Five pillars for stakeholder analyses in sustainability transformations: The global case of phosphorus.

    Get PDF
Phosphorus is a critical agricultural nutrient and a major pollutant in waterbodies due to inefficient use. In the form of rock phosphate it is a finite global commodity vulnerable to price shocks and sourcing challenges. Transforming toward sustainable phosphorus management involves local to global stakeholders. Conventional readings of stakeholders may not reflect system complexity leaving it difficult to see stakeholder roles in transformations. We attempt to remedy this issue with a novel stakeholder analysis method based on five qualitative pillars: stakeholder agency, system roles, power and influence, alignment to the problem, and transformational potential. We argue that our approach suits case studies of individual stakeholders, stakeholder groups, and organisations with relationships to sustainability challenges

Effects of bone morphogenic protein 4 (BMP4) and its inhibitor, Noggin, on in vitro maturation and culture of bovine preimplantation embryos

<p>Abstract</p> <p>Background</p> <p>BMP4 is a member of the transforming growth factor beta (TGFbeta) superfamily and Noggin is a potent BMP inhibitor that exerts its function by binding to BMPs preventing interactions with its receptors. The aim of this work was to investigate the role of BMP4 and Noggin, on oocytes <it>in vitro </it>maturation (m experiments) and embryos <it>in vitro </it>development (c experiments) of bovine.</p> <p>Methods</p> <p>For m experiments, COCs were collected from slaughterhouse ovaries and <it>in vitro </it>matured in TCM with 100 ng/ml of either BMP4 or Noggin. After 24 h, the nuclear stage of the oocytes was determined by staining with Hoechst 33342. In addition, RT-qPCR was performed on MII oocytes to study the relative concentration of <it>ZAR1, GDF9, BAX, MATER </it>and <it>HSP70 </it>transcripts. Treated oocytes were submitted to parthenogenic activation (PA) or <it>in vitro </it>fertilization (IVF) and cultured in CR2. For c experiments, non-treated matured oocytes were submitted to PA or IVF to generate embryos that were exposed to 100 ng/ml of BMP4 or Noggin in CR2 until day nine of culture. Cleavage, blastocyst and hatching rates, expression pattern of the transcription factor Oct-4 in blastocysts and embryo cell number at day two and nine post-activation or fertilization were evaluated.</p> <p>Results</p> <p>We found that Noggin, as BMP4, did not affect oocyte nuclear maturation. Noggin supplementation up-regulated the expression of <it>HSP70 and MATER </it>genes in matured oocytes. Moreover, BMP4 during maturation increased the proportion of Oct-4 positive cells in parthenogenic embryos. On the other hand, when Noggin was added to embryo culture medium, developmental rates of parthenogenic and <it>in vitro </it>fertilized embryos were reduced. However, BMP4 addition decreases the development only for <it>in vitro </it>fertilized embryos. BMP4 and Noggin during culture reduced the proportion of Oct-4-expressing cells.</p> <p>Conclusions</p> <p>Our results show that BMP4 is implicated in bovine oocytes maturation and embryo development. Moreover, our findings demonstrate, for the first time, that a correct balance of BMP signaling is needed for proper pre-implantation development of bovine embryos.</p

Chronic, non-healing, dermal wounds form a worldwide medical problem with limited and inadequate treatment options and high societal burden and costs. With the advent of regenerative therapies exploiting extracellular matrix (ECM) components, its efficacy to augment wound healing is to be explored. This systematic review was performed to assess and compare the current therapeutic efficacy of ECM hydrogels on dermal wound healing. The electronic databases of Embase, Medline Ovid, and Cochrane Central were searched for in vivo and clinical studies on the therapeutic effect of ECM-composed hydrogels on dermal wound healing (April 13, 2021). Two reviewers selected studies independently. Studies were assessed based on ECM content, ECM hydrogel composition, additives, and wound healing outcomes, such as wound size, angiogenesis, and complications. Of the 2102 publications, 9 rodent-based studies were included while clinical studies were not published at the time of the search. Procedures to decellularize tissue or cultured cells and subsequently generate hydrogels were highly variable and in demand of standardization. ECM hydrogels with or without additives reduced wound size and also seem to enhance angiogenesis. Serious complications were not reported. To date, preclinical studies preclude to draw firm conclusions on the efficacy and working mechanism of ECM-derived hydrogels on dermal wound healing. The use of ECM hydrogels can be considered safe. Standardization of decellularization protocols and implementation of quality and cytotoxicity controls will enable obtaining a generic and comparable ECM product. Impact statementExtracellular matrix (ECM)-based hydrogels are biocompatible and harbor growth factors that can instruct tissue healing. Their application is a novelty in (pre)clinical wound healing treatment. This systematic review provides an overview of the current evidence for ECM hydrogels in enhancing wound healing and an extensive overview of the decellularization procedures used. Lastly, challenges and future directions to standardize decellularization procedures and implement quality controls are proposed

An autochthonous case of visceral leishmaniasis is reported in a dog (Canis familiaris) as an apparently natural infection in a non-endemic area. DNA obtained from spleen and liver samples produced the expected fragment in a Leishmania-specific rDNA-based nested-PCR assay. The PCR product, a 490 bp fragment, was sequenced and the nucleotide sequence was identical to that of Leishmania (Leishmania) infantum chagasi. These results are surprising since no autochthonous human or canine cases of visceral leishmaniasis have ever been reported in this municipality. This case suggests that natural transmission of this disease is occurring in this area.

The aim of this study was evaluating the influence of sewage-sludge addition on the quality of organic matter (OM) of a Brazilian Oxisol by Laser Induced Fluorescence (LIF). LIF was used to analyze OM of whole soil and different soil fractions separated by chemical and physical methods. The high fluorescence contribution of humin fraction to the fluorescence of whole soils was shown, stressing the importance of studying OM associated to mineral matrix of soil. Spectra with different shapes were obtained for every particle size fraction, indicating differences in organic compounds bounded to them. The fraction with the higher carbon content was the 2-20 µm, that contains ~5% C and represents only 10% in soil, but stores 34-39 % of total C and shows the highest fluorescence intensity. The spectrum of this fraction shows its heterogeneity and a higher concentration of compounds which fluorescence is centered at 510 nm. This indicates a higher concentration of unsaturated bond systems capable of high degree of resonance, increased conjugation of the electron pi system, and higher aromaticity comparing with other fractions. No differences were detected for treatments of sewage-sludge applications. LIF spectroscopy is a promising technique for OM studies in whole soils, allowing to study spatial distribution of OM within the soil's mineral matrix, including Oxisols
