    Práce se zabývá areálem pozdně barokní zahrady vzniklé kolem poloviny 18. století při zámku ve středočeském Smilkově, jenž tehdy patřil hraběti Janu Jindřichovi z Bissingenu. Hlavním cílem práce je komplexní prostorová analýza zahrady a pokus o rekonstrukci tzv. teatronu, který představoval zcela unikátní realizaci svého druhu u nás. Pozornost je věnována i širšímu evropskému kontextu pozdně barokních a rokokových zahrad a zaměřuje se rovněž na otázku možného autorství celku. Součástí práce je vizualizace navrhované rekonstrukce a podrobná fotografická dokumentace.The work deals with the area of the late baroque garden which was built approximately in the middle of the 18th century at the chateau in the central Bohemian town of Smilkov, which then belonged to Count Jan Jindřich of Bissingen. The thesis focuses on a complex spatial analysis of the garden and also it attempts to reconstruct the so-called theatron as it represented a totally unique realization of its kind in our country. Attention is also paid to the wider European context of late baroque and rococo gardens as well as to the question of possible authorship of the whole complex.     Nicotinic acetylcholine receptors modulate osteoclastogenesis

    Background: Our aim was to investigate the role of nicotinic acetylcholine receptors (nAChRs) in in-vitro osteoclastogenesis and in in-vivo bone homeostasis. Methods: The presence of nAChR subunits as well as the in-vitro effects of nAChR agonists were investigated by ex vivo osteoclastogenesis assays, real-time polymerase chain reaction, Western blot and flow cytometry in murine bone marrow-derived macrophages differentiated in the presence of recombinant receptor activator of nuclear factor kappa B ligand (RANKL) and macrophage colony-stimulating factor (M-CSF). The bone phenotype of mice lacking various nAChR subunits was investigated by peripheral quantitative computed tomography and histomorphometric analysis. Oscillations in the intracellular calcium concentration were detected by measuring the Fura-2 fluorescence intensity. Results: We could demonstrate the presence of several nAChR subunits in bone marrow-derived macrophages stimulated with RANKL and M-CSF, and showed that they are capable of producing acetylcholine. nAChR ligands reduced the number of osteoclasts as well as the number of tartrate-resistant acidic phosphatase-positive mononuclear cells in a dose-dependent manner. In vitro RANKL-mediated osteoclastogenesis was reduced in mice lacking α7 homomeric nAChR or β2-containing heteromeric nAChRs, while bone histomorphometry revealed increased bone volume as well as impaired osteoclastogenesis in male mice lacking the α7 nAChR. nAChR ligands inhibited RANKL-induced calcium oscillation, a well-established phenomenon of osteoclastogenesis. This inhibitory effect on Ca2+ oscillation subsequently led to the inhibition of RANKL-induced NFATc1 and c-fos expression after long-term treatment with nicotine. Conclusions: We have shown that the activity of nAChRs conveys a marked effect on osteoclastogenesis in mice. Agonists of these receptors inhibited calcium oscillations in osteoclasts and blocked the RANKL-induced activation of c-fos and NFATc1. RANKL-mediated in-vitro osteoclastogenesis was reduced in α7 knockout mice, which was paralleled by increased tibial bone volume in male mice in vivo. © 2016 Mandl et al

    Předkládaná práce se zabývá gnozeologickými a komunikačními funkcemi konceptuální tvorby a možnostmi jejich využití ve vzdělávání, přičemž se zaměřuje na téma času. V první části se zabývám srovnáním a interpretací vybraných konceptuálních děl z hlediska jejich práce s fenoménem času, přičemž mi jde především o postižení možného poznání (vyplývajícího z kontaktu s těmito díly) týkajícího se časové dimenze lidské existence. Ve druhé části se snažím uvést některé poznatky z předchozí analýzy do námětů využitelných ve vzdělávací praxi. The first part of the thesis inquires into comparison and interpretation of selected conceptual art works in light of its operation with time phenomenon. The prime intention is to reflect and describe the possibilities of understanding time dimension of human existence through the contact with conceptual art works. The essay is anchored in their rich experi- ence with different kinds of ego-documents from that time, especially correspondence, daybooks, memoirs, small glosses and imaginative literature. The model of a student as a socially unanchored and financially dependent person, which can be found in the history from medieval times, these authors confront with the new situation of so-called modern times. Using concrete examples, the authors illustrate the fact, that from the 19th century students' financiall indemnity started to increase and gradually shaped some kind of system. The basic methods of acquiring money for school were still conditions. In connection with emergence of new newspapers and publishers the possibilities of external work for money broadened. Fundations were also very important; some of them had a character of assem- blage. Universities started to provide a pension system as well (school reference and social pension). After the creation of independent state possibilities of travelling pension also broadened.
