    Transcriptional regulation of the grape cytochrome P450 monooxygenase gene CYP736B expression in response to Xylella fastidiosa infection

    <p>Abstract</p> <p>Background</p> <p>Plant cytochrome P450 monooxygenases (CYP) mediate synthesis and metabolism of many physiologically important primary and secondary compounds that are related to plant defense against a range of pathogenic microbes and insects. To determine if cytochrome P450 monooxygenases are involved in defense response to <it>Xylella fastidiosa </it>(<it>Xf</it>) infection, we investigated expression and regulatory mechanisms of the cytochrome P450 monooxygenase <it>CYP736B </it>gene in both disease resistant and susceptible grapevines.</p> <p>Results</p> <p>Cloning of genomic DNA and cDNA revealed that the <it>CYP736B </it>gene was composed of two exons and one intron with GT as a donor site and AG as an acceptor site. <it>CYP736B </it>transcript was up-regulated in PD-resistant plants and down-regulated in PD-susceptible plants 6 weeks after <it>Xf </it>inoculation. However, <it>CYP736B </it>expression was very low in stem tissues at all evaluated time points. 5'RACE and 3'RACE sequence analyses revealed that there were three candidate transcription start sites (TSS) in the upstream region and three candidate polyadenylation (PolyA) sites in the downstream region of <it>CYP736B</it>. Usage frequencies of each transcription initiation site and each polyadenylation site varied depending on plant genotype, developmental stage, tissue, and treatment. These results demonstrate that expression of <it>CYP736B </it>is regulated developmentally and in response to <it>Xf </it>infection at both transcriptional and post-transcriptional levels. Multiple transcription start and polyadenylation sites contribute to regulation of <it>CYP736B </it>expression.</p> <p>Conclusions</p> <p>This report provides evidence that the cytochrome P450 monooxygenase <it>CYP736B </it>gene is involved in defense response at a specific stage of <it>Xf </it>infection in grapevines; multiple transcription initiation and polyadenylation sites exist for <it>CYP736B </it>in grapevine; and coordinative and selective use of transcription initiation and polyadenylation sites play an important role in regulation of <it>CYP736B </it>expression during growth, development and response to <it>Xf </it>infection.</p

    Closed orbit correction at synchrotrons for symmetric and near-symmetric lattices

    This contribution compiles the benefits of lattice symmetry in the context of closed orbit correction. A symmetric arrangement of BPMs and correctors results in structured orbit response matrices of Circulant or block Circulant type. These forms of matrices provide favorable properties in terms of computational complexity, information compression and interpretation of mathematical vector spaces of BPMs and correctors. For broken symmetries, a nearest-Circulant approximation is introduced and the practical advantages of symmetry exploitation are demonstrated with the help of simulations and experiments in the context of FAIR synchrotrons

    Axion-photon Couplings in Invisible Axion Models

    We reexamine the axion-photon couplings in various invisible axion models motivated by the recent proposal of using optical interferometry at the ASST facility in the SSCL to search for axion. We illustrate that the assignment of U(1)PQU(1)_{PQ} charges for the fermion fields plays an important role in determining the couplings. Several simple non-minimal invisible axion models with suppressed and enhanced axion-photon couplings are constructed, respectively. We also discuss the implications of possible new experiments to detect solar axions by conversion to XX-rays in a static magnetic apparatus tracking the sun.Comment: 14 pages, LaTeX fil

    The crest phenotype in domestic chicken is caused by a 197 bp duplication in the intron of HOXC10

    The Crest mutation in chicken shows incomplete dominance and causes a spectacular phenotype in which the small feathers normally present on the head are replaced by much larger feathers normally present only in dorsal skin. Using whole-genome sequencing, we show that the crest phenotype is caused by a 197 bp duplication of an evolutionarily conserved sequence located in the intron of HOXC10 on chromosome 33. A diagnostic test showed that the duplication was present in all 54 crested chickens representing eight breeds and absent from all 433 non-crested chickens representing 214 populations. The mutation causes ectopic expression of at least five closely linked HOXC genes, including HOXC10, in cranial skin of crested chickens. The result is consistent with the interpretation that the crest feathers are caused by an altered body region identity. The upregulated HOXC gene expression is expanded to skull tissue of Polish chickens showing a large crest often associated with cerebral hernia, but not in Silkie chickens characterized by a small crest, both homozygous for the duplication. Thus, the 197 bp duplication is required for the development of a large crest and susceptibility to cerebral hernia because only crested chicken show this malformation. However, this mutation is not sufficient to cause herniation because this malformation is not present in breeds with a small crest, like Silkie chickens

    X-ray observations of RX J0822-4300 and Puppis-A

    Based on observations with the X-ray observatories Chandra and XMM-Newton we present results from a detailed spectro-imaging and timing analysis of the central compact X-ray source RX J0822-4300 in the supernova remnant Puppis-A. The superior angular resolution of Chandra allows for the first time to pinpoint the point source nature of this object down to 0.59±0.010.59\pm0.01 arcsec (FWHM) and to determine its position:{RA=08h21m57.40s08^{\rm h}21^{\rm m}57.40^{\rm s}, Dec=430016.69-43^{\circ}00^{'}16.69^{''} (J2000)} with sub-arcsecond accuracy. Spectral fits based on Chandra and XMM-Newton data provide a tight constraint on the emission properties of RX J0822-4300. Most of its X-ray emission seems to be of thermal origin. A model spectrum consisting of two blackbody components with T12.6×106T_{1}\simeq 2.6\times10^{6} K, T25.0×106T_{2}\simeq 5.0\times10^{6} K and R13.3R_{1}\simeq 3.3 km, R20.75R_2 \simeq 0.75 km for the blackbody temperatures and the size of the projected emitting regions, respectively, provides the best model description of its spectrum. A search for X-ray pulsations from RX J0822-4300, revealed an interesting periodicity candidate which, if confirmed, does not support a scenario of steady spin-down.Comment: Accepted by A&A, 20 pages, 10 figures, 3 table

    Studies of the motion and decay of axion walls bounded by strings

    We discuss the appearance at the QCD phase transition, and the subsequent decay, of axion walls bounded by strings in N=1 axion models. We argue on intuitive grounds that the main decay mechanism is into barely relativistic axions. We present numerical simulations of the decay process. In these simulations, the decay happens immediately, in a time scale of order the light travel time, and the average energy of the radiated axions is 7ma \simeq 7 m_a for va/ma500v_a/m_a \simeq 500. is found to increase approximately linearly with ln(va/ma)\ln(v_a/m_a). Extrapolation of this behaviour yields 60ma \sim 60 m_a in axion models of interest. We find that the contribution to the cosmological energy density of axions from wall decay is of the same order of magnitude as that from vacuum realignment, with however large uncertainties. The velocity dispersion of axions from wall decay is found to be larger, by a factor 10310^3 or so, than that of axions from vacuum realignment and string decay. We discuss the implications of this for the formation and evolution of axion miniclusters and for the direct detection of axion dark matter on Earth. Finally we discuss the cosmology of axion models with N>1N>1 in which the domain wall problem is solved by introducing a small UPQ_{PQ}(1) breaking interaction.     Phenomenological Consequences of Singlet Neutrinos

    In this paper, we study the phenomenology of right-handed neutrino isosinglets. We consider the general situation where the neutrino masses are not necessarily given by mD2/Mm_D^2/M, where mDm_D and MM are the Dirac and Majorana mass terms respectively. The consequent mixing between the light and heavy neutrinos is then not suppressed, and we treat it as an independent parameter in the analysis. It turns out that μe\mu-e conversion is an important experiment in placing limits on the heavy mass scale (MM) and the mixing. Mixings among light neutrinos are constrained by neutrinoless double beta decay, as well as by solar and atmospheric neutrino experiments.     Sepsis-associated microvascular dysfunction measured by peripheral arterial tonometry: an observational study

    INTRODUCTION: Sepsis has a high mortality despite advances in management. Microcirculatory and endothelial dysfunction contribute to organ failure, and better tools are needed to assess microcirculatory responses to adjunctive therapies. We hypothesised that peripheral arterial tonometry (PAT), a novel user-independent measure of endothelium-dependent microvascular reactivity, would be impaired in proportion to sepsis severity and related to endothelial activation and plasma arginine concentrations. METHODS: Observational cohort study in a 350-bed teaching hospital in tropical Australia. Bedside microvascular reactivity was measured in 85 adults with sepsis and 45 controls at baseline and 2-4 days later by peripheral arterial tonometry. Microvascular reactivity was related to measures of disease severity, plasma concentrations of L-arginine (the substrate for nitric oxide synthase), and biomarkers of endothelial activation. RESULTS: Baseline reactive hyperaemia index (RH-PAT index), measuring endothelium-dependent microvascular reactivity; (mean [95% CI]) was lowest in severe sepsis (1.57 [1.43-1.70]), intermediate in sepsis without organ failure (1.85 [1.67-2.03]) and highest in controls (2.05 [1.91-2.19]); P < 0.00001. Independent predictors of baseline RH-PAT index in sepsis were APACHE II score and mean arterial pressure, but not plasma L-arginine or markers of endothelial activation. Low baseline RH-PAT index was significantly correlated with an increase in SOFA score over the first 2-4 days (r = -0.37, P = 0.02). CONCLUSIONS: Endothelium-dependent microvascular reactivity is impaired in proportion to sepsis severity and suggests decreased endothelial nitric oxide bioavailability in sepsis. Peripheral arterial tonometry may have a role as a user-independent method of monitoring responses to novel adjunctive therapies targeting endothelial dysfunction in sepsis
