Généralisation du diagramme de Voronoï et placement de formes géométriques complexes dans un nuage de points.
La géométrie algorithmique est une discipline en pleine expansion dont l'objet est la conception d'algorithmes résolvant des problèmes géométriques. De tels algorithmes sont très utiles notamment dans l'ingénierie, l'industrie et le multimédia. Pour être performant, il est fréquent qu'un algorithme géométrique utilise des structures de données spécialisées.Nous nous sommes intéressés à une telle structure : le diagramme de Voronoï et avons proposé une généralisation de celui-ci. Ladite généralisation résulte d'une extension du prédicat du disque vide (prédicat propre à toute région de Voronoï) à une union de disques. Nous avons analysé les régions basées sur le prédicat étendu et avons proposé des méthodes pour les calculer par ordinateur.Par ailleurs, nous nous sommes intéressés aux problèmes de placement de formes , thème récurrent en géométrie algorithmique. Nous avons introduit un formalisme universel pour de tels problèmes et avons, pour la première fois, proposé une méthode de résolution générique, en ce sens qu'elle est apte à résoudre divers problèmes de placement suivant un même algorithme.Nos travaux présentent, d'une part, l'avantage d'élargir le champ d'application de structures de données basées sur Voronoï. D'autre part, ils facilitent de manière générale l'utilisation de la géométrie algorithmique, en unifiant définitions et algorithmes associés aux problèmes de placement de formes.Computational geometry is an active branch of computer science whose goal is the design of efficient algorithms solving geometric problems. Such algorithms are useful in domains like engineering, industry and multimedia. In order to be efficient, algorithms often use special data structures.In this thesis we focused on such a structure: the Voronoi diagram. We proposed a new generalized diagram. We have proceeded by extending the empty disk predicate (satisfied by every Voronoi region) to an arbitrary union of disks. We have analyzed the new plane regions based on the extended predicate, and we designed algorithms for computing them.Then, we have considered another topic, which is related to the first one: shape placement problems. Such problems have been studied repeatedly by researchers in computational geometry. We introduced new notations along with a global framework for such problems. We proposed, for the first time a generic method, which is able to solve various placement problems using a single algorithm.Thus, our work extend the scope of Voronoi based data structures. Characterization and sequence mapping of large RNA and mRNA therapeutics using mass spectrometry
Large RNA including mRNA (mRNA) has emerged as an important new class of therapeutics. Recently, this has been demonstrated by two highly efficacious vaccines based on mRNA sequences encoding for a modified version of the SARS-CoV-2 spike protein. There is currently significant demand for the development of new and improved analytical methods for the characterization of large RNA including mRNA therapeutics. In this study, we have developed an automated, high-throughput workflow for the rapid characterization and direct sequence mapping of large RNA and mRNA therapeutics. Partial RNase digestions using RNase T1 immobilized on magnetic particles were performed in conjunction with high-resolution liquid chromatography–mass spectrometry analysis. Sequence mapping was performed using automated oligoribonucleotide annotation and identifications based on MS/MS spectra. Using this approach, a >80% sequence of coverage of a range of large RNAs and mRNA therapeutics including the SARS-CoV-2 spike protein was obtained in a single analysis. The analytical workflow, including automated sample preparation, can be completed within 90 min. The ability to rapidly identify, characterize, and sequence map large mRNA therapeutics with high sequence coverage provides important information for identity testing, sequence validation, and impurity analysis
Polytypism and polysomatism in mixed-layer chalcogenides: characterization of PbBi(4)Te(4)S(3) and inferences for ordered phases in the aleksite series
Bi-Pb-chalcogenides of the aleksite series represent homologous mixed-layer compounds derived from tetradymite (Bi₂Te₂S). Considering tetradymite as composed of five-atom (Bi₂Te₂S) layers, the named minerals of the aleksite homologous series, aleksite (PbBi₂Te₂S₂) and saddlebackite, (Pb₂Bi₂Te₂S₃) have been considered as phases formed by regular stacking of longer seven- and nine-atom layers. High-angle annular dark-field scanning transmission electron microscope (HAADF-STEM) imaging of thinned foils prepared in-situ on domains deemed homogeneous from electron probe microanalysis, STEM energy-dispersive X-ray spectrometry (EDS) element mapping and fast Fourier transforms (FFTs) from the images offer new insights into these structures. The hitherto-unnamed phase, PbBi₄Te₄S₃, previously interpreted as regular intergrowths of five- and seven-atom layers, is characterized in terms of regular repeats of five- and seven-atom layers over distances of at least 350 nm, defining the (57), or 24H polytype. Imaging of other domains in the same lamella with identical composition at the electron microprobe scale reveals the presence of two additional polytypes: (5559), or 48H; and (557.559) or 72H. Unit cell dimensions for all three polytypes of PbBi₄Te₄S₃ can be both measured and predicted from the HAADF STEM imaging and FFTs. STEM EDS mapping of each polytype confirm the internal structure of each layer. Lead and S occur within the centre of the layers, i.e., Te–Bi–S–Pb–S–Bi–Te in the seven-atom layer, Te–Bi–S–Pb–S–Pb–S–Bi–Te in the nine-atom layer, and so on. Polytypism is an intrinsic feature of the aleksite series, with each named mineral or unnamed phase, except the simple five-atom layer, defined by several alternative stacking sequences of different length, readily explaining the differing c values given in previous work. Homology is defined in terms of layer width and the stacking arrangement of those layers. Coherent structures of the same composition need not only be built of layers of adjacent size (i.e., five- and seven-atom layers) but, as exemplified by the (5559) polytype, may also contain non-adjacent layers, a finding not anticipated in previous work. New polysomes remain to be discovered in nature and each potentially exists as multiple polytypes. The present study further emphasizes the utility of HAADF STEM imaging and atomic-scale STEM EDS element mapping as an optimal tool for tracking stacking sequences and characterising structures in mixed-layer compounds.Nigel J. A pre-post study of behavioural determinants and practice change in Ugandan clinical officers
Background. Understanding the drivers of ‘provider behaviour’ has been highlighted as one of the six domains of behaviour change in strengthening healthcare systems.Objectives. To assess changes in healthcare provider behaviour, i.e. use of the Airway, Breathing, Circulation, Disability, Exposure (ABCDE) approach in acute illness management, after participating in a 1-day course on the assessment and management of acutely ill patients. We aimed to assess whether changes in psychological determinants of the ABCDE approach were associated with changes in the use of the approach.Methods. We used a pre-post design to study self-reported change in behaviour after a 1-day training course from pre-course to follow-up 1 month later. We also measured psychological determinants of behaviour immediately before and after and at 1-month follow-up. We explored if changes in psychological determinants were associated with change in practice 1 month later.Results. We found the following: firstly, use of the ABCDE approach increased at 1 month post-course from a median use of 50 - 90%. Secondly, the increase in the ABCDE approach was associated with a positive change in only one of the determinants of practice from pre- to post-course: perception of environmental determinants (r=0.323; p<0.05). Finally, there were no other significant associations with practice change or practice at follow-up.Conclusions. Change in perceptions of availability of resources was associated with increased use of an ABCDE approach, but evidence was limited owing to the pre-post design
Fat mass and obesity associated (FTO) gene influences skeletal muscle phenotypes in non-resistance trained males and elite rugby playing position
Background FTO gene variants have been associated with obesity phenotypes in sedentary and obese populations, but rarely with skeletal muscle and elite athlete phenotypes. Methods In 1089 participants, comprising 530 elite rugby athletes and 559 non-athletes, DNA was collected and genotyped for the FTO rs9939609 variant using real-time PCR. In a subgroup of non-resistance trained individuals (NT; n = 120), we also assessed structural and functional skeletal muscle phenotypes using dual energy x-ray absorptiometry, ultrasound and isokinetic dynamometry. In a subgroup of rugby athletes (n = 77), we assessed muscle power during a countermovement jump. Results In NT, TT genotype and T allele carriers had greater total body (4.8% and 4.1%) and total appendicular lean mass (LM; 3.0% and 2.1%) compared to AA genotype, with greater arm LM (0.8%) in T allele carriers and leg LM (2.1%) for TT, compared to AA genotype. Furthermore, the T allele was more common (94%) in selected elite rugby union athletes (back three and centre players) who are most reliant on LM rather than total body mass for success, compared to other rugby athletes (82%; P = 0.01, OR = 3.34) and controls (84%; P = 0.03, OR = 2.88). Accordingly, these athletes had greater peak power relative to body mass than other rugby athletes (14%; P = 2 x 10-6). Conclusion Collectively, these results suggest that the T allele is associated with increased LM and elite athletic success. This has implications for athletic populations, as well as conditions characterised by low LM such as sarcopenia and cachexia
