Non-Coding RNA Sequencing of Equine Endometrium During Maternal Recognition of Pregnancy.
Maternal recognition of pregnancy (MRP) in the mare is not well defined. In a non-pregnant mare, prostaglandin F2α (PGF) is released on day 14 post-ovulation (PO) to cause luteal regression, resulting in loss of progesterone production. Equine MRP occurs prior to day 14 to halt PGF production. Studies have failed to identify a gene candidate for MRP, so attention has turned to small, non-coding RNAs. The objective of this study was to evaluate small RNA (<200 nucleotides) content in endometrium during MRP. Mares were used in a cross-over design with each having a pregnant and non-mated cycle. Each mare was randomly assigned to collection day 11 or 13 PO (n = 3/day) and endometrial biopsies were obtained. Total RNA was isolated and sequencing libraries were prepared using a small RNA library preparation kit and sequenced on a HiSeq 2000. EquCab3 was used as the reference genome and DESeq2 was used for statistical analysis. On day 11, 419 ncRNAs, representing miRNA, snRNA, snoRNA, scaRNA, and vaultRNA, were different between pregnancy statuses, but none on day 13. Equine endometrial ncRNAs with unknown structure and function were also identified. This study is the first to describe ncRNA transcriptome in equine endometrium. Identifying targets of these ncRNAs could lead to determining MRP
Resistance of uveal melanoma to the interstrand cross-linking agent mitomycin C is associated with reduced expression of CYP450R
background: Uveal melanoma (UM) is the most common primary intraocular tumour of adults, frequently metastasising to the liver. Hepatic metastases are difficult to treat and are mainly unresponsive to chemotherapy. To investigate why UM are so chemo-resistant we explored the effect of interstrand cross-linking agents mitomycin C (MMC) and cisplatin in comparison with hydroxyurea (HU).
methods: Sensitivity to MMC, cisplatin and HU was tested in established UM cell lines using clonogenic assays. The response of UM to MMC was confirmed in MTT assays using short-term cultures of primary UM. The expression of cytochrome P450 reductase (CYP450R) was analysed by western blotting, and DNA cross-linking was assessed using COMET analysis supported by γ-H2AX foci formation.
results: Both established cell lines and primary cultures of UM were resistant to the cross-linking agent MMC (in each case P<0.001 in Student's t-test compared with controls). In two established UM cell lines, DNA cross-link damage was not induced by MMC (in both cases P<0.05 in Students's t-test compared with damage induced in controls). In all, 6 out of 6 UMs tested displayed reduced expression of the metabolising enzyme CYP450R and transient expression of CYP450R increased MMC sensitivity of UM.
conclusion: We suggest that reduced expression of CYP450R is responsible for MMC resistance of UM, through a lack of bioactivation, which can be reversed by complementing UM cell lines with CYP450R
The effects of disk and dust structure on observed polarimetric images of protoplanetary disks
Imaging polarimetry is a powerful tool for imaging faint circumstellar
material. For a correct analysis of observations we need to fully understand
the effects of dust particle parameters, as well as the effects of the
telescope, atmospheric seeing, and assumptions about the data reduction and
processing of the observed signal. Here we study the major effects of dust
particle structure, size-dependent grain settling, and instrumental properties.
We performed radiative transfer modeling using different dust particle models
and disk structures. To study the influence of seeing and telescope diffraction
we ran the models through an instrument simulator for the ExPo dual-beam
imaging polarimeter mounted at the 4.2m William Herschel Telescope (WHT).
Particle shape and size have a strong influence on the brightness and
detectability of the disks. In the simulated observations, the central
resolution element also contains contributions from the inner regions of the
protoplanetary disk besides the unpolarized central star. This causes the
central resolution element to be polarized, making simple corrections for
instrumental polarization difficult. This effect strongly depends on the
spatial resolution, so adaptive optics systems are needed for proper
polarization calibration. We find that the commonly employed homogeneous sphere
model gives results that differ significantly from more realistic models. For a
proper analysis of the wealth of data available now or in the near future, one
must properly take the effects of particle types and disk structure into
account. The observed signal depends strongly on the properties of these more
realistic models, thus providing a potentially powerful diagnostic. We conclude
that it is important to correctly understand telescope depolarization and

The First Science Results from SPHERE: Disproving the Predicted Brown Dwarf around V471 Tau
Variations of eclipse arrival times have recently been detected in several
post common envelope binaries consisting of a white dwarf and a main sequence
companion star. The generally favoured explanation for these timing variations
is the gravitational pull of one or more circumbinary substellar objects
periodically moving the center of mass of the host binary. Using the new
extreme-AO instrument SPHERE, we image the prototype eclipsing post-common
envelope binary V471 Tau in search of the brown dwarf that is believed to be
responsible for variations in its eclipse arrival times. We report that an
unprecedented contrast of 12.1 magnitudes in the H band at a separation of 260
mas was achieved, but resulted in a non-detection. This implies that there is
no brown dwarf present in the system unless it is three magnitudes fainter than
predicted by evolutionary track models, and provides damaging evidence against
the circumbinary interpretation of eclipse timing variations. In the case of
V471 Tau, a more consistent explanation is offered with the Applegate
mechanism, in which these variations are prescribed to changes in the
quadrupole moment within the main-sequence sta
Quantifying the speech-gesture relation with massive multimodal datasets: informativity in time expressions
The development of large-scale corpora has led to a quantum leap in our understanding of speech in recent years. By contrast, the analysis of massive datasets has so far had a lim- ited impact on the study of gesture and other visual communicative behaviors. We utilized the UCLA-Red Hen Lab multi-billion-word repository of video recordings, all of them showing communicative behavior that was not elicited in a lab, to quantify speech-gesture co-occur- rence frequency for a subset of linguistic expressions in American English. First, we objec- tively establish a systematic relationship in the high degree of co-occurrence between gesture and speech in our subset of expressions, which consists of temporal phrases. Sec- ond, we show that there is a systematic alignment between the informativity of co-speech gestures and that of the verbal expressions with which they co-occur. By exposing deep, systematic relations between the modalities of gesture and speech, our results pave the way for the data-driven integration of multimodal behavior into our understanding of human communication
The role of thiol species in the hypertolerance of Aspergillus sp. P37 to arsenic.
Aspergillus sp. P37 is an arsenate-hypertolerant fungus isolated from a river in Spain with a long history of contamination with metals. This strain is able to grow in the presence of 0.2 M arsenate, i.e. 20-fold higher than the reference strain, Aspergillus nidulans TS1. Although Aspergillus sp. P37 reduces As(V) to As(III), which is slowly pumped out of the cell, the measured efflux of oxyanions is insufficient to explain the high tolerance levels of this strain. To gain an insight into this paradox, the accumulation of acid-soluble thiol species in Aspergillus sp. P37 when exposed to arsenic was compared with that of the arsenic-sensitive A. nidulans TS1 strain. Increasing levels of arsenic in the medium did not diminish the intracellular pool of reduced glutathione in Aspergillus sp. P37, in sharp contrast with the decline of glutathione in A. nidulans under the same conditions. Furthermore, concentrations of arsenic that were inhibitory for the sensitive A. nidulans strain (e.g. 50 mM and above) provoked a massive formation of vacuoles filled with thiol species. Exocomet signatures around the A-shell star Leo?
We present an intensive monitoring of high-resolution spectra of the Ca {\sc
ii} K line in the A7IV shell star Leo at very short (minutes, hours),
short (night to night), and medium (weeks, months) timescales. The spectra show
remarkable variable absorptions on timescales of hours, days, and months. The
characteristics of these sporadic events are very similar to most that are
observed toward the debris disk host star Pic, which are commonly
interpreted as signs of the evaporation of solid, comet-like bodies grazing or
falling onto the star. Therefore, our results suggest the presence of solid
bodies around Leo. To our knowledge, with the exception of Pic,
our monitoring has the best time resolution at the mentioned timescales for a
star with events attributed to exocomets. Assuming the cometary scenario and
considering the timescales of our monitoring, our results indicate that
Leo presents the richest environment with comet-like events known to date,
