    Trust-Based Social Behaviour in Role-Playing Games

    Although previous artificial intelligence research has done much to advance video games, not as much has been done to integrate that research into commercially viable titles. In this study, the game Barrel Smasher was developed, demonstrating a socially aware model of Non-Player Characters based on internal trust scores of other characters. The game allows the player to interact with these characters through dialogue, and it provides elements of Role-Playing Games like quests, items, and combat. The result is a game that combines social interactions and other forms of gameplay into a single, connected system. In doing so, the game creates interactive quest progression with more variation than is found in traditional quest systems

    Polymorphism in glutathione S-transferase P1 is associated with susceptibility to chemotherapyinduced leukemia

    Glutathione S-transferases (GSTs) detoxify potentially mutagenic and toxic DNA-reactive electrophiles, including metabolites of several chemotherapeutic agents, some of which are suspected human carcinogens. Functional polymorphisms exist in at least three genes that encode GSTs, including GSTM1, GSTT1, and GSTP1. We hypothesize, therefore, that polymorphisms in genes that encode GSTs alter susceptibility to chemotherapy-induced carcinogenesis, specifically to therapy-related acute myeloid leukemia (t-AML), a devastating complication of long-term cancer survival. Elucidation of genetic determinants may help to identify individuals at increased risk of developing t-AML. To this end, we have examined 89 cases of t-AML, 420 cases of de novo AML, and 1,022 controls for polymorphisms in GSTM1, GSTT1, and GSTP1. Gene deletion of GSTM1 or GSTT1 was not specifically associated with susceptibility to t-AML. Individuals with at least one GSTP1 codon 105 Val allele were significantly over-represented in t-AML cases compared with de novo AML cases [odds ratio (OR), 1.81; 95% confidence interval (CI), 1.11–2.94]. Moreover, relative to de novo AML, the GSTP1 codon 105 Val allele occurred more often among t-AML patients with prior exposure to chemotherapy (OR, 2.66; 95% CI, 1.39–5.09), particularly among those with prior exposure to known GSTP1 substrates (OR, 4.34; 95% CI, 1.43–13.20), and not among those t-AML patients with prior exposure to radiotherapy alone (OR,1.01; 95% CI, 0.50–2.07). These data suggest that inheritance of at least one Val allele at GSTP1 codon 105 confers a significantly increased risk of developing t-AML after cytotoxic chemotherapy, but not after radiotherapy

    Dielectric characterization of Plasmodium falciparum infected red blood cells using microfluidic impedance cytometry

    Although malaria is the world’s most life-threatening parasitic disease, there is no clear understanding of how certain biophysical properties of infected cells change during the malaria infection cycle. In this article, we use microfluidic impedance cytometry to measure the dielectric properties of Plasmodium falciparum-infected red blood cells (i-RBCs) at specific time-points during the infection cycle. Individual parasites were identified within i-RBCs using Green Fluorescent Protein (GFP) emission. The dielectric properties of cell sub-populations were determined using the multi-shell model. Analysis showed that the membrane capacitance and cytoplasmic conductivity of i-RBCs increased along the infection time-course, due to membrane alterations caused by parasite infection. The volume ratio occupied by the parasite was estimated to vary from <10% at earlier stages, to ~90% at later stages. This knowledge could be used to develop new label-free cell sorting techniques for sample pre-enrichment, improving diagnosis

    Electron impact excitations of S2 molecules

    Low-energy electron impact excitations of S_2 molecules are studied using the fixed-bond R-matrix method based on state-averaged complete active space SCF orbitals. Integral cross sections are calculated for elastic electron collision as well as impact excitation of the 7 lowest excited electronic states. Also, differential cross sections are obtained for elastic collision and excitation of the a^1 Delta_g, b^1 Sigma_g^+ and B^3 Sigma_u^- states.     The SLoWPoKES Catalog of Low-mass Ultra-wide Binaries: A Cool Stars Resource for Testing Fundamental Properties and for Constraining Binary Formation Theory

    We present results from the Sloan Low-mass Wide Pairs of Kinematically Equivalent Stars (SLoWPoKES) catalog of ultra-wide (10^3-10^5.5 AU), low-mass (K5-M7) common proper motion binaries. We constructed a Galactic model, based on empirical stellar number density and 3D velocity distributions, to select bona fide pairs with probability of chance alignment <5%, making SLoWPoKES an efficient sample for followup observations. Our initial catalog contains 1342 disk dwarf, subdwarf, and white dwarf-red dwarf systems and is the largest collection of low-mass, wide binaries ever assembled. The diversity---in mass, metallicity, age, and evolutionary states---of SLoWPoKES pairs makes it a valuable resource of coeval laboratories to examine and constrain the physical properties of low-mass stars. SLoWPoKES pairs show signatures of two (or more) formation modes in the distribution of the physical separation and higher-order multiplicity. Neither dynamical dissipation of primordial triples/quadruples or dynamical capture of ejected stars can explain the observed populations by itself. We use followup spectroscopic observations to recalibrate the metallicity-sensitive {\zeta}_(TiO/CaH) index by assuming that both members of the binary system have the same composition. Our new formulation is a significantly better tracer of absolute metallicity, particularly for the early-type M dwarfs.     Prevalence and characterization of human mecC methicillin-resistant Staphylococcus aureus isolates in England

    Objectives: There are limited data available on the epidemiology and prevalence of methicillin-resistant Staphylococcus aureus (MRSA) in the human population that encode the recently described mecA homologue, mecC. To address this knowledge gap we undertook a prospective prevalence study in England to determine the prevalence of mecC among MRSA isolates.&lt;p&gt;&lt;/p&gt; Patients and methods: Three hundred and thirty-five sequential MRSA isolates from individual patients were collected from each of six clinical microbiology laboratories in England during 2011–12. These were tested by PCR or genome sequencing to differentiate those encoding mecA and mecC. mecC-positive isolates were further characterized by multilocus sequence typing, spa typing, antimicrobial susceptibility profile and detection of PBP2a using commercially available kits.&lt;p&gt;&lt;/p&gt; Results: Nine out of the 2010 MRSA isolates tested were mecC positive, indicating a prevalence among MRSA in England of 0.45% (95% CI 0.24%–0.85%). The remainder were mecA positive. Eight out of these nine mecC MRSA isolates belonged to clonal complex 130, the other being sequence type 425. Resistance to non-β-lactam antibiotics was rare among these mecC MRSA isolates and all were phenotypically identified as MRSA using oxacillin and cefoxitin according to BSAC disc diffusion methodology. However, all nine mecC isolates gave a negative result using three different commercial PBP2a detection assays. Conclusions: mecC MRSA are currently rare among MRSA isolated from humans in England and this study provides an important baseline prevalence rate to monitor future changes, which may be important given the increasing prevalence of mecC MRSA reported in Denmark.&lt;p&gt;&lt;/p&gt

    Probing the anomalous extinction of four young star clusters: the use of colour-excess, main sequence fitting and fractal analysis

    Four young star clusters were studied in order to characterize their anomalous extinction or variable reddening that could be due to a possible contamination by dense clouds or circumstellar effects. The extinction law (Rv) was evaluated by adopting two methods: (i) the use of theoretical expressions based on the colour-excess of stars with known spectral type, and (ii) the analysis of two-colour diagrams, where the slope of observed colours distribution is compared to the normal distribution. An algorithm to reproduce the zero age main sequence (ZAMS) reddened colours was developed in order to derive the average visual extinction (Av) that provides the best fitting of the observational data. The structure of the clouds was evaluated by means of statistical fractal analysis, aiming to compare their geometric structure with the spatial distribution of the cluster members. The cluster NGC 6530 is the only object of our sample showing anomalous extinction. In average, the other clusters are suffering normal extinction, but several of their members, mainly in NGC 2264, seem to have high Rv, probably due to circumstellar effects. The ZAMS fitting provides Av values that are in good agreement with those found in the literature. The fractal analysis shows that NGC 6530 has a centrally concentrated distribution of stars that is different of the sub-structures found in the density distribution of the cloud projected in the Av map, suggesting that the original cloud has been changed with the cluster formation. On the other hand, the fractal dimension and the statistical parameters of Berkeley 86, NGC 2244, and NGC 2264 indicate a good cloud-cluster correlation, when compared to other works based on artificial distribution of points.Comment: 13 pages, 7 figure
