    Amelanotic melanoma in dogs : a retrospective study of 35 cases (2004-2010) and immunohistochemical characterization

    Melanomas são tumores agressivos de melanócitos que ocorrem principalmente na cavidade oral, nas junções mucocutâneas e na pele de cães. Este tipo de neoplasma pode apresentar diversos graus de pigmentação melânica, incluindo total ausência (melanomas amelanóticos [MA]). Os arquivos de biópsia do SPV-UFRGS, que compreendem o período de 2004 a 2010, foram revisados e levantados os casos de neoplasias melanocíticas em cães. Realizou-se estudo retrospectivo de 35 casos de MA e caracterização pela imuno-histoquímica (IHQ). As principais raças acometidas foram o Poodle, Dachshund e Cocker Spaniel, mas o maior número de casos foi observado em cães sem raça definida (SRD). A idade média desses cães foi de 10,7 anos (variação de 5 a 18 anos) e não houve predileção por sexo. As principais localizações incluíram cavidade oral (57,1%) e dígitos (17,1%). Histologicamente, 40% dos MA foram classificados como epitelioides, 34,3% como mistos e 25,7% como fusiformes. Na avaliação IHQ, 86,6% dos casos foram positivos para a vimentina, 70% para a proteína S-100 e 56,6% para o melan-A. Os resultados obtidos neste trabalho possibilitam concluir que os cães com MA caracterizavam-se por serem velhos. A forma celular mais observada foi a epitelioide. Devido a pouca diferenciação desses tumores, ressalta-se a importância da realização do painel imuno-histoquímico, sobretudo da proteína S-100, que apresentou melhor marcação que o melan-A.Melanomas are aggressive tumors of melanocytes. They are common in dogs and involve mainly the oral cavity, mucocutaneous junction, and skin. Furthermore, these tumors could be highly pigmented or lack pigment. The biopsies archives from SPV-UFRGS, 2004 to 2010, were retrieved and melanocytic neoplasms in dogs were revised. A retrospective study of 35 cases of amelanotic melanomas (AM) was performed, also immunohistochemistry (IHC) characterization was evaluated. The dogs more affected were mixed breed followed by Poodle, Dachshund and Cocker Spaniel. The average age of the dogs was 10.7 years (5-18 years in age) and there was no sex predilection. The locations of the neoplasms were the oral cavity (57.1%) and digits (17.1%). Histologically, 40% were classified as epithelioid, 34.3% mixed and 25.7% spindle. The positive immunostaining for vimentin, S-100 protein and melan-A were 86.6%, 70%, and 56.6% respectively. These results indicated the most affected dogs with AM were elderly. Epiteliod classification was the most observed histologically. It is important to perform IHC, due to lacking of differentiation of AM, mainly, anti S-100 protein that showed to be the best option of positive marker, even better to Melan-A

    Prevalência de asma e fatores de risco em escolares da cidade de São Paulo

    OBJECTIVE: To assess asthma prevalence and potential risk factors associated. METHODS: Cross-sectional study part of the International Study of Asthma and Allergies in Childhood. A total of 561 schoolchildren aged 6-7 years from 35 public schools in the city of São Paulo (Southeastern Brazil) were drawn to participate in the study, in 2002. The sample consisted of 168 asthmatic and 393 non-asthmatic children who answered a questionnaire comprising 33 questions on personal, family and environmental information. The association between asthma and the risk factors studied was assessed by logistic regression analysis at a 5% statistical significance. RESULTS: Among the schoolchildren studied, 31.2% reported wheezing in the 12 months preceding the interview. The following risk factors were significantly associated with asthma: male gender (OR=2.4; 95% CI: 1.4;4.2), maternal smoking in the child's first year of life (OR=2.0; 95% CI: 1.1;3.8), eczema on characteristic body areas (OR=3.0; 95% CI: 1.2;7.6) and rhinoconjunctivitis (OR=2.4; 95% CI: 1.2;4.8). CONCLUSIONS: Asthma prevalence in the study area was high and the risk factors identified were male gender, rhinoconjunctivitis in last year, maternal smoking in the child's first year of life and eczema on characteristic body areas.OBJETIVO: Analisar a prevalência de asma e possíveis fatores de risco associados. CONCLUSÕES: A prevalência de asma na região estudada foi elevada e os fatores de risco relacionados foram: sexo masculino, sintomas de rinoconjuntivite no último ano, mãe fumante no primeiro ano de vida e presença de eczema em locais característicos

    The Lipid Mediator Resolvin D1 Reduces the Skin Inflammation and Oxidative Stress Induced by UV Irradiation in Hairless Mice

    UV irradiation-induced oxidative stress and inflammation contribute to the development of skin diseases. Therefore, targeting oxidative stress and inflammation might contribute to reduce skin diseases. Resolvin D1 (RvD1) is a bioactive metabolite generated during inflammation to actively orchestrate the resolution of inflammation. However, the therapeutic potential of RvD1 in UVB skin inflammation remains undetermined, which was, therefore, the aim of the present study. The intraperitoneal treatment with RvD1 (3-100 ng/mouse) reduced UVB irradiation-induced skin edema, myeloperoxidase activity, matrix metalloproteinase 9 activity, and reduced glutathione depletion with consistent effects observed with the dose of 30 ng/mouse, which was selected to the following experiments. RvD1 inhibited UVB reduction of catalase activity, and hydroperoxide formation, superoxide anion production, and gp91phox mRNA expression. RvD1 also increased the Nrf2 and its downstream targets NQO1 and HO-1 mRNA expression. Regarding cytokines, RvD1 inhibited UVB-induced production of IL-1β, IL-6, IL-33, TNF-α, TGF-β, and IL-10. These immuno-biochemical alterations by RvD1 treatment had as consequence the reduction of UVB-induced epidermal thickness, sunburn and mast cell counts, and collagen degradation. Therefore, RvD1 inhibited UVB-induced skin oxidative stress and inflammation, rendering this resolving lipid mediator as a promising therapeutic agent

    INTOXICAÇÃO POR NITRATO/NITRITO NO OESTE DE SANTA CATARINA - ESTUDO RETROSPECTIVO (2013-2017)

    A intoxicação por nitrato/nitrito é uma enfermidade comum no estado de Santa Catarina, e que leva a morte súbita em bovinos. Trata-se de um distúrbio ocasionado pela utilização excessiva de adubação nitrogenada nas pastagens e condições climáticas adversas, como diminuição de luminosidade ou déficit hídrico, levando ao acúmulo de nitrato na pastagem. Este trabalho teve como objetivo relatar a ocorrência dos casos de intoxicação por nitrato/nitrito em bovinos na região oeste de Santa Catarina, diagnosticados pelo Laboratório de Patologia Veterinária (LPV) do Instituto Federal Catarinense – Campus Concórdia. Entre os anos de 2013 e 2017, foram diagnosticados por necropsia 10 surtos de intoxicações por nitrato/nitrito. De um total de 641 bovinos necropsiados pelo LPV nesse período, 19 correspondem a essa intoxicação, representando 2,96% da casuística de necropsias e 2,20% da casuística total de diagnósticos (necropsias e histopatológico). Ainda que seja um número expressivo de causa mortis, acredita-se que existam muitos outros casos não diagnosticados pelo LPV, visto que nem todos os animais são remetidos para necropsia. Conclui-se que as intoxicações por nitrato/nitrito são expressiva causa de morte nos rebanhos bovinos da região, devendo serem adotadas medidas de controle e prevenção. Palavras-chave: metemoglobina, patologia, bovino, diagnóstico, necropsia. 

    DOENÇAS DIAGNOSTICADAS PELO LABORATÓRIO DE PATOLOGIA VETERINÁRIA NO QUINQUÊNIO 2013-2017

    Este trabalho apresenta a casuística de diagnósticos nos anos de 2013 a 2017 do Laboratório de Patologia Veterinária do IFC Campus Concórdia. Foram 865 diagnósticos em bovinos, 131 em ovinos e 367 em suínos, totalizando 1363. Destes, 1000 (80,4%) foram através de necropsias e 363 (19,6%) através de amostras formolizadas enviadas por veterinários. Em bovinos as doenças mais incidentes foram miosite clostridial com 34 casos (3,8%); babesiose com 31 casos cada (3,6%); leucose enzoótica bovina com 28 casos (3,2%); anaplasmose com 27 casos (3,1%); hemoncose com 26 casos (3,0%); intoxicação por Pteridium arachnoideumcom 26 casos (3,0%); peritonite com 23 casos (2,7%) e endocardite com 23 casos (2,7%). Já em ovinos hemoncose com 28 casos (21,4%) e suínos doença de Glässer com 27 casos (7,3%) e circovirose com 22 casos (6,0%)

    INTOXICAÇÃO POR PTERIDIUM ARACHNOIDEUM NO OESTE DE SANTA CATARINA - ESTUDO RETROSPECTIVO (2013-2017)

    Samambaia (Pteridium arachnoideum) é uma das plantas tóxicas mais importantes na criação de bovinos de todo o Brasil. O objetivo desse trabalho foi descrever os diagnósticos de intoxicação por samambaia, em bovinos na região oeste de Santa Catarina, realizados pelo Laboratório de Patologia Veterinária (LPV) do IFC – Campus Concórdia, no período entre 2013 e 2017. No período, foram realizadas 641 necropsias de bovinos, sendo que 22 dessas tiveram o diagnóstico de intoxicação por samambaia, totalizando 3,4% da casuística total de necropsias em bovinos. Foram 13 casos de síndrome hemorrágica aguda, seis casos de carcinomas de células escamosas na base da língua, esôfago ou rúmen, e três casos de hematúria enzoótica bovina. Apesar dos inúmeros trabalhos científicos a respeito da toxicidade do P. arachnoideum, o número de diagnósticos dessa intoxicação é alto na região, e a planta ainda é facilmente encontrada. A melhor forma de diminuir as intoxicações é o adequado manejo das pastagens e a correta adubação do solo, evitando com que a planta esteja disponível ao consumo dos animais, porém essa informação precisa ser difundida aos produtores rurais.  Palavras-chave: diagnóstico, patologia, samambaia, hematúria, carcinoma.

    Characterization of the degree of food processing in the European Prospective Investigation into Cancer and Nutrition: Application of the Nova classification and validation using selected biomarkers of food processing

    Background: Epidemiological studies have demonstrated an association between the degree of food processing in our diet and the risk of various chronic diseases. Much of this evidence is based on the international Nova classification system, which classifies food into four groups based on the type of processing: (1) Unprocessed and minimally processed foods, (2) Processed culinary ingredients, (3) Processed foods, and (4) “Ultra-processed” foods (UPF). The ability of the Nova classification to accurately characterise the degree of food processing across consumption patterns in various European populations has not been investigated so far. Therefore, we applied the Nova coding to data from the European Prospective Investigation into Cancer and Nutrition (EPIC) in order to characterize the degree of food processing in our diet across European populations with diverse cultural and socio-economic backgrounds and to validate this Nova classification through comparison with objective biomarker measurements. Methods: After grouping foods in the EPIC dataset according to the Nova classification, a total of 476,768 participants in the EPIC cohort (71.5% women; mean age 51 [standard deviation (SD) 9.93]; median age 52 [percentile (p)25– p75: 58–66] years) were included in the cross-sectional analysis that characterised consumption patterns based on the Nova classification. The consumption of food products classified as different Nova categories were compared to relevant circulating biomarkers denoting food processing, measured in various subsamples (N between 417 and 9,460) within the EPIC cohort via (partial) correlation analyses (unadjusted and adjusted by sex, age, BMI and country). These biomarkers included an industrial transfatty acid (ITFA) isomer (elaidic acid; exogenous fatty acid generated during oil hydrogenation and heating) and urinary 4-methyl syringol sulfate (an indicator for the consumption of smoked food and a component of liquid smoke used in UPF). Results: Contributions of UPF intake to the overall diet in % grams/day varied across countries from 7% (France) to 23% (Norway) and their contributions to overall % energy intake from 16% (Spain and Italy) to >45% (in the UK and Norway). Differences were also found between sociodemographic groups; participants in the highest fourth of UPF consumption tended to be younger, taller, less educated, current smokers, more physically active, have a higher reported intake of energy and lower reported intake of alcohol. The UPF pattern as defined based on the Nova classification (group 4;% kcal/day) was positively associated with blood levels of industrial elaidic acid (r = 0.54) and 4-methyl syringol sulfate (r = 0.43). Associations for the other 3 Nova groups with these food processing biomarkers were either inverse or non-significant (e.g., for unprocessed and minimally processed foods these correlations were –0.07 and –0.37 for elaidic acid and 4-methyl syringol sulfate, respectively). Conclusion: These results, based on a large pan-European cohort, demonstrate sociodemographic and geographical differences in the consumption of UPF. Furthermore, these results suggest that the Nova classification can accurately capture consumption of UPF, reflected by stronger correlations with circulating levels of industrial elaidic acid and a syringol metabolite compared to diets high in minimally processed foods
