DARPins detect the formation of hetero-tetramers of p63 and p73 in epithelial tissues and in squamous cell carcinoma
The two p53 homologues p63 and p73 regulate transcriptional programs in epithelial tissues and several cell types in these tissues express both proteins. All members of the p53 family form tetramers in their active state through a dedicated oligomerization domain that structurally assembles as a dimer of dimers. The oligomerization domain of p63 and p73 share a high sequence identity, but the p53 oligomerization domain is more divergent and it lacks a functionally important C-terminal helix present in the other two family members. Based on these structural differences, p53 does not hetero-oligomerize with p63 or p73. In contrast, p63 and p73 form hetero-oligomers of all possible stoichiometries, with the hetero-tetramer built from a p63 dimer and a p73 dimer being thermodynamically more stable than the two homo-tetramers. This predicts that in cells expressing both proteins a p63/p73 hetero-tetramer is formed. So far, the tools to investigate the biological function of this hetero-tetramer have been missing. Here we report the generation and characterization of Designed Ankyrin Repeat Proteins (DARPins) that bind with high affinity and selectivity to the p63/p73 hetero-tetramer. Using these DARPins we were able to confirm experimentally the existence of this hetero-tetramer in epithelial mouse and human tissues and show that its level increases in squamous cell carcinoma
Insights into mantle composition and mantle melting beneath mid-ocean ridges from postspreading volcanism on the fossil Galapagos Rise
New major and trace element and Sr, Nd, and Pb isotope data, together with 39Ar-40Ar ages for lavas from the extinct Galapagos Rise spreading center in the eastern Pacific reveal the evolution in magma compositions erupted during slowdown and after the end of active spreading at a mid-ocean ridge. Lavas erupted at 9.2 Ma, immediately prior to the end of spreading are incompatible element depleted mid-ocean ridge tholeiitic basalts, whereas progressively younger (7.5 to 5.7 Ma) postspreading lavas are increasingly alkalic, have higher concentrations of incompatible elements, higher La/Yb, K/Ti, 87Sr/86Sr, and lower 143Nd/144Nd ratios and were produced by smaller degrees of mantle melting. The large, correlated variations in trace element and isotope compositions can only be explained by melting of heterogenous mantle, in which incompatible trace element enriched lithologies preferentially contribute to smaller degree mantle melts. The effects of variable degrees of melting of heterogeneous mantle on lava compositions must be taken into account when using mid-ocean ridge basalt (MORB) to infer the conditions of melting beneath active spreading ridges. For example, the stronger âgarnet signatureâ inferred from Sm/Nd and 143Nd/144Nd ratios for postspreading lavas from the Galapagos Rise results from a larger contribution from enriched lithologies with high La/Yb and Sm/Yb, rather than from a greater proportion of melting in the stability field of garnet peridotite. Correlations between ridge depth and Sm/Yb and fractionation-corrected Na concentrations in MORB worldwide could result from variations in mantle fertility and/or variations in the average degree of melting, rather than from large variations in mantle temperature. If more fertile mantle lithologies are preferentially melted beneath active spreading ridges, then the upper mantle may be significantly more âdepletedâ than is generally inferred from the compositions of MORB

Responsivity of the striatal dopamine system to methylphenidate â a within-subject I-123-Ă-CIT-SPECT study in children and adolescents with Attention-Deficit/Hyperactivity Disorder
Background Methylphenidate (MPH) is the first-line pharmacological treatment of attention-deficit/hyperactivity disorder (ADHD). MPH binds to the dopamine (DA) transporter (DAT), which has high density in the striatum. Assessments of the striatal dopamine transporter by single positron emission computed tomography (SPECT) in childhood and adolescent patients are rare but can provide insight in how effects of MPH affect DAT availability. The aim of our within-subject study was to investigate the effect of MPH on DAT availability and how responsivity to MPH in DAT availability is linked to clinical symptoms and cognitive functioning.
Methods Thirteen adolescent male patients (9-16 years) with diagnosis of ADHD according to DSM-IV and long-term stimulant medication (for at least 6 months) with MPH were assessed twice within 7 days using SPECT after application of I-123-Ă-CIT to examine DAT binding potential (DAT BP). SPECT measures took place in on and off-MPH status balanced for order across participants. A virtual-reality continuous-performance test was performed at each time point. Further clinical symptoms were assessed for baseline off-MPH.
Results On-MPH status was associated with a highly significant decrease (â27,6%) of striatal DAT BP as compared to off-MPH (t=4.93, p<0.001). More pronounced decrease in striatal DAT BP was associated with higher off-MPH attentional and externalizing symptom ratings (Pearson r=0.68, p=0.01). Striatal DAT BP off-MPH, but not on-MPH, was associated with higher symptom ratings off-MPH (Pearson r=0.56, p=0.04). In further exploratory analysis in left vs. right striatal sub-regions, stronger decrease in DAT BP in the right caudate nucleus was weakly associated with improved performance in the continuous-performance test (Pearson r= - 0.54, p=0.07).
Conclusion Our findings corroborate previous reports from mainly adult samples that MPH reduces striatal DAT BP availability and suggest higher off-MPH DAT BP, likely reflecting low baseline DA levels, as a marker of symptom severity. More speculatively, regional specific responsivity of DAT BP to MPH may reflect treatment response with respect to cognitive functioning. However, implications from this small patient sample should be treated with caution and warrant replication

A more pronounced change in striatal DAT BP was associated with higher off-MPH attentional and externalizing symptom ratings (Pearson r = 0.68, p = 0.01). Striatal DAT BP off-MPH, but not on-MPH, was associated with higher symptom ratings (Pearson r = 0.56, p = 0.04). Results
On-MPH status was associated with a highly significant change (-29.9%) of striatal DAT BP as compared to off-MPH (t = -4.12, p = 0.002). A more pronounced change in striatal DAT BP was associated with higher off-MPH attentional and externalizing symptom ratings (Pearson r = 0.68, p = 0.01). Striatal DAT BP off-MPH, but not on-MPH, was associated with higher symptom ratings (Pearson r = 0.56, p = 0.04).
Conclusion
Our findings corroborate previous reports from mainly adult samples that MPH changes striatal DAT BP availability and suggest higher off-MPH DAT BP, likely reflecting low baseline DA levels, as a marker of symptom severity
Responsivity of the striatal dopamine system to methylphenidate â A within-subject I-123-ÎČ-CIT-SPECT study in male children and adolescents with attention-deficit/hyperactivity disorder
Background:
Methylphenidate (MPH) is the first-line pharmacological treatment of attention-deficit/hyperactivity disorder (ADHD). MPH binds to the dopamine (DA) transporter (DAT), which has high density in the striatum. Assessments of the striatal dopamine transporter by single positron emission computed tomography (SPECT) in childhood and adolescent patients are rare but can provide insight on how the effects of MPH affect DAT availability. The aim of our within-subject study was to investigate the effect of MPH on DAT availability and how responsivity to MPH in DAT availability is linked to clinical symptoms and cognitive functioning.
Methods
Thirteen adolescent male patients (9â16 years) with a diagnosis of ADHD according to the DSM-IV and long-term stimulant medication (for at least 6 months) with MPH were assessed twice within 7 days using SPECT after application of I-123-ÎČ-CIT to examine DAT binding potential (DAT BP). SPECT measures took place in an on- and off-MPH status balanced for order across participants. A virtual reality continuous performance test was performed at each time point. Further clinical symptoms were assessed for baseline off-MPH.

TREATgermany, ein Register fĂŒr Patienten mit moderater bis schwerer atopischer Dermatitis (AD), etablierte im FrĂŒhjahr 2020 einen zusĂ€tzlichen Fragebogen, um Auswirkungen der Corona-Pandemie auf die Lebenssituation von Patienten mit AD zu untersuchen.
Methodik
Anhand eines Fragebogens wurden allgemeine Informationen zum Erleben der Corona-Pandemie und, unter Anwendung des Inventars lebensverÀndernder Ereignisse, die resultierenden Belastungen der Patienten untersucht. Zur Analyse möglicher Assoziationen zwischen Erkrankungsschwere (EASI-Score, oSCORAD, IGA, PGA, POEM), LebensqualitÀt (DLQI) und persönlichen Belastungen wurden t-Tests, Varianzanalysen und Korrelationen kontrolliert nach Geschlecht und Alter genutzt.
Ergebnisse
58 % (N = 233) der einbezogenen 400 Registerpatienten berichteten eine hohe Belastung durch die Corona-Pandemie, unabhĂ€ngig von einer tatsĂ€chlichen Infektion. MĂ€nner zeigten signifikant höhere Belastungswerte als Frauen sowie jĂŒngere höhere als Ă€ltere Befragte (jeweils P = 0,03). Es gab keine Unterschiede der Belastungswerte bezogen auf die Ă€rztliche Beurteilung der Erkrankungsschwere. Jedoch reflektierten Patienten mit höherer EinschrĂ€nkung der LebensqualitĂ€t und ausgeprĂ€gterer Krankheitslast die Belastung durch die Corona-Pandemie als weniger hoch (DLQI P = 0,019, PGA P = 0,044).
Schlussfolgerungen
Es konnte gezeigt werden, dass die Registerpatienten die Corona-Pandemie als lebensverĂ€nderndes Ereignis reflektieren und die Belastung unterschiedlich wahrnehmen. Dies sollte sowohl bei der Behandlung von Patienten mit moderater bis schwerer AD berĂŒcksichtigt als auch in weiterfĂŒhrenden Untersuchungen vertieft werden.BACKGROUND: TREATgermany, a registry for patients with moderate to severe atopic dermatitis (AD), established an additional questionnaire in spring 2020 to investigate the effects of the coronavirus pandemic on the daily life of patients with AD.; MATERIAL AND METHODS: A questionnaire was used to analyze general information regarding a patient's experience of the coronavirus pandemic and, using the Inventory of Life-Changing Events, the resulting personal burden. To analyze possible associations between disease severity (EASI score, oSCORAD, IGA, PGA, POEM), quality of life (DLQI) and personal burden, t-tests, analyses of variance and correlations were evaluated, controlled for sex and age.; RESULTS: 58% (n=233) of the included 400 registry patients reported high burden scores caused by the coronavirus pandemic, regardless of an actual infection. Men showed significantly higher burden scores than women, and younger than older respondents (both P=0.03). There were no differences in burden scores related to the physician's assessment of disease severity. However, patients with higher quality of life impairments and higher disease severity perceived the burden of the coronavirus pandemic as less severe (DLQI P=0.019, PGA P=0.044).; CONCLUSIONS: Our data show that registry patients considered the coronavirus pandemic as a life-changing event and perceived the burden differently.
