lifetime of a downstream cloud decreases with increasing cloud-to-intercloud mass ratio.     MSK1 is required for the beneficial synaptic and cognitive effects of enriched experience across the lifespan

    Positive experiences, such as social interaction, cognitive training and physical exercise, have been shown to ameliorate some of the harms to cognition associated with ageing. Animal models of positive interventions, commonly known as environmental enrichment, strongly influence neuronal morphology and synaptic function and enhance cognitive performance. While the profound structural and functional benefits of enrichment have been appreciated for decades, little is known as to how the environment influences neurons to respond and adapt to these positive sensory experiences. We show that adult and aged male wild-type mice that underwent a 10-week environmental enrichment protocol demonstrated improved performance in a variety of behavioural tasks, including those testing spatial working and spatial reference memory, and an enhancement in hippocampal LTP. Aged animals in particular benefitted from enrichment, performing spatial memory tasks at levels similar to healthy adult mice. Many of these benefits, including in gene expression, were absent in mice with a mutation in an enzyme, MSK1, which is activated by BDNF, a growth factor implicated in rodent and human cognition. We conclude that enrichment is beneficial across the lifespan and that MSK1 is required for the full extent of these experience-induced improvements of cognitive abilities, synaptic plasticity and gene expression

    CREB serine 133 is necessary for spatial cognitive flexibility and long-term potentiation

    The transcription factor cAMP response element binding protein (CREB) is widely regarded as orchestrating the genomic response that underpins a range of physiological functions in the central nervous system, including learning and memory. Of the means by which CREB can be regulated, emphasis has been placed on the phosphorylation of a key serine residue, S133, in the CREB protein, which is required for CREB-mediated transcriptional activation in response to a variety of activity-dependent stimuli. Understanding the role of CREB S133 has been complicated by molecular genetic techniques relying on over-expression of either dominant negative or activating transgenes that may distort the physiological role of endogenous CREB. A more elegant recent approach targeting S133 in the endogenous CREB gene has yielded a mouse with constitutive replacement of this residue with alanine (S133A), but has generated results (no behavioural phenotype and no effect on gene transcription) at odds with contemporary views as to the role of CREB S133, and which may reflect compensatory changes associated with the constitutive mutation. To avoid this potential complication, we generated a post-natal and forebrain-specific CREB S133A mutant in which the expression of the mutation was under the control of CaMKIIα promoter. Using male and female mice we show that CREB S133 is necessary for spatial cognitive flexibility, the regulation of basal synaptic transmission, and for the expression of long-term potentiation (LTP) in hippocampal area CA1. These data point to the importance of CREB S133 in neuronal function, synaptic plasticity and cognition in the mammalian brain

    Experimental Study of the Shortest Reset Word of Random Automata

    In this paper we describe an approach to finding the shortest reset word of a finite synchronizing automaton by using a SAT solver. We use this approach to perform an experimental study of the length of the shortest reset word of a finite synchronizing automaton. The largest automata we considered had 100 states. The results of the experiments allow us to formulate a hypothesis that the length of the shortest reset word of a random finite automaton with nn states and 2 input letters with high probability is sublinear with respect to nn and can be estimated as $1.95 n^{0.55}.

    What are the beliefs, attitudes and practices of front-line staff in long-term care (LTC) facilities related to osteoporosis awareness, management and fracture prevention?

    <p>Abstract</p> <p>Background</p> <p>Compared to the general elderly population, those institutionalized in LTC facilities have the highest prevalence of osteoporosis and subsequently have higher incidences of vertebral and hip fractures. The goal of this study is to determine how well nurses at LTC facilities are educated to properly administer bisphosphonates. A secondary question assessed was the nurse's and PSW's attitudes and beliefs regarding the role and benefits of vitamin D for LTC patients.</p> <p>Methods</p> <p>Eight LTC facilities in Hamilton were surveyed, and all nurses were offered a survey. A total 57 registered nurses were surveyed. A 21 item questionnaire was developed to assess existing management practices and specific osteoporosis knowledge areas.</p> <p>Results</p> <p>The questionnaire assessed the nurse's and personal support worker's (PSWs) education on how to properly administer bisphosphonates by having them select all applicable responses from a list of options. These options included administering the drug before, after or with meals, given with or separate from other medications, given with juice, given with or without water, given with the patient sitting up, or finally given with the patient supine. Only 52% of the nurses and 8.7% of PSWs administered the drug properly, where they selected the options: (given before meals, given with water, given separate from all other medications, and given in a sitting up position). If at least one incorrect option was selected, then it was scored as an inappropriate administration. Bisphosphonates were given before meals by 85% of nurses, given with water by 90%, given separately from other medication by 71%, and was administered in an upright position by 79%. Only 52% of the nurses and 8.7% of PSWs surveyed were administering the drug properly. Regarding the secondary question, of the 57 nurses surveyed, 68% strongly felt their patients should be prescribed vitamin D supplements. Of the 124 PSWs who completed the survey, 44.4% strongly felt their patients should be prescribed vitamin D supplementation.</p> <p>Conclusion</p> <p>Bisphosphonates are quite effective in increasing the bone mineral density of LTC patients, and may reduce fracture rates, but it is only effective if properly administered. In our study, proper administration of bisphosphonate therapy was less than optimal. In summary, although the education of health providers has improved since the mid-1990's, this area still requires further attention and the subject of future quality assurance research.</p
