    Prevention of folate deficiency by food fortification

    The antimegaloblastic effect of cooked folic acid-fortified maize meal was tested by feeding the meal to 5 lactating patients with folate-deficient megaloblastic anaemia. The dose of folic acid in the meal before cooking varied from 100 to ·500 µg daily. One patient who received 100 µg daily had a suboptimal haematological response. Meal containing 300 µg and 500 µg daily produced optimal haematological response in all 4 patients so treated, and there was no secondary reticulocyte response in the 2 patients subsequently given pharmacological doses of folic acid.     In silico assessment of genetic variation in KCNA5 reveals multiple mechanisms of human atrial arrhythmogenesis

    A recent experimental study investigating patients with lone atrial fibrillation identified six novel mutations in the KCNA5 gene. The mutants exhibited both gain- and loss-of-function of the atrial specific ultra-rapid delayed rectifier K+ current, IKur. The aim of this study is to elucidate and quantify the functional impact of these KCNA5 mutations on atrial electrical activity. A multi-scale model of the human atria was updated to incorporate detailed experimental data on IKur from both wild-type and mutants. The effects of the mutations on human atrial action potential and rate dependence were investigated at the cellular level. In tissue, we assessed the effects of the mutations on the vulnerability to unidirectional conduction patterns and dynamics of re-entrant excitation waves. Gain-of-function mutations shortened the action potential duration in single cells, and stabilised and accelerated re-entrant excitation in tissue. Loss-of-function mutations had heterogeneous effects on action potential duration and promoted early-after-depolarisations following beta-adrenergic stimulation. In the tissue model, loss-of-function mutations facilitated breakdown of excitation waves at more physiological excitation rates than the wild-type, and the generation of early-after-depolarisations promoted unidirectional patterns of excitation. Gain- and loss-of-function IKur mutations produced multiple mechanisms of atrial arrhythmogenesis, with significant differences between the two groups of mutations. This study provides new insights into understanding the mechanisms by which mutant IKur contributes to atrial arrhythmias. In addition, as IKur is an atrial-specific channel and a number of IKur-selective blockers have been developed as anti-AF agents, this study also helps to understand some contradictory results on both pro- and anti-arrhythmic effects of blocking IKur

    Enhanced electrochemiluminescence from a stoichiometric ruthenium(II)-iridium(III) complex soft salt

    The authors thank NSERC, CFI, FQRNT, PREA, and The University of Western Ontario for generous financial support to this research.Electrochemiluminescence (ECL) and electrochemistry are reported for a heterometallic soft salt, [Ru(dtbubpy)3][Ir(ppy)2(CN)2]2 ([Ir][Ru][Ir]), consisting of a 2:1 ratio of complementary charged Ru and Ir complexes possessing two different emission colors. The [Ru]2+ and [Ir]− moieties in the [Ir][Ru][Ir] greatly reduce the energy required to produce ECL. Though ECL intensity in the annihilation path was enhanced 18× relative to that of [Ru(bpy)3]2+, ECL in the co-reactant path with tri-n-propylamine was enhanced a further 4×. Spooling spectroscopy gives insight into ECL mechanisms: the unique light emission at 634 nm is due to the [Ru]2+* excited state and no [Ir]−* was generated in either route. Overall, the soft salt system is anticipated to be attractive and suitable for the development of efficient and low-energy-cost ECL detection systems.PostprintPeer reviewe

    Analysis of United Kingdom Off-Highway Construction Machinery Market and Its Consumers Using New-Sales Data

    The off-highway construction machinery market and its consumers have attracted minimal previous research. This study addresses that void by analyzing annual United Kingdom (UK) (volume/portfolio) new-sales data for the 10 most popular products within that market, 1990–2010 inclusive. Graphical, descriptive statistical, Pearson-correlational, autocorrelational, and elementary modeling are employed to identify contrasts in sales regarding (1) high- and low-volume items; (2) growth trends and significant recessionary effects on volumes; (3) a demand change point circa 1997, since when annual product portfolio has changed little; and (4) product associations in consumer demand. Significant association is demonstrated between demand and construction output, especially with the value of new housing. Subsequently, consumption of wheeled loaders is modeled using construction volume, and demand for mini and crawler excavators is modeled using new-housing data. Time series trends for these machinery types are presented and forecast through 2015. The primary contribution of this study is a deeper understanding of the UK new-machinery market and the predilections of its consumers over the last two decades (to present)

    Red cell folate levels were measured for 6 weeks in an index member and the oldest member of each of 6 families who had been given folic acid-fortified maize meal for use in the home. Five of the index subjects were pregnant and one was lactating. The amount of folic acid added to the maize was calculated so that each adult would receive 500 /Lg folic acid daily. In the index subjects of 5 of the 6 families studied, red cell folate levels rose significantly. The changes in red cell folate levels suggest that pregnant women consume more maize meal than elderly subjects. This possibility would tend to increase the margin of safety when folic acid-fortified foods are consumed by populations in which vitamin B12 deficient megaloblastic anaemia is found.S. Afr. Med.     Diet, Digestion and Energy Intake in Captive Common Marmosets (Callithrix jacchus): Research and Management Implications

    Common marmosets (Callithrix jacchus) are susceptible to intestinal inflammation which leads to chronic diarrhea, weight loss, and vitamin D deficiency. We examined food intake and digestion in three mixed-sex groups of adult marmosets maintained on three commercial base diets. Animals underwent two consecutive 4-day digestion trials. Body mass stayed constant. Feces and diet were assayed for Mn, fat, and gross energy (GE). Apparent digestibility of dry matter (ADDM) was calculated by the total collection method and from dietary and fecal Mn; the methods produced correlated results (r = 0.658, p \u3c 0.001). Apparent digestibility of energy (ADE) was calculated from ADDM and the GE of feces and diet; apparent digestibility of fat (ADfat) was calculated from ADDM and fecal fat. ADDM and ADE varied by diet (p \u3c 0.001). We found poor digesters on all three diets. The concentration of fecal fat was inversely related to ADE (r = −0.729, p \u3c 0.001). High fecal fat (\u3e10%) was associated with ADfat of zero, consistent with lipid malabsorption. Mean digestible energy intake (DEI) was equal to 1.5 the estimated metabolic rate, but varied widely between individuals. The diet with the fewest animals with high fecal fat had the highest mean DEI and most animals above 450 g, suggesting it may be obesogenic

    Optimized unconventional superconductivity in a molecular Jahn-Teller metal

    Understanding the relationship between the superconducting, the neighboring insulating, and the normal metallic state above Tc is a major challenge for all unconventional superconductors. The molecular A3C60 fulleride superconductors have a parent antiferromagnetic insulator in common with the atom-based cuprates, but here, the C603– electronic structure controls the geometry and spin state of the structural building unit via the on-molecule Jahn-Teller effect. We identify the Jahn-Teller metal as a fluctuating microscopically heterogeneous coexistence of both localized Jahn-Teller–active and itinerant electrons that connects the insulating and superconducting states of fullerides. The balance between these molecular and extended lattice features of the electrons at the Fermi level gives a dome-shaped variation of Tc with interfulleride separation, demonstrating molecular electronic structure control of superconductivity
