Who benefits from school-to-work linkages in the labour market?:A comparison between natives, migrants educated abroad, and those educated domestically
Recent research suggests that people with more occupation-specific qualifications (i.e. qualifications that link to a smaller set of occupations) experience greater benefits in the labour market. Based on human capital, signalling and credentialing theory, we argue that these benefits may vary between the native majority population, individuals with a migration background who hold a foreign qualification, and individuals with a migration background with a domestic qualification. Using data from the German Microcensus, we find that for both the native majority and immigrants with a domestic qualification, holding a more occupation-specific qualification relates to a higher chance of working in a position the individual is educated for in terms of both level and field. Holding a more occupation-specific qualification also relates to higher labour market returns (i.e. income and occupational status) for those who work in a job for which they are educated, yet is mostly negatively related to the labour market returns for those who do not work in a job for which they are educated. Migrants with a foreign qualification profit less from holding occupation-specific qualifications and suffer more from their associated disadvantages

Circulating adrenomedullin estimates survival and reversibility of organ failure in sepsis: the prospective observational multinational Adrenomedullin and Outcome in Sepsis and Septic Shock-1 (AdrenOSS-1) study
Background: Adrenomedullin (ADM) regulates vascular tone and endothelial permeability during sepsis. Levels of circulating biologically active ADM (bio-ADM) show an inverse relationship with blood pressure and a direct relationship with vasopressor requirement. In the present prospective observational multinational Adrenomedullin and Outcome in Sepsis and Septic Shock 1 (, AdrenOSS-1) study, we assessed relationships between circulating bio-ADM during the initial intensive care unit (ICU) stay and short-term outcome in order to eventually design a biomarker-guided randomized controlled trial. Methods: AdrenOSS-1 was a prospective observational multinational study. The primary outcome was 28-day mortality. Secondary outcomes included organ failure as defined by Sequential Organ Failure Assessment (SOFA) score, organ support with focus on vasopressor/inotropic use, and need for renal replacement therapy. AdrenOSS-1 included 583 patients admitted to the ICU with sepsis or septic shock. Results: Circulating bio-ADM levels were measured upon admission and at day 2. Median bio-ADM concentration upon admission was 80.5 pg/ml [IQR 41.5-148.1 pg/ml]. Initial SOFA score was 7 [IQR 5-10], and 28-day mortality was 22%. We found marked associations between bio-ADM upon admission and 28-day mortality (unadjusted standardized HR 2.3 [CI 1.9-2.9]; adjusted HR 1.6 [CI 1.1-2.5]) and between bio-ADM levels and SOFA score (p < 0.0001). Need of vasopressor/inotrope, renal replacement therapy, and positive fluid balance were more prevalent in patients with a bio-ADM > 70 pg/ml upon admission than in those with bio-ADM †70 pg/ml. In patients with bio-ADM > 70 pg/ml upon admission, decrease in bio-ADM below 70 pg/ml at day 2 was associated with recovery of organ function at day 7 and better 28-day outcome (9.5% mortality). By contrast, persistently elevated bio-ADM at day 2 was associated with prolonged organ dysfunction and high 28-day mortality (38.1% mortality, HR 4.9, 95% CI 2.5-9.8). Conclusions: AdrenOSS-1 shows that early levels and rapid changes in bio-ADM estimate short-term outcome in sepsis and septic shock. These data are the backbone of the design of the biomarker-guided AdrenOSS-2 trial. Trial registration: ClinicalTrials.gov, NCT02393781. Registered on March 19, 2015

Democratic core values in high school - Year 4 2021-2022:Adolescent panel Democratic Core Values and School Trajectories
Dit rapport brengt de opvattingen van jongeren over de democratische rechtsstaat in kaart voor leerlingen van het vierde jaar van de middelbare school. Het rust op data die zijn verzameld in het Adolescentenpanel Democratische Kernwaarden en Schoolloopbanen (ADKS), een meerjarig onderzoek waarbij duizenden Nederlandse leerlingen in tientallen scholen gedurende het voortgezet onderwijs worden gevolgd om de ontwikkeling van democratische kernwaarden bij jongeren in kaart te brengen. Dit rapport volgt op drie eerdere, waaraan dezelfde leerlingen meewerkten toen zij nog in het eerste, tweede en/of derde leerjaar zaten. De data van jaar 4 zijn verzameld in het voorjaar van 2022. Hoewel de meeste beperkingen vanwege de Covid19-pandemie versoepeld waren, waren ze niet verdwenen. Ze hebben de dataverzameling in jaar 4 bemoeilijkt. We trekken twee overkoepelende conclusies. Ten eerste blijven de grote verschuivingen in steun voor democratische kernwaarden in jaar 3 grotendeels bestaan in jaar 4. De hechting aan de democratie is hoog. Leerlingen vinden het net zo belangrijk om in een democratie te leven als in jaar 3. De vertegenwoordigende democratie is nog altijd de populairste bestuursvorm. Ook lijkt het jongeren leuker om in de politiek te werken, hoewel het politiek zelfvertrouwen constant blijft. Daarnaast praten jongeren meer over politiek en samenleving met ouders, leerkrachten en vrienden dan vorige jaren. Alleen het eerder gestegen voornemen om later te gaan stemmen daalde terug tot het niveau van jaar 2. Tegelijk blijft het vertrouwen in ambtsdragers lager dan in jaar 1 en 2. Het vertrouwen in politici daalt zelfs verder. Het vertrouwen van jongeren dat politici luisteren naar mensen zoals zij en hun ouders is wat gestegen, maar nog steeds veel lager dan in de eerste twee jaren. Tot slot zien we dat jongeren in jaar 4 nog wat meer schuiven naar nadruk op het individu boven de gemeenschap dan in jaar 3: meer privacy boven veiligheid, meer vrijheid van meningsuiting boven anderen niet kwetsen, meer je eigen zin krijgen boven je aan de wet houden. Deze trend moet begrepen worden tegen de achtergrond dat jongeren veelal geen uitgesproken posities innemen. De tweede overkoepelende conclusie betreft de aanhoudende verschillen tussen groepen. De grote verschillen tussen opleidingstypes (vwo, havo en vmbo), die al sinds het begin van de middelbare school aanwezig zijn, blijven het belangrijkste groepsverschil. Vwo-leerlingen zijn het meest gehecht aan de democratie, hebben het meeste politiek vertrouwen en zelfvertrouwen, hebben de grootste neiging te gaan stemmen, en hebben de meeste politieke ambitie. Jongeren op het vmbo scoren op al deze kenmerken in vergelijking een stuk lager. Deze verschillen bestaan al aan de start van de middelbare school, en kunnen niet goed toegewezen worden aan de school. Ze kunnen voor een deel in stand gehouden worden door het vriendennetwerk van jongeren: het grootste deel van de beste vrienden komt uit dezelfde klas. Ook geven jongeren aan grotendeels dezelfde mening te hebben over politiek en maatschappij als hun vrienden
Remodeling of cholinergic input to the hippocampus after noise exposure and tinnitus induction in Guinea pigs
Here, we investigate remodeling of hippocampal cholinergic inputs after noise exposure and determine the relevance of these changes to tinnitus. To assess the effects of noise exposure on the hippocampus, guinea pigs were exposed to unilateral noise for 2 hr and 2 weeks later, immunohistochemistry was performed on hippocampal sections to examine vesicular acetylcholine transporter (VAChT) expression. To evaluate whether the changes in VAChT were relevant to tinnitus, another group of animals was exposed to the same noise band twice to induce tinnitus, which was assessed using gapâprepulse Inhibition of the acoustic startle (GPIAS) 12âweeks after the first noise exposure, followed by immunohistochemistry. Acoustic Brainstem Response (ABR) thresholds were elevated immediately after noise exposure for all experimental animals but returned to baseline levels several days after noise exposure. ABR wave I amplitudeâintensity functions did not show any changes after 2 or 12âweeks of recovery compared to baseline levels. In animals assessed 2âweeks following noiseâexposure, hippocampal VAChT puncta density decreased on both sides of the brain by 20â60% in exposed animals. By 12âweeks following the initial noise exposure, changes in VAChT puncta density largely recovered to baseline levels in exposed animals that did not develop tinnitus, but remained diminished in animals that developed tinnitus. These tinnitusâspecific changes were particularly prominent in hippocampal synapseârich layers of the dentate gyrus and areas CA3 and CA1, and VAChT density in these regions negatively correlated with tinnitus severity. The robust changes in VAChT labeling in the hippocampus 2 weeks after noise exposure suggest involvement of this circuitry in auditory processing. This large, international dataset contains survey responses from N = 12,570 students from 100 universities in 35 countries, collected in 21 languages. We measured anxieties (statistics, mathematics, test, trait, social interaction, performance, creativity, intolerance of uncertainty, and fear of negative evaluation), self-efficacy, persistence, and the cognitive reflection test, and collected demographics, previous mathematics grades, self-reported and official statistics grades, and statistics module details. Data reuse potential is broad, including testing links between anxieties and statistics/mathematics education factors, and examining instrumentsâ psychometric properties across different languages and contexts

IL-1ÎČ-Mediated Activation of Adipose-Derived Mesenchymal Stromal Cells Results in PMN Reallocation and Enhanced Phagocytosis: A Possible Mechanism for the Reduction of Osteoarthritis Pathology
Background: Injection of adipose-derived mesenchymal stromal cells (ASCs) into murine knee joints after induction of inflammatory collagenase-induced osteoarthritis (CiOA) reduces development of joint pathology. This protection is only achieved when ASCs are applied in early CiOA, which is characterized by synovitis and high S100A8/A9 and IL-1ÎČ levels, suggesting that inflammation is a prerequisite for the protective effect of ASCs. Our objective was to gain more insight into the interplay between synovitis and ASC-mediated amelioration of CiOA pathology.Methods: CiOA was induced by intra-articular collagenase injection. Knee joint sections were stained with hematoxylin/eosin and immunolocalization of polymorphonuclear cells (PMNs) and ASCs was performed using antibodies for NIMP-R14 and CD271, respectively. Chemokine expression induced by IL-1ÎČ or S100A8/A9 was assessed with qPCR and Luminex. ASC-PMN co-cultures were analyzed microscopically and with Luminex for inflammatory mediators. Migration of PMNs through transwell membranes toward conditioned medium of non-stimulated ASCs (ASCNS-CM) or IL-1ÎČ-stimulated ASCs (ASCIL-1ÎČ-CM) was examined using flow cytometry. Phagocytic capacity of PMNs was measured with labeled zymosan particles.Results: Intra-articular saline injection on day 7 of CiOA increased synovitis after 6 h, characterized by PMNs scattered throughout the joint cavity and the synovium. ASC injection resulted in comparable numbers of PMNs which clustered around ASCs in close interaction with the synovial lining. IL-1ÎČ-stimulation of ASCs in vitro strongly increased expression of PMN-attracting chemokines CXCL5, CXCL7, and KC, whereas S100A8/A9-stimulation did not. In agreement, the number of clustered PMNs per ASC was significantly increased after 6 h of co-culturing with IL-1ÎČ-stimulated ASCs. Also migration of PMNs toward ASCIL-1ÎČ-CM was significantly enhanced (287%) when compared to ASCNS-CM. Interestingly, association of PMNs with ASCs significantly diminished KC protein release by ASCs (69% lower after 24 h), accompanied by reduced release of S100A8/A9 protein by the PMNs. Moreover, phagocytic capacity of PMNs was strongly enhanced after priming with ASCIL-1ÎČ-CM.Conclusions: Local application of ASCs in inflamed CiOA knee joints results in clustering of attracted PMNs with ASCs in the synovium, which is likely mediated by IL-1ÎČ-induced up-regulation of chemokine release by ASCs. This results in enhanced phagocytic capacity of PMNs, enabling the clearance of debris to attenuate synovitis
