    Model transcriptional networks with continuously varying expression levels

    <p>Abstract</p> <p>Background</p> <p>At a time when genomes are being sequenced by the hundreds, much attention has shifted from identifying genes and phenotypes to understanding the networks of interactions among genes. We developed a gene network developmental model expanding on previous models of transcription regulatory networks. In our model, each network is described by a matrix representing the interactions between transcription factors, and a vector of continuous values representing the transcription factor expression in an individual.</p> <p>Results</p> <p>In this work we used the gene network model to look at the impact of mating as well as insertions and deletions of genes in the evolution of complexity of these networks. We found that the natural process of diploid mating increases the likelihood of maintaining complexity, especially in higher order networks (more than 10 genes). We also show that gene insertion is a very efficient way to add more genes to a network as it provides a much higher chance of developmental stability.</p> <p>Conclusions</p> <p>The continuous model affords a more complete view of the evolution of interacting genes. The notion of a continuous output vector also incorporates the reality of gene networks and graded concentrations of gene products.</p

We are introducing here the Blood Ontology, an ongoing initiative designed to serve as a controlled vocabulary for use in organizing information about blood. Para isso, será realizada uma pesquisa exploratória fundamenta-se no referencial teórico. Partindo se do princípio de que o marketing e a principal ferramenta de que dispõe o varejo mercadológico para atrair e os clientes, sugere-se a implementação de uma estratégia de marketing mediante a qual os supermercados devem desenvolver o recrutamento de pessoas para entregas de produtos em horários ociosos, ou seja, em que o fluxo de clientes nos supermercados e reduzido. Assim sendo, o consumidor ganha na medida em que não se ausenta de seu trabalho e o supermercado, ao oferecer urn serviço diversificado. Essa medida se deve ao fato de que o consumidor normalmente responde de forma positiva ao marketing principalmente no que se refere a conveniência de pregos e localização estratégica da loja varejista, mostra-se comprometido com relação aos supermercados que frequenta, de forma que, a fidelização continuará sendo um atributo que almeja alcançar serviços que visem a comodidade do consumido

    In this article we present the Educational Platform of the collaborative exhibition “Resistance Now! Strengthening and uniting indigenous cultures. Kaingang, Guarani Nhandewa and Terena”, developed by the three indigenous groups in the Midwest of the State of São Paulo in partnership with the MAE-USP. The discussion about collaborative curatorial processes in the area of education in museums contributes to extend this agenda in the field of museums. The ethical challenge of actions to expand and respect indigenous voices must be a constant search by the team of professionals, and in this sense, the inclination to listen and foster the relationship is a path in potential&nbsp; to positive results. Working in interculturality presents possibilities for the implementation of decolonial practices.Apresentamos neste artigo a Plataforma Educativa da exposição colaborativa “Resistência Já! Fortalecimento e união das culturas indígenas. Kaingang, Guarani Nhandewa e Terena”, desenvolvida pelos três grupos indígenas do centro-oeste do Estado de São Paulo em parceria com o MAE-USP. A discussão de processos curatoriais colaborativos a partir da área da educação em museus contribui para estendermos essa pauta no campo museal. O desafio ético das ações para ampliação e respeito das vozes indígenas deve ser uma busca constante da equipe de profissionais, e nesse sentido, o movimento de escuta e o fomento da relação são caminhos potenciais para resultados positivos.     Arthroscopic double- bundle reconstruction of anterior cruciate ligament using hamstring tendon grafts: fixation with two interference screws

    Surgical procedures for double-bundle reconstruction of anterior cruciate ligament, which currently use semitendinous and gracilis tendon grafts, have been described in the last decade. Most of the techniques utilize twice the hardware used in single-bundle reconstructions. We report an original anterior cruciate ligament double-bundle reconstruction technique using semitendinous and gracilis tendon grafts, maintaining their tibial bone insertions with two tibial and two femoral tunnels. A simplified and precise outside-in femoral drilling technique is utilized, and the graft fixation is made utilizing only two interference screws.Procedimentos cirúrgicos de reconstrução do ligamento cruzado anterior com duplo feixe dos tendões dos músculos semitendíneo e grácil têm sido descritos na última década. A maioria das técnicas descritas utiliza o dobro de material de síntese empregado na reconstrução com feixe único. Relatamos uma técnica original para a reconstrução do ligamento cruzado anterior com duplo feixe, na qual mantemos as inserções tibiais dos tendões dos músculos semitendíneo e grácil e realizamos dois túneis tibiais e dois túneis femorais.     Green Extraction Processes for Complex Samples from Vegetable Matrices Coupled with On-Line Detection System: A Critical Review

    The detection of analytes in complex organic matrices requires a series of analytical steps to obtain a reliable analysis. Sample preparation can be the most time-consuming, prolonged, and error-prone step, reducing the reliability of the investigation. This review aims to discuss the advantages and limitations of extracting bioactive compounds, sample preparation techniques, automation, and coupling with on-line detection. This review also evaluates all publications on this topic through a longitudinal bibliometric analysis, applying statistical and mathematical methods to analyze the trends, perspectives, and hot topics of this research area. Furthermore, state-of-the-art green extraction techniques for complex samples from vegetable matrices coupled with analysis systems are presented. Among the extraction techniques for liquid samples, solid-phase extraction was the most common for combined systems in the scientific literature. In contrast, for on-line extraction systems applied for solid samples, supercritical fluid extraction, ultrasound-assisted extraction, microwave-assisted extraction, and pressurized liquid extraction were the most frequent green extraction techniques

    Physico-chemical characterization of centrifuged sludge from the Tamanduá water treatment plant (Foz do Iguaçu, PR)

    The water treatment process generates a residue called water treatment plant (WTP) sludge, which needs tobe correctly characterized to ensure appropriate disposal or reuse. This study aimed to characterize the centrifugedsludge produced at the Tamanduá WTP, Iguaçu Falls City, Brazil, and considered opportunities for itsreuse in the production of concrete for the civil construction industry. Wet sludge (sludge in its natural form)analysis included the determination of total solids, moisture content, density, pH, and inorganic parameters(As, Al, Ba, Cd, Pb, Cr, F, Hg, Ag, and Se) through thermogravimetric analysis, X-ray diffraction, and chemicalanalysis by X-ray fluorescence and loss ignition. For calcined WTP sludge, chemical and mineralogicalcomposition and laser granulometry were evaluated. The results indicated that calcined sludge had the potentialto be used in the production of cement materials; conversely wet sludge did not reach the appropriatesafety standards due to the high quantity of organic matter.Keywords: Water treatment plant, water sludge, coagulant, solid waste

    An experience in teaching virtue ethics in administration

    Teaching ethics in business involves numerous methods and perspectives. Virtues are stable character dispositions that have supported ethical actions. This article aims to discuss how virtues can be taught in Business, considering the case of graduate students in a doctoral elective discipline. The perspective of virtue ethics developed in business ethics composes the theoretical framework of this study, offering teaching and learning methods to cultivate moral and intellectual virtues. Through a qualitative case study approach, participant observation, and focus group interviews were conducted with doctoral students, providing data coded and interpreted with the support of Atlas.ti. Results show that theoretical discussions articulated to experiential activities, such as outdoor adventure education in the natural environment and film analysis, allow students to express a repertoire of virtues, as well as to be virtuous in the activities conducted. The importance of theoretical knowledge, leadership, and methods in conducting the practices is highlighted. The articulation between methods, involving theory and practical experience, contributes to the students' sensitivity to ethical issues, adding knowledge, decisions, and real actions that demand virtues such as courage and prudence. The research brings together methods for teaching-learning ethics in Management, indicating that the articulation between methods contributes to enriching knowledge, reflection, and perception about virtues

    Semiconductor-based DNA sequencing of histone modification states

    The recent development of a semiconductor-based, non-optical DNA sequencing technology promises scalable, low-cost and rapid sequence data production. The technology has previously been applied mainly to genomic sequencing and targeted re-sequencing. Here we demonstrate the utility of Ion Torrent semiconductor-based sequencing for sensitive, efficient and rapid chromatin immunoprecipitation followed by sequencing (ChIP-seq) through the application of sample preparation methods that are optimized for ChIP-seq on the Ion Torrent platform. We leverage this method for epigenetic profiling of tumour tissues
