    A Simple Device for Measuring the Minimum Current Velocity to Maintain Semi-Buoyant Fish Eggs in Suspension

    Pelagic broadcast spawning cyprinids are common to Great Plains rivers and streams. This reproductive guild produces non-adhesive semi-buoyant eggs that require sufficient current velocity to remain in suspension during development. Although studies have shown that there may be a minimum velocity needed to keep the eggs in suspension, this velocity has not been estimated directly nor has the influence of physicochemical factors on egg buoyancy been determined. We developed a simple, inexpensive flow chamber that allowed for evaluation of minimum current velocity needed to keep semi-buoyant eggs in suspension at any time frame during egg development. The device described here has the capability of testing the minimum current velocity needed to keep semi-buoyant eggs in suspension at a wide range of physicochemical conditions. We used gellan beads soaked in freshwater for 0, 24, and 48 hrs as egg surrogates and evaluated minimum current velocities necessary to keep them in suspension at different combinations of temperature (20.0 ± 1.0 °C, 25.0 ± 1.0 °C, and 28.0 ± 1.0 °C) and total dissolved solids (TDS; 1,000 mg L-1, 3,000 mg L-1, and 6,000 mg L-1). We found that our methodology generated consistent, repeatable results within treatment groups. Current velocities ranging from 0.001–0.026 needed to keep the gellan beads in suspension were negatively correlated to soak times and TDS and positively correlated with temperature. The flow chamber is a viable approach for evaluating minimum current velocities needed to keep the eggs of pelagic broadcast spawning cyprinids in suspension during development

    The MEGAPIE-TEST Project

    The goals of the MEGAPIE initiative are to design, build and operate a 1MW heavy liquid metal target. The first step towards the realization of the MEGAPIE target was the feasibility studies, which outlined the entire project. Contextually to the feasibility studies the conceptual design phase started with the establishment of R&D working groups assisting the design and validation of both the target and its ancillary systems. In this framework the EU project MEGAPIE-TEST has been structured in three work packages with tasks concerning the finalization of the engineering design, the components and subsystem testing, the integral test and the first irradiation period. The MEGAPIE-TEST consortium is composed by the 14 partners: FZK, PSI, CEA, ENEA, SCK-CEN, CNRS /IDFE, IN2P3, LMPGM, EMN, ISMRA, UNIV-NANTES, U-PSUDXI, USTL. Currently the engineering design of the target has been finalized, its manufacturing has been launched and the design activities on the ancillary systems were almost completed. R&D activities in the fields of materials, thermal – hydraulics, structural mechanics, neutronic and nuclear assessment and liquid metal technologies were performed in order to assist specific design issues. Some Subsystem and component tests were also performed and the preparation of the integral test is an ongoing activity

    Paradigms in multiple sclerosis: time for a change, time for a unifying concept

    It has recently been suggested that, rather than being an autoimmune disease, multiple sclerosis (MS) is an example of a neurocristopathy, a pathological process resulting from a faulty development of the neural crest. Whilst several characteristics of the disease suggest a neurocristopathy, other aetiological factors require consideration, including hygiene-related factors that alter the immune responses to common pathogens resulting in an eclipse of immune reactivity that could protect against MS, the possible role of human endogenous retroviruses (HERVs) in pathogenesis and autoimmune phenomena, HLA polymorphism, vitamin D levels before and after birth and immune repair mechanisms. A postulated aetiological factor in MS, associated with altered vitamin D metabolism and abnormal HERV expression, is a long-lasting disturbed redox regulation in the biosynthesis of a melanoma-like melanin pigment. Although intensive further studies on melanin pigments in nerve tissue in MS are required, the known properties of a pathological form of such pigments in melanoma could explain a number of observations in MS, including the impact of light, UV-light, and vitamin D, and could explain the clinical manifestations of MS on the basis of an oscillating process of oxidative charge and discharge of the pigments and a threshold phenomenon with a change of the quasi-catalytic function of the pigment from destroying reactive oxygen radicals or species to transforming them to more harmful long-persisting highly reactive species. Taken together with the consequences of an adaptive process in partly demyelinated neurons, resulting in an increase in number of mitochondria, and the impact of stressful life events, these conditions are necessary and sufficient to explain the disease process of MS with its spatial (plaques) and temporal (attacks and remissions) characteristics. This suggested unifying concept of the pathogenesis of MS may open perspectives for prevention, diagnosis and therapy. In particular, prevention may be achieved by vaccinating against Epstein-Barr virus in early childhood

    Assessment of motor functioning in the preschool period

    The assessment of motor functioning in young children has become increasingly important in recent years with the acknowledgement that motor impairment is linked with cognitive, language, social and emotional difficulties. However, there is no one gold standard assessment tool to investigate motor ability in children. The aim of the current paper was to discuss the issues related to the assessment of motor ability in young pre-school children and to provide guidelines on the best approach for motor assessment. The paper discusses the maturational changes in brain development at the preschool level in relation to motor ability. Other issues include sex differences in motor ability at this young age, and evidence for this in relation to sociological versus biological influences. From the previous literature it is unclear what needs to be assessed in relation to motor functioning. Should the focus be underlying motor processes or movement skill assessment? Several key assessment tools are discussed that produce a general measure of motor performance followed by a description of tools that assess specific skills, such as fine and gross motor, ball and graphomotor skills. The paper concludes with recommendations on the best approach in assessing motor function in pre-school children

    Neuroimaging standards for research into small vessel disease and its contribution to ageing and neurodegeneration: A united approach

    Item does not contain fulltextCerebral small vessel disease (SVD) is a common accompaniment of ageing. Features seen on neuroimaging include recent small subcortical infarcts, lacunes, white matter hyperintensities, perivascular spaces, microbleeds, and brain atrophy. SVD can present as a stroke or cognitive decline, or can have few or no symptoms. SVD frequently coexists with neurodegenerative disease, and can exacerbate cognitive deficits, physical disabilities, and other symptoms of neurodegeneration. Terminology and definitions for imaging the features of SVD vary widely, which is also true for protocols for image acquisition and image analysis. This lack of consistency hampers progress in identifying the contribution of SVD to the pathophysiology and clinical features of common neurodegenerative diseases. We are an international working group from the Centres of Excellence in Neurodegeneration. We completed a structured process to develop definitions and imaging standards for markers and consequences of SVD. We aimed to achieve the following: first, to provide a common advisory about terms and definitions for features visible on MRI; second, to suggest minimum standards for image acquisition and analysis; third, to agree on standards for scientific reporting of changes related to SVD on neuroimaging; and fourth, to review emerging imaging methods for detection and quantification of preclinical manifestations of SVD. Our findings and recommendations apply to research studies, and can be used in the clinical setting to standardise image interpretation, acquisition, and reporting. This Position Paper summarises the main outcomes of this international effort to provide the STandards for ReportIng Vascular changes on nEuroimaging (STRIVE)

    Development and evaluation of a novel fast broad-range PCR and sequencing assay (FBR-PCR/S) using dual priming oligonucleotides targeting the ITS/LSU gene regions for rapid diagnosis of invasive fungal diseases: multi-year experience in a large Canadian healthcare zone

    Abstract Background This study evaluated the performance of a novel fast broad range PCR and sequencing (FBR-PCR/S) assay for the improved diagnosis of invasive fungal disease (IFD) in high-risk patients in a large Canadian healthcare region. Methods A total of 114 clinical specimens (CS) including bronchoalveolar lavages (BALs) were prospectively tested from 107 patients over a 2-year period. Contrived BALs (n = 33) inoculated with known fungi pathogens were also tested to increase diversity. Patient characteristics, fungal stain and culture results were collected from the laboratory information system. Dual-priming oligonucleotide (DPO) primers targeted to the internal transcribed spacer (ITS) (~ 350 bp) and large subunit (LSU) (~ 550 bp) gene regions were used to perform FBR-PCR/S assays on extracted BALs/CS. The performance of the molecular test was evaluated against standard microbiological methods and clinical review for the presence of IFD. Results The 107 patients were predominantly male (67, 62.6%) with a mean age of 59 years (range = 0–85 years): 74 (69.2%) patients had at least one underlying comorbidity: 19 (34.5%) had confirmed and 12 (21.8%) had probable IFD. Culture recovered 66 fungal isolates from 55 BALs/CS with Candida spp. and Aspergillus spp. being most common. For BALs, the molecular assay vs. standard methods had sensitivity, specificity, positive predictive value (PPV) and negative predictive value (NPV), and efficiency of 88.5% vs.100%, 100% vs. 61.1%, 100% vs. 88.5%, 61.1% vs. 100%, and 90.2% for both. For other CS, the molecular assay had similar performance to standard methods with sensitivity, specificity, PPV, NPV and efficiency of 66.7%, 87.0%, 66.7%, 87.0% and 81.3% for both methods. Both methods also performed similarly, regardless of whether CS stain/microscopy showed yeast/fungal elements. FBR-PCR/S assays results were reported in ~ 8 h compared to fungal cultures that took between 4 and 6 weeks. Conclusions Rapid molecular testing compared to standard methods have equivalent diagnostic efficiency but improves clinical utility by reporting a rapid species-level identification the same dayshift (~ 8 h)
