    Low incidence of toxoplasma infection during pregnancy and in newborns in Sweden

    To estimate the burden of disease due to congenital toxoplasmosis in Sweden the incidence of primary infections during pregnancy and birth prevalence of congenital toxoplasmosis in 40978 children born in two regions in Sweden was determined. Women possibly infected during pregnancy were identified based on: 1, detection of specific IgG based on neonatal screening of the phenylketonuria (PKU) card blood spot followed by retrospective testing of stored prenatal samples to detect women who acquired infection during pregnancy and follow up of their children to 12 months; 2, detection of specific IgM on the PKU blood spot. The birth prevalence of congenital toxoplasmosis was 0·73/10000 (95% CI 0·15–2·14) (3/40978). The incidence of primary infection during pregnancy was 5·1/10000 (95% CI 2·6–8·9) susceptible pregnant women. The seroprevalence in the southern part was 25·7% and in the Stockholm area 14·0%. The incidence of infection during pregnancy was low, as the birth prevalence of congenital toxoplasmosis. Neonatal screening warrants consideration in view of the low cost and feasibility

    The challenges and benefits of analyzing feedback comments in surveys: Lessons from a cross-national online survey of small-scale cannabis growers

    It is common practice in survey questionnaires to include a general open and non-directive feedback question at the end, but the analysis of this type of data is rarely discussed in the methodological literature. While these open-ended comments can be useful, most researchers fail to report on this issue. The aim of this article is to illustrate and reflect upon the benefits and challenges of analyzing responses to open-ended feedback questions. The article describes the experiences of coding and analyzing data generated through a feedback question at the end of an international online survey with small-scale cannabis cultivators carried out by the Global Cannabis Cultivation Research Consortium. After describing the design and dataset of the web survey, the analytical approach and coding frame are presented. The analytical strategies chosen in this study illustrate the diversity and complexity of feedback comments which pose methodological challenges to researchers wishing to use them for data analyses. In this article, three types of feedback comments (political/policy comments, general comments of positive and negative appreciation, and methodological comments) are used to illustrate the difficulties and advantages of analyzing this type of data. The advantages of analyzing feedback comments are well known, but they seem to be rarely exploited. General feedback questions at the end of surveys are typically non-directive. If researchers want to use these data for research and analyses, they need a clear strategy. They ought to give enough thought to why they are including this type of question, and develop an analytical strategy at the design stage of the study

    Provenance in Open Data Entity-Centric Aggregation

    An increasing number of web services these days require combining data from several data providers into an aggregated database. Usually this aggregation is based on the linked data approach. On the other hand, the entity-centric model is a promising data model that outperforms the linked data approach because it solves the lack of explicit semantics and the semantic heterogeneity problems. However, current open data which is available on the web as raw datasets can not be used in the entity-centric model before processing them with an import process to extract the data elements and insert them correctly in the aggregated entity-centric database. It is essential to certify the quality of these imported data elements, especially the background knowledge part which acts as input to semantic computations, because the quality of this part affects directly the quality of the web services which are built on top of it. Furthermore, the aggregation of entities and their attribute values from different sources raises three problems: the need to trace the source of each element, the need to trace the links between entities which can be considered equivalent and the need to handle possible conflicts between different values when they are imported from various data sources. In this thesis, we introduce a new model to certify the quality of a back ground knowledge base which separates linguistic and language independent elements. We also present a pipeline to import entities from open data repositories to add the missing implicit semantics and to eliminate the semantic heterogeneity. Finally, we show how to trace the source of attribute values coming from different data providers; how to choose a strategy for handling possible conflicts between these values; and how to keep the links between identical entities which represent the same real world entity

    Evolution from adherent to suspension: systems biology of HEK293 cell line development

    The need for new safe and efficacious therapies has led to an increased focus on biologics produced in mammalian cells. The human cell line HEK293 has bio-synthetic potential for human-like production attributes and is currently used for manufacturing of several therapeutic proteins and viral vectors. Despite the increased popularity of this strain we still have limited knowledge on the genetic composition of its derivatives. Here we present a genomic, transcriptomic and metabolic gene analysis of six of the most widely used HEK293 cell lines. Changes in gene copy and expression between industrial progeny cell lines and the original HEK293 were associated with cellular component organization, cell motility and cell adhesion. Changes in gene expression between adherent and suspension derivatives highlighted switching in cholesterol biosynthesis and expression of five key genes (RARG, ID1, ZIC1, LOX and DHRS3), a pattern validated in 63 human adherent or suspension cell lines of other origin

    The rapid development of the magnetic tunnel junction (MTJ) spin torque oscillator (STO) technology demands an analytical model to enable building MTJ STO-based circuits and systems so as to evaluate and utilize MTJ STOs in various applications. In Part I of this paper, an analytical model based on the macrospin approximation has been introduced and verified by comparing it with the measurements of three different MTJ STOs. In Part II, the full Verilog-A implementation of the proposed model is presented. To achieve a reliable model, an approach to reproducing the phase noise generated by the MTJ STO has been proposed and successfully employed. The implemented model yields a time domain signal, which retains the characteristics of operating frequency, linewidth, oscillation amplitude, and DC operating point, with respect to the magnetic field and applied DC current. The Verilog-A implementation is verified against the analytical model, providing equivalent device characteristics for the full range of biasing conditions. Furthermore, a system that includes an MTJ STO and CMOS RF circuits is simulated to validate the proposed model for system- and circuit-level designs. The simulation results demonstrate that the proposed model opens the possibility to explore STO technology in a wide range of applications

    Globally Distributed Drug Discovery of New Antibiotics: Design and Combinatorial Synthesis of Amino Acid Derivatives in the Organic Chemistry Laboratory

    An experiment for the synthesis of N-acyl derivatives of natural amino acids has been developed as part of the Distributed Drug Discovery (D3) program. Students use solid-phase synthesis techniques to complete a three-step, combinatorial synthesis of six products, which are analyzed using LC–MS and NMR spectroscopy. This protocol is suitable for introductory organic laboratory students and has been successfully implemented at multiple academic sites internationally. Accompanying prelab activities introduce students to SciFinder and to medicinal chemistry design principles. Pairing of these activities with the laboratory work provides students an authentic and cohesive research project experience
