    The dynamics of critical Kauffman networks under asynchronous stochastic update

    We show that the mean number of attractors in a critical Boolean network under asynchronous stochastic update grows like a power law and that the mean size of the attractors increases as a stretched exponential with the system size.     We investigate the influence of a deterministic but non-synchronous update on Random Boolean Networks, with a focus on critical networks. Knowing that ``relevant components'' determine the number and length of attractors, we focus on such relevant components and calculate how the length and number of attractors on these components are modified by delays at one or more nodes.     Hsc70-induced changes in clathrin-auxilin cage structure suggest a role for clathrin light chains in cage disassembly

    The molecular chaperone, Hsc70, together with its co-factor, auxilin, facilitates the ATP-dependent removal of clathrin during clathrin-mediated endocytosis in cells. We have used cryo-electron microscopy to determine the 3D structure of a complex of clathrin, auxilin401-910 and Hsc70 at pH 6 in the presence of ATP, frozen within 20 seconds of adding Hsc70 in order to visualize events that follow the binding of Hsc70 to clathrin and auxilin before clathrin disassembly. In this map, we observe density beneath the vertex of the cage that we attribute to bound Hsc70. This density emerges asymmetrically from the clathrin vertex, suggesting preferential binding by Hsc70 for one of the three possible sites at the vertex. Statistical comparison with a map of whole auxilin and clathrin previously published by us reveals the location of statistically significant differences which implicate involvement of clathrin light chains in structural rearrangements which occur after Hsc70 is recruited. Clathrin disassembly assays using light scattering suggest that loss of clathrin light chains reduces the efficiency with which auxilin facilitates this reaction. These data support a regulatory role for clathrin light chains in clathrin disassembly in addition to their established role in regulating clathrin assembly

    Determinants of food insecurity among smallholder farmer households in Central America: recurrent versus extreme weather-driven events

    To ensure food security among rural communities under a changing climate, policymakers need information on the prevalence and determinants of food insecurity, the role of extreme weather events in exacerbating food insecurity,and the strategies that farmers use to cope with food insecurity. Using household surveys in Guatemala and Honduras, we explore the prevalence of food insecurity among smallholder farmers on both a recurrent (seasonal)and episodic (resulting from extreme weather events) basis, analyze the factors associated with both types of food insecurity, and document farmer coping strategies. Of the 439 households surveyed, 56% experienced recurrent foodinsecurity, 36% experienced episodic food insecurity due to extreme weather events, and 24% experienced both types. Food insecurity among smallholder farmers was correlated with sociodemographic factors (e.g., age, education,migration) and asset ownership. The factors affecting food insecurity differed between type and prevalence of food insecurity. Our results highlight the urgent need for policies and programs to help smallholder farmers improve theiroverall food security and resilience to extreme weather shocks. Such policies should focus on enhancing farmer education levels, securing land tenure, empowering women, promoting generational knowledge exchange, and providing emergency food support in the lean season or following extreme weather events

    Percepciones de cambio climático y respuestas adaptativas de caficultores costarricenses de pequeña escala

El cambio climático afectará la distribución, productividad y rentabilidad del cultivo de café en América Central, perjudicando las economías nacionales y los medios de vida de los pequeños productores. Es necesario entender cómo el cambio climático está afectando a los pequeños caficultores en la región para promover medidas que permitan enfrentar estos cambios y adaptarse a ellos. Objetivo. El objetivo de este trabajo fue describir los sistemas cafetaleros costarricenses de pequeña escala en dos zonas productivas y explorar los esfuerzos de adaptación al cambio climático que realizan los productores. Materiales y métodos. Entre marzo y mayo de 2014, se realizaron encuestas estructuradas en hogares cafetaleros de dos paisajes vulnerables al cambio climático: Turrialba y Los Santos. El estudio se centró en las percepciones de los productores sobre los cambios en temperatura y lluvia, los impactos reportados como consecuencia de dichos cambios y las acciones de adaptación implementadas en las fincas. Resultados. Los resultados indicaron que el 98 % de los productores percibió cambios en el clima, gran parte de ellos los asociaron con impactos sobre su producción de café, y el 60 % de los encuestados realizó cambios en el manejo de sus cafetales para reducir los efectos del cambio climático. Las prácticas de adaptación más frecuentes fueron sembrar árboles e incrementar el uso de agroquímicos, en mayor medida en respuesta al incremento de temperatura. Conclusión. Este estudio destaca la necesidad de incrementar el apoyo técnico, financiero y político orientado a fomentar y diversificar la implementación de prácticas de adaptación entre pequeños caficultores e incrementar su resiliencia ante el cambio climático

    Dietary Inflammatory Index and Non-Communicable Disease Risk: A Narrative Review

    There are over 1,000,000 publications on diet and health and over 480,000 references on inflammation in the National Library of Medicine database. In addition, there have now been over 30,000 peer-reviewed articles published on the relationship between diet, inflammation, and health outcomes. Based on this voluminous literature, it is now recognized that low-grade, chronic systemic inflammation is associated with most non-communicable diseases (NCDs), including diabetes, obesity, cardiovascular disease, cancers, respiratory and musculoskeletal disorders, as well as impaired neurodevelopment and adverse mental health outcomes. Dietary components modulate inflammatory status. In recent years, the Dietary Inflammatory Index (DII®), a literature-derived dietary index, was developed to characterize the inflammatory potential of habitual diet. Subsequently, a large and rapidly growing body of research investigating associations between dietary inflammatory potential, determined by the DII, and risk of a wide range of NCDs has emerged. In this narrative review, we examine the current state of the science regarding relationships between the DII and cancer, cardiometabolic, respiratory and musculoskeletal diseases, neurodevelopment, and adverse mental health outcomes. We synthesize the findings from recent studies, discuss potential underlying mechanisms, and look to the future regarding novel applications of the adult and children’s DII (C-DII) scores and new avenues of investigation in this field of nutritional research

    Abnormal repetitive behaviors in zebrafish and their relevance to human brain disorders

    Abnormal repetitive behaviors (ARBs) are a prominent symptom of numerous human brain disorders and are commonly seen in rodent models as well. While rodent studies of ARBs continue to dominate the field, mounting evidence suggests that zebrafish (Danio rerio) also display ARB-like phenotypes and may therefore be a novel model organism for ARB research. In addition to clear practical research advantages as a model species, zebrafish share high genetic and physiological homology to humans and rodents, including multiple ARB-related genes and robust behaviors relevant to ARB. Here, we discuss a wide spectrum of stereotypic repetitive behaviors in zebrafish, data on their genetic and pharmacological modulation, and the overall translational relevance of fish ARBs to modeling human brain disorders. Overall, the zebrafish is rapidly emerging as a new promising model to study ARBs and their underlying mechanisms. © 2019 Elsevier B.V

    To uncover the circuit-level alterations that underlie atypical sensory processing associated with autism, we adopted a symptom-to-circuit approach in the Fmr1-knockout (Fmr1-/-) mouse model of Fragile X syndrome. Using a go/no-go task and in vivo two-photon calcium imaging, we find that impaired visual discrimination in Fmr1-/- mice correlates with marked deficits in orientation tuning of principal neurons and with a decrease in the activity of parvalbumin interneurons in primary visual cortex. Restoring visually evoked activity in parvalbumin cells in Fmr1-/- mice with a chemogenetic strategy using designer receptors exclusively activated by designer drugs was sufficient to rescue their behavioral performance. Strikingly, human subjects with Fragile X syndrome exhibit impairments in visual discrimination similar to those in Fmr1-/- mice. These results suggest that manipulating inhibition may help sensory processing in Fragile X syndrome

    Recent studies have recognized that signs of functional disability in schizophrenia are evident in early phases of the disorder, and, as a result, can potentially serve as vulnerability markers of future illness. However, functional measures in the psychosis prodrome have focused exclusively on real-world achievements, rather than on the skills required to carry-out a particular real-world function (ie, capacity). Despite growing evidence that diminished capacity is critical to the etiology of the established disorder, virtually no attention has been directed towards assessing functional capacity in the pre-illness stages.
