    Высоковольтная система инжекции для компактного сильноточного бетатрона

    Система высоковольтной инжекции является одним из основных узлов сильноточного бетатрона, определяя в известной мере вес и габаритные размеры всего ускорителя в целом. В статье описываются конструктивные изменения элементов высоковольтной инжекции, которые, однако, не снижают амплитуду напряжения и мощность в импульсе по сравнению с генераторами инжекции предыдущих разработок. Указанные меры позволяют сократить размеры и вес высоковольтного источника ускоренных электронов с целью применения в системе инжекции компактного сильноточного бетатрона. Объединение электронной пушки торцового типа с импульсным трансформатором, питающим одновременно с пушкой и инфлектор электростатического типа, позволяет повысить мобильность и упростить режим настройки на излучение сильноточного бетатрона. Поскольку для условий максимального токопрохождения пучка электронов через инфлектор требуется определенное согласование по фазе импульсов, подаваемых на пушку и инфлектор, в статье рассматриваются схемы для совмещенного питания, удовлетворяющие этим требованиям. Повышение экономичности системы инжекции для компактного бетатрона достигается за счет применения резонансного способа заряда накопителя комбинированного типа. Приводятся основные соотношения для расчета и выбора параметров элементов высоковольтного модулятора

    Disruption of the plant-specific CFS1 gene impairs autophagosome turnover and triggers EDS1-dependent cell death

    Cell death, autophagy and endosomal sorting contribute to many physiological, developmental and immunological processes in plants. They are mechanistically interconnected and interdependent, but the molecular basis of their mutual regulation has only begun to emerge in plants. Here, we describe the identification and molecular characterization of CELL DEATH RELATED ENDOSOMAL FYVE/SYLF PROTEIN 1 (CFS1). The CFS1 protein interacts with the ENDOSOMAL SORTING COMPLEX REQUIRED FOR TRANSPORT I (ESCRT-I) component ELCH (ELC) and is localized at ESCRT-I-positive late endosomes likely through its PI3P and actin binding SH3YL1 Ysc84/Lsb4p Lsb3p plant FYVE (SYLF) domain. Mutant alleles of cfs1 exhibit auto-immune phenotypes including spontaneous lesions that show characteristics of hypersensitive response (HR). Autoimmunity in cfs1 is dependent on ENHANCED DISEASE SUSCEPTIBILITY 1 (EDS1)-mediated effector-triggered immunity (ETI) but independent from salicylic acid. Additionally, cfs1 mutants accumulate the autophagy markers ATG8 and NBR1 independently from EDS1. We hypothesize that CFS1 acts at the intersection of autophagosomes and endosomes and contributes to cellular homeostasis by mediating autophagosome turnover

    Phase-ordering dynamics of the Gay-Berne nematic liquid crystal

    Phase-ordering dynamics in nematic liquid crystals has been the subject of much active investigation in recent years in theory, experiments and simulations. With a rapid quench from the isotropic to nematic phase a large number of topological defects are formed and dominate the subsequent equilibration process. We present here the results of a molecular dynamics simulation of the Gay-Berne model of liquid crystals after such a quench in a system with 65536 molecules. Twist disclination lines as well as type-1 lines and monopoles were observed. Evidence of dynamical scaling was found in the behavior of the spatial correlation function and the density of disclination lines. However, the behavior of the structure factor provides a more sensitive measure of scaling, and we observed a crossover from a defect dominated regime at small values of the wavevector to a thermal fluctuation dominated regime at large wavevector.Comment: 18 pages, 16 figures, animations available at http://www.physics.brown.edu/Users/faculty/pelcovits/lc/coarsening.htm

    Sea level changes during the last and present interglacials in Sal Island (Cape Verde archipelago)

    Last interglacial and Holocene deposits are particularly well developed in the southern parts of Sal Island (Cape Verde Archipelago). They primarily consist of low-elevation (≤2 m above sea level [a.s.l.]) marine deposits made of a basal conglomerate embedded in carbonate mud, passing upwards to calcarenites. All deposits contain an abundant fauna with corals, algae and molluscs with Strombus latus Gmelin and accompanying warm water species of the “Senegalese” fauna. Small scale geomorphological mapping with detailed morphosedimentary analysis revealed lateral facies changes and imbricate (offlapping) structures that suggest small-scale oscillations of paleo-sealevels during high sea stand intervals. U-series measurements (in coral fragments) allowed unequivocal identification of Marine Isotope Substage (MIS) 5.5 units, but were not precise enough to date the sea level oscillations of the interval. However, geomorphological data and sedimentary facies analysis suggest a double sea level highstand during the peak of the last interglacial. MIS 5.5 age deposits occur at Sal and the Canary Islands at low topographic elevations, between 1 and 2 masl. However, these values are lower than the elevations measured for the correlative terraces outcropping at the western tropical Atlantic islands, widely considered to be tectonically stable. Combining the results in this paper with earlier investigations of the “Senegalese” fauna distribution as far north as the Mediterranean basin, it is suggested that the last-interglacial oceanic temperatures in this basin, as well as the temperatures in other islands of the Eastern Atlantic and the coasts of Morocco, were warmer than modern temperatures

    A Gene Regulatory Network for Root Epidermis Cell Differentiation in Arabidopsis

    The root epidermis of Arabidopsis provides an exceptional model for studying the molecular basis of cell fate and differentiation. To obtain a systems-level view of root epidermal cell differentiation, we used a genome-wide transcriptome approach to define and organize a large set of genes into a transcriptional regulatory network. Using cell fate mutants that produce only one of the two epidermal cell types, together with fluorescence-activated cell-sorting to preferentially analyze the root epidermis transcriptome, we identified 1,582 genes differentially expressed in the root-hair or non-hair cell types, including a set of 208 “core” root epidermal genes. The organization of the core genes into a network was accomplished by using 17 distinct root epidermis mutants and 2 hormone treatments to perturb the system and assess the effects on each gene's transcript accumulation. In addition, temporal gene expression information from a developmental time series dataset and predicted gene associations derived from a Bayesian modeling approach were used to aid the positioning of genes within the network. Further, a detailed functional analysis of likely bHLH regulatory genes within the network, including MYC1, bHLH54, bHLH66, and bHLH82, showed that three distinct subfamilies of bHLH proteins participate in root epidermis development in a stage-specific manner. The integration of genetic, genomic, and computational analyses provides a new view of the composition, architecture, and logic of the root epidermal transcriptional network, and it demonstrates the utility of a comprehensive systems approach for dissecting a complex regulatory network

    Global and local sea level during the Last Interglacial: A probabilistic assessment

    The Last Interglacial (LIG) stage, with polar temperatures likely 3-5 C warmer than today, serves as a partial analogue for low-end future warming scenarios. Based upon a small set of local sea level indicators, the Intergovernmental Panel on Climate Change (IPCC) inferred that LIG global sea level (GSL) was about 4-6 m higher than today. However, because local sea levels differ from GSL, accurately reconstructing past GSL requires an integrated analysis of globally distributed data sets. Here we compile an extensive database of sea level indicators and apply a novel statistical approach that couples Gaussian process regression of sea level to Markov Chain Monte Carlo modeling of geochronological errors. Our analysis strongly supports the hypothesis that LIG GSL was higher than today, probably peaking at 6-9 m.     Multivariate analysis of mineral constituents of edible Parasol Mushroom (Macrolepiota procera) and soils beneath fruiting bodies collected from Northern Poland

    Caps and stipes of 141 fruiting bodies of Parasol Mushroom (Macrolepiota procera) and surface layer of soils collected from 11 spatially distant and background (pristine) areas in Northern Poland were analyzed for Ag, Al, Ba, Ca, Cd, Co, Cr, Cu, Fe, Hg, K, Mg, Mn, Na, Ni, P, Pb, Rb, Sr, and Zn by inductively coupled plasma optical emission spectroscopy and cold vapor atomic absorption spectroscopy. In terms of bioconcentration and bioexclusion concept, K, Ag, Cu, Rb, and P were highly bioconcentrated in caps, and their bioconcentration factor values varied for the 11 sites between 120 and 500—67–420, 70–220, 10–170, and 45–100, respectively. Cd, Zn, Mg, and Na showed bioconcentration factors (BCFs) between 3.3 and 36, 3.7–15, 0.92–6.3, and 1.4–44 while Al, Ba, Ca, Co, Cr, Mn, Ni, Pb, and Sr were excluded (BCF < 1). The Parasol Mushroom is a species harvested in the wild, and its caps are of unique taste and can contain a spectrum of essential and hazardous mineral compounds accumulated at elevated concentrations, even if collected at the background (pristine) areas. These elevated mineral concentrations of the caps are due to the efficient bioconcentration potential of the species (K, Ag, Cu, Rb, P, Cd, Zn, Mg, and Na) and abundance in the soil substrates (Al, Ca, Fe, Mn). The estimated intake rates of Cd, Hg, and Pb contained in Parasol Mushroom’s caps show a cause for concern associated with these metals resulting from the consumption of between 300- and 500-g caps daily, on a frequent basis in the mushrooming season

    Sea level and climate changes during OIS 5e in the Western Mediterranean

    Palaeontological, geomorphological and sedimentological data supported by isotopic dating on Oxygen Isotopic Stage (OIS) 5e deposits from the Spanish Mediterranean coast, are interpreted with the aim of reconstructing climatic instability in the Northern Hemisphere. Data point to marked climatic instability during the Last Interglacial (OIS 5e), with a change in meteorological conditions and, consequently, in the sedimentary environment. The oolitic facies generated during the first part of OIS 5e (ca. 135 kyr) shift into reddish conglomeratic facies during the second part (ca. 117 kyr). Sea surface Temperature (SST) and salinity are interpreted mainly on the basis of warm Senegalese fauna, which show chronological and spatial differential distribution throughout the Western Mediterranean. Present hydrological and meteorological conditions are used also as modern analogues to reconstruct climatic variability throughout the Last Interglacial, and this variability is interpreted within the wider framework of the North Atlantic record. All the available data indicate an increase in storminess induced by an increase in the influence of northwesterlies, a slight drop of SST in the northern Western Mediterranean, and an important change in meteorological conditions at the end of OIS 5e (117 kyr). These changes correlate well with the decrease in summer insolation and with the climatic instability recorded in North Atlantic high latitudes

    Endoreplication Controls Cell Fate Maintenance

    Cell-fate specification is typically thought to precede and determine cell-cycle regulation during differentiation. Here we show that endoreplication, also known as endoreduplication, a specialized cell-cycle variant often associated with cell differentiation but also frequently occurring in malignant cells, plays a role in maintaining cell fate. For our study we have used Arabidopsis trichomes as a model system and have manipulated endoreplication levels via mutants of cell-cycle regulators and overexpression of cell-cycle inhibitors under a trichome-specific promoter. Strikingly, a reduction of endoreplication resulted in reduced trichome numbers and caused trichomes to lose their identity. Live observations of young Arabidopsis leaves revealed that dedifferentiating trichomes re-entered mitosis and were re-integrated into the epidermal pavement-cell layer, acquiring the typical characteristics of the surrounding epidermal cells. Conversely, when we promoted endoreplication in glabrous patterning mutants, trichome fate could be restored, demonstrating that endoreplication is an important determinant of cell identity. Our data lead to a new model of cell-fate control and tissue integrity during development by revealing a cell-fate quality control system at the tissue level

    Endocytic regulation of alkali metal transport proteins in mammals, yeast and plants

    The relative concentrations of ions and solutes inside cells are actively maintained by several classes of transport proteins, in many cases against their concentration gradient. These transport processes, which consume a large portion of cellular energy, must be constantly regulated. Many structurally distinct families of channels, carriers, and pumps have been characterized in considerable detail during the past decades and defects in the function of some of these proteins have been linked to a growing list of human diseases. The dynamic regulation of the transport proteins present at the cell surface is vital for both normal cellular function and for the successful adaptation to changing environments. The composition of proteins present at the cell surface is controlled on both the transcriptional and post-translational level. Post-translational regulation involves highly conserved mechanisms of phosphorylation- and ubiquitylation-dependent signal transduction routes used to modify the cohort of receptors and transport proteins present under any given circumstances. In this review, we will summarize what is currently known about one facet of this regulatory process: the endocytic regulation of alkali metal transport proteins.
