Development of lipid-polymer hybrid nanoparticles as a strategy to treat Alzheimer’s Disease

Apesar da grande incidência da AD na população mundial, ainda não existe uma abordagem de tratamento eficaz para a patologia. A terapia farmacológica que existe para a doença de AD apenas trata os sintomas, não sendo capaz de induzir ou prevenir regressão da doença.
Devido aos muitos fatores que estão envolvidos na patologia da AD, uma terapia de múltiplos alvos seria a ideal para esta patologia. Isto, aliado ao facto de a AD ser uma doença do sistema nervoso central (CNS) e de o CNS estar protegido por uma barreira altamente seletiva, a barreira hematoencefálica (BBB), explica a ausência de um tratamento eficaz da doença.
Outra limitação para tratar a perturbação é a falta de um diagnóstico preciso. No passado, um diagnóstico preciso desta patologia era mais difícil. No entanto, hoje em dia o uso de técnicas de neuroimagem como a ressonância magnética (MRI) podem dar importantes informações sobre biomarcadores.
Devido a todas as limitações existentes para criar um tratamento eficaz e local da AD, esta tese de mestrado propõe o desenvolvimento de um nanossistema composto por nanopartículas híbridas de tamanho suficientemente pequeno para passar pelo BBB, que encapsulam, por sua vez, nanopartículas superparamagnéticas de óxidos de ferro (SPIONs) e um fármaco (curcumina). O nanossistema incluirá também a funcionalização destas nanopartículas com um péptido penetrante de células (CPP) que permite a translocação de membranas celulares.
O nanossistema proposto pode ser considerado um agente de teranóstico, uma vez que é capaz de ter tanto um efeito terapêutico como de diagnóstico na AD. O efeito terapêutico é dado pela curcumina presente nas nanopartículas, e o efeito de diagnóstico é dado pelas SPIONs encapsuladas uma vez que podem atuar como agentes de contraste para ressonância magnética, permitindo o diagnóstico da AD.
Todas as nanopartículas foram caracterizadas durante as diferentes fases do trabalho. As SPIONs produzidas foram analisadas por microscopia eletrónica de transmissão e o tamanho médio obtido foi de 12,3 ± 2,3 nm. Em seguida, todas as nanopartículas produzidas durante a otimização das nanopartículas híbridas foram caracterizadas utilizando a técnica DLS. Os diâmetros hidrodinâmicos obtidos para as nanopartículas não funcionalizadas foram de 91,6 ± 1,6 nm e para o nanossistema final (funcionalizado) foram de 120,1± 0,1 nm.
Para avaliar a eficiência de internalização das nanopartículas nas células, foi utilizada uma linha celular de células endoteliais humanas do cérebro. A mesma linha celular foi também utilizada para realizar ensaios de citotoxicidade. Os ensaios celulares mostraram que as nanopartículas conseguem internalizar nas células, sendo que o nanossistema mostrou uma capacidade de internalização mais elevada do que as nanopartículas híbridas + SPIONs. Os dois tipos de nanopartículas testados também mostraram uma tendência para se tornarem citotóxicos nas concentrações testadas mais elevadas.
As características do nanossistema produzido nesta tese de mestrado mostram-se promissoras para atravessar a BBB devido à sua capacidade de internalização numa linha celular de células endoteliais do cérebro humano in vitro.Alzheimer’s disease (AD) is a chronic neurodegenerative disorder responsible for most
cases of dementia in the world. Despite the large incidence of AD in the world population, there
is still no effective treatment approach to the pathology. The pharmacologic therapy that exists
for AD only treat the symptoms, it is not able to induce or prevent disease regression.
Due to the many factors that are involved in AD pathology, a multiple-target therapy
would be the preferred treatment for this pathology. This, allied to the fact that AD is a central
nervous system (CNS) disorder and that the CNS is protected by a highly selective barrier, the
blood-brain barrier (BBB), explains the absence of effective treatment for the disorder.
Another limitation in treating the disorder is the lack of an accurate diagnosis. In the past,
an accurate diagnosis of this pathology was more difficult. Nowadays the use of neuroimaging
techniques such as magnetic resonance imaging (MRI) can give important biomarker
information.
Due to all the existing limitations to creating an effective and local treatment for AD, this
master thesis proposes the development of a nanosystem composed of hybrid nanoparticles with
a small enough size to pass through the BBB, that encapsulates superparamagnetic iron oxide
nanoparticles (SPIONs) and a drug (curcumin). The nanosystem will also include the
functionalization of these nanoparticles with a cell-penetrating peptide (CPP) that allows the
translocation of cell membranes.
The proposed nanosystem can be considered a theranostics agent, since it is able to both
have a therapeutic and diagnostic effect on AD. The therapeutic effect is given by the curcumin
present in the nanoparticles, and the diagnostic effect is given by the SPIONs encapsulated
since they can act as contrast agents in MRI, allowing AD diagnosis.
All nanoparticles were characterized during the different stages of the work. The
produced SPIONs were analyzed by transmission electron microscopy and the obtained average
size was 12.3 ± 2.3 nm. Then, all the nanoparticles produced during the optimization of the
hybrid nanoparticles were characterized using DLS technique. The hydrodynamic diameters
obtained for the nanoparticles not functionalized was 91.6 ± 1.6 nm and for the final nanosystem
(functionalized) was 120.1± 0.1 nm.
To assess the efficiency of the nanoparticles in internalizing in cells, a cell line of human
brain endothelial cells was used. The same cell line was also used to perform cytotoxicity
assays. Cellular assays showed that the nanoparticles can internalize in the cells. However, the
nanosystem showed a higher internalization capacity when compared to the hybrid
nanoparticles + SPIONs. Both types of nanoparticles tested also showed a tendency to become
cytotoxic at the higher concentrations tested.
The characteristics of the produced nanosystem in this master thesis are promising to
overcome the BBB due to their ability to internalize in a cell line of human brain endothelial

Improving writing through creativity
Trabalho de Projecto apresentado para cumprimento dos requisitos necessários à obtenção do grau de Mestre em Ensino do InglêsThis project is the result of an attempt to answer the question “Can Creative Writing Be Taught?” proposed by Francine Prose (2007), in the first chapter of her book Reading Like a Writer. It also aims to show the difficulties of implementing such a project in the classroom, due to factors such as lack of time, the pressure of teaching a very tight syllabus, and the working conditions students and teachers have in order to succeed.
In order to implement the project, a series of activities was set up. Through music, paintings, drawings, hand paintings and other materials, the students produced words and simple sentences, which were enlarged so as to become complex sentences and paragraphs. Each piece of material was prepared bearing in mind the ideas the students had produced during the previous session, so that a connection could be established between all elements. The main purpose of these strategies was to lead the students into writing a story.
Lastly, this project aims at showing how the different creative writing activities contributed to a greater awareness of individual writing errors and error-correction, improving the students‟ writing skill as a whole

be structured, what returns it could provide, and what exit
strategies can be pursued. In addition, we will focus on analysing
and forecasting the business plan and defining the optimal capital
structure. The conclusion is that VAA is an attractive investment
opportunity, able to leverage the market's growth

The following report has the objective of assessing the remote health care market and defining how José de Mello Saúde can sustain its leadership position by establishing are mote clinical relationship with its clients. For that purpose, a market assessment was conducted, followed by an extensive analysis of the clients’ demand and of the health care professionals’ willingness to provide remote clinical services. Finally, the team focused its attention on there commendation of three services, which are the Teleconsultation ,the Clinical Contact Centre and the Direct Contact to Doctor. The three main marine depositional cycles of the Neogene of Portugal
The present work follows a stratigraphic model for the marine Neogene of Portugal based on the definition of three main marine
sedimentary cycles. Conceptually the I, II and III Neogene Cycles can be defined as 2nd order sedimentary sequences with duration
ranging from 5 to 8 Ma. The I Neogene Cycle is fully represented only in the Lower Tagus Basin. Ranging from the Early Aquitanian to the Late Burdigalian the I Neogene Cycle testify a transgressive episode in the region of Lisbon and Setúbal Peninsula. Rapid
lateral facies variations suggest a shallowmarine basin. This cycle ends with an important Late Burdigalian tectonic compressive event expressed by uplift of the surrounding areas and deformation affecting the Early Miocene deposits of the Arrábida Chain.
The II Neogene Cycle includes thick sedimentary sequences covering Paleozoic and Mesozoic formations in the Algarve and
Alvalade-Melides regions and it extends as far north as Santarém in the Lower Tagus Basin. Mainly controlled by global eustasy, it was generated by the important positive eustatic trend that characterized the Middle Miocene worldwide to which the Portuguese
continental margin acted more or less passively. This cycle ended with a second and the most important compression event starting after the end of the Serravallian affecting the entire Portuguese onshore and shelf areas. This led to an important depositional hiatus of marine sediments for more than 2.5 Ma.
During the Early and the Middle Tortonian occurred the clockwise rotation of the Guadalquivir Basin. The thickmarine units
deposited afterwards in this basin produced a litostatic load, which seems to have induced subsidence farther west resuming the
Neogene marine sedimentation in the Cacela region (Eastern Algarve), during the Late Tortonian. This marks the beginning of the
III Neogene Cycle. To the north, in the Sado Basin (Alvalade-Melides region), a similar depositional sequence starts its sedimentation during the Messinian. Further north, in the Pombal-Caldas da Rainha region, marine sedimentation started during the Late Pliocene (Piacenzian). The migration in time, from south to north for the beginning of the marine sedimentation of this cycle is interpreted as reflecting a visco-elastic propagation of the deformation from the Betic chain northwards
Desempenho de sistema de monitorização capacitivo em implante ósseo instrumentado
Musculoskeletal disorders are becoming an ever-growing societal burden
and, as a result, millions of bone replacements surgeries are performed per
year worldwide. Although total joint replacements are recognized among the
most successful surgeries of the last century, implant failure rates exceeding
10% are still reported. These numbers highlight the necessity of technologies
to provide an accurate monitoring of the bone-implant interface state.
This work aims to identify the performance of an instrumented implant to
monitor implant stability using a planar capacitive technology. A 5x10x0.8
mm printed circuit with two 5x2 mm electrodes was fabricated to be integrated
in an implantable device, with the objective of assessing the effect of
a fully implantation into a biological specimen. The implant was fabricated
with a conic geometry, to achieve a press-fit fixation, with 55 mm of length
and a minimum/maximum diameters of 12 and 15 mm, respectively. After
implantation, the system was put under compression and decompression
cycles, so the bone-implant interface could be altered. In the compression
cycle, the observed capacitance values decreased, indicating the sensor was
moving away from the bone; and contrarily, in the decompression cycle, the
capacitance increased with the progressive unloading. Values were obtained
in intervals of [2.2090; 3.0764] pF for the compression and [1.9806; 3.1841]
pF for the decompression. The mean percentage of capacitance change for
the compression cycle was 3.67% and 5.06% for the decompression, indicating
a greater change rate in the decompression cycle. Additional tests
were carried where the implant and the sensor were rotated 90 and 180º,
to show the influence of different interfaces in the measured capacitance.
The latter tests allowed to support the results obtained without rotation,
as different sensor positions provided different behaviors of the capacitance
change. Further development is still needed related to the experimental
setup, more specifically the in vitro specimens fixation and the environment
control of the experiment room. In addition, energy harvesting to create self
powering systems to avoid exernal links or finite-life alternatives are also a
necessity for future instrumented implants. This work further demonstrated
the potential of capacitive technologies to monitor the bone-implant fixation.
Therefore, it also contributed towards the design of a new era of
high-sophisticated implantable medical devices.Distúrbios musculares estão a tornar-se um fardo cada vez maior para a
sociedade atual, e, como resultado, milhões de artroplastias são realizadas
anualmente por todo o mundo. Apesar da artroplastia estar reconhecida
entre os procedimentos mais bem sucedidos do último século, ainda se observa
uma taxa de falha em implantes de cerca de 10%. Estes números
realçam a necessidade das tecnologias conseguirem fornecer um diagnóstico
preciso da interface osso-implante, podendo reduzir significativamente a necessidade
de cirurgias de revisão. Este trabalho tem como objetivo avaliar
o desempenho de um implante instrumentado para monitorizar a estabilidade
em implantes, utilizando uma tecnologia capacitiva planar. De forma
a verificar o efeito de uma inserção completa em espécimes biológicos, um
circuito impresso de dimensões 5x10x0.8 mm com dois elétrodos de 5x2 mm
foi fabricado com o objetivo de ser integrado dentro de um implante. O
implante foi projetado com uma geometria cónica, de forma a obter uma
fixação press-fit, com diâmetros mínimo/máximo de 12 e 15 mm, respetivamente,
e um comprimento de 55 mm. Depois de implantado, o sistema
foi posto sobre ciclos de compressão e de descompressão de forma a alterar
a interface osso-implante. Nos ciclos de compressão, os valores observados
da capacidade decresceram, indicando que o sensor se estava a afastar do
osso; contrariamente, nos ciclos de descompressão, a capacidade tendia a
aumentar com o descarregamento. Os valores foram obtidos no intervalo
de [2.2090; 3.0764] pF para os ciclos de compressão e de [1.9806; 3.1841]
pF para a descompressão. A percentagem média de variação da capacidade
para os ciclos de compressão foi de 3.67% e de 5.06% para os de descompressão,
indicando uma maior taxa de variação nos ciclos de descompressão.
Adicionalmente, foram realizados testes em que o implante e o sensor foram
rodados 90 e 180 de forma a verificar o efeito de interfaces diferentes na
capacidade medida. Estes últimos testes permitiram também corroborar a
validade dos testes de compressão e descompressão, visto que orientações
diferentes do sensor deveriam dar tendências diferentes nas curvas de capacidade.
No entanto ainda é necessário um desenvolvimento adicional
relativamente ao setup experimental, mais concretamente na fixação dos
espécimes biológicos durante os ensaios in vitro, assim como no controlo
das condições ambiente do laboratório. Adicionalmente, o desenvolvimento
de sistemas de energy harvesting são uma necessidade para o futuro dos
implantes instrumentados de forma a ter um sistema auto-sustentável, evitando
ligações com o exterior ou soluções de vida limitada. Este trabalho
permitiu comprovar o potencial de tecnologias capacitivas para a monitorização
do estado da interface osso-implante. Assim, também contribuiu
para o desenvolvimento de uma nova era de dispositivos médicos implantáveis

Certificação de idoneidade formativa do serviço de medicina interna do Hospital de São Bernardo
Na formação inicial de Enfermagem os ensinos clínicos são um momento de consolidação de saberes teóricos e simultaneamente um momento de aquisição de saberes práticos em contexto real de trabalho. Este momento de formação é supervisionado por Enfermeiros da prática clínica que assumem um importante papel no processo de formação dos estudantes de enfermagem.
Para consolidar este processo de ensino/aprendizagem a Ordem dos Enfermeiros desenvolveu um extenso trabalho que visa a certificação de Enfermeiros Supervisores, em que propõe que os Licenciados em Enfermagem, realizem um período de Exercício Profissional Tutelado, acompanhados por esse mesmo Enfermeiro Supervisor.
Para que esse período ocorra é necessário a convergência de dois fatores, um é a existência de um Enfermeiro Supervisor certificado pela Ordem dos Enfermeiros, outro é a certificação do serviço onde esse exercício profissional tutelado decorra, num espaço onde exista Certificação de Serviço com Idoneidade Formativa certificada pela Ordem dos Enfermeiros. Para que tal aconteça é necessário que o supervisor e o serviço cumpram os critérios preconizados pela Ordem dos Enfermeiros.
Este trabalho surge com o objetivo de certificar o Serviço de Medicina Interna do Hospital de São Bernardo como unidade de saúde com Idoneidade Formativa certificada pela Ordem dos Enfermeiros para o Exercício Profissional Tutelado, naquele que é inerente ao Modelo de Desenvolvimento Profissional proposto pela Ordem dos EnfermeirosThe initial training of nursing clinical teaching is a time of consolidation of academic knowledge and simultaneously a moment of acquisition of practical skills in a real work environment. This time of training is supervised by nurses in clinical practice that play an important role in the training of nursing students process.
To consolidate this process of teaching/learning the Nursing Council developed an extensive work aimed at Supervisors Nurses certification, and propose that the Licensed Nursing, undertake a period of Exercise Tutored Professional, accompanied by the same nurse supervisor.
For this period occurs is necessary convergence of two factors, one is the existence of a Nurse Supervisor Certified by the Nursing Council, the other is the certification of service where that tutored professional practice arises in a space where there is service certification with the reputation Formative certified by the Nursing Council. For this to happen it is necessary that the supervisor and the service meet the criteria established by the Nursing Council.
This work appears in order to make the Internal Medicine Service of the Hospital de São Bernardo as a health unit with Formative Reputation certified by the Nursing Council for the Professional Practice Tutored in what is inherent in the Professional Development Model proposed by the Nursing Council
A revisão textual em contexto institucional: relatório de estágio
O presente relatório tem como objetivo apresentar o trabalho desenvolvido
durante o estágio realizado no Gabinete de Imagem e Comunicação da Unidade
Hospitalar de Portimão do Centro Hospitalar do Algarve, no âmbito do Mestrado em
Ciências da Linguagem da Faculdade de Ciências Humanas e Sociais da Universidade
do Algarve.
Com duração de 400 horas de trabalho, o estágio foi realizado entre setembro e
dezembro de 2016. Durante este período, foram realizadas atividades de revisão textual
e tradução, bem como tarefas do foro da comunicação institucional, entre as quais
contacto com a comunicação social e organização de eventos.
O relatório começará por apresentar o enquadramento do estágio, explicando o
seu funcionamento e apresentando o Gabinete de Imagem e Comunicação, seguindo-se
a exposição do trabalho realizado.
Este estágio foi realizado na área da Revisão Textual e irá começar com uma
apresentação do estado da arte. De seguida serão apresentadas duas questões
selecionadas para figurarem neste relatório, pela sua relevância e por se terem provado
problemáticas ao longo dos processos de revisão: a pontuação e a aplicação do Acordo
Ortográfico da Língua Portuguesa de 1990 (AO90). Serão ainda apresentados alguns
dilemas do foro estilístico, que se detetaram com maior frequência ou que suscitaram
dúvidas. Para os problemas expostos, serão apresentadas sugestões de correção
devidamente justificadas.
Para concluir, será feita uma reflexão sobre o trabalho realizado durante o
estágio e uma análise sobre a necessidade da existência de um especialista em ciências
da linguagem numa instituição deste género
Estudo do desempenho a longo prazo de aterros
Nos últimos anos na área dos aterros viários foram desenvolvidas essencialmente duas linhas de investigação. A primeira corresponde à utilização de equipamentos de pequeno porte para controlo da compactação dos aterros. A segunda serve-se da teoria dos solos não saturados para explicar as deformações dos aterros a curto e a longo prazo. A presente tese é constituída por duas partes que juntam estas duas vertentes. Na primeira parte é avaliada a eficácia de novas metodologias de avaliação do desempenho em aterros viários, na ótica da sua deformabilidade e resistência, na segunda parte é apresentada a caracterização experimental e a modelação hidromecânica da resposta de um aterro considerando o efeito da sucção.
Na primeira parte são utilizados os equipamentos de ensaio DIP e geogauge, que caracterizam a compacidade e a rigidez (deformabilidade) do meio, e o equipamento PDL, que possibilita estimar a compacidade e resistência do terreno. No aterro de uma barragem em construção foram realizados ensaios com estes equipamentos e com os métodos clássicos de controlo de aterros. A análise integrada destes resultados evidencia algumas tendências estatísticas entre os métodos clássicos e os resultados dos ensaios com o DIP, geogauge e PDL, bem como afasta a possibilidade, para o solo estudado, de estabelecer outras. Nos resultados obtidos é exposta a
dualidade existente entre a compactação no lado seco e no lado húmido do ótimo Proctor. Esta dualidade conduz ao efeito do estado hídrico do solo – logo da sucção então instalada – nas propriedades estimadas experimentalmente.
Na segunda parte é estudado o efeito da sucção na resposta hidromecânica laboratorial de uma areia bem graduada (SW, segundo os critérios da CUS). É demostrada a simulação do comportamento a longo prazo de aterros viários construídos nesse material, sob condições climáticas diversas e contrastantes, através de um modelo temporal de elementos finitos calculado no programa Code-Bright. Na modelação foi utilizado o modelo constitutivo BBM, de raiz inserida na Teoria dos Estados Críticos, cujos parâmetros elásticos, elastoplásticos e hídricos foram estimados com base num extenso programa laboratorial. Neste, foram realizados ensaios avançados para determinação da curva característica e da sua histerese, bem como ensaios de compressão isotrópica e triaxial, com ciclos de descarga-recarga
Compositional data analysis as a potential tool to study the (paleo)ecology of calcareous nannoplankton from the central portuguese submarine canyons (W off Portugal)
Submarine canyons are deep and steep incisions on the continental margins. The physical forcing
mechanisms linked with these marine systems, such as the enhancement of upwelling and bottom
sediment resuspension, are expected to provide a nutrient source that will increase phytoplankton
density (Hickey, 1995, Kampf,2006).
Coccolithophores are the predominant phytoplanktonic group within the calcareous
nannoplankton and their sensitivity to a variety of surface water environmental parameters makes
them important markers of oceanographic processes and proxies of sea surface water masses and
temperatures, productivity and past climate changes (e.g. Ziveri et al., 2004; Silva et al., 2008).
In the present work we propose to test compositional analysis (Buccianti & Esposito, 2004;
Pawlowsky-Glahn & Egozcue, 2006) as a tool to: a) achieve a clearer distinction between
opportunistic coastal-neritic species (r-strategists) and typical oceanic species (k-strategists) in the
central Portuguese margin, and b) to identify a coccolith assemblage that might reflect favorable
environmental conditions found in the vicinity of the canyon that promotes the productivity of
calcareous nannoplankton. Our main difficulty will be in distinguishing the ecological signal from
the effects of other environmental factors mentioned before (i.e. advection, dissolution, bottom
resuspension). One way to infer the species’ ecological inter-relationships is by determining species
relative percentages. The main concern is how the closure problem and the inconsistency of
percentage determinations will affect our results. Compositional analysis was designed to provide
more reliable and thus representative results, since the inference made on the coccolith assemblage
features from which the data are drawn is correctly performed from a theoretical point of view
(Buccianti & Esposito, 2004). Here, we present the first insights from applying compositional data
analysis to coccolith assemblages from 85 surface sediment samples collected from the central
