    Floral Variation and Breeding System in Distylous and Homostylous Species of Clonal Aquatic 'Nymphoides' (Menyanthaceae)

    This study focuses on floral variation and the breeding system of two species of the clonal aquatic 'Nymphoides' (Menyanthaceae), in particular, the phenomena of distyly, loss of sex and homostyly. Distyly is a floral design that promotes efficient pollen transfer while reducing self-interference and self-pollination. Populations of distylous species contain two floral morphs that differ in the reciprocal positioning of the sex organs; i.e. reciprocal herkogamy. Distylous species usually possess a dimorphic incompatibility system in which only pollination between flowers of different morphs produce seed. There are, however, atypical features of distyly, which provide excellent examples to study the maintenance or evolutionary transition of this breeding system. 'Nymphoides montana' is a perennial clonal aquatic species, native to Australia, found in shallow water on the edges of wetlands. Selective factors that maintain distyly were sought by examining floral morphologies, compatibility relationships, morph frequencies and reproductive success in three 'N. montana' populations

    Integrating value of research into NCI Clinical Trials Cooperative Group research review and prioritization: A pilot study

    BackgroundThe Institute of Medicine has called for approaches to help maximize the return on investments (ROI) in cancer clinical trials. Value of Research (VOR) is a health economics technique that estimates ROI and can inform research prioritization. Our objective was to evaluate the impact of using VOR analyses on the clinical trial proposal review process within the SWOG cancer clinical trials consortium.MethodsWe used a previously developed minimal modeling approach to calculate VOR estimates for 9 phase II/III SWOG proposals between February 2015 and December 2016. Estimates were presented to executive committee (EC) members (N = 12) who determine which studies are sent to the National Cancer Institute for funding consideration. EC members scored proposals from 1 (best) to 5 based on scientific merit and potential impact before and after receiving VOR estimates. EC members were surveyed to assess research priorities, proposal evaluation process satisfaction, and the VOR process.ResultsValue of Research estimates ranged from −2.1Bto2.1B to 16.46B per proposal. Following review of VOR results, the EC changed their score for eight of nine proposals. Proposal rankings were different in pre‐ vs postscores (P value: 0.03). Respondents had mixed views of the ultimate utility of VOR for their decisions with most supporting (42%) or neutral (41%) to the idea of adding VOR to the evaluation process.ConclusionsThe findings from this pilot study indicate use of VOR analyses may be a useful adjunct to inform proposal reviews within NCI Cooperative Clinical Trials groups.The Instiztute of Medicine has called for approaches to help maximize the return on investments in cancer clinical trials.     Island questions: the chronology of the Brochtorff Circle at Xagħra, Gozo, and its significance for the Neolithic sequence on Malta

    Bayesian chronological modelling of radiocarbon dates from the Brochtorff Circle at Xagħra, Gozo, Malta (achieved through the ToTL and FRAGSUS projects), provides a more precise chronology for the sequence of development and use of a cave complex. Artefacts show that the site was in use from the Żebbuġ period of the late 5th/early 4th millennium cal BC to the Tarxien Cemetery phase of the later 3rd/early 2nd millennia cal BC. Absolutely dated funerary activity, however, starts with a small rock-cut tomb, probably in use in the mid to late fourth millennium cal BC, in the Ġgantija period. After an interval of centuries, burial resumed on a larger scale, probably in the 30th century cal BC, associated with Tarxien cultural material, with the use of the cave for collective burial and other depositions, with a series of structures, most notably altar-like settings built from massive stone slabs, which served to monumentalise the space. This process continued at intervals until the deposition of the last burials, probably in the 24th century cal BC; ceremonial activity may have ended at this time or a little later, to be followed by occupation in the Tarxien Cemetery period. The implications for the development of Neolithic society on Malta are discussed, as well as the changing character of Neolithic Malta in comparison to contemporary communities in Sicily, peninsular Italy and southern Iberia. It is argued that underground settings and temples on Malta may have served to reinforce locally important values of cooperation and consensus, against a wider tide of differentiation and accumulation, but that there could also have been increasing control of the treatment of the dead through time.     Tn-Seq reveals hidden complexity in the utilization of host-derived glutathione in \u3cem\u3eFrancisella tularensis\u3c/em\u3e

    Host-derived glutathione (GSH) is an essential source of cysteine for the intracellular pathogen Francisella tularensis. In a comprehensive transposon insertion sequencing screen, we identified several F. tularensis genes that play central and previously unappreciated roles in the utilization of GSH during the growth of the bacterium in macrophages. We show that one of these, a gene we named dptA, encodes a proton-dependent oligopeptide transporter that enables growth of the organism on the dipeptide Cys-Gly, a key breakdown product of GSH generated by the enzyme γ-glutamyltranspeptidase (GGT). Although GGT was thought to be the principal enzyme involved in GSH breakdown in F. tularensis, our screen identified a second enzyme, referred to as ChaC, that is also involved in the utilization of exogenous GSH. However, unlike GGT and DptA, we show that the importance of ChaC in supporting intramacrophage growth extends beyond cysteine acquisition. Taken together, our findings provide a compendium of F. tularensis genes required for intracellular growth and identify new players in the metabolism of GSH that could be attractive targets for therapeutic intervention

    A20, a modulator of smooth muscle cell proliferation and apoptosis, prevents and induces regression of neointimal hyperplasia

    A20 is a NF‐κB‐dependent gene that has dual anti‐inflammatory and antiapoptotic functions in endothelial cells (EC). The function of A20 in smooth muscle cells (SMC) is unknown. We demonstrate that A20 is induced in SMC in response to inflammatory stimuli and serves an anti‐inflammatory function via blockade of NF‐κB and NF‐κB‐dependent proteins ICAM‐1 and MCP‐1. A20 inhibits SMC proliferation via increased expression of cyclin‐dependent kinase inhibitors p21waf1 and p27kip1. Surprisingly, A20 sensitizes SMC to cytokine‐ and Fas‐mediated apoptosis through a novel NO‐dependent mechanism. In vivo, adenoviral delivery of A20 to medial rat carotid artery SMC after balloon angioplasty prevents neointimal hyperplasia by blocking SMC proliferation and accelerating re‐endothelialization, without causing apoptosis. However, expression of A20 in established neointimal lesions leads to their regression through increased apoptosis. This is the first demonstration that A20 exerts two levels of control of vascular remodeling and healing. A20 prevents neointimal hyperplasia through combined anti‐inflammatory and antiproliferative functions in medial SMC. If SMC evade this first barrier and neointima is formed, A20 has a therapeutic potential by uniquely sensitizing neointimal SMC to apoptosis.     Rate Effects on Timing, Key Velocity, and Finger Kinematics in Piano Performance

    We examined the effect of rate on finger kinematics in goal-directed actions of pianists. In addition, we evaluated whether movement kinematics can be treated as an indicator of personal identity. Pianists' finger movements were recorded with a motion capture system while they performed melodies from memory at different rates. Pianists' peak finger heights above the keys preceding keystrokes increased as tempo increased, and were attained about one tone before keypress. These rate effects were not simply due to a strategy to increase key velocity (associated with tone intensity) of the corresponding keystroke. Greater finger heights may compensate via greater tactile feedback for a speed-accuracy tradeoff that underlies the tendency toward larger temporal variability at faster tempi. This would allow pianists to maintain high temporal accuracy when playing at fast rates. In addition, finger velocity and accelerations as pianists' fingers approached keys were sufficiently unique to allow pianists' identification with a neural-network classifier. Classification success was higher in pianists with more extensive musical training.     Oxygen isotope dendrochronology of Llwyn Celyn; One of the oldest houses in Wales

    We report the application of oxygen isotope dendrochronology to date a high-status and remarkably unaltered late medieval hall house on the eastern border of South Wales. The oak timbers have either short and complacent ring series, or very strong growth disturbance, and none were suitable for ring-width dendrochronology. By using stable oxygen isotopes from the latewood cellulose, rather than ring widths, it was possible to cross-match and date all 14 timber samples and to provide felling dates related to several phases of building. The hall and solar cross-wing were constructed shortly after 1420CE, which is remarkably early. The house was upgraded using timbers felled in the winter of 1695/6CE by ceiling over of the hall and inserting a chimney. A separate small domestic building was added at the same time and the addition of the kitchen is likely to be contemporaneous. A substantial beast house was added a few years before the house was refurbished, emphasising the importance of cattle as the main source of wealth. A small barn with timbers felled in spring 1843 CE was added later. Llwyn Celyn is one of the most important domestic buildings in Wales, but without the new approach none of the phases of its evolution could have been dated precisely. Oxygen isotope dendrochronology has enormous potential for dating timbers that have small numbers of rings and/or show severe growth disturbance and it works well in regions where tree growth is not strongly constrained by climate.     The ASTRO-H X-ray Observatory

    The joint JAXA/NASA ASTRO-H mission is the sixth in a series of highly successful X-ray missions initiated by the Institute of Space and Astronautical Science (ISAS). ASTRO-H will investigate the physics of the high-energy universe via a suite of four instruments, covering a very wide energy range, from 0.3 keV to 600 keV. These instruments include a high-resolution, high-throughput spectrometer sensitive over 0.3-2 keV with high spectral resolution of Delta E < 7 eV, enabled by a micro-calorimeter array located in the focal plane of thin-foil X-ray optics; hard X-ray imaging spectrometers covering 5-80 keV, located in the focal plane of multilayer-coated, focusing hard X-ray mirrors; a wide-field imaging spectrometer sensitive over 0.4-12 keV, with an X-ray CCD camera in the focal plane of a soft X-ray telescope; and a non-focusing Compton-camera type soft gamma-ray detector, sensitive in the 40-600 keV band.
