    Structural determinants of specific DNA-recognition by the THAP zinc finger

    Human THAP1 is the prototype of a large family of cellular factors sharing an original THAP zinc-finger motif responsible for DNA binding. Human THAP1 regulates endothelial cell proliferation and G1/S cell-cycle progression, through modulation of pRb/E2F cell-cycle target genes including rrm1. Recently, mutations in THAP1 have been found to cause DYT6 primary torsion dystonia, a human neurological disease. We report here the first 3D structure of the complex formed by the DNA-binding domain of THAP1 and its specific DNA target (THABS) found within the rrm1 target gene. The THAP zinc finger uses its double-stranded ÎČ-sheet to fill the DNA major groove and provides a unique combination of contacts from the ÎČ-sheet, the N-terminal tail and surrounding loops toward the five invariant base pairs of the THABS sequence. Our studies reveal unprecedented insights into the specific DNA recognition mechanisms within this large family of proteins controlling cell proliferation, cell cycle and pluripotency

    A Fixed Point Semantics for the ATMS

    In [CTW90], the ATMS resolution mechanisms have been formalized, using the concept of DK-tree. We propose in this paper a new and simple approach based on the fixed point theory and an original demonstration of the completeness of these mechanisms. We first introduce the basic definitions of terms, sets and functions which are used in the paper and build a lattice. Then, the basic formal tools to describe the ATMS mechanisms are introduced: The notion of DK-Clash (in fact, the inference rule), and a monotonic function, which represents the deduction process and whose least fixed point corresponds to the output of an ATMS. We prove the soundness and the completeness (for specific clauses) of these mechanisms. This demonstration brings to light an original and interesting property, whose applications are discussed at the end of the paper. En tĂȘte des ventes sur Amazon, le premier tome des MĂ©moires de Jean-Marie Le Pen a Ă©tĂ© Ă©puisĂ© avant sa mise en vente dans les librairies prĂ©vue ce jeudi, et un nouveau tirage de 50 000 exemplaires a Ă©tĂ© lancĂ© selon son Ă©diteur. IntitulĂ© "Fils de la nation", ce premier volume de l'ancien prĂ©sident et cofondateur du Front national alterne souvenirs familiaux, rĂšglements de comptes et testament politique. Ainsi au fil des 450 pages, l’eurodĂ©putĂ© se raconte de sa naissance en Bretagne en 1928 Ă  la crĂ©ation du FN en 1972. Il revient sur diffĂ©rentes rencontres politiques et sur plusieurs Ă©pisodes, notamment la Seconde Guerre mondiale, les guerres d’Indochine et d’AlgĂ©rie, en passant par la perte de son Ɠil. Il s'attarde aussi sur ses thĂšmes de prĂ©dilection : son anticommunisme viscĂ©ral, sa haine du gĂ©nĂ©ral de Gaulle, sa dĂ©nonciation de "l'invasion" migratoire ou sa volontĂ© de rĂ©habilitation du marĂ©chal PĂ©tain.     The Sm-like RNA chaperone Hfq mediates transcription antitermination at Rho-dependent terminators.

    International audienceIn Escherichia coli, the essential motor protein Rho promotes transcription termination in a tightly controlled manner that is not fully understood. Here, we show that the general post-transcriptional regulatory protein Hfq associates with Rho to regulate Rho function. The Hfq:Rho complex can be further stabilized by RNA bridging both factors in a configuration that inhibits the ATP hydrolysis and duplex unwinding activities of Rho and that mediates transcription antitermination at Rho-dependent terminators in vitro and in vivo. Antitermination at a prototypical terminator (λtR1) requires Hfq binding to an A/U-rich transcript region directly upstream from the terminator. Antitermination is modulated by trans-acting factors (NusG or nucleic acid competitors) that affect Hfq association with Rho or RNA. These data unveil a new Hfq function and a novel transcription regulatory mechanism with potentially important implications for bacterial RNA metabolism, gene silencing, and pathogenicity
