    Toolkit for Simulation Modeling of Logistics Warehouse in Distributed Computing Environment

    We address an important problem of an automation of logistics warehouses simulation modeling in distributed service-oriented computing environments. To this end, we propose new approach for adjusting management system parameters of real warehouse in production use. It is based on the integration of the conceptual, wireframe and service-oriented programming used to develop parameter sweep applications and data analysis in the simulation modeling process. We develop a toolkit for supporting modeling of warehouse logistics. The practical experiments are focused upon the refrigerated warehouse.     Probing New Physics Models of Neutrinoless Double Beta Decay with SuperNEMO

    The possibility to probe new physics scenarios of light Majorana neutrino exchange and right-handed currents at the planned next generation neutrinoless double beta decay experiment SuperNEMO is discussed. Its ability to study different isotopes and track the outgoing electrons provides the means to discriminate different underlying mechanisms for the neutrinoless double beta decay by measuring the decay half-life and the electron angular and energy distributions.Comment: 17 pages, 14 figures, to be published in E.P.J.

    Introduction. Despite advances in the treatment of melanoma, the results of therapy cannot be considered satisfactory, and the search for new drugs and effective combinations of medicine continues. The drugs are being developed aimed at reducing the metastatic tumor potential – migrastatics. The targets of the drugs can be cytoskeletal proteins of tumor cells – cytokeratin (CK) intermediate filaments and microtubule protein beta-III tubulin (TUBB3).Aim. To estimate of the CK and TUBB3 expression in melanoma cell lines to form an informative in vitro cell model for screening and studying migrastatics.Materials and methods. The molecular phenotype of 21 human melanoma cell lines from the collection of N. N. Blokhin National Medical Research Center of Oncology, and 18 of which were isolated from tumor metastases in the lymph nodes, soft tissues or subcutaneously. The level of TUBB3 expression and de novo expression of CKs in vimentin-expressing cells (CK + Vim) were assessed by an immunofluorescent method and flow cytometry.Results. Beta-III tubulin expression was detected in all cultures studied, de novo expression of CKs was found in 20 / 21 lines. The exception was primary uveal melanoma 92-1, that did not express CK + Vim. Both parameters significantly differed between the cells of the studied panel: CK + Vim co-expression – from 0 to 91 %, TUBB3 – from 18 to 86 %. No correlation was found between the expression level of TUBB3 and CK + Vim (Pearson’s correlation coefficient r = 0.11; p = 0.65). Three groups of the cell lines with different ratio of TUBB3 expression and CK + Vim co-expression were identified: 1) similar level of expression of both markers; 2) the level of co-expression of CK + Vim more or less high than the index for TUBB3; 3) the level of TUBB3 expression more or less high than the index for CK + Vim co-expression.Conclusion. A panel of 21 human melanoma cell lines was formed with quantitatively estimated expression of cytoske-letal proteins responsible for the migration activity of tumor cells – CKs and TUBB3. Groups of the lines with different expression ratio of the markers can be used for screening and preclinical evaluation potential migrastatics that reduce the metastatic potential of melanoma and may reduce resistance to taxanes.Введение. Несмотря на достижения в лечении меланомы, результаты терапии нельзя признать удовлетворительными, и поиск новых препаратов и эффективных комбинаций лекарств продолжается. Разрабатываются препараты, направленные на снижение метастатического потенциала опухолей, – миграстатики. Точкой приложения этой группы препаратов могут являться белки цитоскелета опухолевой клетки, к которым относятся промежуточные филаменты – цитокератины (Цк) и белок микротрубочек бета-III тубулин (TUBB3).Цель исследования – формирование панели культур клеток меланомы с охарактеризованной экспрессией Цк и TUBB3 для создания информативной клеточной модели in vitro для скрининга и изучения миграстатиков.Материалы и методы. Исследован молекулярный фенотип 21 культуры клеток меланомы из коллекции клеточных линий ФГБУ «Национальный медицинский исследовательский центр онкологии им. Н. Н. Блохина» Минздрава России, из которых 18 культур получены из метастазов опухоли в лимфатические узлы, мягкие ткани или подкожно. Оценка уровня экспрессии TUBB3 и de novo экспрессии Цк в клетках, экспрессирующих виментин (Вим) (Цк + Вим), проведена иммунофлуоресцентным методом, ассоциированным с проточной цитометрией.Результаты. экспрессия TUBB3 выявлена во всех исследованных культурах, de novo экспрессия Цк – в 20 из 21 культуры. Исключение составила первичная увеальная меланома 92-1, не экспрессирующая Цк + Вим. Оба показателя значительно различались между клетками исследованной панели: коэкспрессия Цк + Вим – от 0 до 91 %, TUBB3 – от 18 до 86 %. корреляции между уровнями экспрессии TUBB3 и Цк + Вим не выявлено (коэффициент корреляции пирсона r = 0,11; p = 0,65). Выделены 3 группы культур клеток с разным соотношением уровня экспрессии TUBB3 и коэкспрессии Цк + Вим: 1) сходный уровень показателей экспрессии маркеров; 2) уровень коэкспрессии Цк + Вим в большей или меньшей степени превышает показатель для TUBB3; 3) уровень экспрессии TUBB3 в разной степени превышает показатель коэкспрессии Цк + Вим.Заключение. Сформирована панель из 21 культуры клеток меланомы человека, в которых количественно охарактеризована экспрессия Цк и TUBB3 – белков цитоскелета, ответственных за миграционную активность опухолевых клеток. Группы культур с разным соотношением показателей экспрессии этих маркеров могут быть использованы для скрининга и доклинической оценки потенциальных миграстатиков, которые уменьшают метастатический потенциал меланомы и могут снижать резистентность к таксанам

    Measurement of the 2νββ decay half-life of 150Nd and a search for 0νββ decay processes with the full exposure from the NEMO-3 detector

    We present results from a search for neutrinoless double-β (0νββ) decay using 36.6 g of the isotope 150Nd with data corresponding to a live time of 5.25 y recorded with the NEMO-3 detector. We construct a complete background model for this isotope, including a measurement of the two-neutrino double-β decay half-life of T2ν 1=2 ¼ ½9.34 0.22ðstatÞ þ0.62 −0.60 ðsystÞ × 1018 y for the ground state transition, which represents the most precise result to date for this isotope. We perform a multivariate analysis to search for 0νββ decays in order to improve the sensitivity and, in the case of observation, disentangle the possible underlying decay mechanisms. As no evidence for 0νββ decay is observed, we derive lower limits on half-lives for several mechanisms involving physics beyond the standard model. The observed lower limit, assuming light Majorana neutrino exchange mediates the decay, is T0ν 1=2 > 2.0 × 1022 y at the 90% C.L., corresponding to an upper limit on the effective neutrino mass of hmνi < 1.6–5.3 eV

    The Deep Underground Neutrino Experiment (DUNE) is an international, world-class experiment aimed at exploring fundamental questions about the universe that are at the forefront of astrophysics and particle physics research. DUNE will study questions pertaining to the preponderance of matter over antimatter in the early universe, the dynamics of supernovae, the subtleties of neutrino interaction physics, and a number of beyond the Standard Model topics accessible in a powerful neutrino beam. A critical component of the DUNE physics program involves the study of changes in a powerful beam of neutrinos, i.e., neutrino oscillations, as the neutrinos propagate a long distance. The experiment consists of a near detector, sited close to the source of the beam, and a far detector, sited along the beam at a large distance. This document, the DUNE Near Detector Conceptual Design Report (CDR), describes the design of the DUNE near detector and the science program that drives the design and technology choices. The goals and requirements underlying the design, along with projected performance are given. It serves as a starting point for a more detailed design that will be described in future documents

    Design, construction and operation of the ProtoDUNE-SP Liquid Argon TPC

    The ProtoDUNE-SP detector is a single-phase liquid argon time projection chamber (LArTPC) that was constructed and operated in the CERN North Area at the end of the H4 beamline. This detector is a prototype for the first far detector module of the Deep Underground Neutrino Experiment (DUNE), which will be constructed at the Sandford Underground Research Facility (SURF) in Lead, South Dakota, U.S.A. The ProtoDUNE-SP detector incorporates full-size components as designed for DUNE and has an active volume of 7 × 6 × 7.2 m3. The H4 beam delivers incident particles with well-measured momenta and high-purity particle identification. ProtoDUNE-SP's successful operation between 2018 and 2020 demonstrates the effectiveness of the single-phase far detector design.
