    International audienceThe Au-Ag epithermal mineralization of the Shila Cordillera is dated at about 10.7 Ma (K/Ar on adularia). The vein system is characterized by the association of a major east-west vein and N120-135°E secondary fractures. The strike-slip faults controlling the veins indicate an initial NE-SW to ENE-WSW shortening direction, which is compatible with that generally accepted for this period. These structures were reopened during a second phase and channelized mineralizing fluids, the circulation of which may have began at the end of stage 1.Les minéralisations épithermales à Au-Ag de la Cordillera Shila sont datées à environ 10,7 Ma (K/Ar sur adulaire). Le système de veines est caractérisé par l'association entre une veine principale sensiblement est-ouest et des fractures satellites N120 à N135°E.     EU-Raw Materials Intelligence Capacity Platform (or EU-RMICP) integrates metadata on data sources related to primary and secondary mineral resources and brings the end users an expertise on the methods and tools used in mineral intelligence. The system is capable of bringing relevant user ‘answers’ of the type 'how to proceed for …' on almost any question related to mineral resources, on the whole supply chain, from prospecting to recycling, taking into account the environmental, political and social dimensions. EU-RMICP is based on an ontology of the domain of mineral resources (coupled with more generic cross-functional ontologies, relative to commodities, time and space), which represents the domain of the questions of the users (experts and non-experts). The user navigates in the ontology by using a Dynamic Graph of Decision (DDG), which allows him/her to discover the solutions which he/she is looking for without having to formulate any question. The system is coupled with a 'RDF Triple Store' (a database storing the ontologies), factSheets, doc-Sheets and flowSheets (i.e., specific formatted forms) related to methods and documentation, scenarios and metadata.JRC.B.6-Digital Econom

    The epithermal Shila-Paula Au–Ag district is characterized by numerous veins hosted in Tertiary volcanic rocks of the Western Cordillera (southern Peru). Field studies of the ore bodies reveal a systematic association of a main E–W vein with secondary N55–60°W veins—two directions that are also reflected by the orientation of fluid-inclusion planes in quartz crystals of the host rock. In areas where this pattern is not recognized, such as the Apacheta sector, vein emplacement seems to have been guided by regional N40°E and N40°W fractures. Two main vein-filling stages are identified. stage 1 is a quartz–adularia–pyrite–galena–sphalerite–chalcopyrite–electrum–Mn silicate–carbonate assemblage that fills the main E–W veins. stage 2, which contains most of the precious-metal mineralization, is divided into pre-bonanza and bonanza substages. The pre-bonanza substage consists of a quartz–adularia–carbonate assemblage that is observed within the secondary N45–60°W veins, in veinlets that cut the stage 1 assemblage, and in final open-space fillings. The two latter structures are finally filled by the bonanza substage characterized by a Fe-poor sphalerite–chalcopyrite–pyrite–galena–tennantite–tetrahedrite–polybasite–pearceite–electrum assemblage. The ore in the main veins is systematically brecciated, whereas the ore in the secondary veins and geodes is characteristic of open-space crystallization. Microthermometric measurements on sphalerite from both stages and on quartz and calcite from stage 2 indicate a salinity range of 0 to 15.5 wt% NaCl equivalent and homogenization temperatures bracketed between 200 and 330°C. Secondary CO2-, N2- and H2S-bearing fluid inclusions are also identified. The age of vein emplacement, based on 40Ar/39Ar ages obtained on adularia of different veins, is estimated at around 11 Ma, with some overlap between adularia of stage 1 (11.4±0.4 Ma) and of stage 2 (10.8±0.3 Ma). A three-phase tectonic model has been constructed to explain the vein formation. Phase 1 corresponds to the assumed development of E–W sinistral shear zones and associated N60°W cleavages under the effects of a NE–SW shortening direction that is recognized at Andean scale. These structures contain the stage 1 ore assemblage that was brecciated during ongoing deformation. Phase 2 is a reactivation of earlier structures under a NW–SE shortening direction that allowed the reopening of the preexisting schistosity and the formation of scarce N50°E-striking S2-cleavage planes filled by the stage 2 pre-bonanza minerals. Phase 3 coincides with the bonanza ore emplacement in the secondary N45–60°W veins and also in open-space in the core of the main E–W veins. Our combined tectonic, textural, mineralogical, fluid-inclusion, and geochronological study presents a complete model of vein formation in which the reactivation of previously formed tectonic structures plays a significant role in ore formation

    This Report presents the recommendations arising from the ProSUM Project. It contains recommendations designed to continue to improve the knowledge base for secondary raw materials with the overarching objective of increasing recycling and supply of such materials. It specifically addresses data availability, data quality, data harmonisation, data structure and data presentation.The report contains a complete list of recommendations arising from the work undertaken in the project covering the ‘urban mine’ of electrical and electronic equipment, batteries and vehicles, their wastes, and mining wastes. It is the culmination of three years’ work which has resulted in:• The characterisation of products in terms of CRM content;• A comprehensive review and screening of all available data to characterise products;• An assessment of the factors affecting CRM content in products and the future trends for products;• A comprehensive review of existing and development of new methodologies for sampling and analysisof products;• An assessment of the current stocks of products held in households and business;• The quantification of flows of waste products not captured by national reporting on producer compliance;• A new model to quantify stocks and flows of products, their waste and material flows;• A comprehensive review and screening of all available data to characterise waste flows;• A comprehensive review of existing and development of new methodologies for sampling and analysisof wastes;• An evaluation of relevant product waste flows and mining wastes deposits;• Creation of the Urban Mine Platform (UMP) including a unified data model and code lists and meta datasystem;• Expansion of the Minerals Knowledge Data Platform (MKDP) for mining wastes;• And a new harmonised classification system to describe data in the urban mine

    ABSTRACT: GeoERA Minerals projects have produced data aimed at supporting Europe’s minerals sector and to assist the European Commission to realise its goals for raw materials. Data has been compiled on mineral occurrences and mineral provinces across Europe, in particular, areas with potential to host Critical Raw Materials. Anecdotal evidence from the minerals sector provides an indication of the likelihood of exploration leading to mine development. For every 1,000 mineral showings examined, only 100 may receive further exploration work and of those 100, only 10 may warrant more detailed sampling either through trenching, drilling or other means and of those 10 only 1 may proceed to an evaluation through a full feasibility study which itself has only 50% chance of being positive. Following this, any project for which a mine proposal is made must undergo a full evaluation and permitting by authorities including full public consultation. The proposal may or may not pass this scrutiny.     Batteries, electrical and electronic equipment, vehicles and mining waste contain both significant amounts and a large variety of raw materials, ranging from base metals to plastics, as well as precious metals and critical raw materials (CRMs). The EU is reliant on imports for many of these raw materials and aims to a Circular Economy. Securing responsible sourcing of those materials as well as increasing recycling rates is a complex societal challenge, partly because of the lack of structured data on the quantities, concentrations, trends and final whereabouts in different waste flows of these secondary raw materials in the Urban Mine in Europe. Currently, data on primary and secondary raw materials are available in Europe, but scattered amongst a variety of institutions including government agencies, universities, NGOs and industry. The aim of the ProSUM project was to provide a state of the art knowledge base, using best available data in a harmonised and updateable format, which allows the recycling industry and policymakers to make more informed investment and policy decisions to increase the supply and recycling of secondary raw materials

    Divergent Pathways in COS-7 Cells Mediate Defective Internalization and Intracellular Routing of Truncated G-CSFR Forms in SCN/AML

    Expression of truncated G-CSFR forms in patients with SCN/AML induces hyperproliferation and prolonged cell survival. Previously, we showed that ligand internalization is delayed and degradation of truncated G-CSFR forms is defective in patients with SCN/AML.In this study, we investigated the potential roles of dileucine and tyrosine-based motifs within the cytoplasmic domain of the G-CSFR in modulating ligand/receptor internalization. Using standard binding assays with radiolabeled ligand and COS-7 cells, substitutions in the dileucine motif or deletion of tyrosine residues in the G-CSFR did not alter internalization. Attachment of the transferrin receptor YTRF internalization motif to a truncated G-CSFR form from a patient with SCN/AML corrected defective internalization, but not receptor degradation suggesting that receptor internalization and degradation occur independently via distinct domains and/or processes.Our data suggest that distinct domains within the G-CSFR mediate separate processes for receptor internalization and degradation. Our findings using standard binding assays differ from recently published data utilizing flow cytometry

    Use of humanised rat basophilic leukaemia cell line RS-ATL8 for the assessment of allergenicity of Schistosoma mansoni proteins.

    BACKGROUND Parasite-specific IgE is thought to correlate with protection against Schistosoma mansoni infection or re-infection. Only a few molecular targets of the IgE response in S. mansoni infection have been characterised. A better insight into the basic mechanisms of anti-parasite immunity could be gained from a genome-wide characterisation of such S. mansoni allergens. This would have repercussions on our understanding of allergy and the development of safe and efficacious vaccinations against helminthic parasites. METHODOLOGY/PRINCIPAL FINDINGS A complete medium- to high-throughput amenable workflow, including important quality controls, is described, which enables the rapid translation of S. mansoni proteins using wheat germ lysate and subsequent assessment of potential allergenicity with a humanised Rat Basophilic Leukemia (RBL) reporter cell line. Cell-free translation is completed within 90 minutes, generating sufficient amounts of parasitic protein for rapid screening of allergenicity without any need for purification. Antigenic integrity is demonstrated using Western Blotting. After overnight incubation with infected individuals' serum, the RS-ATL8 reporter cell line is challenged with the complete wheat germ translation mixture and Luciferase activity measured, reporting cellular activation by the suspected allergen. The suitability of this system for characterization of novel S. mansoni allergens is demonstrated using well characterised plant and parasitic allergens such as Par j 2, SmTAL-1 and the IgE binding factor IPSE/alpha-1, expressed in wheat germ lysates and/or E. coli. SmTAL-1, but not SmTAL2 (used as a negative control), was able to activate the basophil reporter cell line. CONCLUSION/SIGNIFICANCE This method offers an accessible way for assessment of potential allergenicity of anti-helminthic vaccine candidates and is suitable for medium- to high-throughput studies using infected individual sera.
