Ătude phytochimique et Ă©valuation de lâactivitĂ© anti-oxydante de Thymus CIliatus ssp. Coloratus
Thymus ciliatus ssp. Coloratus, plante aromatique, spontanĂ©e et rĂ©pandue en rĂ©gions mĂ©diterranĂ©ennes et dans le nord de lâAlgĂ©rie est trĂšs utilisĂ©e par les populations locales pour ses vertus mĂ©dicinales. Les tests phytochimiques appliquĂ©s au Thymus ciliatus ssp coloratus ont montrĂ© la prĂ©sence de quelques familles de composĂ©s chimiques et notamment les flavonoĂŻdes. L'activitĂ© antioxydante in vitro de lâextrait de flavonoĂŻdes a Ă©tĂ© Ă©valuĂ©e par la technique de rĂ©duction du fer « FRAP » (ferric reducing antioxydant power) et par le test de DPPH. La premiĂšre mĂ©thode a montrĂ© que la fraction acĂ©tate dâĂ©thyle de la partie aĂ©rienne a une capacitĂ© Ă rĂ©duire le fer plus marquĂ©e que celles de lâacide ascorbique et le BHA et que celle des racines, est nettement infĂ©rieure. Quant aux fractions butanoliques des flavonoĂŻdes de la partie aĂ©rienne et des racines, elles ont une capacitĂ© Ă rĂ©duire le fer largement infĂ©rieure Ă celles de lâacide ascorbique et le BHA. LâactivitĂ© antioxydante, obtenue par la second procĂ©dĂ© et relative Ă la fraction acĂ©tate dâĂ©thyle des flavonoĂŻdes de la partie aĂ©rienne de la plante est plus importante (I.Câ

Coloratus Thymus ciliatus ssp. coloratus, aromatic plant, spontaneous and widespread in the Mediterranean and in northern Algeria is widely used by local people for its medicinal properties. Phytochemical tests applied for Thymus sciliatus ssp coloratus showed the presence of several families of chemical compounds including flavonoĂŻds. The in vitro antioxidant activity of flavonoĂŻds extract was evaluated by the iron reduction technique "FRAP" (ferric reducing antioxidant power) and DPPH test. The first method showed that the ethyl acetate fraction of the aerial part has an ability to reduce the iron greater than ascorbic acid and BHA and the roots, is significantly lower. As for butanol fractions of flavonoĂŻds from the aerial part and roots, they have an ability to reduce iron significantly lower than those of ascorbic acid and BHA. The antioxidant activity obtained by the second method and on the ethyl acetate fraction of flavonoĂŻds from the aerial part of the plant is more important (I.Câ

Postendovascular thoracic aortic repair subclavian steal syndrome revealed by severe headache
AbstractSubclavian steal syndrome (SCSS) has been known since 1960 in the medical literature. Its principal cause is atherosclerosis responsible of occlusion of the subclavian artery (SCA). It is the pathological process in which blood flows in reverse direction from the vertebral artery (VA) to the SCA. Usually asymptomatic, but a variety of symptoms may develop involving the vertebro-basilar and/or the carotid territories and may be precipitated by exercise of the upper extremity. In some circumstances it can be iatrogenic complicating the course of a thoracic endovascular aortic repair (TEVAR) when the left SCA is covered by the endoprosthesis, which is a necessity many times giving the frequent proximity of the acute thoracic pathologies to the origin of this vessel.We present a case of severe headache occurring after a TEVAR with intentional coverage of the origin of the left SCA. This headache was the only symptom from which the patient complained, and which disappeared immediately after carotid-SCA bypass. Other devastating complications can happen, which gave as a concern about the management of SCA when decision to practice a TEVAR is taken
Evaluation de l'impact des nouvelles Directives et recommandations architecturales des établissements médico-sociaux vaudois (DAEMS) sur la satisfaction des usagers
L'environnement construit contribue Ă influencer la santĂ© des individus, selon l'Organisation Mondiale de la SantĂ©. L'architecture des Ă©tablissements mĂ©dico-sociaux (EMS) peut, elle aussi, produire un impact sur le bien-ĂȘtre et la satisfaction des rĂ©sident-e-s et des autres usagers. Dans le canton de Vaud, des normes architecturales de construction des EMS (les Directives et recommandations architecturales des Ă©tablissements mĂ©dico-sociaux, DAEMS) ont Ă©tĂ© crĂ©Ă©es en 2003 afin d'optimiser la conception des EMS par rapport aux besoins des usagers. Notre Ă©tude a pour but de vĂ©rifier si les EMS construits selon les DAEMS sont associĂ©s avec une satisfaction accrue des usagers par rapport aux EMS qui n'ont pas Ă©tĂ© construits selon ces directives.
"Le but de la présente étude est de vérifier si le fait de construire les EMS selon les DAEMS est associé avec une plus grande satisfaction chez les usagers par rapport aux EMS qui n'ont pas été construits selon les DAEMS. Nous avons effectué une comparaison de la satisfaction auprÚs des trois-publics qui sont les principaux usagers des EMS, à savoir les résident-e-s, leurs proches de référence, et les professionnel-le-s en EMS. Trente EMS ont participé à l'étude, à savoir dix construits selon les DAEMS (EMS « DAEMS »), et vingt qui n'ont pas été construits selon les DAEMS (EMS « non-DAEMS »). Ce rapport a été défini pour garantir une bonne validité statistique des résultats

The implementation of hygiene system in the school catering is based on the application of good hygienic practice and regulations of food safety (security). These measures of hygiene contribute absolutely, in the elimination of the potential food risks susceptible to be at the origin of several diseases. This article presents the results of a study carried out to evaluate the degree of the respect for the sanitary conditions at the level of three school restaurants. The result of our study aims to determine the level of compliance of their hygienic systems to international hygienic standards. The rates of conformity of the sites of the study were of the order of 33, 60 %, 33, 00 % and of 48, 58 % respectively at the level of the establishment n°1, n°2 and n°3. The recorded results show apparently, the level of conformity with regard to the standards. In front of this situation, it is essential to plan and to implement relevant actions of improvement and to engage ways and resources to eliminate the noticed gaps and increase the level of conformit
The Edinburgh Cognitive and Behavioral ALS Screen (ECAS) in frontotemporal dementia
To examine the usefulness of the Edinburgh Cognitive and Behavioral Amyotrophic Lateral Sclerosis (ALS) Screen (ECAS) as a cognitive screening tool for the detection of behavioral variant frontotemporal dementia (bvFTD). A secondary aim was to determine whether people with FTD combined with ALS (ALS-FTD) exhibit a similar ECAS profile to that of people with bvFTD alone. Methods: Patients with ALS-FTD and bvFTD and healthy controls were recruited. Participants were administered the ECAS, which comprises tests of language, verbal fluency, executive functions, memory, and visual-spatial functions. They also carried out analogous, full-length cognitive tests that examine naming, spelling, sentence completion, and social cognition skills. Results: The study cohort comprised 20 ALS-FTD patients, 23 with bvFTD, and 30 controls. Highly significant group differences were elicited for all cognitive domains, reflecting poorer performance in patients compared to controls. No significant differences in overall test scores were found between ALS-FTD and bvFTD patients, although ALS-FTD patients showed a higher frequency of impairment on verbal fluency. Correlative analyses revealed inter-relationships in patients (but not controls) between scores in different domains, most marked in bvFTD. There were strong correlations between performance on ECAS subtests and analogous cognitive tasks. Conclusion: The ECAS is a sensitive and valuable tool for the assessment of FTD. Executive, language and behavioral breakdown may, however, compromise performance in other cognitive domains, reducing the specificity of the âfrontotemporalâ cognitive profile. Development of a LAMP assay for detection of Leishmania infantum infection in dogs using conjunctival swab samples
Background: Leishmania infantum infections in dogs play a crucial role in the transmission of pathogens causing visceral leishmaniasis to humans in the Gansu province, northwest China. To be able to control zoonotic transmission of the parasite to humans, a non-invasive loop-mediated isothermal amplification (LAMP) assay to specifically detect L. infantum infections in dogs was developed. Methods: The primers used in the LAMP assay were designed to target kinetoplast DNA minicircle sequences of the L. infantum isolate MCAN/CN/90/SC and tested using DNA isolated from promastigotes of different Leishmania species. The LAMP assay was evaluated with conjunctional swab samples obtained from 111 and 33 dogs living in an endemic and a non-endemic region of zoonotic visceral leishmaniasis in the Gansu province, respectively. The LAMP assay was also compared with conventional PCR, ELISA and microscopy using conjunctional swab, serum and bone marrow samples from the dogs, respectively. Results: The LAMP assay detected 1 fg of L. infantum DNA purified from cultured promastigotes which was 10-fold more sensitive than a conventional PCR test using Leishmania genus-specific primers. No cross reaction was observed with DNA isolated from promastigotes of L. donovani, L. major, L. tropica, and L. braziliensis, and the L. infantum reference strain MHOM/TN/80/IPT1. The L. infantum-positive rates obtained for field-collected samples were 61.3%, 58.6%, 40.5% and 10.8% by LAMP, PCR, ELISA and microscopy, respectively. As only one out of the 33 samples from control dogs from the non-endemic region of zoonotic visceral leishmaniasis was positive by the LAMP assay and the PCR test, the observed true negative rate (specificity) was 97% for both methods. Conclusion: This study has shown that the non-invasive, conjunctional swab-based LAMP assay developed was more sensitive in the detection of leishmaniasis in dogs than PCR, ELISA and microscopy. The findings indicate that the LAMP assay is a sensitive and specific method for the field surveillance of domestic dogs, particularly of asymptomatic canines, in ZVL-endemic areas in western China
Regenerative potential of human muscle stem cells in chronic inflammation
International audienceABSTRACT: INTRODUCTION: Chronic inflammation is a profound systemic modification of the cellular microenvironment which could affect survival, repair and maintenance of muscle stem cells. The aim of this study was to define the role of chronic inflammation on the regenerative potential of satellite cells in human muscle. METHODS: As a model for chronic inflammation, 11 patients suffering from rheumatoid arthritis (RA) were included together with 16 patients with osteoarthritis (OA) as controls. The mean age of both groups was 64 years, with more females in the RA group compared to the OA group. During elective knee replacement surgery, a muscle biopsy was taken from the distal musculus vastus medialis. Cell populations from four RA and eight OA patients were used for extensive phenotyping because these cell populations showed no spontaneous differentiation and myogenic purity greater than 75% after explantation. RESULTS: After mononuclear cell explantation, myogenic purity, viability, proliferation index, number of colonies, myogenic colonies, growth speed, maximum number of population doublings and fusion index were not different between RA and OA patients. Furthermore, the expression of proteins involved in replicative and stress-induced premature senescence and apoptosis, including p16, p21, p53, hTERT and cleaved caspase-3, was not different between RA and OA patients. Mean telomere length was shorter in the RA group compared to the OA group. CONCLUSIONS: In the present study we found evidence that chronic inflammation in RA does not affect the in vitro regenerative potential of human satellite cells. Identification of mechanisms influencing muscle regeneration by modulation of its microenvironment may, therefore, be more appropriate
Mutations in the Mitochondrial Citrate Carrier SLC25A1 are Associated with Impaired Neuromuscular Transmission.
BACKGROUND AND OBJECTIVE: Congenital myasthenic syndromes are rare inherited disorders characterized by fatigable weakness caused by malfunction of the neuromuscular junction. We performed whole exome sequencing to unravel the genetic aetiology in an English sib pair with clinical features suggestive of congenital myasthenia. METHODS: We used homozygosity mapping and whole exome sequencing to identify the candidate gene variants. Mutant protein expression and function were assessed in vitro and a knockdown zebrafish model was generated to assess neuromuscular junction development. RESULTS: We identified a novel homozygous missense mutation in the SLC25A1 gene, encoding the mitochondrial citrate carrier. Mutant SLC25A1 showed abnormal carrier function. SLC25A1 has recently been linked to a severe, often lethal clinical phenotype. Our patients had a milder phenotype presenting primarily as a neuromuscular (NMJ) junction defect. Of note, a previously reported patient with different compound heterozygous missense mutations of SLC25A1 has since been shown to suffer from a neuromuscular transmission defect. Using knockdown of SLC25A1 expression in zebrafish, we were able to mirror the human disease in terms of variable brain, eye and cardiac involvement. Importantly, we show clear abnormalities in the neuromuscular junction, regardless of the severity of the phenotype. CONCLUSIONS: Based on the axonal outgrowth defects seen in SLC25A1 knockdown zebrafish, we hypothesize that the neuromuscular junction impairment may be related to pre-synaptic nerve terminal abnormalities. Our findings highlight the complex machinery required to ensure efficient neuromuscular function, beyond the proteomes exclusive to the neuromuscular synapse
