    Comparing American soccer dialogues: social media commentary Surrounding the 2014 US men’s and 2015 US women’s World Cup teams

    Mega sporting events such as the World Cup have been found to stimulate categorization of in-groups and out-groups among fans. While self-categorization correlates with gender, the sport of soccer also facilitates nationalistic categorization. The World Cup features nation vs. nation competition while making gender a non-variable as the men and women compete in separate tournaments in separate years. This study examined 33,529 tweets illustrating social media match commentary involving US teams and opponents on Twitter during the 2014 and 2015 World Cups. Results revealed US teams were more likely to be described in regard to attributions of success and failure, while opposition teams were more likely to receive personal and physical attributions. Conversely, no differences were found between US Men’s and Women’s teams in regard to characterizations of success and failure, but revealed the Women’s team was more likely to receive personal and physical characterizations

    ‘Get back to the kitchen, cos u talk s*** on tv’: gendered online abuse and trigger events in sport

    Research question: Online abuse is prevalent in sport and can be the by-product of trigger events–reactive social media posts that motivate online hate. Little is known about what triggers online abuse, types of content, and how this impacts certain groups. The current research examined how online behaviour emerges, and evolves during a trigger event, through a gendered lens. Research methods: This research employed a two phase, mixed methods approach of a digital netnography with participation observation through social network analysis and thematic content analysis of 1332 (N = 1332) tweets in the United Kingdom. The trigger event examined abusive content toward Karen Carney following post-match football commentary on 29 December 2020. Results and findings: Results identified 590 individuals who formed two distinct groups. Directed network visualisation indicated Carney was the focus of the trigger event. Thematic time series analysis revealed emotional maltreatment (i.e. ridiculing, humiliating, belittling) progressing to overt gendered discriminatory maltreatment. Implications: Findings support the need for safeguarding policies for target groups, as trigger events escalate quickly, and group affiliations impact abusive content. From a theoretical standpoint, in-group and out-group affiliations resulted in rhetoric highlighting persistent, gendered socio-normative issues within sport, amplified in an online environment

    Optimising experimental design for high-throughput phenotyping in mice: a case study

    To further the functional annotation of the mammalian genome, the Sanger Mouse Genetics Programme aims to generate and characterise knockout mice in a high-throughput manner. Annually, approximately 200 lines of knockout mice will be characterised using a standardised battery of phenotyping tests covering key disease indications ranging from obesity to sensory acuity. From these findings secondary centres will select putative mutants of interest for more in-depth, confirmatory experiments. Optimising experimental design and data analysis is essential to maximise output using the resources with greatest efficiency, thereby attaining our biological objective of understanding the role of genes in normal development and disease. This study uses the example of the noninvasive blood pressure test to demonstrate how statistical investigation is important for generating meaningful, reliable results and assessing the design for the defined research objectives. The analysis adjusts for the multiple-testing problem by applying the false discovery rate, which controls the number of false calls within those highlighted as significant. A variance analysis finds that the variation between mice dominates this assay. These variance measures were used to examine the interplay between days, readings, and number of mice on power, the ability to detect change. If an experiment is underpowered, we cannot conclude whether failure to detect a biological difference arises from low power or lack of a distinct phenotype, hence the mice are subjected to testing without gain. Consequently, in confirmatory studies, a power analysis along with the 3Rs can provide justification to increase the number of mice used

    Finishing the euchromatic sequence of the human genome

    The sequence of the human genome encodes the genetic instructions for human physiology, as well as rich information about human evolution. In 2001, the International Human Genome Sequencing Consortium reported a draft sequence of the euchromatic portion of the human genome. Since then, the international collaboration has worked to convert this draft into a genome sequence with high accuracy and nearly complete coverage. Here, we report the result of this finishing process. The current genome sequence (Build 35) contains 2.85 billion nucleotides interrupted by only 341 gaps. It covers ∼99% of the euchromatic genome and is accurate to an error rate of ∼1 event per 100,000 bases. Many of the remaining euchromatic gaps are associated with segmental duplications and will require focused work with new methods. The near-complete sequence, the first for a vertebrate, greatly improves the precision of biological analyses of the human genome including studies of gene number, birth and death. Notably, the human enome seems to encode only 20,000-25,000 protein-coding genes. The genome sequence reported here should serve as a firm foundation for biomedical research in the decades ahead

    Immunoglobulin, glucocorticoid, or combination therapy for multisystem inflammatory syndrome in children: a propensity-weighted cohort study.

    BACKGROUND: Multisystem inflammatory syndrome in children (MIS-C), a hyperinflammatory condition associated with SARS-CoV-2 infection, has emerged as a serious illness in children worldwide. Immunoglobulin or glucocorticoids, or both, are currently recommended treatments. METHODS: The Best Available Treatment Study evaluated immunomodulatory treatments for MIS-C in an international observational cohort. Analysis of the first 614 patients was previously reported. In this propensity-weighted cohort study, clinical and outcome data from children with suspected or proven MIS-C were collected onto a web-based Research Electronic Data Capture database. After excluding neonates and incomplete or duplicate records, inverse probability weighting was used to compare primary treatments with intravenous immunoglobulin, intravenous immunoglobulin plus glucocorticoids, or glucocorticoids alone, using intravenous immunoglobulin as the reference treatment. Primary outcomes were a composite of inotropic or ventilator support from the second day after treatment initiation, or death, and time to improvement on an ordinal clinical severity scale. Secondary outcomes included treatment escalation, clinical deterioration, fever, and coronary artery aneurysm occurrence and resolution. This study is registered with the ISRCTN registry, ISRCTN69546370. FINDINGS: We enrolled 2101 children (aged 0 months to 19 years) with clinically diagnosed MIS-C from 39 countries between June 14, 2020, and April 25, 2022, and, following exclusions, 2009 patients were included for analysis (median age 8·0 years [IQR 4·2-11·4], 1191 [59·3%] male and 818 [40·7%] female, and 825 [41·1%] White). 680 (33·8%) patients received primary treatment with intravenous immunoglobulin, 698 (34·7%) with intravenous immunoglobulin plus glucocorticoids, 487 (24·2%) with glucocorticoids alone; 59 (2·9%) patients received other combinations, including biologicals, and 85 (4·2%) patients received no immunomodulators. There were no significant differences between treatments for primary outcomes for the 1586 patients with complete baseline and outcome data that were considered for primary analysis. Adjusted odds ratios for ventilation, inotropic support, or death were 1·09 (95% CI 0·75-1·58; corrected p value=1·00) for intravenous immunoglobulin plus glucocorticoids and 0·93 (0·58-1·47; corrected p value=1·00) for glucocorticoids alone, versus intravenous immunoglobulin alone. Adjusted average hazard ratios for time to improvement were 1·04 (95% CI 0·91-1·20; corrected p value=1·00) for intravenous immunoglobulin plus glucocorticoids, and 0·84 (0·70-1·00; corrected p value=0·22) for glucocorticoids alone, versus intravenous immunoglobulin alone. Treatment escalation was less frequent for intravenous immunoglobulin plus glucocorticoids (OR 0·15 [95% CI 0·11-0·20]; p<0·0001) and glucocorticoids alone (0·68 [0·50-0·93]; p=0·014) versus intravenous immunoglobulin alone. Persistent fever (from day 2 onward) was less common with intravenous immunoglobulin plus glucocorticoids compared with either intravenous immunoglobulin alone (OR 0·50 [95% CI 0·38-0·67]; p<0·0001) or glucocorticoids alone (0·63 [0·45-0·88]; p=0·0058). Coronary artery aneurysm occurrence and resolution did not differ significantly between treatment groups. INTERPRETATION: Recovery rates, including occurrence and resolution of coronary artery aneurysms, were similar for primary treatment with intravenous immunoglobulin when compared to glucocorticoids or intravenous immunoglobulin plus glucocorticoids. Initial treatment with glucocorticoids appears to be a safe alternative to immunoglobulin or combined therapy, and might be advantageous in view of the cost and limited availability of intravenous immunoglobulin in many countries.     Communicating via photographs: A gendered analysis of Olympic athletes’ visual self-presentation on Instagram

    The purpose of this study was to examine Olympic athletes’ self-presentation on Instagram, a social media outlet on which users post photographs as their primary communication mechanism, in order to develop an understanding of the ways in which athletes use this medium as a communication and marketing tool to build their personal brand. Additionally, this research sought to understand whether differences existed between the utilisation of Instagram by male and female athletes. Drawing on Goffman's (1959) theory of self-presentation, this study examined eight purposively selected Olympic athletes’ Instagram photos. Findings revealed similarities to previous research on athletes’ use of written social media outlets such as Facebook and Twitter, as the majority of photos posted by athletes were personal in nature, thus reflecting backstage performance strategies according to self-presentation theory. The results indicated that female athletes were more likely to share photos of themselves and photos taken in private settings, but male athletes who posted a wider variety of photos encouraged greater engagement from their followers. Personal brand management implications for athletes and sport organisations are examined in greater detail in the discussion.</p
