    Nutritional and functional advantages of the use of fermented black chickpea flour for semolina-pasta fortification

    Pasta represents a dominant portion of the diet worldwide and its functionalization with high nutritional value ingredients, such as legumes, is the most ideal solution to shape consumers behavior towards healthier food choices. Aiming at improving the nutritional quality of semolina pasta, semi-liquid dough of a Mediterranean black chickpea flour, fermented with Lactiplantibacillus plantarum T0A10, was used at a substitution level of 15% to manufacture fortified pasta. Fermentation with the selected starter enabled the release of 20% of bound phenolic compounds, and the conversion of free compounds into more active forms (dihydrocaffeic and phloretic acid) in the dough. Fermented dough also had higher resistant starch (up to 60% compared to the control) and total free amino acids (almost 3 g/kg) contents, whereas antinutritional factors (raffinose, condensed tannins, trypsin inhibitors and saponins) significantly decreased. The impact of black chickpea addition on pasta nutritional, technological and sensory features, was also assessed. Compared to traditional (semolina) pasta, fortified pasta had lower starch hydrolysis rate (ca. 18%) and higher in vitro protein digestibility (up to 38%). Moreover, fortified cooked pasta, showing scavenging activity against DPPH and ABTS radicals and intense inhibition of linoleic acid peroxidation, was appreciated for its peculiar organoleptic profile. Therefore, fermentation technology appears to be a promising tool to enhance the quality of pasta and promote the use of local chickpea cultivars while preventing their genetic erosion

    Single-shot hybrid photoacoustic-fluorescent microendoscopy through a multimode fiber with wavefront shaping

    We present a minimally-invasive endoscope based on a multimode fiber that combines photoacoustic and fluorescence sensing. From the measurement of a transmission matrix during a prior calibration step, a focused spot is produced and raster-scanned over a sample at the distal tip of the fiber by use of a fast spatial light modulator. An ultra-sensitive fiber-optic ultrasound sensor for photoacoustic detection placed next to the fiber is combined with a photodetector to obtain both fluorescence and photoacoustic images with a distal imaging tip no larger than 250 µm. The high signal-to-noise ratio provided by wavefront shaping based focusing and the ultra-sensitive ultrasound sensor enables imaging with a single laser shot per pixel, demonstrating fast two-dimensional hybrid in vitro imaging of red blood cells and fluorescent beads

    Effect of Porcine circovirus 2 (PCV-2) maternally derived antibodies on performance and PCV-2 viremia in vaccinated piglets under field conditions

    Background: Nowadays, the most common presentation of PCV-2 is the subclinical infection in piglets after weaning. The success of PCV-2 vaccination is associated with the control of the clinical disease as well as the improvement of production parameters. In consequence, the objective of the present study was to analyse the effect of PCV-2 maternally derived antibody (MDA) levels on vaccine efficacy in piglets vaccinated at three weeks of age with a commercial PCV-2 subunit vaccine. The study was performed analysing a database with 6112 wean-to-slaughter piglets from 4 different European regions. Results: Results showed that the use of the vaccine was able to decrease the PCV-2 viremia calculated as area under the curve (AUC = 60.29 ± 3.73), increase average daily weight gain (ADWG = 0.65 ± 0.01 kg/day) and reduce mortality (7%) in vaccinated piglets compared to non-vaccinated ones (AUC of 198.27 ± 6.14, 0.62 ± 0.01 kg/day and 11% respectively). The overall difference of ADWG between both groups was close to 30 g per day (p < 0.05), also when they were split for low and high levels of MDA titres. Moreover, the animals with the highest ADWG were observed in the group of piglets vaccinated with high or extremely high antibody titres (0.66 and 0.65 kg/day respectively). Considering only animals with extremely high antibody titres, both study groups performed similar, however there was a numerical difference of 10 g/day in favour of vaccinated piglets. Likewise, lack of correlation between ADWG and MDA was observed suggesting that no maternal antibody interference was present with the tested vaccine because the vaccinated animals grew faster compared to unvaccinated control animals, regardless of the level of maternal antibodies present at the time of vaccination. Conclusions: The results of the present study demonstrated that the MDA against PCV-2 transferred through the colostrum intake has a protective effect against this viral infection. The vaccine used in the present study (Ingelvac CircoFLEX®) was effective when applied at three weeks of age and was not affected by the level of MDA at the time of vaccination

    Aquest treball pren com a objecte d'estudi el videojoc japonès Dragon Quest (Enix, 1986) i el seu procés d'adaptació al mercat estatunidenc. Per entendre les decisions preses per modificar l'apartat estètic original del producte per part dels agents de distribució es contextualitzarà l'origen de Dragon Quest a la indústria del videojoc japonès, en el gènere dels videojocs de rol i en com encaixa dins de l'ecosistema de consum dels productes culturals japonesos. Un cop presentat el producte i l'entorn local en què es consumeix s'analitzarà quins elements van modificar la seva figura per tal de disminuir-ne l'aroma cultural. Posteriorment, després d'analitzar l'impacte de Dragon Quest al Japó i als Estats Units, s'investigarà per què fora del Japó gaudeix de menys popularitat que una altra franquícia de videojocs japoneses del mateix gènere anomenada Final Fantasy (Square, Square Enix, 1987-).Este trabajo de investigación toma como objeto de estudio el videojuego japonés Dragon Quest (Enix, 1986) y su proceso de adaptación para el mercado norteamericano. Para entender las decisiones tomadas para modificar el apartado estético original del producto por parte de los agentes de distribución se contextualizará el origen de Dragon Quest en la industria del videojuego japonés, en el género de los videojuegos de rol y en cómo encaja dentro del ecosistema de consumo de productos culturales japoneses. Una vez introducido el producto y el entorno local en el que se consume se analizará qué elementos se modificaron en su figura con tal de disminuir su aroma cultural. Posteriormente, tras analizar el impacto de Dragon Quest en Japón y en Estados Unidos, se investigará por qué fuera de Japón goza de menor popularidad que otra franquicia de videojuegos japoneses del mismo género llamada Final Fantasy (Square, Square Enix, 1987-).This research takes as its object of study the Japanese videogame Dragon Quest (Enix, 1986) and its adaptation process to the US market. To understand the decisions made by the distribution agents in order to modify the original art style of the product, the origin of Dragon Quest in the Japanese videogame industry, and its role in the role-playing videogame genre will be contextualized, as well as the way it fits in the consumption system of Japanese cultural products. After that, the elements that were modified in its figure in order to erase its cultural odor will be analyzed. Subsequently, after analyzing the impact of Dragon Quest in Japan and in the United States, research will be focused on why it is less popular than another Japanese videogame franchise, that belongs to the same genre, which is called Final Fantasy (Square, Square Enix, 1987-)

    Casein SNP in Norwegian goats: additive and dominance effects on milk composition and quality

    <p>Abstract</p> <p>Background</p> <p>The four casein proteins in goat milk are encoded by four closely linked casein loci (<it>CSN1S1</it>, <it>CSN2</it>, <it>CSN1S2 </it>and <it>CSN3</it>) within 250 kb on caprine chromosome 6. A deletion in exon 12 of <it>CSN1S1</it>, so far reported only in Norwegian goats, has been found at high frequency (0.73). Such a high frequency is difficult to explain because the national breeding goal selects against the variant's effect.</p> <p>Methods</p> <p>In this study, 575 goats were genotyped for 38 Single Nucleotide Polymorphisms (SNP) located within the four casein genes. Milk production records of these goats were obtained from the Norwegian Dairy Goat Control. Test-day mixed models with additive and dominance fixed effects of single SNP were fitted in a model including polygenic effects.</p> <p>Results</p> <p>Significant additive effects of single SNP within <it>CSN1S1 </it>and <it>CSN3 </it>were found for fat % and protein %, milk yield and milk taste. The allele with the deletion showed additive and dominance effects on protein % and fat %, and overdominance effects on milk quantity (kg) and lactose %. At its current frequency, the observed dominance (overdominance) effects of the deletion allele reduced its substitution effect (and additive genetic variance available for selection) in the population substantially.</p> <p>Conclusions</p> <p>The selection pressure of conventional breeding on the allele with the deletion is limited due to the observed dominance (overdominance) effects. Inclusion of molecular information in the national breeding scheme will reduce the frequency of this deletion in the population.</p

    Measurement of the 8B solar neutrino flux in SNO+ with very low backgrounds

    A measurement of the 8B solar neutrino flux has been made using a 69.2 kt-day dataset acquired with the SNO+ detector during its water commissioning phase. At energies above 6 MeV the dataset is an extremely pure sample of solar neutrino elastic scattering events, owing primarily to the detector’s deep location, allowing an accurate measurement with relatively little exposure. In that energy region the best fit background rate is 0.25+0.09−0.07  events/kt−day, significantly lower than the measured solar neutrino event rate in that energy range, which is 1.03+0.13−0.12  events/kt−day. Also using data below this threshold, down to 5 MeV, fits of the solar neutrino event direction yielded an observed flux of 2.53+0.31−0.28(stat)+0.13−0.10(syst)×106  cm−2 s−1, assuming no neutrino oscillations. This rate is consistent with matter enhanced neutrino oscillations and measurements from other experiments

    Changes in the Diversity of Soil Arbuscular Mycorrhizal Fungi after Cultivation for Biofuel Production in a Guantanamo (Cuba) Tropical System

    The arbuscular mycorrhizal fungi (AMF) are a key, integral component of the stability, sustainability and functioning of ecosystems. In this study, we characterised the AMF biodiversity in a native vegetation soil and in a soil cultivated with Jatropha curcas or Ricinus communis, in a tropical system in Guantanamo (Cuba), in order to verify if a change of land use to biofuel plant production had any effect on the AMF communities. We also asses whether some soil properties related with the soil fertility (total N, Organic C, microbial biomass C, aggregate stability percentage, pH and electrical conductivity) were changed with the cultivation of both crop species. The AM fungal small sub-unit (SSU) rRNA genes were subjected to PCR, cloning, sequencing and phylogenetic analyses. Twenty AM fungal sequence types were identified: 19 belong to the Glomeraceae and one to the Paraglomeraceae. Two AMF sequence types related to cultured AMF species (Glo G3 for Glomus sinuosum and Glo G6 for Glomus intraradices-G. fasciculatum-G. irregulare) did not occur in the soil cultivated with J. curcas and R. communis. The soil properties (total N, Organic C and microbial biomass C) were higher in the soil cultivated with the two plant species. The diversity of the AMF community decreased in the soil of both crops, with respect to the native vegetation soil, and varied significantly depending on the crop species planted. Thus, R. communis soil showed higher AMF diversity than J. curcas soil. In conclusion, R. communis could be more suitable for the long-term conservation and sustainable management of these tropical ecosytems
