Extracellular vesicles in cancer: therapeutic implications
Trabalho Final de Mestrado Integrado, Ciências Farmacêuticas, 2021, Universidade de Lisboa, Faculdade de Farmácia.O termo vesículas extracelulares engloba diversos tipos de biopartículas muito diversas, compostas por uma bicamada lipídica, sem núcleo funcional e que contêm informação na forma de proteínas, lípidos e ácidos nucleicos, como DNA, RNA ou microRNAs.
As vesículas extracelulares são uma importante forma de comunicação entre as células. Podem mediar a comunicação intercelular em condições fisiológicas e patológicas, tendo uma ampla gama de funções. É já reconhecido que as vesículas extracelulares podem promover a coagulação e a regeneração óssea, permitir a maturação de reticulócitos, modular a resposta imune, e também, mediar a neurodegeneração, doenças cardiovasculares e a permissividade do hospedeiro durante infeções.
As vesículas extracelulares parecem apresentar um papel relevante na progressão do cancro, através da sinalização oncogénica, da modulação do microambiente tumoral e da imunomodulação, que podem influenciar o crescimento tumoral, a capacidade de invasão, a angiogénese e a metastização. Descobrir mais sobre a sua origem, conteúdo e funções no cancro poderá ser útil no futuro para o desenvolvimento de novas estratégias de inibição do desenvolvimento oncológico e de progressão da doença. É necessária mais investigação nesta área, uma vez que várias das funções destas vesiculas ainda estão totalmente estudadas.
Com os avanços recentes na investigação da área das vesículas extracelulares e do seu papel no cancro, parece evidente que elas terão um papel cada vez mais importante no diagnóstico e prognóstico do cancro, através de novos métodos, como a biópsia líquida. As vesículas extracelulares poderão em breve ser consideradas como biomarcadores relevantes para o diagnóstico e prognóstico do cancro, assim como para a monitorização da resposta terapêutica. São alvos de pesquisa promissores como agentes de distribuição de fármacos, sendo ainda hipoteticamente possível que as vesículas extracelulares também possam ter um papel ativo contra o cancro. As terapêuticas baseadas em vesiculas extracelulares apenas começaram agora a ser desenvolvidas e investigadas, ainda existe um longo caminho por percorrer. Veremos no futuro até onde nos podem levar na luta contra o cancro.Extracellular vesicles encompass several types of very diverse bioparticles composed by a lipid bilayer, without a functional nucleus, which contain information in the form of proteins, lipids, and nucleic acids, such as DNA, RNA, or microRNA.
Extracellular vesicles are an important way of communication between cells. They can mediate intercellular communication, with a very wide range of functions in both physiological and pathological conditions. It is already recognized that extracellular vesicles can promote coagulation and bone regeneration, allow reticulocyte maturation, modulate immune response, and mediate neurodegeneration, cardiovascular diseases, and host permissiveness during infections.
These vesicles seem to present a relevant role in cancer progression, through oncogenic signalling, tumour microenvironment modulation and immunomodulation, which might influence tumour growth, invasiveness, angiogenesis, and metastasis. A better understanding of their origin, cargo, and functions in cancer, may be useful to develop new strategies for inhibiting oncologic development and the disease progression. Further research is needed in this area since several of extracellular vesicles’ roles are not yet fully studied.
Considering the recent advances in the field of research of extracellular vesicles in cancer, it seems evident that they will play an increasingly important role in cancer diagnosis and prognosis through non-invasive diagnostic methods, such as liquid biopsy. Extracellular vesicles could be soon considered relevant biomarkers for cancer diagnosis and prognosis, as well as to therapeutic monitoring.
Extracellular vesicles are also promising research targets as drug delivery agents. It is even hypothesized that extracellular vesicles could work actively against cancer. Extracellular vesicles-based therapies have just started being developed and researched, there is still a long way to go. We will see in the future how far they can take us in the fight against cancer

Web 2.0 e padrões na aprendizagem da matemática
Mestrado em Multimédia em EducaçãoO ensino continua a obedecer a lugares definidos no tempo e no espaço, ou
seja, o professor continua a ser o principal responsável pela transmissão de
conteúdos e o papel do aluno continua a ser bastante passivo. As Tecnologias
de Informação e Comunicação (TIC) podem portanto influenciar e transformar a
aprendizagem.
As plataformas de gestão da aprendizagem e applets permitem definir uma nova
metodologia de ensino e aprendizagem, organizando o espaço de interacção, de
acordo com uma dada intencionalidade, incentivando a auto-aprendizagem dos
alunos recorrendo a uma rede de colaboração. No entanto, apesar da divulgação
de plataformas e da formação de professores na sua utilização, o seu uso ainda
não é generalizado e não está a ser efectuado da forma mais adequada.
Por outro lado, há necessidade de compreender os conceitos algébricos, as
estruturas e os princípios que regem as manipulações simbólicas e como estes
símbolos podem ser utilizados para traduzir ideias matemáticas e a exploração
de padrões e regularidades ajuda os alunos a desenvolver o pensamento
algébrico. Apesar da maioria dos professores e investigadores, na área da
Matemática, reconhecerem a importância dos padrões no desenvolvimento e na
compreensão das estruturas algébricas, a sua abordagem, em contexto de sala
de aula, é ainda pouco explorada, sendo importante uma investigação nesta área
e numa perspectiva inovadora.
Neste contexto, desenvolveu-se este estudo com o primordial objectivo de avaliar
o impacto da utilização de recursos da Web 2.0 (applets e plataforma de gestão
da aprendizagem) na abordagem do domínio temático da Álgebra, mais
concretamente, no estudo das funções, centrada nos padrões e regularidades ao
nível do desenvolvimento de: competências tecnológicas; apetências,
conhecimentos e capacidades matemáticas, transversais e específicas e
apetências e capacidades de trabalho colaborativo.
Para tentar dar resposta às questões de investigação subjacentes aos objectivos
que se perseguem, optou-se por um estudo de caso desenvolvido num contexto
de investigação-acção uma vez que se pretendia analisar, de modo intensivo,
uma situação particular, em contexto real, e mais focada nos processos do que
nos resultados finais, não seguindo uma linha rígida de orientação. A parte
empirica do estudo decorreu em ambiente académico normal, em contexto de
sala de aula e a distância, numa escola dos 2.º e 3º Ciclos do Ensino Básico com
Ensino Secundário do distrito de Leiria e incidiu sobre alunos de uma turma do
8.º ano de escolaridade.
Ad principais técnicas de recolha de dados foram a observação, a inquirição e a
análise documental.
A análise dos dados recolhidos foi, essencialmente, de natureza qualitativa, não
pondo de parte, no entanto, elementos quantificáveis envolvidos, com o objectivo
de suportar a análise qualitativa.
A análise de conteúdo a que os dados foram submetidos permitiu concluir que a
integração de recursos da Web 2.0 (applets e plataforma de gestão de
aprendizagem) nas actividades lectivas, em diferentes contextos espaçotemporais
- nas aulas de Matemática, em sessões de Estudo Acompanhado (EA)
e a distância - contribuiu para motivar os alunos para uma aprendizagem mais
profunda, significativa e eficaz e desenvolver e/ou construir conhecimentos
tecnológicos e algébricos, uma visão mais positiva e dinâmica da matemática,
capacidades de resolução de problemas, de raciocinio e de comunicação
matemática de forma mais autónoma e responsável. A utilização de uma
componente a distância, como complemento das aulas, estimulou e favoreceu o
processo de ensino e aprendizagem e promoveu a partilha de informação, a
construção de conhecimento partilhado e colaborativo e interacções mais ricas
entre os diferentes intervenientes (saber, alunos e professora/investigadora).The education continue to obey to defined positions in time and space, that
means, the teacher still to be the main responsable for the contents transmition
and the student role continues to be quite passive. The Information and
Communication Technologies (ICT) can therefore influence and transform the
learning.
The learn mannagement systems and the applets allow to define a new
methodology of teaching and learning, by organizing the interaction space in
accordance with a given intencionality, encouraging self-learning of the
students recurring to a colaboration network. However, despite the diffusion of
the platforms and the training of the teachers in their use, their use is not yet
generalised and is not being made in the more apropriate manner.
By other way, there is the need to understand the algebric concepts, the
structures and principals that rule the symbolic manipulations and how these
symbols can be used to translate mathematical ideas and the exploration of the
patterns and regularities helps the students to develop algebric thinking.Despite
the majority of teachers and investigators in the Mathematical field, does
recognize the importance of patterns in the development and comprehension
of the algebric structures, their approach, in a classroom context, is yet less
explored, being important a research in this area and in a innovative
perspective.
In this context, this study has been developed with the primordal purpose of
evaluating the impact of the use of resources Web 2.00 (applets and learn
mannagement systems) in the approach of themathic domain of the Algebra ,
more concretely, in the study of functions, centred in patterns and regularities to
the development level of: technologic competences, appetites, knowledgments
and transversal and specific mathematical capabilities appetites and
capabilities of colaborative work.
To Try to give an answer to the questions of the underlying investigation to the
purposes wich are pursue, was decided for a case study developed in a context
of action-investigation once that it was intended to do, an analise in an intensive
way, a particular situation, in a real context, and more focused in the process
than in the outcomes, not been followed a strict line of orientation.
The empirical part of the study took place in normal academic environment, in
classroom context and the distance, in a school of the second and third Cycles
of Basic Education with elementary education in the district of Leiria and
focused on students from the 8 degree of education.
The main techniques of data’s collection were the watching, the inquiry and the
documental analysis.
The analise of the collected data, was essentialy on qualitative nature, not
disregarding, however,quantifiable elements envolved , with the aim to support
the qualitative analise. The content analyses that those data’s were submitted
allowed to conclude that the interaction of resources Web 2.00 (applets and
learn mannagement systems) in the academic activities,
in different space-temporals context in the mathematics classes in Oriented
study sessions (OS) and distance, contributed to motivate the students for a
more deep learning, significant and effective, and develop and/or to build
Technological and algebric knowledegments;a more positive view and dynamic
of the mathematic; to develop the capabilities of problems resolution,
mathematical reasoning and mathematical communication; a more positive
view and dynamic of the mathematic;capabilities of problems resolution, of
reasoning and of mathematical communication, in a more autonomic and
responsible way. The usage of one component the distance, complementary to
the lessons, favoured and encouraged the process of education and learning
and fostered the sharing of information, the construction of sharing
knowledgement and collaborative and more rich interactions between the
different intervenient (knowledge, students and teacher/ researcher)
Aplicação de um modelo internacional de auditoria ambiental um em hotel de Curitiba

Assim, é crescente a necessidade de intervir e tornar o setor de turismo cada vez mais viável ambientalmente, socialmente e economicamente. Este trabalho teve como objetivo a adaptação seguida de aplicação de uma lista de verificação ambiental, originado na Suíça, em um Hotel de Luxo em Curitiba. Após o diagnóstico, foi apresentada uma proposta de plano de ação para o Hotel colocar em prática para, desse modo, tornar-se um empreendimento mais sustentável. Com o índice gerado em cada área, foi possível analisar o perfil do empreendimento, que obteve uma média geral de 79%, se encaixando na avaliação como um perfil bom. A partir deste resultado, o plano de ação foi elaborado para que o Hotel passe para perfil ótimo, e possa manter este patamar
Acolhimento institucional : estratégias desenvolvidas pelos cuidadores formais
As transformações da estrutura demográfica, social, económica e familiar
trouxeram, entre as suas diversas consequências, uma nova visibilidade social das
pessoas idosas, numa altura em que os conceitos de velhice e de envelhecimento sofrem
mudanças decorrentes dos desafios que a sociedade atual enfrenta.
O aumento progressivo da institucionalização de idosos surge como uma das
várias respostas que o Estado criou para proporcionar a satisfação das necessidades e o
bem-estar aos idosos.
A entrada para um contexto residencial implica um confronto com uma realidade
até então desconhecida, que inclui, nomeadamente, a partilha de espaços, os horários, o
regulamento interno, a ausência de privacidade, convívio com uma heterogeneidade de
pessoas, entre outros. A adaptação a esta nova realidade implica estratégias de
adaptação individualizadas e personalizadas por parte dos cuidadores formais e uma
capacidade de resiliência por parte do idoso que deverá recorrer a estratégias internas
e/ou externas de forma a conseguir a melhor ambientação possível.
É fundamental o respeito pela individualidade, personalidade, gostos, hábitos e
história de vida do utente, para evitar situações de isolamento, depressão e insatisfação
com a vida. As situações de dependência, a falta de conhecimentos e a incapacidade,
motivadas por fatores familiares e económicos, são razões que levam o idoso e/ou os
cuidadores informais a optarem por esta resposta social. O idoso, a família e o lar
devem trabalhar em conjunto para permitir que a ambientação decorra de forma natural,
respeitando a vontade, sentimentos e valores do utente.The changes in the demographic, social, economic and family structure, brought
to its various consequences, a new social visibility of the elderly, at a time when the
concept of old age and aging undergoes changes resulting from the challenges that
modern society faces.
The progressive increase of elderly in care emerged as one of several decisions
that the Government created to provide the satisfaction of needs and provide welfare for
the elderly.
The entrance to a residential care context implies a confrontation with a reality
previously unknown particular the sharing of spaces, times, the rules, the lack of
privacy and living with different people. The new reality implies individualized
adaptation strategies and customized by the formal caregivers and involves resilience
capacity of the elderly who should use internal and / or external strategies to get the best
adaption.
It is essential respect for individuality, personality, tastes, habits, and life history
of the wearer to avoid situations of isolation, depression and dissatisfaction with life.
The situations of dependency, lack of knowledge and unavailability motivated by
family and economic factors are the reasons why the elderly and / or informal caregivers
to choose this social response. The elderly, the family and the Residential care must
work together to allow the setting takes place naturally respecting the will, feelings and
values of the weare
Caracterização da interação APP/HB-EGF
Mestrado em Biomedicina MolecularAlzheimer’s disease (AD) is the most prevalent neurodegenerative disorder worldwide and the leading cause of dementia in the elderly. Abnormal processing of Alzheimer’s amyloid precursor protein (APP) and increased generation of its amyloid beta (Aβ) fragment are central events in the AD pathogenesis, propelling major studies on APP biology. APP is thought to be involved in important processes such as cell adhesion, survival, migration and differentiation. Therefore, because of the imperative need to study APP biological functions, the search for APP-binding partners has stand out. Recently in our laboratory, a plethora of putative APP-binding proteins was unraveled through the use of the Yeast Two Hybrid (YTH) system. Among them, the heparin-binding epidermal growth factor-like growth factor (HB-EGF) has emerged as an interesting target of study.
HB-EGF is a heparin-binding member of the EGF family of growth factors that stimulates growth and differentiation. It has been purposed has an important trophic factor in the developing and adult central nervous system (CNS), being expressed at much higher levels than EGF in the CNS, which indicates that HB-EGF may serve as a major ligand for EGFR in neurons. HB-EGF is synthesized as a pre-pro-form of 208 amino acids in length and is expressed at the cell surface as a 20-30kDa type I transmembrane precursor, named proHB-EGF. This larger membrane-anchored precursor is then proteolytically processed, generating the mature soluble HB-EGF (sHB-EGF) that is released to the extracellular medium. Very interestingly, HB-EGF presents autocrine, paracrine and juxtacrine biological activities, with proHB-EGF evidencing unique biological characteristics distinct from sHB-EGF.
In the work here described, we first validated the APP/HB-EGF interaction by yeast co-transformation, and unraveled that the interaction is not mediated by the APP intracellular domain (AICD). We further confirmed it as an in vivo interaction by GFP-Trap pull-down assays and accessed the physiological relevance of this novel interaction through co-localization and signaling studies in HeLa cells transfected with the APP-GFP and HB-EGF cDNAs. Although the functional role of the APP/HB-EGF complex was not determined, results suggest that these proteins physically and functionally interact, having potential value in regulating each other signal pathways, with a role for APP in inducing the activation of the MAPK signaling being evidenced. In addition, a putative novel interaction with a proEGF species was also detected and APP was shown to act synergistically with EGF to activate the MAPK signaling. These results deepen our understanding of the APP biology, a crucial protein in cerebral physiology and AD pathophysiology.A Doença de Alzheimer (DA) é a doença neurodegenerativa mais prevalente a nível mundial, e a principal causa de demência na população sénior. O processamento anormal da Proteína Precursora de Amilóide de Alzheimer (PPA) e a produção aumentada do seu fragmento beta amilóide (Aβ) constituem eventos centrais na patogénese da DA, o que tem fomentado a investigação da PPA. Esta tem sido descrita como uma proteína envolvida em processos celulares determinantes, como adesão, migração, diferenciação, e sobrevivência celular. Como tal, devido à necessidade imperativa de caracterizar as suas funções biológicas, a investigação de proteínas que interagem com a PPA é de vital importância. Recentemente, diversas proteínas foram identificadas no nosso laboratório como putativos interatores da PPA, através da técnica de Yeast Two Hybrid (YTH). De entre estes, o fator de crescimento de ligação à heparina semelhante ao fator de crescimento epidermal (HB-EGF) revelou-se um interessante alvo de estudo.
O HB-EGF é um membro da família do fator de crescimento epidermal (EGF) com capacidade de ligação à heparina, que se destaca pelas suas capacidades de estimular o crescimento e diferenciação celulares, tendo sido proposto como um relevante fator trófico para o desenvolvimento e manutenção do sistema nervoso central (SNC). Na verdade, o HB-EGF é mais abundante no SNC do que o próprio EGF, o que sugere que o HB-EGF é o principal ligando neuronal para o recetor do EGF (EGFR). O HB-EGF é sintetizado como um precursor de 208 aminoácidos (pre-proHB-EGF), sendo exposto na membrana celular como um precursor transmembranar de 20-30 kDa, o proHB-EGF. Esta forma é subsequentemente proteoliticamente processada, gerando um péptido solúvel que é libertado para o meio extracelular (sHB-EGF). Curiosamente, o HB-EGF apresenta atividades biológicas parácrinas, autócrinas e justácrinas.
O presente trabalho teve como principal objetivo a validação da interação entre a PPA e o HB-EGF, primeiramente alcançado através da técnica de YTH, que revelou também que a interação entre as duas proteínas não é mediada pelo domínio intracelular da PPA. Esta interação foi subsequentemente confirmada por ensaios de GFP-Trap pull-down em cultura de células de mamífero, e a sua relevância fisiológica estudada através de estudos de co-localização e sinalização celulares, usando células HeLa transfectadas com os cDNAs da PPA e do HB-EGF. Apesar do papel funcional do complexo PPA/HB-EGF não ter sido determinado, os resultados obtidos sugerem que as duas proteínas interagem física e funcionalmente, influenciando a sinalização mediada por cada uma delas separadamente, como a ativação da via de sinalização MAPK que verificámos ser também induzida pela PPA. Adicionalmente, descrevemos uma nova interação proteica entre a PPA e uma forma precursora do EGF, e demonstrámos que a PPA atua de forma sinérgica com o EGF. Estes resultados levam a uma maior compreensão da biologia da PPA, uma proteína importante na fisiologia cerebral e na fisiopatologia da DA
Contribution of Animal Models to Contemporary Understanding of Attention Deficit Hyperactivity Disorder
Attention Deficit Hyperactivity Disorder (ADHD) is a poorly understood neurodevelopmental disorder of multifactorial origin. Animal-based research has been used to investigate ADHD etiology, pathogenesis and treatment, but the efficacy of this research for patients has not yet been systematically evaluated. Such evaluation is important given the resource consumption and ethical concerns incurred by animal use.
We used the citation tracking facility within Web of Science to locate citations of original research papers on animal models related to ADHD published prior to 2010 identified in PubMed by relevant search terms. Human medical papers citing those animal studies were carefully analyzed by two independent raters to evaluate the contribution of the animal data to the human studies.
211 publications describing relevant animal studies were located. Approximately half (3,342) of their 6,406 citations were by other animal studies. 446 human medical papers cited 121 of these 211 animal studies, a total of 500 times. 254 of these 446 papers were human studies of ADHD. However, only eight of the cited animal papers (cited 10 times) were relevant to the hypothesis of the human medical study in question. Three of these eight papers described results from both human and animal studies, but their citations solely referred to the human data. Five animal research papers were relevant to the hypotheses of the applicable human medical papers.
Citation analysis indicates that animal research has contributed very little to contemporary understanding of ADHD. To ensure optimal allocation of Research & Development funds targeting this disorder the contribution of other research methods should be similarly evaluated
