    Peroxisomes in intestinal and gallbladder epithelial cells of the stickleback, Gasterosteus aculeatus L. (Teleostei)

    The occurrence of microbodies in the epithelial cells of the intestine and gallbladder of the stickleback, Gasterosteus aculeatus L., is described. In the intestine the organelles are predominantly located in the apical and perinuclear zone of the cells and may contain small crystalline cores. In gallbladder epithelial cells the microbodies are distributed randomly. The latter organdies are characterized by the presence of large crystalloids. Cytochemical and biochemical experiments show that catalase and D-amino acid oxidase are main matrix components of the microbodies in both the intestinal and gallbladder epithelia. These organelles therefore are considered peroxisomes. In addition, in intestinal mucosa but not in gallbladder epithelium a low activity of palmitoyl CoA oxidase was detected biochemically. Urate oxidase and L-α hydroxy acid oxidase activities could not be demonstrated.

    OBJECTIVE—Type 2 diabetes is the most common form of diabetes worldwide. Some of its complications, such as retinop-athy and neuropathy, are long-term and protracted, with an un-clear etiology. Given this problem, genetic model systems, such as in flies where type 2 diabetes can be modeled and studied, offer distinct advantages. RESEARCH DESIGN AND METHODS—We used individual flies, control, and mutant individuals with partial loss-of-function insulin pathway genes. We measured wing size and tested body weight for growth phenotypes, the latter by means of a microbal-ance. We studied total lipid and carbohydrate content, lipids by a reaction in single fly homogenates with vanillin-phosphoric acid, and carbohydrates with an anthrone-sulfuric acid reaction.     Large-scale changes of the cloud coverage in the ε Indi Ba,Bb system

    We present the results of 14 nights of I-band photometric monitoring of the nearby brown dwarf binary, ε Indi Ba,Bb. Observations were acquired over 2 months, and total close to 42 hours of coverage at a typically high cadence of 1.4 minutes. At a separation of just 0.7″, we do not resolve the individual components, and so effectively treat the binary as if it were a single object. However, ε Indi Ba (spectral type T1) is the brightest known T-type brown dwarf, and is expected to dominate the photometric signal. We typically find no strong variability associated with the target during each individual night of observing, but see significant changes in mean brightness - by as much as 0.10 magnitudes - over the 2 months of the campaign. This strong variation is apparent on a timescale of at least 2 days. We detect no clear periodic signature, which suggests we may be observing the T1 brown dwarf almost pole-on, and the days-long variability in mean brightness is caused by changes in the large-scale structure of the cloud coverage. Dynamic clouds will very likely produce lightning, and complementary high cadence V-band and Hα images were acquired to search for the emission signatures associated with stochastic ‘strikes’.     <p>Abstract</p> <p>Background</p> <p>Dyslipidemia, particularly hypertriglyceridemia is common in uremia, and represents an independent risk factor for atherosclerosis.</p> <p>Methods</p> <p>To investigate the effects of hemodialysis (HD) duration on very low density lipoprotein (VLDL) and low density lipoprotein (LDL) compositions and lipopolytic activities, 20 patients on 5 to 7 years hemodialysis were followed-up during 9 years. Blood samples were drawn at T0 (beginning of the study), T1 (3 years after initiating study), T2 (6 years after initiating study) and T3 (9 years after initiating study). T0 was taken as reference.</p> <p>Results</p> <p>Triacylglycerols (TG) values were correlated with HD duration (r = 0.70, P < 0.05). An increase of total cholesterol was noted at T2 and T3. Lowered activity was observed for lipoprotein lipase (LPL) (-44%) at T3 and hepatic lipase (HL) (-29%) at T1, (-64%) at T2 and (-73%) at T3. Inverse relationships were found between HD duration and LPL activity (r = -0.63, P < 0.05), and HL activity (r = -0.71, P < 0.01). At T1, T2 and T3, high VLDL-amounts and VLDL-TG and decreased VLDL-phospholipids values were noted. Increased LDL-cholesteryl esters values were noted at T1 and T2 and in LDL-unesterified cholesterol at T2 and T3.</p> <p>Conclusion</p> <p>Despite hemodialysis duration, VLDL-LDL metabolism alterations are aggravated submitting patients to a greater risk of atherosclerosis.</p

    The Role of the Substantia Nigra Pars Compacta in Regulating Sleep Patterns in Rats

    Background. As of late, dopaminergic neurotransmission has been recognized to be involved in the generation of sleep disturbances. Increasing evidence shows that sleep disturbances in Parkinson's disease (PD) patients are mostly related to the disease itself, rather than being a secondary phenomenon. Evidence contained in the literature lends support to the hypothesis that the dopaminergic nigrostriatal pathway is closely involved in the regulation of sleep patterns. Methodology/Principal Findings. To test this hypothesis we examined the electrophysiological activity along the sleep-wake cycle of rats submitted to a surgically induced lesion of the SNpc by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP). We demonstrated that a 50% lesion of the substantia nigra pars compacta (SNpc) suffices to produce disruptions of several parameters in the sleep-wake pattern of rats. A robust and constant decrease in the latency to the onset of slow wave sleep (SWS) was detected throughout the five days of recording in both light [F((22.16)) = 72.46, p<0.0001] and dark [F((22.16)) = 75.0, p<0.0001] periods. Also found was a pronounced increase in the percentage of sleep efficiency during the first four days of recording [F((21.15)) = 21.48, p<0.0001], in comparison to the sham group. Additionally, the reduction in the SNpc dopaminergic neurons provoked an ablation in the percentage of rapid eye movement sleep (REM) during three days of the sleep-wake recording period with a strong correlation (r = 0.91; p<0.0001) between the number of dopaminergic neurons lost and the percentage decrease of REM sleep on the first day of recording. On day 4, the percentage of REM sleep during the light and dark periods was increased, [F((22.16)) = 2.46, p<0.0007], a phenomenon consistent with REM rebound. Conclusions/Significance.     Ubiquitous presence of gluconeogenic regulatory enzyme, fructose-1,6-bisphosphatase, within layers of rat retina

    To shed some light on gluconeogenesis in mammalian retina, we have focused on fructose-1,6-bisphosphatase (FBPase), a regulatory enzyme of the process. The abundance of the enzyme within the layers of the rat retina suggests that, in mammals in contrast to amphibia, gluconeogenesis is not restricted to one specific cell of the retina. We propose that FBPase, in addition to its gluconeogenic role, participates in the protection of the retina against reactive oxygen species. Additionally, the nuclear localization of FBPase and of its binding partner, aldolase, in the retinal cells expressing the proliferation marker Ki-67 indicates that these two gluconeogenic enzymes are involved in non-enzymatic nuclear processes

    Evidence for alternative electron sinks to photosynthetic carbon assimilation in the high mountain plant species Ranunculus glacialis

    International audienceThe high mountain plant species Ranunculus glacialis has a low antioxidative scavenging capacity and a low activity of thermal dissipation of excess light energy despite its growth under conditions of frequent light and cold stress. In order to examine whether this species is protected from over-reduction by matching photosystem II (PSII) electron transport (ETR) and carbon assimilation, both were analysed simultaneously at various temperatures and light intensities using infrared gas absorption coupled with chlorophyll fluorescence. ETR exceeded electron consumption by carbon assimilation at higher light intensities and at all temperatures tested, necessitating alternative electron sinks. As photorespiration might consume the majority of excess electrons, photorespiration was inhibited by either high internal leaf CO2 molar ratio (C-i), low oxygen partial pressure (0.5% oxygen), or both. At 0.5% oxygen ETR was significantly lower than at 21% oxygen. At 21% oxygen, however, ETR still exceeded carbon assimilation at high C-i, suggesting that excess electrons are transferred to another oxygen consuming reaction when photorespiration is blocked. Nevertheless, photorespiration does contribute to electron consumption. While the activity of the water -water cycle to electron consumption is not known in leaves of R. glacialis, indirect evidence such as the high sensitivity to oxidative stress and the low initial NADP-malate dehydrogenase (NADP-MDH) activity suggests only a minor contribution as an alternative electron sink. Alternatively, the plastid terminal oxidase (PTOX) may transfer excess electrons to oxygen. This enzyme is highly abundant in R. glacialis leaves and exceeds the PTOX content of every other plant species so far examined, including those of transgenic tomato leaves overexpressing the PTOX protein. Finally, PTOX contents strongly declined during deacclimation of R. glacialis plants, suggesting their important role in photoprotection. Ranunculus glacialis is the first reported plant species with such a high PTOX protein content

    Inter-Observer Agreement on Subjects' Race and Race-Informative Characteristics

    Health and socioeconomic disparities tend to be experienced along racial and ethnic lines, but investigators are not sure how individuals are assigned to groups, or how consistent this process is. To address these issues, 1,919 orthodontic patient records were examined by at least two observers who estimated each individual's race and the characteristics that influenced each estimate. Agreement regarding race is high for African and European Americans, but not as high for Asian, Hispanic, and Native Americans. The indicator observers most often agreed upon as important in estimating group membership is name, especially for Asian and Hispanic Americans. The observers, who were almost all European American, most often agreed that skin color is an important indicator of race only when they also agreed the subject was European American. This suggests that in a diverse community, light skin color is associated with a particular group, while a range of darker shades can be associated with members of any other group. This research supports comparable studies showing that race estimations in medical records are likely reliable for African and European Americans, but are less so for other groups. Further, these results show that skin color is not consistently the primary indicator of an individual's race, but that other characteristics such as facial features add significant information
