    A Dissociation of Attention and Awareness in Phase-sensitive but Not Phase-insensitive Visual Channels

    The elements most vivid in our conscious awareness are the ones to which we direct our attention. Scientific study confirms the impression of a close bond between selective attention and visual awareness, yet the nature of this association remains elusive. Using visual afterimages as an index, we investigate neural processing of stimuli as they enter awareness and as they become the object of attention. We find evidence of response enhancement accompanying both attention and awareness, both in the phase-sensitive neural channels characteristic of early processing stages and in the phase-insensitive channels typical of higher cortical areas. The effects of attention and awareness on phase-insensitive responses are positively correlated, but in the same experiments, we observe no correlation between the effects on phase-sensitive responses. This indicates independent signatures of attention and awareness in early visual areas yet a convergence of their effects at more advanced processing stages

    Viscoelastic liquid crystal colloids for the continuous processing of twisted nematic electro-optical cells

    Liquid crystal colloid materials are described based on the liquid crystal (LC) E7 and submicron sized poly(methyl methacrylate-co-divinylbenzene) particles. Application of a thermal treatment to the composite material produces a finely dispersed network of the internally crosslinked polymeric inclusions in the LC-E7. Dynamic rheological measurements on the LC colloids show that the presence of this network imposes pronounced viscoelastic behavior on the material, which may be exploited in the manufacturing of large-area twisted nematic (TN) electro-optical cells via continuous methods as an alternative to the currently available batchwise routes. The electro-optical characteristics of TN cells based on the composite material are approximately comparable to the electro-optical characteristics of a reference cell filled with pure LC E7, which ensures that the largely increased viscoelasticity of the composite does not lead to a degradation of electro-optical propertie

    Morphosyntactic processing in late second-language learners

    The goal of the present study was to investigate the electro- physiological correlates of second-language (L2) morphosyn- tactic processing in highly proficient late learners of an L2 with long exposure to the L2 environment. ERPs were col- lected from 22 English–Spanish late learners while they read sentences in which morphosyntactic features of the L2 present or not present in the first language (number and gender agree- ment, respectively) were manipulated at two different sentence positions—within and across phrases. The results for a control group of age-matched native-speaker Spanish participants in- cluded an ERP pattern of LAN-type early negativity followed by P600 effect in response to both agreement violations and for both sentence positions. The late L2 learner results included a similar pattern, consisting of early negativity followed by P600, in the first sentence position (within-phrase agreement viola- tions) but only P600 effects in the second sentence position (across-phrase agreement violation), as well as significant am- plitude and onset latency differences between the gender and the number violation effects in both sentence positions. These results reveal that highly proficient learners can show electro- physiological correlates during L2 processing that are qualita- tively similar to those of native speakers, but the results also indicate the contribution of factors such as age of acquisition and transfer processes from first language to L

    Serum Iron Parameters, HFE C282Y Genotype, and Cognitive Performance in Older Adults: Results From the FACIT Study

    Although iron homeostasis is essential for brain functioning, the effects of iron levels on cognitive performance in older individuals have scarcely been investigated. In the present study, serum iron parameters and hemochromatosis (HFE) C282Y genotype were determined in 818 older individuals who participated in a 3-year randomized, placebo-controlled double-blind trial examining the effects of folic acid on carotid intima-media thickness. All participants had slightly elevated homocysteine levels and were vitamin B12 replete. Cognitive functioning was assessed at baseline and after 3 years by means of a neuropsychological test battery. At baseline, increased serum ferritin was associated with decreased sensorimotor speed, complex speed, and information-processing speed and increased serum iron was associated with decreased sensorimotor speed. Cognitive performance over 3 years was not associated with HFE C282Y genotype or iron parameters. In conclusion, serum iron parameters do not show a straightforward relationship with cognitive functioning, although elevated iron levels may decrease cognitive speed in older individuals susceptible to cognitive impairmen

    Human cytomegalovirus (HCMV) is the most frequent viral cause of congenital defects and can trigger devastating disease in immune-suppressed patients. Cytotoxic lymphocytes (CD8+ T cells and NK cells) control HCMV infection by releasing interferon-γ and five granzymes (GrA, GrB, GrH, GrK, GrM), which are believed to kill infected host cells through cleavage of intracellular death substrates. However, it has recently been demonstrated that the in vivo killing capacity of cytotoxic T cells is limited and multiple T cell hits are required to kill a single virus-infected cell. This raises the question whether cytotoxic lymphocytes can use granzymes to control HCMV infection in a noncytotoxic manner. Here, we demonstrate that (primary) cytotoxic lymphocytes can block HCMV dissemination independent of host cell death, and interferon-α/β/γ. Prior to killing, cytotoxic lymphocytes induce the degradation of viral immediate-early (IE) proteins IE1 and IE2 in HCMV-infected cells. Intriguingly, both IE1 and/or IE2 are directly proteolyzed by all human granzymes, with GrB and GrM being most efficient. GrB and GrM cleave IE1 after Asp398 and Leu414, respectively, likely resulting in IE1 aberrant cellular localization, IE1 instability, and functional impairment of IE1 to interfere with the JAK-STAT signaling pathway. Furthermore, GrB and GrM cleave IE2 after Asp184 and Leu173, respectively, resulting in IE2 aberrant cellular localization and functional abolishment of IE2 to transactivate the HCMV UL112 early promoter. Taken together, our data indicate that cytotoxic lymphocytes can also employ noncytotoxic ways to control HCMV infection, which may be explained by granzyme-mediated targeting of indispensable viral proteins during lytic infection

    Melanocortin Receptor 4 Deficiency Affects Body Weight Regulation, Grooming Behavior, and Substrate Preference in the Rat

    Obesity is caused by an imbalance between energy intake and expenditure and has become a major health-care problem in western society. The central melanocortin system plays a crucial role in the regulation of feeding and energy expenditure, and functional loss of melanocortin receptor 4 (MC4R) is the most common genetic cause of human obesity. In this study, we present the first functional Mc4r knockout model in the rat, resulting from an N-ethyl-N-nitrosourea mutagenesis–induced point mutation. In vitro observations revealed impaired membrane-binding and subsequent nonfunctionality of the receptor, whereas in vivo observations showed that functional loss of MC4R increased body weight, food intake, white adipose mass, and changed substrate preference. In addition, intracerebroventricular (ICV) administration of Agouti-Related Protein79–129 (AgRP79–129), an MC4R inverse agonist, or Melanotan-II (MTII), an MC4R agonist, did affect feeding behavior in wild-type rats but not in homozygous mutant rats, confirming complete loss of MC4R function in vivo. Finally, ICV administration of MTII induced excessive grooming behavior in wild-type rats, whereas this effect was absent in homozygous mutant rats, indicating that MTII-induced grooming behavior is exclusively regulated via MC4R pathways. Taken together, we expect that the MC4R rat model described here will be a valuable tool for studying monogenic obesity in humans. More specifically, the relative big size and increased cognitive capacity of rats as compared to mice will facilitate complex behavioral studies and detailed mechanistic studies regarding central function of MC4R, both of which ultimately may help to further understand the specific mechanisms that induce obesity during loss of MC4R function

    Impaired perception of facial motion in autism spectrum disorder

We used animated average faces to examine the ability of adults with autism spectrum disorders (ASD) to perceive facial motion. Participants completed increasingly difficult tasks involving the discrimination of (1) sequences of facial motion, (2) the identity of individuals based on their facial motion and (3) the gender of individuals. Stimuli were presented in both upright and upside-down orientations to test for the difference in inversion effects often found when comparing ASD with controls in face perception. The ASD group’s performance was impaired relative to the control group in all three tasks and unlike the control group, the individuals with ASD failed to show an inversion effect. These results point to a deficit in facial biological motion processing in people with autism, which we suggest is linked to deficits in lower level motion processing we have previously reported
