Allergen immunotherapy on the way to product-based evaluation - a WAO statement
Allergen immunotherapy (AIT) is widely used in clinical practice for patients with moderate to severe allergic rhinitis due to inhalant allergens and may be delivered via subcutaneous (SCIT) and sublingual routes (SLIT). However, the quality of evidence for individual AIT products is very heterogeneous, and extensions of overall conclusions ("class effects") on the efficacy and disease-modifying effects to all AIT products are unjustified. In contrast, each product needs to be evaluated individually, based on available study results, to justify efficacy and specific claims on sustained and disease modifying effects per allergen and targeted patient group (children vs. adults, allergic rhinitis vs. asthma). WAO intends to support the current development to evidence-based AIT, which ultimately will lead to a more efficacious treatment of allergic patients and the appropriate recognition of AIT
Exploring CEvNS with NUCLEUS at the Chooz Nuclear Power Plant
Coherent elastic neutrino-nucleus scattering (CENS) offers a unique way
to study neutrino properties and to search for new physics beyond the Standard
Model. Nuclear reactors are promising sources to explore this process at low
energies since they deliver large fluxes of (anti-)neutrinos with typical
energies of a few MeV. In this paper, a new-generation experiment to study
CENS is described. The NUCLEUS experiment will use cryogenic detectors
which feature an unprecedentedly low energy threshold and a time response fast
enough to be operated in above-ground conditions. Both sensitivity to
low-energy nuclear recoils and a high event rate tolerance are stringent
requirements to measure CENS of reactor antineutrinos. A new experimental
site, denoted the Very-Near-Site (VNS) at the Chooz nuclear power plant in
France is described. The VNS is located between the two 4.25 GW
reactor cores and matches the requirements of NUCLEUS. First results of on-site
measurements of neutron and muon backgrounds, the expected dominant background
contributions, are given. In this paper a preliminary experimental setup with
dedicated active and passive background reduction techniques is presented.
Furthermore, the feasibility to operate the NUCLEUS detectors in coincidence
with an active muon-veto at shallow overburden is studied. The paper concludes
with a sensitivity study pointing out the promising physics potential of
NUCLEUS at the Chooz nuclear power plant
The CUORE Cryostat: A 1-Ton Scale Setup for Bolometric Detectors
The cryogenic underground observatory for rare events (CUORE) is a 1-ton
scale bolometric experiment whose detector consists of an array of 988 TeO2
crystals arranged in a cylindrical compact structure of 19 towers. This will be
the largest bolometric mass ever operated. The experiment will work at a
temperature around or below 10 mK. CUORE cryostat consists of a cryogen-free
system based on pulse tubes and a custom high power dilution refrigerator,
designed to match these specifications. The cryostat has been commissioned in
2014 at the Gran Sasso National Laboratories and reached a record temperature
of 6 mK on a cubic meter scale. In this paper, we present results of CUORE
commissioning runs. Details on the thermal characteristics and cryogenic
performances of the system will be also given.Comment: 7 pages, 2 figures, LTD16 conference proceedin

Altered Hippocampal-Prefrontal Neural Dynamics in Mouse Models of Down Syndrome
Altered neural dynamics in medial prefrontal cortex (mPFC) and hippocampus may contribute to cognitive impairments in the complex chromosomal disorder, Down Syndrome (DS). Here, we demonstrate non-overlapping behavioural differences associated with distinct abnormalities in hippocampal and mPFC electrophysiology during a canonical spatial memory task in three partially trisomic mouse models of DS (Dp1Tyb, Dp10Yey, Dp17Yey) that together cover all regions of homology with human chromosome 21 (Hsa21). Dp1Tyb mice showed slower decision-making (unrelated to the gene dose of DYRK1A, which has been implicated in DS cognitive dysfunction) and altered theta dynamics (reduced frequency, increased hippocampal-mPFC coherence, increased modulation of hippocampal high gamma); Dp10Yey mice showed impaired alternation performance and reduced theta modulation of hippocampal low gamma; while Dp17Yey mice were no different from wildtype mice. These results link specific hippocampal and mPFC circuit dysfunctions to cognitive deficits in DS models and, importantly, map them to discrete regions of Hsa21
ARIA 2016 Executive Summary Integrated care pathways for predictive medicine across the life cycle
The Allergic Rhinitis and its Impact on Asthma (ARIA) initiative commenced during a World Health Organization (WHO) workshop in 1999. The initial goals were (i) to propose a new allergic rhinitis classification, (ii) to promote the concept of multi-morbidity in asthma and rhinitis and (iii) to develop guidelines with all stakeholders for global use in all countries and populations. ARIA - disseminated and implemented in over 70 countries globally - is now focusing on the implementation of emerging technologies for individualized and predictive medicine. MASK (MACVIA (Contre les MAladies Chroniques pour un VIeillissement Actif)-ARIA Sentinel NetworK) uses mobile technology to develop care pathways in order to enable the management of rhinitis and asthma by a multi-disciplinary group or by patients themselves. An App (Android and iOS) is available in 20 countries and 15 languages. It uses a visual analogue scale to assess symptom control and work productivity as well as a clinical decision support system. It is associated with an inter-operable tablet for physicians and other health care professionals. The scaling up strategy uses the recommendations of the European Innovation Partnership on Active and Healthy Ageing. The aim of the novel ARIA approach is to provide an active and healthy life to rhinitis sufferers, whatever their age, sex or socio-economic status, in order to reduce health and social inequalities incurred by the disease. Keywords: ARIA, rhinitis, ICT, EIP on AHA, mobile technology, AIRWAYS ICP
The CUORE cryostat: an infrastructure for rare event searches at millikelvin temperatures
The CUORE experiment is the world's largest bolometric experiment. The
detector consists of an array of 988 TeO2 crystals, for a total mass of 742 kg.
CUORE is presently taking data at the Laboratori Nazionali del Gran Sasso,
Italy, searching for the neutrinoless double beta decay of 130Te. A large
custom cryogen-free cryostat allows reaching and maintaining a base temperature
of about 10 mK, required for the optimal operation of the detector. This
apparatus has been designed in order to achieve a low noise environment, with
minimal contribution to the radioactive background for the experiment. In this
paper, we present an overview of the CUORE cryostat, together with a
description of all its sub-systems, focusing on the solutions identified to
satisfy the stringent requirements. We briefly illustrate the various phases of
the cryostat commissioning and highlight the relevant steps and milestones
achieved each time. Finally, we describe the successful cooldown of CUORE
Nrf2-interacting nutrients and COVID-19: time for research to develop adaptation strategies.
There are large between- and within-country variations in COVID-19 death rates. Some very low death rate settings such as Eastern Asia, Central Europe, the Balkans and Africa have a common feature of eating large quantities of fermented foods whose intake is associated with the activation of the Nrf2 (Nuclear factor (erythroid-derived 2)-like 2) anti-oxidant transcription factor. There are many Nrf2-interacting nutrients (berberine, curcumin, epigallocatechin gallate, genistein, quercetin, resveratrol, sulforaphane) that all act similarly to reduce insulin resistance, endothelial damage, lung injury and cytokine storm. They also act on the same mechanisms (mTOR: Mammalian target of rapamycin, PPARγ:Peroxisome proliferator-activated receptor, NFκB: Nuclear factor kappa B, ERK: Extracellular signal-regulated kinases and eIF2α:Elongation initiation factor 2α). They may as a result be important in mitigating the severity of COVID-19, acting through the endoplasmic reticulum stress or ACE-Angiotensin-II-AT1R axis (AT1R) pathway. Many Nrf2-interacting nutrients are also interacting with TRPA1 and/or TRPV1. Interestingly, geographical areas with very low COVID-19 mortality are those with the lowest prevalence of obesity (Sub-Saharan Africa and Asia). It is tempting to propose that Nrf2-interacting foods and nutrients can re-balance insulin resistance and have a significant effect on COVID-19 severity. It is therefore possible that the intake of these foods may restore an optimal natural balance for the Nrf2 pathway and may be of interest in the mitigation of COVID-19 severity
First results from the CRESST-III low-mass dark matter program
The CRESST experiment is a direct dark matter search which aims to measure
interactions of potential dark matter particles in an earth-bound detector.
With the current stage, CRESST-III, we focus on a low energy threshold for
increased sensitivity towards light dark matter particles. In this manuscript
we describe the analysis of one detector operated in the first run of
CRESST-III (05/2016-02/2018) achieving a nuclear recoil threshold of 30.1eV.
This result was obtained with a 23.6g CaWO crystal operated as a cryogenic
scintillating calorimeter in the CRESST setup at the Laboratori Nazionali del
Gran Sasso (LNGS). Both the primary phonon/heat signal and the simultaneously
emitted scintillation light, which is absorbed in a separate
silicon-on-sapphire light absorber, are measured with highly sensitive
transition edge sensors operated at ~15mK. The unique combination of these
sensors with the light element oxygen present in our target yields sensitivity
to dark matter particle masses as low as 160MeV/c.Comment: 9 pages, 9 figure
