    Diplomová práce se zabývá tvorbou návrhu komplexní plynotěsné bariéry v rámci uzavíracích hrází zabraňující prostupu důlních vzdušin do prostoru jámy Je-1 na dole Jeremenko v Ostravě - Vítkovicích. V práci jsem se soustředil nejen na vytvoření plynonepropustné bariéry v tělese uzavíracích hrází, ale také v okolním horninovém prostředí, přičemž hloubka rozrušení následkem redistribuce byla zjištěna výpočtem matematického modelu v softwaru Plaxis 2D.The diploma thesis deals with the design of complex gas-tight barrier within horizontal permanent stoppings preventing the penetration of mine air into the Je-1 pit at the Jeremenko mine in Ostrava – Vítkovice.     Import 02/11/2016Bakalářská práce podává výčet několika známých zachovalých historických geotechnických monumentálních zděných a sypaných staveb, které dokumentují vysokou technickou úroveň jejich stavitelů a mohou být zdrojem poučení a vzorem současným stavitelům geotechnických staveb. Jednotlivé kapitoly práce věnují pozornost a rozebírají geotechnické a konstrukční okolnosti včetně rozboru technologie výstavby těchto staveb. Následně se zabývá odkazem těchto zachovalých staveb pro stavby nové, ať už se jedná o poučení z negativních zkušeností, nebo pozitivních zkušeností, což je hlavním cílem tohoto tématu. V poslední části práce uvádí několik příkladů srovnání analogických historických staveb a novodobých staveb, popřípadě srovnání téže technologie užité na historické a novodobé stavbě.The bachelor thesis provides a list of several known preserved geotechnical historical monumental masonry and earth structures, which document high technical level of their builders and can be a source of knowledge and model for builders of current geotechnical structures. Individual chapters pay attention and analyze geotechnical and structural factors, including an analysis of the technology of construction of these buildings. Subsequently we deal with the legacy of these preserved buildings for new construction, whether it is the lessons from negative experiences or positive experiences, which is the main objective of this theme.     Tato práce je konstrukčním řešením inteligentní rychlonabíječky olověných akumulátorů. Jsou tu uvedeny výpočty jednotlivých prvků. Základ nabíječky tvoří spínaný zdroj, proto je nabíječka snadno přenosná, lehká a její rozměry jsou malé. Nabíjí akumulátor pomocí metody konstantního napětí, s proudovým omezením 0,5A, 5A, 10A, 50A. Proud 50A využijeme v zimě při startování automobilu, kdy baterie nedodá dostatečný proud. Pro zpětnou kontrolu jsou v přední straně umístěny 3 kontrolní LED diody popisující různé stavy nabíječky. Zařízení také obsahuje bezpečnostní ochrany pro samotnou nabíječku, napájecí síť i baterii.This thesis is an engineering design of an intelligent fast-charger for lead-acid accumulators. It contains calculations of each component. The switching power supply is the mainstay of the charger that makes the charger portable, lightweight and small. The Intelligent fast-charger for lead-acid accumulators charges the accumulator by method of constant voltage with current limitation of 0,5A, 5A, 10A, 50A. 50A current is used in winter during starting of a car, when the battery can´t provide sufficient current. There are three LEDs placed on the front side of the charger reflecting status of the device. The device comprises safety protection for the charger, supply network and battery.

    Leaf Eh and pH: A Novel Indicator of Plant Stress. Spatial, Temporal and Genotypic Variability in Rice (Oryza sativa L.)

    A wealth of knowledge has been published in the last decade on redox regulations in plants. However, these works remained largely at cellular and organelle levels. Simple indicators of oxidative stress at the plant level are still missing. We developed a method for direct measurement of leaf Eh and pH, which revealed spatial, temporal, and genotypic variations in rice. Eh (redox potential) and Eh@pH7 (redox potential corrected to pH 7) of the last fully expanded leaf decreased after sunrise. Leaf Eh was high in the youngest leaf and in the oldest leaves, and minimum for the last fully expanded leaf. Leaf pH decreased from youngest to oldest leaves. The same gradients in Eh-pH were measured for various varieties, hydric conditions, and cropping seasons. Rice varieties differed in Eh, pH, and/or Eh@pH7. Leaf Eh increases and leaf pH decreases with plant age. These patterns and dynamics in leaf Eh-pH are in accordance with the pattern and dynamics of disease infections. Leaf Eh-pH can bring new insight on redox processes at plant level and is proposed as a novel indicator of plant stress/health. It could be used by agronomists, breeders, and pathologists to accelerate the development of crop cultivation methods leading to agroecological crop protection

    Abstract Titanium dioxide nanoparticles (TiO2NPs) are revolutionizing biomedicine due to their potential application as diagnostic and therapeutic agents. However, the TiO2NP immune-compatibility remains an open issue, even for ethical reasons. In this work, we investigated the immunomodulatory effects of TiO2NPs in an emergent proxy to human non-mammalian model for in vitro basic and translational immunology: the sea urchin Paracentrotus lividus. To highlight on the new insights into the evolutionarily conserved intracellular signaling and metabolism pathways involved in immune-TiO2NP recognition/interaction we applied a wide-ranging approach, including electron microscopy, biochemistry, transcriptomics and metabolomics. Findings highlight that TiO2NPs interact with immune cells suppressing the expression of genes encoding for proteins involved in immune response and apoptosis (e.g. NF-κB, FGFR2, JUN, MAPK14, FAS, VEGFR, Casp8), and boosting the immune cell antioxidant metabolic activity (e.g. pentose phosphate, cysteine-methionine, glycine-serine metabolism pathways). TiO2NP uptake was circumscribed to phagosomes/phagolysosomes, depicting harmless vesicular internalization. Our findings underlined that under TiO2NP-exposure sea urchin innate immune system is able to control inflammatory signaling, excite antioxidant metabolic activity and acquire immunological tolerance, providing a new level of understanding of the TiO2NP immune-compatibility that could be useful for the development in Nano medicines

    Plant-associated microbiota as a source of antagonistic bacteria against the phytopathogen Erwinia amylovora

    Control of bacterial plant diseases is a major concern, as they affect economically important species and spread easily, such as the case of fire blight of rosaceous caused by Erwinia amylovora. In the search for alternatives to the use of agrochemicals and antibiotics, this work presents a screening of natural bacterial antagonists of this relevant and devastating phytopathogen. We recovered bacterial isolates from different plant tissues and geographical origins and then selected those with the strongest ability to reduce fire blight symptoms ex vivo and remarkable in vitro antagonistic activity against E. amylovora. None of them elicited a hypersensitivity reaction in tobacco leaves, most produced several hydrolytic enzymes and presented other biocontrol and/or plant growth-promoting activities, such as siderophore production and phosphate solubilization. These isolates, considered as biocontrol candidates, were identified by 16S rRNA sequencing as Pseudomonas rhizosphaerae, Curtobacterium flaccumfaciens, Enterobacter cancerogenus, Pseudomonas azotoformans, Rosenbergiella epipactidis and Serratia plymuthica. This is the first time that the last five bacterial species are reported to have biocontrol potential against E. amylovora

    LocZ is a new cell division protein involved in proper septum placement in Streptococcus pneumoniae

    How bacteria control proper septum placement at midcell, to guarantee the generation of identical daughter cells, is still largely unknown. Although different systems involved in the selection of the division site have been described in selected species, these do not appear to be widely conserved. Here, we report that LocZ (Spr0334), a newly identified cell division protein, is involved in proper septum placement in Streptococcus pneumoniae. We show that locZ is not essential but that its deletion results in cell division defects and shape deformation, causing cells to divide asymmetrically and generate unequally sized, occasionally anucleated, daughter cells. LocZ has a unique localization profile. It arrives early at midcell, before FtsZ and FtsA, and leaves the septum early, apparently moving along with the equatorial rings that mark the future division sites. Consistently, cells lacking LocZ also show misplacement of the Z-ring, suggesting that it could act as a positive regulator to determine septum placement. LocZ was identified as a substrate of the Ser/Thr protein kinase StkP, which regulates cell division in S. pneumoniae. Interestingly, homologues of LocZ are found only in streptococci, lactococci, and enterococci, indicating that this close phylogenetically related group of bacteria evolved a specific solution to spatially regulate cell division. IMPORTANCE Bacterial cell division is a highly ordered process regulated in time and space. Recently, we reported that the Ser/ Thr protein kinase StkP regulates cell division in Streptococcus pneumoniae, through phosphorylation of several key proteins. Here, we characterized one of the StkP substrates, Spr0334, which we named LocZ. We show that LocZ is a new cell division protein important for proper septum placement and likely functions as a marker of the cell division site. Consistently, LocZ supports proper Z-ring positioning at midcell. LocZ is conserved only among streptococci, lactococci, and enterococci, which lack homologues of the Min and nucleoid occlusion effectors, indicating that these bacteria adapted a unique mechanism to find their middle, reflecting their specific shape and symmetry

    Variability of aggressiveness and virulence of Erwinia carotovora subsp. carotovorum causing the soft rot on potato tubers in the western of Algeria

    Soft rot symptoms were observed on potato plants in several potato cultivars in the western part of Algeria. A total of four strains of Erwinia are devided as follow: i) three strains of bacteria isolated from diseased tissues and soil, identified as Erwinia carotovora subsp carotovorum using conventional bacteriological and biochemical methods; and ii) one strain as Erwinia sp, not pathogens. In vitro tests, on tuber slices were set up to determine slices weight lost, which allows to find differences in cultivar susceptibility and isolate aggressiveness. Among the five cultivars, Laura was the most susceptible than the others tested cultivars. Moreover, it was found that MAI isolate was the most virulent than the other bacterial isolates. The results of this study should allow an optimization of the potato storage, after considering the susceptibility of a given cultivar to soft rot development and the aggressiveness
