Attention explores space periodically at the theta frequency
Voluntary attention is at the core of a wide variety of cognitive functions. Attention can be oriented to and sustained at a location or reoriented in space to allow processing at other locations—critical in an ever-changing environment. Numerous studies have investigated attentional orienting in time and space, but little is known about the spatiotemporal dynamics of attentional reorienting. Here we explicitly manipulated attentional reorienting using a cuing procedure in a two- alternative forced-choice orientation-discrimination task. We interrogated attentional distribution by flashing two probe stimuli with various delays between the precue and target stimuli. Then we used the probabilities that both probes and neither probe were correctly reported to solve a second-degree equation, which estimates the report probability at each probe location. We demonstrated that attention reorients periodically at ~4 Hz (theta) between the two stimulus locations. We further characterized the processing dynamics at each stimulus location, and demonstrated that attention samples each location periodically at ;11 Hz (alpha). Finally, simulations support our findings and show that this method is sufficiently powered, making it a valuable tool for studying the spatiotemporal dynamics of attention
Resolving desorption of complex organic molecules in a hot core: Transition from non-thermal to thermal desorption or two-step thermal desorption?
Using the high angular resolution provided by the ALMA interferometre we want
to resolve the COM emission in the hot molecular core Sagittarius B2(N1) and
thereby shed light on the desorption process of Complex Organic Molecules
(COMs) in hot cores. We use data taken as part of the 3 mm spectral line survey
Re-exploring Molecular Complexity with ALMA (ReMoCA) to investigate the
morphology of COM emission in Sagittarius B2(N1). Spectra of ten COMs are
modelled under the assumption of LTE and population diagrams are derived for
positions at various distances to the south and west from the continuum peak.
Based on this analysis, resolved COM rotation temperature and COM abundance
profiles are derived. Based on the morphology, a rough separation into O- and
N-bearing COMs can be done. Temperature profiles are in agreement with
expectations of protostellar heating of an envelope with optically thick dust.
Abundance profiles reflect a similar trend as seen in the morphology and, to a
great extent, agree with results of astrochemical models that, besides the
co-desorption with water, predict that O-bearing COMs are mainly formed on dust
grain surfaces at low temperatures while at least some N-bearing COMs and
CHCHO are substantially formed in the gas phase at higher temperatures. Our
observational results, in comparison with model predictions, suggest that COMs
that are exclusively or to a great extent formed on dust grains desorb
thermally at ~100 K from the grain surface likely alongside water. Non-zero
abundance values below ~100 K suggest that another desorption process is at
work at these low temperatures: either non-thermal desorption or partial
thermal desorption related to lower binding energies experienced by COMs in the
outer, water-poor ice layers. In either case, this is the first time that the
transition between two regimes of COM desorption has been resolved in a hot

Nucleocapsid Protein Zinc-Finger Mutants of Simian Immunodeficiency Virus Strain Mne Produce Virions That Are Replication Defectivein Vitroandin Vivo
AbstractAll retroviruses (except the spumaretroviruses) contain a nucleocapsid (NC) protein that encodes one or two copies of the Zn2+-finger sequence -Cys-X2-Cys-X4-His-X4-Cys-. This region has been shown to be essential for recognition and packaging of the genomic RNA during virion particle assembly. Additionally, this region has been shown to be involved in early infection events in a wide spectrum of retroviruses, including mammalian type C [e.g., murine leukemia virus (MuLV)], human immunodeficiency virus type 1 (HIV-1),Rous sarcomavirus, and other retroviruses. Mutations in the two Zn2+-fingers of the NC protein of simian immunodeficiency virus strain Mne [SIV(Mne)] have been generated. The resulting virions contained the normal complement of processed viral proteins with densities indistinguishable from wild-type SIV(Mne). All of the mutants had electron micrograph morphologies similar to those of immature particles observed in wild-type preparations. RNA packaging was less affected by mutations in the NC protein of SIV(Mne) than has been observed for similar mutants in the MuLV and HIV-1 systems. Nevertheless,in vitroreplication of SIV(Mne) NC mutants was impaired to levels comparable to those observed for MuLV and HIV-1 NC mutants; replication defective NC mutants are typically 105- to 106-fold less infectious than similar levels of wild-type virus. One mutant, ΔCys33–Cys36, was also found to be noninfectiousin vivowhen mutant virus was administered intravenously to a pig-tailed macaque. NC mutations can therefore be used to generate replication defective virions for candidate vaccines in the SIV macaque model for primate lentiviral diseases
Genome-wide association study of hepatitis C virus- and cryoglobulin-related vasculitis
The host genetic basis of mixed cryoglobulin vasculitis is not well understood and has not been studied in large cohorts. A genome-wide association study was conducted among 356 hepatitis C virus (HCV) RNA-positive individuals with cryoglobulin-related vasculitis and 447 ethnically matched, HCV RNA-positive controls. All cases had both serum cryoglobulins and a vasculitis syndrome. A total of 899 641 markers from the Illumina HumanOmni1-Quad chip were analyzed using logistic regression adjusted for sex, as well as genetically determined ancestry. Replication of select single-nucleotide polymorphisms (SNPs) was conducted using 91 cases and 180 controls, adjusting for sex and country of origin. The most significant associations were identified on chromosome 6 near the NOTCH4 and MHC class II genes. A genome-wide significant association was detected on chromosome 6 at SNP rs9461776 (odds ratio=2.16, P=1.16E-07) between HLA-DRB1 and DQA1: this association was further replicated in additional independent samples (meta-analysis P=7.1 × 10(-9)). A genome-wide significant association with cryoglobulin-related vasculitis was identified with SNPs near NOTCH4 and MHC Class II genes. The two regions are correlated and it is difficult to disentangle which gene is responsible for the association with mixed cryoglobulinemia vasculitis in this extended major histocompatibility complex region

Using human genetics to understand the disease impacts of testosterone in men and women.
Testosterone supplementation is commonly used for its effects on sexual function, bone health and body composition, yet its effects on disease outcomes are unknown. To better understand this, we identified genetic determinants of testosterone levels and related sex hormone traits in 425,097 UK Biobank study participants. Using 2,571 genome-wide significant associations, we demonstrate that the genetic determinants of testosterone levels are substantially different between sexes and that genetically higher testosterone is harmful for metabolic diseases in women but beneficial in men. For example, a genetically determined 1 s.d. higher testosterone increases the risks of type 2 diabetes (odds ratio (OR) = 1.37 (95% confidence interval (95% CI): 1.22-1.53)) and polycystic ovary syndrome (OR = 1.51 (95% CI: 1.33-1.72)) in women, but reduces type 2 diabetes risk in men (OR = 0.86 (95% CI: 0.76-0.98)). We also show adverse effects of higher testosterone on breast and endometrial cancers in women and prostate cancer in men. The Difficulty of Detecting Occult Metastases in Patients with Potentially Resectable Pancreatic Cancer:Development and External Validation of a Preoperative Prediction Model
Occult metastases are detected in 10–15% of patients during exploratory laparotomy for pancreatic cancer. This study developed and externally validated a model to predict occult metastases in patients with potentially resectable pancreatic cancer. Model development was performed within the Dutch Pancreatic Cancer Audit, including all patients operated for pancreatic cancer (January 2013–December 2017). Multivariable logistic regression analysis based on the Akaike Information Criteria was performed with intraoperative pathologically proven metastases as the outcome. The model was externally validated with a cohort from the University Hospital of Verona (January 2013–December 2017). For model development, 2262 patients were included of whom 235 (10%) had occult metastases, located in the liver (n = 143, 61%), peritoneum (n = 73, 31%), or both (n = 19, 8%). The model included age (OR 1.02, 95% CI 1.00–1.03), BMI (OR 0.96, 95% CI 0.93–0.99), preoperative nutritional support (OR 1.73, 95% CI 1.01–2.74), tumor diameter (OR 1.60, 95% CI 1.04–2.45), tumor composition (solid vs. cystic) (OR 2.33, 95% CI 1.20–4.35), and indeterminate lesions on preoperative imaging (OR 4.01, 95% CI 2.16–7.43). External validation showed poor discrimination with a C-statistic of 0.56. Although some predictor variables were significantly associated with occult metastases, the model performed insufficiently at external validation.</p

One way to assess whether the results obtained depend on the analysis strategy chosen is to employ multiple analysts and leave each of them free to follow their own approach. Here, we present consensus-based guidance for conducting and reporting such multi-analyst studies, and we discuss how broader adoption of the multi-analyst approach has the potential to strengthen the robustness of results and conclusions obtained from analyses of datasets in basic and applied research
