    Background The immunological escape of tumors represents one of the main ob- stacles to the treatment of malignancies. The blockade of PD-1 or CTLA-4 receptors represented a milestone in the history of immunotherapy. However, immune checkpoint inhibitors seem to be effective in specific cohorts of patients. It has been proposed that their efficacy relies on the presence of an immunological response. Thus, we hypothesized that disruption of the PD-L1/PD-1 axis would synergize with our oncolytic vaccine platform PeptiCRAd. Methods We used murine B16OVA in vivo tumor models and flow cytometry analysis to investigate the immunological background. Results First, we found that high-burden B16OVA tumors were refractory to combination immunotherapy. However, with a more aggressive schedule, tumors with a lower burden were more susceptible to the combination of PeptiCRAd and PD-L1 blockade. The therapy signifi- cantly increased the median survival of mice (Fig. 7). Interestingly, the reduced growth of contralaterally injected B16F10 cells sug- gested the presence of a long lasting immunological memory also against non-targeted antigens. Concerning the functional state of tumor infiltrating lymphocytes (TILs), we found that all the immune therapies would enhance the percentage of activated (PD-1pos TIM- 3neg) T lymphocytes and reduce the amount of exhausted (PD-1pos TIM-3pos) cells compared to placebo. As expected, we found that PeptiCRAd monotherapy could increase the number of antigen spe- cific CD8+ T cells compared to other treatments. However, only the combination with PD-L1 blockade could significantly increase the ra- tio between activated and exhausted pentamer positive cells (p= 0.0058), suggesting that by disrupting the PD-1/PD-L1 axis we could decrease the amount of dysfunctional antigen specific T cells. We ob- served that the anatomical location deeply influenced the state of CD4+ and CD8+ T lymphocytes. In fact, TIM-3 expression was in- creased by 2 fold on TILs compared to splenic and lymphoid T cells. In the CD8+ compartment, the expression of PD-1 on the surface seemed to be restricted to the tumor micro-environment, while CD4 + T cells had a high expression of PD-1 also in lymphoid organs. Interestingly, we found that the levels of PD-1 were significantly higher on CD8+ T cells than on CD4+ T cells into the tumor micro- environment (p < 0.0001). Conclusions In conclusion, we demonstrated that the efficacy of immune check- point inhibitors might be strongly enhanced by their combination with cancer vaccines. PeptiCRAd was able to increase the number of antigen-specific T cells and PD-L1 blockade prevented their exhaus- tion, resulting in long-lasting immunological memory and increased median survival

    Agrobiodiversidad y servicios ecosistémicos : una revisión de los componentes y prácticas de manejo

    En los últimos años se ha documentado las consecuencias que genera la pérdida de la biodiversidad agrícola en el funcionamiento de los agroecosistemas, así como su rol en la generación de servicios ecosistémicos. Por lo tanto, con el fin de analizar la relación entre la agrobiodiversidad y los servicios ecosistémicos, se realizó una revisión sistemática de literatura científica a nivel global, a través de la construcción de un patrón de búsqueda y la consulta de diferentes bases de datos de impacto. La cual, permitió la identificación de 71 servicios ecosistémicos provistos por los componentes de la agrobiodiversidad asociada, planeada y circundante, en sistemas agrícolas diversificados. Esta revisión documental señala que los servicios de regulación son los más evaluados (49%), seguido de los servicios culturales (25%), de suministro (13%) y soporte (13%). La provisión de servicios ecosistémicos en agroecosistemas diversificados se da gracias a la implementación de prácticas de manejo que proveen, favorecen y permiten la conservación de la biodiversidad agrícola. Respecto al componente planeado, la producción de alimentos de calidad es el servicio de mayor relevancia para los agricultores. Los servicios de regulación son provistos especialmente por los componentes circundantes, mientras que los servicios culturales, por el componente asociado. Finalmente, los resultados obtenidos permiten concluir que los agroecosistemas cumplen un rol más allá se producir alimento. Se logró una compresión de la relación entre agrobiodiversidad y servicios ecosistémicos gracias a factores determinantes como la diversificación de cultivos, la asociación de sistemas agrícolas y la implementación de prácticas sostenibles.In recent years, the consequences of loss of agricultural biodiversity on the functioning of agroecosystems have been documented, as well as their role in generating ecosystem services. Therefore, to analyze the relationship between agrobiodiversity and ecosystem services, a systematic review of scientific literature was carried out at the global level, through the construction of a search pattern and the consultation of different databases of impact. This allowed the identification of 71 ecosystem services provided by the components of associated, planned and surrounding agrobiodiversity in diversified agricultural systems. This documentary review indicates that regulation services are the most evaluated (49%), followed by cultural services (25%), supply (13%) and support (13%). The provision of ecosystem services in diversified agroecosystems is due to the implementation of management practices that provide, favor and allow the conservation of agricultural biodiversity. About the planned component, the production of quality food is the most important service for farmers. Regulation services are provided especially by the surrounding components, while cultural services, by the associated component. Finally, the results obtained allow us to conclude that agroecosystems play a role beyond food production. The understanding of the relationship between agrobiodiversity and ecosystem services was achieved through key factors such as crop diversification, the association of agricultural systems and the implementation of sustainable practices

    Memory and Identity: Citizen Perception in the Processes of Heritage Enhancement and Regeneration in Obsolete Neighborhoods—The Case of Polígono de San Pablo, Seville

    The designation of which resources are valued as emerging heritage is at a turning point. This has resulted in urban heritage regeneration processes designed from the top down, neglecting the perception of the local. This article highlights the importance of generating participatory multi-actor spaces where the knowledge of agents involved in the same process of urban regeneration, in this case linked to heritage in obsolete neighborhoods, can be incorporated and contrasted. The San Pablo neighborhood in Seville, built in the 1960s, is chosen as a case study. Actor–network theory is taken as a methodological basis for articulating a network with the voices of agents involved in heritage regeneration processes in obsolete neighborhoods. A methodology designed from the bottom up is put into practice, having as a base the social agents, followed by the academy, and finally with the technical knowledge of official institutions. This research concludes that incorporating the elderly population as social agents in regenerative actions in the San Pablo neighborhood is a determining factor in characterizing its uniqueness. Collective memory naturally associates resources that are part of the social identity of the neighborhood. Due to its deep roots, the older population recognizes different elements that could be protected and possibly recognized as emerging heritage, and that technical agents should consider them to achieve sustainable regeneration

    Objetivo: identificar evidencia bibliográfica del uso de la música como herramienta en procesos de inclusión de personas con discapacidad, que permitiera ofrecer sustento teórico al proyecto del convenio establecido entre la Fundación Saldarriaga Concha y la Fundación Nacional Batuta. Materiales y métodos: como estrategia de búsqueda se utilizaron los temas “música”, “inclusión” y “discapacidad” y el booleano AND en tres bases de datos: Ebsco, Medline y Lilacs. Los análisis fueron realizados mediante el establecimiento de frecuencias para variables como revista, país, tipo de estudio y años de publicación. Se localizaron veintidós artículos que incluyeron los criterios de búsqueda definidos por los autores. Resultados: los resultados muestran que la mayoría de publicaciones se encuentra en la base de datos Ebsco, en la revista Music Educators Journal, en el período comprendido entre el año 2004 a 2006 (40,9%). Estados Unidos es el país que más publicaciones ha realizado (81,8%). El tipo de estudio más frecuente es la revisión de tema (86,4%) y la temática más utilizada incorpora aspectos como música-discapacidad. De acuerdo con Rumbo, Aranton y García, el grado de evidencia de los documentos analizados es 3, lo que indica que son principalmente estudios descriptivos y de opinión de expertos. Conclusiones: después de realizar la búsqueda sistemática se puede concluir que las publicaciones que incorporan el uso de la música como herramienta para facilitar procesos de inclusión social de niños y adolescentes es escasa, por lo que se sugiere sistematizar y publicar resultados de las intervenciones en este campo de la Terapia Ocupacional. El presente documento proporciona una posible ruta para el desarrollo de futuras investigaciones referentes al tema.Objective: to identify literature and bibliographical evidence regarding, the use of music as a tool during social inclusion processes of people with disabilities; this was done in order to obtain and supply theoretical bases to the project which pertains and arises from the agreement between the Fundación Saldarriaga Concha and the Fundación Nacional Batuta. Materials and methods: as the research strategy implemented during the investigation, “music”, “inclusion” and “disability” with the Boolean AND were used as the search criteria in three different databases: Ebsco, Medline and Lilacs. The analysis was elaborated considering the frequency of different variables/ characteristics of the articles such as, journal, country, type of study and the year it was published. Twenty two articles met the search criteria defined by the authors. Results: the results indicated that the database which contained the most amount of evidence is Ebsco, specifically in the Music Educators Journal, during the years of 2004-2006 40,9%) and the majority of articles were published in the United States (81,8%). The most common and frequent type of study found was the subject review (86,4%), mainly incorporating search topics like music-disability. Agreeing with Rumbo, Aranton and García, the level of evidence of the analyzed documents is generally 3, indicating that, above all, the studies tend to be descriptive and based on the opinion of experts. Conclusions: after systematically carrying out the research, it is possible to conclude that the publications which consider the use of music as a tool to facilitate social inclusion processes in children and teenagers, is low. Due to the results obtained, it is strongly suggested that, writers begin to systemize and publish the results evidenced in this field of Occupational Therapy.
