    Polyfunctional HCV-specific T-cell responses are associated with effective control of HCV replication

    Rapport de synthèse : L'infection par le virus de l'hépatite C (VHC) a une évolution sévère chez les patients co-infectés par VIH/VHC, de même que chez les patients transplantés hépatiques. Toutefois, les mécanismes impliqués dans cette évolution restent peu clairs. Dans ce travail, nous étudions le profil fonctionnel des cellules T spécifiques dirigées contre le virus de l'hépatite C chez 86 patients mono-infectés par VHC, 48 patients co-infectés par VIH/VHC et 42 patients ayant bénéficié d'une transplantation hépatique. La production d'IFN-Y et d'IL-2 et la capacité de proliférer des cellules T CD4+ et CD8+ sont évaluées après stimulation par des peptides dérivés du VHC. Chez les patients mono-infectés par le VHC, les cellules T spécifiques au VHC sont polyfonctionnelles du point de vue de la sécrétion de cytokines, avec trois profils de sécrétion pour les cellules T CD4+: sécrétion uniquement de IL-2, sécrétion de IL-2 et IFN-y et sécrétion uniquement de IFN-gamma, et de deux profils pour les cellules T CD8+: sécrétion de IL-2 et IFN-y et sécrétion uniquement de IFN-gamma. En revanche, les cellules T spécifiques au VHC chez les individus coinfectés par VIH/VHC et chez les patients transplantés hépatiques ont un profil de sécrétions de cytokines marqué par l'absence de cellules CD4+ sécrétant uniquement l'Il-2 et l'absence de cellules CD8+ sécrétant à la fois IL-2 et IFN-gamma. De plus, la prolifération de cellules T CD4+ et CD8+ spécifiques au VHC est considérablement réduite chez les patients co-infectés par VIH/VHC, comme chez les transplantés hépatiques. La présence de cellules T effectrices uniquement (définies par l'absence de cellules T CD4+ sécrétants uniquement de l'IL-2 et l'absence de cellules T CD8+ sécrétant à la fois IL-2 et IFN-gamma et altération de la capacité proliférative) est associée avec une charge virale VHC significativement plus élevée et une fibrose hépatique plus sévère. Par conséquent, les présents résultats suggèrent la participation de mécanismes immunitaires dans l'évolution accélérée de l'hépatite C chez les patients co-infectés par VIH-1 et chez les patients greffés hépatiques

    Geodetic, teleseismic, and strong motion constraints on slip from recent southern Peru subduction zone earthquakes

    We use seismic and geodetic data both jointly and separately to constrain coseismic slip from the 12 November 1996 M_w 7.7 and 23 June 2001 M_w 8.5 southern Peru subduction zone earthquakes, as well as two large aftershocks following the 2001 earthquake on 26 June and 7 July 2001. We use all available data in our inversions: GPS, interferometric synthetic aperture radar (InSAR) from the ERS-1, ERS-2, JERS, and RADARSAT-1 satellites, and seismic data from teleseismic and strong motion stations. Our two-dimensional slip models derived from only teleseismic body waves from South American subduction zone earthquakes with M_w > 7.5 do not reliably predict available geodetic data. In particular, we find significant differences in the distribution of slip for the 2001 earthquake from models that use only seismic (teleseismic and two strong motion stations) or geodetic (InSAR and GPS) data. The differences might be related to postseismic deformation or, more likely, the different sensitivities of the teleseismic and geodetic data to coseismic rupture properties. The earthquakes studied here follow the pattern of earthquake directivity along the coast of western South America, north of 5°S, earthquakes rupture to the north; south of about 12°S, directivity is southerly; and in between, earthquakes are bilateral. The predicted deformation at the Arequipa GPS station from the seismic-only slip model for the 7 July 2001 aftershock is not consistent with significant preseismic motion

    Adaptive density estimation for stationary processes

    We propose an algorithm to estimate the common density ss of a stationary process X1,...,XnX_1,...,X_n. We suppose that the process is either β\beta or τ\tau-mixing. We provide a model selection procedure based on a generalization of Mallows' CpC_p and we prove oracle inequalities for the selected estimator under a few prior assumptions on the collection of models and on the mixing coefficients. We prove that our estimator is adaptive over a class of Besov spaces, namely, we prove that it achieves the same rates of convergence as in the i.i.d framework

    A Rare Case of Diffuse Polyarthritis in the Context of an Epididymoorchitis due to Mumps Infection

    Mumps is a childhood disease with declining incidence in the western world and arthritis is a rare complication associated to the disease. Various presentations exist making diagnosis even more challenging. The mechanisms responsible for the joint involvement remain largely unknown but the timing of onset of the symptoms usually coincide with the rise in antibody titers arguing for an immunologic mediated response. We hereby report a rare case of polyarthritis in the onset of epididymoorchitis due to mumps infection in a HIV infected male patient. Elevated IL-6 serum level in our patient suggests that this cytokine may be an interesting biomarker for the diagnosis of mumps related arthritis

    Multi-Scale Threat Assessment of Riverine Ecosystems in the Colorado River Basin

    Freshwater ecosystems are facing a deepening biodiversity crisis. Developing robust indicators to assess ecological integrity across large spatial scales and identifying the specific threats and pathways of impairment are thus critically needed if we are to inform freshwater conservation strategies. Here we present the first comprehensive threat assessment across the Colorado River Basin – one of the largest and most endangered river basins in North America – using a spatial framework accounting for the wide range of human activities (land uses, transportation infrastructure, exploitative activities, water withdrawals), pathways (local footprint, overland runoff, upstream cumulative effects), and spatial extent of influence (valley bottom, catchment and river network) known to affect the ecological integrity of riverine ecosystems. We quantified and mapped 69 individual threat indices with geospatial tools for each permanent, ephemeral, and intermittent stream segment within the Basin, encompassing a total of \u3e1,067,700 river kilometers. We further aggregated these indices into components of water quality (diffuse and point-source pollution), hydrology (flow regulation/uses and climate change), and physical system (connectivity and geomorphology). To demonstrate the potential of our framework to inform spatial planning decision processes, we examined the typical combinations of threats experienced by different hydrologic areas and stream segment types, identified candidate watersheds for habitat restoration and enhancement where hotspots of biodiversity and threat overlapped, and assessed the associations between threat indices and in situ measurements of ecological integrity describing a suite of biological (benthic macroinvertebrate, fish), chemical (total nitrogen load, water conductivity), hydrological (flow alteration) and physical indicators (streambed stability, instream habitat complexity). Our assessment highlights clear disparities in term of overall degree of threat that result from different combinations and contributions of individual stressors, with different priorities emerging for perennial versus intermittent or ephemeral stream segments, and between the upper and lower parts of the Basin. Importantly, we showed that our threat indices were generally correlated with biological, chemical, hydrological and physical indicators of ecological integrity they were intended to capture. In addition to its implications for the conservation and management of the highly imperiled Colorado River Basin, our case study illustrates how multi-faceted threat mapping can be used to assess the ecological integrity of riverine ecosystems in the absence of spatially extensive in situ measurements

    Spin degree of freedom in two dimensional exciton condensates

    We present a theoretical analysis of a spin-dependent multicomponent condensate in two dimensions. The case of a condensate of resonantly photoexcited excitons having two different spin orientations is studied in detail. The energy and the chemical potentials of this system depend strongly on the spin polarization . When electrons and holes are located in two different planes, the condensate can be either totally spin polarized or spin unpolarized, a property that is measurable.     The Crab pulsar light curve in the soft gamma ray range: FIGARO II results

    The FIGARO II experiment (a large area, balloon borne, crystal scintillator detector working from 0.15 to 4.3 MeV) observed the Crab pulsar on 1990 Jul. 9 for about seven hours. The study of the pulse profile confirms some structures detected with a low significance during the shorter observation of 1986, and adds new important elements to the picture. In particular, between the two main peaks, two secondary peaks appear centered at phase values 0.1 and 0.3, in the energy range 0.38 to 0.49 MeV; in the same energy range, a spectral feature at 0.44 MeV, interpreted as a redshifted positron annihilation line, was observed during the same balloon flight in the phase interval including the second main peak and the neighboring secondary peak. If the phase interval considered is extended to include also the other secondary peak, the significance of the spectral line appears to increase

    We investigate, by means of computer simulations, shapes of nonconvex bodies that maximize resistance to their motion through a rarefied medium, considering that bodies are moving forward and at the same time slowly rotating. A two-dimensional geometric shape that confers to the body a resistance very close to the theoretical supremum value is obtained, improving previous results.Comment: This is a preprint version of the paper published in J.
