: In this article we describe and analyze several impacts that coronavirus has produced over some aboriginal population from America and Argentina, taking into consideration, their responses, understanings and developed strategies, related to state policies. Likewise, we develop an analytical description of possibilities that the ethnographic method, belonging to the anthropological sciences, offers, for a multidimensional approach. This method also makes it possible to advise, evaluate and monitor certain public policies. When it comes to the information obtained, it was acquired using several secondary sources of data and interviews posted on the Internet. Fil: Barandela, Ana Clara. Biotechnological production of taxanes: a molecular approach

Podeu consultar el llibre complet a: http://hdl.handle.net/2445/46988Plant cell cultures constitute a promise for the production of a high number of phytochemicals, although the majority of bioprocesses that have been developed so far have not resulted commercially successful. An overview indicates that most of the research carried out until now is of the empirical type. For this reason, there is a need for a rational approach to the molecular and cellular basis of metabolic pathways and their regulation in order to stimulate future advances. The empirical investigations are based on the optimization of the culture system, exclusively considering input factors such as the selection of cellular lines, type and parameters of culture, bioreactor design and elicitor addition, and output factors such as cellular growth, the uptake system of nutrients, production and yield. In a rational approach towards the elucidation of taxol and related taxane production, our group has studied the relationship between the taxane profile and production and the expression of genes codifying for enzymes that participate in early, intermediate and late steps of their biosynthesis in elicited Taxus spp cell cultures. Our results show that elicitors induce a dramatic reprogramming of gene expression in Taxus cell cultures, which likely accounts for the enhanced production of taxol and related taxanes and we have also determined some genes that control the main flux limiting steps. The application of metabolic engineering techniques for the production of taxol and taxanes of interest is also discussed

Viability-reducing activity of Coryllus avellana L. extracts against human cancer cell lines

The increasing rate of cancer incidence has encouraged the search for novel natural sources of anticancer compounds. The presence of small quantities of taxol and taxanes in Corylus avellana L. has impelled new potential applications for this plant in the field of biomedicine. In the present work, the cell viability-reducing activity of stems and leaves from three different hazel trees was studied against three human-derived cancer cell lines (HeLa, HepG2 and MCF-7). Both leaf and stem extracts significantly reduced viability of the three cell lines either after maceration with methanol or using taxane extraction methods. Since maceration reduced cell viability to a greater extent than taxane extraction methods, we scaled up the maceration extraction process using a method for solid/liquid extraction (Zippertex technology). Methanol leaf extracts promoted a higher reduction in viability of all cell lines assayed than stem extracts. Fractionation of methanol leaf extracts using silica gel chromatography led to the purification and identification of two compounds by HPLC-MS and NMR: (3R,5R)-3,5-dihydroxy-1,7-bis(4-hydroxyphenyl) heptane 3-O-β-d-glucopyranoside and quercetin-3-O-rhamnoside. The isolated compounds decreased viability of HeLa and HepG2 cells to a greater extent than MCF-7 cells. Our results suggest a potential use of C. avellana extracts in the pharmacotherapy of cervical cancer and hepatocarcinoma and, to a lesser extent, breast cancer

Filtering the NMR Spectra of Mixtures by Coordination to Paramagnetic Cu2+

    No full text
The paramagnetic spin relaxation (PSR) filter allows the selective NMR signal suppression of components in mixtures according to their complexation ability to a paramagnetic ion. It relies on the faster relaxation of nuclei in paramagnetic environments and thus is complementary to classical diffusion and relaxation filters. So far, the PSR filter has established Gd3+ as the sole PSR agent, restricting the paramagnetic filtering repertoire. Herein, we present Cu2+ as a robust PSR agent with characteristic filtering properties. While Gd3+ depends on unspecific ion-pair interactions with anionic components, Cu2+ stands out for filtering species via ordered coordination complexes. An evaluation of the paramagnetic effect of Cu2+ over more than 50 small molecules and polymers has unveiled different sensitivities to Cu2+ (especially high for pyridines, diamines, polyamines, and amino alcohols) and precise filtering conditions for mixtures (1H, COSY, and HMQC) that were challenged with a test bed of commercial drugs. The advantage of integrating Cu2+ and Gd3+ for the stepwise PSR filtering of complex mixtures is also shownThis work was supported by the Spanish Ministry of Science and Innovation (and PID2021-127684OB-I00), the Xunta de Galicia (ED431C 2018/30, and Centro singular de investigación de Galicia accreditation 2019–2022, ED431G2019/03), Axencia Galega de Innovación (IN845D 2020/09), and the European Union (European Regional Development Fund-ERDF).S
