    Viral antibody dynamics in a chiropteran host

    1. Bats host many viruses that are significant for human and domestic animal health, but the dynamics of these infections in their natural reservoir hosts remain poorly elucidated.<p></p> 2. In these, and other, systems, there is evidence that seasonal life-cycle events drive infection dynamics, directly impacting the risk of exposure to spillover hosts. Understanding these dynamics improves our ability to predict zoonotic spillover from the reservoir hosts.<p></p> 3. To this end, we followed henipavirus antibody levels of >100 individual E. helvum in a closed, captive, breeding population over a 30-month period, using a powerful novel antibody quantitation method.<p></p> 4. We demonstrate the presence of maternal antibodies in this system and accurately determine their longevity. We also present evidence of population-level persistence of viral infection and demonstrate periods of increased horizontal virus transmission associated with the pregnancy/lactation period.<p></p> 5.The novel findings of infection persistence and the effect of pregnancy on viral transmission, as well as an accurate quantitation of chiropteran maternal antiviral antibody half-life, provide fundamental baseline data for the continued study of viral infections in these important reservoir hosts

    Are in vitro estimates of cell diffusivity and cell proliferation rate sensitive to assay geometry?

    AbstractCells respond to various biochemical and physical cues during wound-healing and tumour progression. in vitro assays used to study these processes are typically conducted in one particular geometry and it is unclear how the assay geometry affects the capacity of cell populations to spread, or whether the relevant mechanisms, such as cell motility and cell proliferation, are somehow sensitive to the geometry of the assay. In this work we use a circular barrier assay to characterise the spreading of cell populations in two different geometries. Assay 1 describes a tumour-like geometry where a cell population spreads outwards into an open space. Assay 2 describes a wound-like geometry where a cell population spreads inwards to close a void. We use a combination of discrete and continuum mathematical models and automated image processing methods to obtain independent estimates of the effective cell diffusivity, D, and the effective cell proliferation rate, λ. Using our parameterised mathematical model we confirm that our estimates of D and λ accurately predict the time-evolution of the location of the leading edge and the cell density profiles for both assay 1 and assay 2. Our work suggests that the effective cell diffusivity is up to 50% lower for assay 2 compared to assay 1, whereas the effective cell proliferation rate is up to 30% lower for assay 2 compared to assay 1

    Sediment removal by prairie filter strips in row-cropped ephemeral watersheds

    Twelve small watersheds in central Iowa were used to evaluate the effectiveness of prairie filter strips (PFS) in trapping sediment from agricultural runoff. Four treatments with PFS of different size and location (100% rowcrop, 10% PFS of total watershed area at footslope, 10% PFS at footslope and in contour strips, 20% PFS at footslope and in contour strips) arranged in a balanced incomplete block design were seeded in July 2007. All watersheds were in bromegrass ( L.) for at least 10 yr before treatment establishment. Cropped areas were managed under a no-till, 2-yr corn ( L.)-soybean [ (L.) Merr.] rotation beginning in 2007. About 38 to 85% of the total sediment export from cropland occurred during the early growth stage of rowcrop due to wet field conditions and poor ground cover. The greatest sediment load was observed in 2008 due to the initial soil disturbance and gradually decreased thereafter. The mean annual sediment yield through 2010 was 0.36 and 8.30 Mg ha for the watersheds with and without PFS, respectively, a 96% sediment trapping efficiency for the 4-yr study period. The amount and distribution of PFS had no significant impact on runoff and sediment yield, probably due to the relatively large width (37-78 m) of footslope PFS. The findings suggest that incorporation of PFS at the footslope position of annual rowcrop systems provides an effective approach to reducing sediment loss in runoff from agricultural watersheds under a no-till system

    Extension to order β23\beta^{23} of the high-temperature expansions for the spin-1/2 Ising model on the simple-cubic and the body-centered-cubic lattices

    Using a renormalized linked-cluster-expansion method, we have extended to order β23\beta^{23} the high-temperature series for the susceptibility χ\chi and the second-moment correlation length ξ\xi of the spin-1/2 Ising models on the sc and the bcc lattices. A study of these expansions yields updated direct estimates of universal parameters, such as exponents and amplitude ratios, which characterize the critical behavior of χ\chi and ξ\xi. Our best estimates for the inverse critical temperatures are βcsc=0.221654(1)\beta^{sc}_c=0.221654(1) and βcbcc=0.1573725(6)\beta^{bcc}_c=0.1573725(6). For the susceptibility exponent we get γ=1.2375(6)\gamma=1.2375(6) and for the correlation length exponent we get ν=0.6302(4)\nu=0.6302(4). The ratio of the critical amplitudes of χ\chi above and below the critical temperature is estimated to be C+/C=4.762(8)C_+/C_-=4.762(8). The analogous ratio for ξ\xi is estimated to be f+/f=1.963(8)f_+/f_-=1.963(8).     Significance Tests for Periodogram Peaks

    We discuss methods currently in use for determining the significance of peaks in the periodograms of time series. We discuss some general methods for constructing significance tests, false alarm probability functions, and the role played in these by independent random variables and by empirical and theoretical cumulative distribution functions. We also discuss the concept of "independent frequencies" in periodogram analysis. We propose a practical method for estimating the significance of periodogram peaks, applicable to all time series irrespective of the spacing of the data.     Anthropogenic Space Weather

    Anthropogenic effects on the space environment started in the late 19th century and reached their peak in the 1960s when high-altitude nuclear explosions were carried out by the USA and the Soviet Union. These explosions created artificial radiation belts near Earth that resulted in major damages to several satellites. Another, unexpected impact of the high-altitude nuclear tests was the electromagnetic pulse (EMP) that can have devastating effects over a large geographic area (as large as the continental United States). Other anthropogenic impacts on the space environment include chemical release ex- periments, high-frequency wave heating of the ionosphere and the interaction of VLF waves with the radiation belts. This paper reviews the fundamental physical process behind these phenomena and discusses the observations of their impacts.Comment: 71 pages, 35 figure

    Protein sequence and structure: Is one more fundamental than the other?

    We argue that protein native state structures reside in a novel "phase" of matter which confers on proteins their many amazing characteristics. This phase arises from the common features of all globular proteins and is characterized by a sequence-independent free energy landscape with relatively few low energy minima with funnel-like character. The choice of a sequence that fits well into one of these predetermined structures facilitates rapid and cooperative folding.
