Motavizumab (MEDI-524) is a monoclonal antibody with enhanced neutralizing activity against RSV. In mice, motavizumab suppressed RSV replication which resulted in significant reduction of clinical parameters of disease severity. We evaluated the effect of motavizumab on the local and systemic immune response induced by RSV in the mouse model. Balb/c mice were intranasally inoculated with 106.5 PFU RSV A2 or medium. Motavizumab was given once intraperitoneally (1.25 mg/mouse) as prophylaxis, 24 h before virus inoculation. Bronchoalveolar lavage (BAL) and serum samples were obtained at days 1, 5 (acute) and 28 (long-term) post inoculation and analyzed with a multiplex assay (Beadlyte Upstate, NY) for simultaneous quantitation of 18 cytokines: IL-1α, IL-1β, IL-2, IL-3, IL-4, IL-5, IL-6, KC (similar to human IL-8), IL-10, IL-12p40, IL-12p70, IL-13, IL-17, TNF-α, MCP-1, RANTES, IFN-γ and GM-CSF. Overall, cytokine concentrations were lower in serum than in BAL samples. By day 28, only KC was detected in BAL specimens at low concentrations in all groups. Administration of motavizumab significantly reduced (p < 0.05) BAL concentrations of IL-1α, IL-12p70 and TNF-α on day 1, and concentrations of IFN-γ on days 1 and 5 compared with RSV-infected untreated controls. In the systemic compartment, the concentrations of IL-10, IFN-γ and KC were significantly reduced in the motavizumab-treated mice compared with the untreated controls. In summary, prophylactic administration of motavizumab was associated with significant reductions on RSV replication and concentrations of cytokine and chemokines, which are likely related to the improvement observed in clinical markers of disease severity

Background: The development of effective COVID-19 vaccines has reduced the impact of COVID-19 on the general population. Our study aims to analyze how vaccination modifies the likelihood of death and length of stay in hospitalized patients with COVID-19; (2) Methods: A retrospective cohort study of 1927 hospitalized patients infected with COVID-19 was conducted. Information was gathered on vaccination status, hospitalization episode, and clinical profile of the patients. The effect of vaccination on mortality was analyzed using a multiple logistic regression model, and length of stay was analyzed using linear regression. The performance and fit of the models were evaluated; (3) Results: In hospitalized patients with COVID-19, the risk of dying during admission in vaccinated patients was half that of non-vaccinated (OR: 0.45; CI 95%: 0.25 to 0.84). In patients who were discharged due to improvement, the reduction in hospital stay in vaccinated patients was 3.17 days (CI 95%: 5.88 to 0.47); (4) Conclusions: Patients who, despite having been vaccinated, acquire the infection by SARS-CoV-2, have a significant reduction of the risk of death during admission and a reduction of hospital stay compared with unvaccinated patients

Background: The altered expression of prolactin (PRL) and its receptor (PRLR) has been implicated in breast and other types of cancer. There are few studies that have focused on the analysis of PRL/PRLR in cervical cancer where the development of neoplastic lesions is influenced by the variation of the hormonal status. The aim of this study was to evaluate the expression of PRL/PRLR and the effect of PRL treatment on cell proliferation and apoptosis in cervical cancer cell lines. Results:High expression of multiple PRLR forms and PRLvariants of 60-80 kDa were observed in cervical cancer cell lines compared with non-tumorigenic keratinocytes evaluated by Western blot, immunofluorecence and real time PCR. Treatment with PRL (200 ng/ml) increased cell proliferation in HeLa cells determined by the MTT assay at day 3 and after 1 day a protective effect against etoposide induced apoptosis in HeLa, SiHa and C-33A cervical cancer cell lines analyzed by the TUNEL assay. Conclusions: Our data suggests that PRL/PRLR signaling could act as an important survival factor for cervical cancer. The use of an effective PRL antagonist may provide a better therapeutic intervention in cervical cancer. 2013 Lopez-Pulido et al.; licensee BioMed Central Ltd

Bacterial meningitis remains a serious cause of morbidity and mortality in childhood, despite the availability of effective vaccines against Haemophilus influenzae type b (Hib) or Streptococcus pneumoniae. The purpose of this study was to analyze data on bacterial meningitis cases in Korea from 1996 through 2005. The information of all hospitalized bacteria-proven meningitis cases was obtained from 17 university hospitals nationwide. A total of 402 cases were identified. Of these, 125 (29.9%) cases were neonates. Streptococcus agalactiae was the most common bacteria responsible for 99 (24.6%) of all cases regardless of age, followed by S. pneumoniae for 91 (22.6%) and H. influenzae for 67 (16.7%) patients. The common etiology beyond the neonatal period was S. pneumoniae for 91 (33.0%) followed by H. influenzae for 63 (22.8%) patients. The overall case fatality rate was 9.4%, which was similar with that in 1986-1995. In conclusion, S. agalactiae, S. pneumoniae and H. influenzae were important etiologic agents of bacterial meningitis in children in the last 10 yrs. It is required to establish the preventive strategy of the three bacteria. The nationwide epidemiologic study should be continued to evaluate immunization strategy and efficacy

Ecological Niche Modeling of Three Species of Stenella Dolphins in the Caribbean Basin, With Application to the Seaflower Biosphere Reserve

Get PDF
Dolphins of the genus Stenella occur in pelagic waters of both tropical and warm-temperate oceans. Three species, the Atlantic spotted dolphin (Stenella frontalis), the pantropical spotted dolphin (S. attenuata), and the spinner dolphin (S. longirostris) are abundant worldwide, but in the Caribbean Basin they have been poorly studied and information on their distribution patterns is scarce. Specifically, in Colombia’s remote Seaflower Biosphere Reserve (SFBR) S. attenuata has been reported occasionally, but S. frontalis and S. longirostris have never been recorded before. To address this information gap, an ecological niche modeling approach was used to determine the potential distribution patterns of these three dolphin species in the region. Records of these species for the Caribbean Basin were compiled, including both published and unpublished data. Environmental information, including bathymetry, bathymetric slope, distance to shore, sea surface temperature, sea surface salinity, and chlorophyll-a concentration was gathered from public databases (MARSPEC and Bio-ORACLE) in raster format. The maximum entropy algorithm (Maxent) for modeling species’ geographic distributions with presence-only data was used. After filtering the data, 210 records of S. attenuata, 204 of S. frontalis, and 80 of S. longirostris were used to run models. The best configuration for each model was chosen based on the ΔAICc criterion. For all three species, the final ecological niche models returned AUC test values higher than 0.8, indicating satisfactory model performance. The resulting potential distribution maps suggested that areas closest to continental shorelines of the Caribbean Basin and surrounding islands had the highest environmental suitability for all species (>70%). All models reported high environmental suitability for S. attenuata and S. longirostris in the SFBR, mainly in the southernmost part surrounding San Andrés and Providence Archipelago. Assessment of niche overlap from the predictions of species distributions using the similarity statistic and pairwise map overlap indicated that S. frontalis and S. longirostris had niches slightly more similar in comparison to S. attenuata. As this was a first effort to fill a gap in our understanding of the distribution of species in the genus Stenella in the Caribbean Basin, further studies are necessary using both niche modeling and biological/ecological approaches
