Dexamethasone-Loaded Lipomers: Development, Characterization, and Skin Biodistribution Studies

Follicular targeting has gained more attention in recent decades, due to the possibility of obtaining a depot effect in topical administration and its potential as a tool to treat hair follicle-related diseases. Lipid core ethyl cellulose lipomers were developed and optimized, following which characterization of their physicochemical properties was carried out. Dexamethasone was encapsulated in the lipomers (size, 115 nm; polydispersity, 0.24; zeta-potential (Z-potential), +30 mV) and their in vitro release profiles against dexamethasone in solution were investigated by vertical diffusion Franz cells. The skin biodistribution of the fluorescent-loaded lipomers was observed using confocal microscopy, demonstrating the accumulation of both lipomers and fluorochromes in the hair follicles of pig skin. To confirm this fact, immunofluorescence of the dexamethasone-loaded lipomers was carried out in pig hair follicles. The anti-inflammatory (via TNFα) efficacy of the dexamethasone-loaded lipomers was demonstrated in vitro in an HEK001 human keratinocytes cell culture and the in vitro cytotoxicity of the nanoformulation was investigated

Dexamethasone-loaded polymer hybrid nanoparticles were developed as a potential tool to treat alopecia areata due to their follicular targeting ability. Freeze drying (FD) is a common technique used to improve nanoparticle stability; however, there are few studies focused on its effect on ethyl cellulose lipid-core nanoparticles. Nanoparticles were lyophilized with different cryoprotectants. Sucrose was selected because it allowed for a good resuspension and provided acceptable physicochemical parameters (374.33 nm, +34.7 mV, polydispersion 0.229%, and 98.87% encapsulation efficiency). The nanoparticles obtained were loaded into a pleasant xanthan gum hydrogel, and the rheological, release, and skin permeation profiles of different formulations were studied. The FD formulation significantly modified the particle size, and the drug release and permeation properties were also altered. In addition, analyses of the cytotoxicity and anti-inflammatory efficacy of FD and non-FD particles on human keratinocytes indicated no differences

Latanoprost (LAT) has been shown to have a hypertrichotic effect, which makes it a promising candidate for alopecia treatments. For the first time, LAT has been encapsulated in nanotransfersomes in order to increase its efficacy. Ex vivo skin biodistribution was studied by confocal laser microscopy both in human scalp and pig skin. Results showed that nanotransfersomes increase the penetration of two different fluorochromes, with similar patterns in both species, compared with fluorochrome solutions containing no nanotransfersomes. Nanotransfersomes were stable under accelerated conditions (40 °C/75% RH) and long-term conditions (25 °C/60% RH) for up to 1 year, with no differences in vesicle size and polydispersity when LAT was loaded. Nanotransfersomes increased the LAT cell proliferation effect in HaCaT cell via MAPK signaling pathway. Collectively, our results demonstrate LAT-nanotransfersomes formulation could be a promising therapy for hair growth disorders

From the sea to the table: the environmental impact assessment of fishing, processing, and end-of-life of albacore in Cantabria

Cantabria, a small coastal region of Northern Spain, is one of the biggest producers of gourmet tuna cans in Europe. The fish capture in the Cantabrian Sea and the subsequent transformation in a local processing plant give distinction to this product, which is widely marketed in cans of 105 g of net weight. This work evaluates for the first time the environmental profile of the whole supply chain of this product, from fishing, processing, and waste valorization to inter-stage transport and packaging management in the end-of-life. To this end, the life cycle assessment methodology was applied considering primary data from the stakeholders involved in the supply chain and analyzing the seven most studied categories in this sector. Results revealed that fishing and processing accounted for the majority of the environmental impacts, while valorization and end-of-life treatments only avoid less than 10% of the burdens. The most important findings are focused on the high dependence on fuel use, identified as a hotspot in most stages although low compared to other fisheries, and on the intensive use of resources, especially sunflower oil, which contributes more than 70% of the impact on the global warming potential of the processing. This current framework forces the enhancement of the efficiency of a sector that attempts to engage the challenge of societal sustainability, by identifying the critical points and guiding policy makers on the path to sustainable development. This work was supported by the EAPA_576/2018NEPTUNUS project. [Abstract] The EMPA-REG OUTCOME (Empagliflozin, Cardiovascular Outcome Event Trial in patients with Type 2 Diabetes Mellitus (T2DM)) trial evidenced the potential of sodium-glucose cotransporter 2 (SGLT2) inhibitors for the treatment of patients with diabetes and cardiovascular disease. Recent evidences have shown the benefits of the SGLT2 inhibitor empagliflozin on improving liver steatosis and fibrosis in patients with T2DM. Metabolomic studies have been shown to be very useful to improve the understanding of liver pathophysiology during the development and progression of metabolic hepatic diseases, and because the effects of empagliflozin and of other SGLT2 inhibitors on the complete metabolic profile of the liver has never been analysed before, we decided to study the impact on the liver of male Zucker diabetic fatty (ZDF) rats of a treatment for 6 weeks with empagliflozin using an untargeted metabolomics approach, with the purpose to help to clarify the benefits of the use of empagliflozin at hepatic level. We found that empagliflozin is able to change the hepatic lipidome towards a protective profile, through an increase of monounsaturated and polyunsaturated glycerides, phosphatidylcholines, phosphatidylethanolamines, lysophosphatidylinositols and lysophosphatidylcholines. Empagliflozin also induces a decrease in the levels of the markers of inflammation IL-6, chemerin and chemerin receptor in the liver. Our results provide new evidences regarding the molecular pathways through which empagliflozin could exert hepatoprotector beneficial effects in T2DM.This work was supported by Boehringer Ingelheim Pharma GmbH and Co., by the National Institute of Health “Fondo de Investigaciones Sanitarias del Instituto de Salud Carlos III” Madrid, Spain (PI15/00681, PI17/00409, PI18/00821, PI20/00902, RETICS Programme RD16/0012/0014 and CIBER de Enfermedades Cardiovasculares (CIBERCV)); European Regional Development Fund (FEDER) and European Union framework MSCA-RISE-H2020 Programme (Project number 734899). For decades, Colombia has suffered the harshness of the armed conflict, the criminal actions of groups outside the law, drug trafficking, social and political nonconformism, the confrontation of powers, ideological positions and violation of rights, bringing with them the victims of the armed conflict, and naturalizing terrifying actions such as displacement, forced disappearance, massacres, threats, recruitment, military harassment, stigmatization, among others. Some of these phenomena, analyzed in this document, from the reality that relate the narratives of individuals and groups, victims of the armed conflict. First, we analyze the story of Camilo, a young man who can show the repercussions of violence on a young Afro-Colombian. Emerging psychosocial, subjective voices and resilient positioning are determined, based on Camilo’s narrative. In the same way, 9 questions are asked, which allow to inquire about the case studied, expanding knowledge, positions and subjective realities. Subsequently, we continue with the analysis of the case of Peñas Coloradas, a community victim of armed conflict, military harassment, displacement and indefinite banishment from its territory, conditions that lead to other difficult situations such as poverty, Overcrowding, abuse and psychosocial dysfunction. Based on this case, an analysis is made and strategies are proposed for a psychosocial approach, in order to support the victims, accompany their process of social reconstruction and mitigate the impact caused by experiencing this situation of violence. To conclude, we present the report that gives an account of the experience of applying photo voice, as a tool for interpreting scenarios permeated by various forms of violence based on projections of individual and collective memories, identifying traces in memory and the ability to rebuild dignity and transform invisible voices
